Effects of Heat Treatment, Sugar Addition and Fermentation on Cytotoxicity of Korean Mistletoe (가열처리, 당의 첨가 및 발효에 의한 한국산 겨우살이의 세포독성변화)

  • As a preliminary study for the development of cancer-preventing functional food using Korean mistletoe, the cytotoxic effects of Korean mistletoe on between non-tumorigenic A3l cell and tumorigenic MSV cell derived from mouse 3T3 fibroblast cell line were investigated. While the raw extract, of which $ID_{50}$, value was $3.94\;{\mu}g/mL$, showed strong cytotoxic effect, its heat-treated extract was not cytotoxic up to $30\;{\mu}g/mL$. On the other hand, the heat-treated extract with law concentration showed an accelerative effect on the proliferation of non-tumorigenic A3l cell and an inhibitory effect on that of tumorigenic MSV cell. In addition, the influences of the addition of carbohydrates, such as galactose, lactose, glucose, mannose, fructose, sucrose and starch, to mistletoe extract were studied. There were not any significant changes with raw extract plus carbohydrate treatment, but the accelerative and inhibitory effects of heat-treated extract on each A3l and MSV cell were increased further by the treatment with sugars such as lactose, galactose, glucose, fructose. In order to investigate the changes of cytotoxicity of fermented Korean mistletoe according to fermentation periods, the raw and heat-treated extract were inoculated with Lactobacillus plantarum. During 1, 3, 5 and 7 fermentation days, the fermented raw mistletoe extract showed gradual accelerative effect on A31 cell proliferation without any changes of cytotoxicity on MSV cell. In case of the fermented heat-treated extract, however, the accelerative effect of heat-treated extract on A31 cell proliferation in early stage was disappeared during the fermentation.

Effects of Concentrated Pig Slurry Using Membrane Filter on the Growth and Yield of Tomato in Nutriculture (막분리 돈분뇨 농축액비를 이용한 양액재배가 토마토의 생육과 수량에 미치는 영향)

  • This experiment was conducted to investigate the effects of concentrated pig slurry using membrane filter on growth of tomato in nutriculture. Pig slurry was filtered by ultra filtration and concentrated by reverse osmosis process. Filtration of pig slurry was necessary to prevent the hose clogging in nutriculture. The concentrated pig slurry (CS) and nutrient solution (NS) were mixed by six different mixing ratios of 0:100, 20:80, 40:60, 60:40%, 80: 20 and 100%:0% based on nitrogen content. The chemical nutrient solution was the solution of National Horticulture Research Station for the growth of tomato. The concentration of nutrient solution was adjusted a range of $1.6{\sim}2.0mS/cm$ in EC. The plant height of tomato treated with CS 20+NS 80% was similar with NS 100% control plot. Plant height was highest in the plot of CS 20+NS 80%. The treatment of 100% concentrated pig slurry was lowest in the gowth characteristics of tomato. Number of cluster was very lower in 100% concentrated pig slurry compared with plot of chemical nutrient solution. In the beginning of growth stage, SPAD reading value was reduced in plot treated with CS 100%, but CS 20+NS 80% plot was higher compared to 100% concentrated pig slurry. SPAD value of tomato leaves was decreased as the amount of CS was increased. The SPAD value also in treatment of concentrated pig slurry was lower in the middle growth stage compared to control plot. The dry weight of stem and leaf were 107.4, 104.2g in plot of NS 100% and CS 20%+NS 80%, respectively. The fruit number and weight were decreased at high application plots of concentrated pig slurry, The fruit setting of tomato showed lowest in the plot treated with 100% concentrated pig slurry, and the growth of tomato severely decreased after application of 100% CS treatment. In conclusion, the growth characteristics such as plant height and fruit weight of tomato were not significantly different between the plots treated with mixture of 20% CS +80%NS and 100% nutrient solution treatment. In conclusion, the mixture solution of 20% of concentrated pig slurry and 80% of nutrient solution could be used as a nutrition solution of tomato nuticulture.

Characteristics of Wheat Flour Dough and Noodles with Amylopectin Content and Hydrocolloids (아밀로펙틴 함량 변화와 하이드로콜로이드 첨가에 의한 밀가루 반죽 및 국수의 특성)

  • The effects of amylopectin and hydrocolloid (locust bean gum and guar gum) content on wheat flour dough and noodle properties were investigated. As the amount of amylopectin increased, the water absorption rate (farinograph), the tension (tension test), the gel stability (freeze-thawing treatment), and the springiness and the cohesiveness (TPA) increased, but the pasting temperature (RVA), the lightness and yellowness (color measurement), and the hardness (TPA) tended to decrease. In sensory evaluations, the scores for cohesiveness, springiness, and acceptability of cooked noodle increased as the proportion of amylopectin increased. The proper combination of amylose/amylopectin ratio and hydrocolloids improved the freeze-thaw stability and the sensory acceptability of wheat flour dough and noodle.

