Aspergillus oryzae 유래의 리파제 Lipolase-100T에 의한 물고기 기름의 가수분해 특성 규명 (Investigation of the Hydrolysis Characteristics of Fish Oil by Means of Aspergillus oryzae Lipase Lipolase-100T)

  • 물고기 기름에 대한 Aspergillus oryzae 유래의 Lipolase-100T의 가수분해 특성을 규명하였다. Lipolase-100T는 트리글리세라이드의 1과 3의 위치에 작용하여 이 위치에 결합되어 있는 아실 체인을 유리지방산으로 가수분해시키는 1,3-위채특이성을 보였다. 또한 Lipolase-100T는 물고기 기름을 구성하는 다중불포화지방산 보다 포화지방산을 쉽게 가수분해시키는 특성도 나타내었다. 이 특성으로 인하여 가수분해 시간에 따라서 생성되는 글리세라이드 혼합물내의 다중 불포화 지방산중 n-3 PUFAs인 C16:4, C20:5 및 C22:6의 농도는 모두 증가하였으며 특히 C22:6의 농도증가가 가장 뚜렷하였다. 이 결과로부터 물고기 기름을 구성하는 n-3 PUFAs는 트리글리세라이드의 2번 위치에 결합하고 있음을 알 수 있었다. 지방산의 결합위치특이성과 Lipolase-100T 1,3-위치특이성으로 인하여 Lipolase-100T를 반응물의 0.4 wt%사용하여 물고기 기름을 120시간 가수분해 시켰을 때 생성된 글리세라이드 혼합물내의 n-3 PUFAs농도는 50 wt%까지 상승하였다.

  • To evaluate the antioxidant effect of flavonoids and a-tocopherol on purified fish oil(up to $40\%$ of n-3 polyunsaturated fatty acids), lipid peroxidation, and fatty acids content during storage and upon heating were determined. The potential of these compounds for inhibiting and delaying both oxidation and lipoxygenase processes was also evaluated. The oxidation of fish oil was effectively inhibited by flavonoids and a-tocopherol. The antioxidizing effect of these compounds increased in proportion to their concentration. The addition of a-tocopherol and catechin-a-tocopherol mixture were prolonged induction period of lipid oxidation by 3.5 to 4 times. All other flavonoids also shown more than twice the prolonging effect. Lipoxygenase activity was decreased by catechin and a-tocopherol effectively.

Effects of dietary rubber seed oil on production performance, egg quality and yolk fatty acid composition of Hy-Line Brown layers

  • Objective: This study aims to evaluate the effects of dietary supplement of rubber seed oil on production performance, egg quality, and yolk fatty acid composition in laying hens during a 16-week feeding trial period. Methods: Forty-eight 25-week-old laying hens of Hy-Line Brown were randomly divided into three groups. Each group comprised four replicates and each replicate had four birds. Rubber seed oil was incorporated into a corn-soybean meal basal diet by 3.5% (group I), 4.5% (group II), or 0 (control group) and equivalent nutrition was supplied for the test groups and the control group. The performance related values were determined using standard or well established methods. Results: No significant difference was found in the production performance, the egg quality, the composition of saturated fatty acids, and the content of cholesterol and monounsaturated fatty acids in the yolk within the three groups. Interestingly, both test groups achieved a significantly higher content of linoleic acid, α-linolenic acid, eicosapentaenoic acid, and docosahexaenoic acid and a significantly lower content of arachidonic acid (p<0.05) compared with the control group. With the increased level of dietary rubber seed oil, there was an increasing trend in the content of n-6 polyunsaturated fatty acids (PUFA), n-3 PUFA and total PUFA, but a declining trend in the n-6/n-3 ratio. Conclusion: These results demonstrate that the rubber seed oil supplemented diet effectively improved the total PUFA content in eggs without impairing the layers' production performance and the egg quality.

건조 고등어 섭취가 마우스의 간 및 신경조직의 지방산 조성에 미치는 영향 (Effect of intake of dried mackerel on fatty acid compositions in liver and nervous tissue)