Effectiveness of Fentanyl Transdermal Patch (Fentanyl-TTS, $Durogegic^{(R)}$) for Radiotherapy Induced Pain and Cancer Pain: Multi-center Trial (방사선치료로 인한 통증 및 암성통증에 대한 듀로제식의 효과: 다기관연구)

  • $\underline{Purpose}$: To evaluate the effectiveness and safety of fentanyl-TTS in the management of radiotherapy induced acute pain and cancer pain treated with radiotherapy. $\underline{Materials\;and\;Methods}$: Our study was open labelled prospective phase IV multi-center study. the study population included patients with more 4 numeric rating scale(NRS) score pain although managed with other analgesics or more than 6 NRS score pain without analgesics. Patients divided into two groups; patients with radiotherapy induced pain (Group A) and patients with cancer pain treated with radiotherapy (Group B). All patients received 25 ug/hr of fentanyl transdermal patch. Primary end point was pain relief; second end points were change in patient quality of life, a degree of satisfaction for patients and clinician, side effects. $\underline{Results}$: Between March 2005 and June 2005, 312 patients from 26 participating institutes were registered, but 249 patients completed this study. Total number of patients in each group was 185 in Group A, 64 in Group B. Mean age was 60 years and male to female ratio was 76:24. Severe pain NRS score at 2 weeks after the application of fentanyl was decreased from 7.03 to 4.01, p=0.003. There was a significant improvement in insomnia, social functioning, and quality of life. A degree of satisfaction for patients and clinician was very high. The most common reasons of patients' satisfactions was good pain control. Ninety six patients reported side effect. Nausea was the most common side effect. There was no serious side effect. $\underline{Conclusion}$: Fentanyl-TTS was effective in both relieving pain with good tolerability and improving the quality of life for patients with radiotherapy induced acute pain and cancer pain treated with radiotherapy. The satisfaction of the patients and doctors was good. There was no major side effect.

Rapid Detection Method for Human Rotavirus from Vegetables by a Combination of Filtration and Integrated Cell Culture/Real-Time Reverse Transcription PCR (Filtration과 Integrated Cell Culture/Real-Time Reverse Transcription PCR 기법을 이용한 채소류에서 Human Rotavirus 신속 검출)

  • The purpose of this study was to evaluate and compare different elution and concentration methods for optimization of human rotavirus (HRV) detection method using real-time RT-PCR and cell culture techniques. The leafy vegetable samples (lettuce, Chinese cabbage) were artificially inoculated with HRV. Viruses were extracted from the vegetables by two different elution buffers, buffer A (100 mM Tris-HCl, 50 mM glycine, 3% beef extract, pH 9.5) and buffer B (250 mM Threonine, 300 mM NaCl, pH 9.5), and the extracted viruses were concentrated by filtration and PEG precipitation sequentially. To determine infectivity of the viruses, the viruses recovered from the samples were infected to the MA-104 cells, and integrated cell culture real-time RT-PCR was performed at 1, 48, 72, 96, 120, 144, 168 h post-infection (p.i.). The elution buffer A was more efficient in extracting the virus from the produce samples tested than the buffer B, 29.54% and 18.32% of recoveries, respectively. The sensitivity of real-time RT-PCR method was markedly improved when the virus was concentrated by the filtration method. When the viruses were eluted and concentrated by buffer A and filtration, respectively, the average recovery rate was approximately 51.89%. When the viruses recovered from samples were infected to MA-104 cell, infectious HRV was detected within 48 h p.i. by ICC/real-time RT-PCR, whereas cytopathic effects were not observed until 72 h p.i. The optimized detection method evaluated in this study could be useful for rapid and reliable detection of HRV in fresh produce products and applied for detection of other food-borne viruses.

Application of the QUAL2E Model and Risk Assessment for Water Quality Management in Namyang Stream in Hwaong Polder (화옹유역 남양천의 수질관리를 위한 QUAL2E적용과 위해성 평가)

  • The Namyang Stream in Hwaong polder was planned for several water uses including recreation, where people can contact the water and consume some amount during the recreational activity. A human health risk was assessed from exposure to E. coli in the Namyang Stream, which receives partially treated wastewater from watershed. The QUAL2E model was applied to simulate stream water quality, and this model was calibrated and verified with field monitoring data. The calibration result showed a high correlation coefficient of greater than 0.9. The mean concentration of E. coli in the Namyang Stream from the QUAL2E output was in the range of 5,000 ${\sim}$ 8,000 MPN 100 mL^{-1}$, which exceeded national and international guidelines. The Beta-Poisson was used to estimate the microbial risk of pathogens ingestion and the Monte-Carlo analysis (10,000 trials) was used to estimate the risk characterization of uncertainty. The Microbial risk assessment showed that the risk ranged from 7.9 ${\times}\;10^{-6}\;to\;9.4\;{\times}10^{-6}$. Based on USEPA guidelines, the range of $10^{-6}\;to\;10^{-8}$ was considered reasonable levels of risk for communicable disease transmission from environmental exposure, and the risk above $10^{-4}$ was considered to be in the danger of infection. Therefore, water quality of the Namyang Stream might not be in the danger of infection although it exceeded national and international guidelines. However, it was in the range of communicable disease transmission, and thorough wastewater collection and treatment at the source is recommended to secure safe recreation water quality.

Recommending Core and Connecting Keywords of Research Area Using Social Network and Data Mining Techniques (소셜 네트워크와 데이터 마이닝 기법을 활용한 학문 분야 중심 및 융합 키워드 추천 서비스)

  • The core service of most research portal sites is providing relevant research papers to various researchers that match their research interests. This kind of service may only be effective and easy to use when a user can provide correct and concrete information about a paper such as the title, authors, and keywords. However, unfortunately, most users of this service are not acquainted with concrete bibliographic information. It implies that most users inevitably experience repeated trial and error attempts of keyword-based search. Especially, retrieving a relevant research paper is more difficult when a user is novice in the research domain and does not know appropriate keywords. In this case, a user should perform iterative searches as follows : i) perform an initial search with an arbitrary keyword, ii) acquire related keywords from the retrieved papers, and iii) perform another search again with the acquired keywords. This usage pattern implies that the level of service quality and user satisfaction of a portal site are strongly affected by the level of keyword management and searching mechanism. To overcome this kind of inefficiency, some leading research portal sites adopt the association rule mining-based keyword recommendation service that is similar to the product recommendation of online shopping malls. However, keyword recommendation only based on association analysis has limitation that it can show only a simple and direct relationship between two keywords. In other words, the association analysis itself is unable to present the complex relationships among many keywords in some adjacent research areas. To overcome this limitation, we propose the hybrid approach for establishing association network among keywords used in research papers. The keyword association network can be established by the following phases : i) a set of keywords specified in a certain paper are regarded as co-purchased items, ii) perform association analysis for the keywords and extract frequent patterns of keywords that satisfy predefined thresholds of confidence, support, and lift, and iii) schematize the frequent keyword patterns as a network to show the core keywords of each research area and connecting keywords among two or more research areas. To estimate the practical application of our approach, we performed a simple experiment with 600 keywords. The keywords are extracted from 131 research papers published in five prominent Korean journals in 2009. In the experiment, we used the SAS Enterprise Miner for association analysis and the R software for social network analysis. As the final outcome, we presented a network diagram and a cluster dendrogram for the keyword association network. We summarized the results in Section 4 of this paper. The main contribution of our proposed approach can be found in the following aspects : i) the keyword network can provide an initial roadmap of a research area to researchers who are novice in the domain, ii) a researcher can grasp the distribution of many keywords neighboring to a certain keyword, and iii) researchers can get some idea for converging different research areas by observing connecting keywords in the keyword association network. Further studies should include the following. First, the current version of our approach does not implement a standard meta-dictionary. For practical use, homonyms, synonyms, and multilingual problems should be resolved with a standard meta-dictionary. Additionally, more clear guidelines for clustering research areas and defining core and connecting keywords should be provided. Finally, intensive experiments not only on Korean research papers but also on international papers should be performed in further studies.