  • 본 연구에서는 n-3계 지방산이 풍부하게 함유된 고등어를 기존의 열풍건조기의 단점을 보완하고 높은 온도에서 손실되는 영양소를 막기 위해 저온진공건조기를 도입하여 건조시킨 후, 건조고등어의 섭취에 의한 간 및 신경조직에서의 지방산 조성 변화를 알아보고자 하였다. 간 지방산 조성의 경우, 고등어군의 총 n-6 지방산 함량은 두 군에서 유의적 차이는 없었으나 총 n-3 지방산의 함량은 고등어군에서 대조군보다 증가하였다. N-3 계열 지방산 중에서 특히 DHA의 함량은 대조군에 비해 229% 증가하였다(p<0.05). 대뇌 피질의 지방산 조성의 경우, 고등어군의 총 monounsaturated 지방산은 대조군에 비해 유의적으로 증가하였으나, 총 n-6 지방산의 함량은 감소하였다(p<0.05). 반면 총 n-3 지방산은 대조군에 비해 12%로 증가하였고 그 중에서 22:5n-3와 DHA의 함량은 고등어군에서 대조군보다 증가하였다(p<0.05). 소뇌의 지방산 조성의 경우, 총 지방산 함량과 총 n-6 지방산의 함량이 고등어군에서 대조군보다 감소하였고, 총 n-3 지방산의 함량은 두 군 간에 유의적 차이는 없었으나 n-3계 지방산중 22:5n-3의 함량은 200%로 증가하였다(p<0.05). 망막의 지방산 조성의 경우, 고등어군의 총 n-6 지방산의 함량은 대조군보다 감소하였으나, 총 n-3 지방산의 함량은 유의적 차이가 나타나지 않았다. 반면, n-3계 지방산 중에서 20:5n-3과 22:5n-3이 각각 200%, 67%로 증가하였다(p<0.05). 이상의 결과로부터 n-3 지방산을 많이 함유한 건조 고등어 식이는 간 및 신경조직의 총 n-3 지방산의 함량을 증가시켰고, 특히 간, 대뇌 피질에서의 DHA의 함량을 증가시켰다. 이상의 결과를 토대로 건조 고등어 섭취는 뇌의 DHA 함량을 증가시켰으므로 향후 공간 기억력 테스트를 행하여 고등어 섭취에 의한 뇌 기능 개선 효과와 뇌의 DHA 함량 증가와의 상호 연관성에 대하여 검토할 예정이다.

Sexual Maturation May Affect the Levels of n-6 PUFA in Muscle Tissues of Male Mice

  • Lipid metabolism in mature male mice may be different from immature male mice, but the relationship of lipid metabolism, especially n-6 fatty acid metabolism, and sexual maturation is not clearly established. This study was carried out to elucidate whether sexual maturation may affect the metabolism of functional n-6 fatty acids of lipid components by investigating the composition of fatty acids in the longissimus muscle tissues of mature and immature male mice with GC and analyzing the expression of genes and proteins for synthesis of n-6 fatty acids with real-time PCR and western blotting, respectively. Mature male mice showed significantly higher testosterone level in the sera. Similarly, n-6 fatty acids, levels of linoleic acid (LA 18:2n-6) and total n-6 PUFA (Polyunsaturated fatty acids) were increased, but the levels of ${\gamma}$-linolenic acid (GLA; 18:3n-6), dihomo-${\gamma}$-linolenic acid (DGLA; 20:3n-6) and arachidonic acid (AA; 20:4 n-6) were decreased in the mature male mice. mRNA levels of ${\Delta}5$-desaturase (FASD1) and elongase (ELOVL5) genes related to n-6 fatty acid metabolism increased. However, the level of FADS1 protein only increased in mature male mice. In conclusion, this study suggested that sexual maturation of male mice affected n-6 fatty acid metabolism by stimulating the expression of enzyme FADS1 of n-6 PUFA metabolism.

n-3 지방산이 유방암세포의 증시과 지질과산화 및 Oncogene 발현에 미치는 영향 (Effects of n-3 Fatty Acids on Proliferation of Human Breast Cancer Cells in Relatino to Lipid Peroxidation and Oncogene Expression)

  • To investigate the effects of n-3 fatty acids on breast cancer, MDA-MB231 human breast cancer cells were cultured in the presence of $\alpha$-linolenic (LNA), eicosapentaenoic(EPA), and docosahexaenoic acid (DHA) at a concentration of 0.5$\mu\textrm{g}$/ml in serum -free IMM medium. Cell growth was monitored and thiobarbituric acid reactive substances (TBARS), $\alpha$-tocopherol contents, and oncogene expression were measured. To compare the effects of n-3 fatty acids with other types of fatty acid, steraic (STA), olieic(OA). linoleic acid(LA) were used. After one day , cell growth was retarded most highly when DHA was in the medium. Cellular TBARS level measured after three days of culture was the highest with DHA in the medium and was also increased by LNA and EPA, compared with STA, OA and LA. Alpha-tocoopherol contents of cells were decreased by DHA but only modestly. There was non significant difference in $\alpha$-tocopherol contents in cells cultured in the presence of the other fatty acids. northern blot hybridization carried out with cells cultured during 24 hours showed that levels of erbB-2 mRNA were not altered by six different fatty acids in the medium but those of c-myc were transiently decreased in the early period by both n-6 and n-3 polyunsaturated fatty acids. The level of tumor suppressor gen p53 mRNA , however, was increased by DHA with time. It is concluded that the cytotoxicity of lipid peroxide and increased expression of tumor suppressor gene p53 are at least partly responsible for the inhibitory effect of DHA on growth of breast cancer cells.