Accuracy Evaluation of Daily-gridded ASCAT Satellite Data Around the Korean Peninsula (한반도 주변 해역에서의 ASCAT 해상풍 격자 자료의 정확성 평가)

  • In order to access the accuracy of the gridded daily Advanced Scatterometer (hereafter DASCAT) ocean surface wind data in the surrounding of Korea, the DASCAT was compared with the wind data from buoys. In addition, the reanalysis data for wind at 10 m provided by European Centre for Medium-Range Weather Forecasts (ECMWF, hereafter ECMWF), National Centers for Environmental Prediction and National Center for Atmospheric Research (NCEP/NCAR, hereafter NCEP), Modern Era Retrospective-analysis for Research and Applications-2 (MERRA-2, hereafter MERRA) were compared and analyzed. As a result, the RMSE of DASCAT for the actual wind speed is about 3 m/s. The zonal components of wind of buoys and the DASCAT have strong correlation more than 0.8 and the meridional components of wind them have lower correlation than that of zonal wind and are the lowest in the Yellow Sea (r=0.7). When the actual wind speed is below 10 m/s, the EMCWF has the highest accuracy, followed by DASCAT, MERRA, and NCEP. However, under the wind speed more than 10 m/s, DASCAT shows the highest accuracy. In the nature of error according to the wind direction, when the zonal wind is strong, all dataset has the error of more than $70^{\circ}$ on the average. On the other hand, the RMSE of wind direction was recorded $50^{\circ}$ under the strong meridional winds. ECMWF shows the highest accuracy in these results. The RMSE of the wind speed according to the wind direction varied depending on the actual wind direction. Especially, MERRA has the highest RMSE under the westerly and southerly wind condition, while the NCEP has the highest RMSE under the easterly and northerly wind condition.

Grain Yield Potential of a Low-Tillering Large Panicle Type in Rice (벼 소얼 수중형 초형의 수량성)

  • For the increase of grain yield potential in rice, a low-tillering large panicled type has been suggested as an ideotype. A low-tillering plant type may have different yield potential and needs different cultural practices from that used in a high-tillering type for the maximum yield. This study was conducted to evaluate the grain yield performances of a low-tillering large panicled rice and high-tillering small panicled rice at different plant spacings, nitrogen(N) levels and seedling numbers per hill. A low-tillering large panicled genotype, IR25588 was compared with a high-tillering small panicled IR58. The grain yield of IR25588 was significantly higher than that of IR58 under a narrow spacing with high N level. The maximum yields of IR58 and IR25588 were reached at about 35,000 and 40,000 spikelets per m$^2$, respectively. The increased grain yield in IR25588 was mainly due to the increase in spikelet number per unit area which is the most precise indicator of grain yield in rice. The optimum spacing for the maximum yield was denser for IR25588 than that for IR58 under high N level. The intra-hill competition of the low-tillering type was lower than that of the high-tillering type. The higher dry matter production and bigger leaf area and culm weight were the main factors for increased grain yield in a low-tillering panicle weight type. Based on the results, the yield potential of a low-tillering panicle weight type was higher than that of a high-tillering panicle number type, especially under a close spacing with high N level.

The Differentiation of the Cholinergic Nerve Cells at the Meynert Basal Nucleus of the Basal Forebrains in the Growth Period of Rat (흰쥐 전뇌 기저부 Meynert기저핵에서 출생 후 발육기간에 따른 콜린성 신경세포의 분화)

  • This study was performed to investigate the distribution and differentiation on the immunoreacted cells of the ChAT (choline acetyltransferase) at the Meynert basal nucleus of the forebrains in the growth periods of rat, using the immunohistochemistric method. According to the cell shape and the ratio of long axis vs short axis of cell soma, the ChAT antibody reacted nerve cells in the Meynert basal nucleus of the rats were classified into six types. In the adult rats, the FD (frequency distributions) of round, oval and elongated cells were maximum. The FD of these types were shown to be progressively decreased during the postnatal development. In addition, the FD of elongated nerve cells in were observed in the adult rats respectively. This was thought to be the same phenomenon as those in the round and oval cells . The total mean volume of ChAT antibody reacted cell somata was lowest in the PND (postnatal days) 7 rats and was highest in the PND 21 rats. But, those were decreased to the adult. These results suggest that ChAT antibody reacted nerve cells grow up to PND 21 and then, differentiate into the various types by neurites outgrowing. On the electron micrography, the adult rat forebrain cells were obtained to be well developed ribosomes, polysomes , rough endoplasmic reticula and mitochondria. The immunreactivities were observed in ribosomes, polysomes, rough endoplasmic reticula and outer membrane of mitochondria. Golgi complexes were poorly developed and not showed jmmunreactivity. The ribosomes , polysomes and endoplasmic reticula are considered to be closely related to the inter cellular localization and biosynthesis of the ChAT but not Golgi complex. According to the results in the present study, it is considered that the ChAT-immunoreacted nerve cells in the Meynert basal nucleus of the rat forebrains are differentiated throughout the postnatal development with following processes of changes; 1) the cholinergic nerve cells develop postnatally 2) cell soma volumes gradually increase during the early postnatal days 3) and then, cells differentiate into the various types by projecting the neurites to the appropriate area after PND 21.