  • The purpose of this study was to investigate the effects of n-3 polyunsaturated fatty acids(PUFA) on glucose and lipids metabolism in high-fat diet rate. Rats were randomly assigned to normal, high-fat with n-3 PUFA and high-fat dietary groups. Experiments were carried out after 5 weeks feeding with prescriptive diets following 7 hrs fasting. Body weight gains tended to be higher in high-fat fed rats than normal. Blood glucose was increased (p<0.05) by high-fat diet compared with normal diet, and decreaseed (p<0.05) to normal level by n-3 PUFA. Plasma insulin level was significcantly higher (p<0.01) in high-fat diet rats than that of normal-diet rats, and also decreased (p<0.01) by n-3 PUFA. Glucose up take of soleus muscle in vitro was decreased markedly in high-fat fed rats than normal diet rats at 0, 1, 10, and 100nM insulin concentration. Therefore insulin sensitivity and responsiveness were decreased by high-fat diet. Omega-3 PUFA made a recover(p<0.01) insulin sensitivity to almost normal level, and improved (p<0.05) insulin responsiveness in some extent. In conclusion, the results suggest that metabolic disorder of glucose and insulin resistance of skeletal muscle are caused by high-fat diet and n-3 PUFA can ameliorate metabolic disorder and insulin resistance.

Fatty Acid Composition of Fry Mirror Carp (Cyprinus carpio) Fed Graded Levels of Sand Smelt (Atherina boyeri) Meal

  • The effect of replacement of fish meal (FM) in diets with sand smelt meal (SSM) on fatty acid composition of carp fry, Cyprinus carpio, was examined. Five isonitrogenous and isoenergetic (38% crude protein, $15.75\;kJ\;g^{-1}$) diets replacing 0, 25, 50, 75, and 100% FM protein by SSM protein were formulated. Each diet was randomly allocated to triplicate groups of fish in aquaria, and each aquarium was stocked with 20 fish (initial average weight of $0.300{\pm}0.65\;g\;fish^{-1}$). Fish were fed twice daily to apparent satiation for 13 weeks. Results indicated that final weight, specific growth rate and feed efficiency ratio of fish fed with different SSM replacement diets did not differ significantly (p>0.05) from fish fed the control diet, except for 100% SSM level. No significant differences were noted among experimental treatments on dry matter, protein, lipid and ash contents of the fish body composition (p>0.05). Fatty acid analysis showed that saturated fatty acids in fish muscle significantly decreased, but monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA) did not change with increasing dietary SSM. However, some changes also could be observed for some particular fatty acids in experimental fish. For example, the amounts of 15:0, 17:0, 18:1n-7, 18:2n-6 and 22:5n-3 significantly increased, but 16:0, 18:1n-9, 18:3n-3 and 20:1 n-9 significantly decreased with increasing dietary SSM. Total n-6 PUFA increased with increasing dietary SSM, but total n-3 PUFA were not changed in muscle of fish fed the experimental diets. The ratio of n-3 to n-6 was not affected significantly in muscle of fish fed the experimental diets containing different proportions of SSM, including the control diet.

Polyunsaturated fatty acids, lung function, and health-related quality of life in patients with chronic obstructive pulmonary disease

  • Background: Dietary polyunsaturated fatty acids (PUFA) are thought to modify systemic inflammation. The present study aimed to evaluate the relationship between PUFA intake, lung function, and health-related quality of life (HRQoL) in patients with chronic obstructive pulmonary disease (COPD). Methods: In this study, we used the dataset of 6th Korea National Health and Nutrition Examination Survey, in which, a total of 22,948 individuals including 573 participants with a high probability of developing COPD were enrolled. Participants with missing data for the investigated variables were excluded. Linear regression analyses were used to evaluate the association between PUFA intake (omega-3 [N3], omega-6 [N6], and total) with lung function, and HRQoL. HRQoL was determined according to the European Quality of Life-5 Dimensions (EQ-5D). Subgroup analysis of older patients was performed. Age, sex, body mass index, smoking, alcohol, education, residence, total calorie intake, and predicted FEV1% were adjusted in all analyses. Results: Although lung function was not associated with PUFA intake, EQ-5D index was remarkably associated with N3, N6, and total PUFA intake in a dose-dependent manner. This association was more pronounced in elderly COPD patients. Mean levels of N3, N6, and total PUFA intake were significantly higher in patients having better HRQoL with respect to mobility, self-care, and usual activities. Conclusion: Our results suggest that N3, N6, and total PUFA intake are associated with HRQoL in COPD patients. This association may be attributed to mobility, self-care, and usual activities. Further longitudinal study is required to clarify this relationship.

Seasonal Variation in the Nutritional Content of Mideodeok Styela clava

  • We evaluated changes in the nutritional composition of Mideodeok Styela clava harvested during the months of January, March, and May 2005. Glutamic acid, aspartic acid, arginine, leucine, and lysine were the most dominant amino acids present. Ratios of essential amino acids to nonessential amino acids were comparable to those of many fish species, with values ranging from 0.55 to 0.61 and 0.66 to 0.67 for muscle of Mideodeok from Geoje and Tongyeong, respectively. Mideodeok seems to be a suitable source of important fatty acids as it contains high levels of polyunsaturated fatty acids. Eicosapentanoic acid EPA; 20:5n-3 and docosahexanoic acid DHA; 22:6n-3 were the most dominant fatty acids, ranging from 20.0 to 22.3% and 16.5 to 17.9% in muscle, and 20.3 to 23.2% and 15.2 to 18.8% in tunic, respectively. The total mineral fraction of Mideodeok was 22.2-27.3% of dry matter. Sodium, calcium, magnesium, and potassium were the most dominant minerals in both muscle and tunic.
