Physicochemical Properties and Antioxidant Activities from Hot-air and Freeze Dried Aged Black Ginger (Zingiber officinale) (열풍 및 동결건조에 따른 숙성 흑생강의 이화학적 특성 및 항산화 활성)

  • Hot-air and freeze dried powder from aged black ginger were mixed under the ratio conditions of sample "A" (8:2), "B" (5:5), "C"(2:8), "D" (10:0) and "E" (0:10), respectively. The physicochemical properties and antioxidant activities in the 5 kinds of powder were compared. The water absorption index was significantly higher in freeze dried powder, and it tended to be significantly increased depend on the ratio of freeze dried sample. The browning intensity was significantly higher in the hot-air dried samples, but the color intensity was not significant. Total and reducing sugar contents were significantly higher in freeze dried sample. Total sugar content in mixed sample tended to significantly increased as the amount of freeze dried sample. Total phenol content was significantly higher in the sample "A". The contents of gingerol and shogaol were significantly higher in mixed sample "A", 6-gingerol content showed no significant difference according to the drying method. Cholesterol absorption activity was significantly higher in the freeze dried sample "E". The DPPH radical scavenging activity showed a little difference according to the drying method, but the ABTS radical scavenging activity and reducing power in the hot-air dried sample were significantly higher than those of freeze dried. And the activity of mixed powder "A" was significantly higher than others. These results suggest that aged black ginger was superior in terms of quality and functionality of the hot-air drying compared to freeze-drying, and it would be economically effective to the mixed sample "A".

Measurement and Assessment of Absolute Quantification from in Vitro Canine Brain Metabolites Using 500 MHz Proton Nuclear Magnetic Resonance Spectroscopy: Preliminary Results (개의 뇌 조직로부터 추출한 대사물질의 절대농도 측정 및 평가: 500 MHz 고자장 핵자기공명분광법을 이용한 예비연구결과)

  • The purpose of this study was to confirm the exactitude of in vitro nuclear magnetic resonance spectroscopy(NMRS) and to complement the defect of in vivo NMRS. It has been difficult to understand the metabolism of a cerebellum using in vivo NMRS owing to the generated inhomogeneity of magnetic fields (B0 and B1 field) by the complexity of the cerebellum structure. Thus, this study tried to more exactly analyze the metabolism of a canine cerebellum using the cell extraction and high resolution NMRS. In order to conduct the absolute metabolic quantification in a canine cerebellum, the spectrum of our phantom included in various brain metabolites (i.e., NAA, Cr, Cho, Ins, Lac, GABA, Glu, Gln, Tau and Ala) was obtained. The canine cerebellum tissue was extracted using the methanol-chloroform water extraction (M/C extraction) and one group was filtered and the other group was not under extract processing. Finally, NMRS of a phantom solution and two extract solution (90% D2O) was progressed using a 500MHz (11.4 T) NMR machine. Filtering a solution of the tissue extract increased the signal to noise ratio (SNR). The metabolic concentrations of a canine cerebellum were more close to rat’s metabolic concentration than human’s metabolic concentration. The present study demonstrates the absolute quantification technique in vitro high resolution NMRS with tissue extraction as the method to accurately measure metabolite concentration.

Chlorophyll Fluorescence, Chlorophyll Content, Graft-taking, and Growth of Grafted Cucumber Seedlings Affected by Photosynthetic Photon Flux of LED Lamps (LED 램프의 광합성유효광양자속이 오이접목묘의 엽록소형광, 엽록소함량, 활착 및 생장에 미치는 영향)

  • Chlorophyll fluorescence, chlorophyll content, graft-taking and growth of grafted cucumber seedlings as affected by photosynthetic photon flux (PPF) of LED lamps were analyzed in this study. Four PPF levels, namely 25, 50, 100, $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ were provided to investigate the effect of light intensity on the chlorophyll fluorescence, chlorophyll content, graft-taking and growth of grafted cucumber seedlings. Air temperature, relative humidity, and photoperiod for graft-taking were maintained at $25^{\circ}C$, 90%, $16h{\cdot}d^{-1}$, respectively. Maximum quantum yield (Fv/Fm) of rootstock as affected by PPF was found to be 0.84-0.85 and there was no significant change in Fv/Fm. Even though Fv/Fm of scion measured at 2 days after grafting was lowered to 0.81-0.82, after then it gradually increased with increasing PPF. At 4 days after grafting, the chlorophyll content extracted from scion increased with increasing PPF. Graft-taking ratio of grafted cucumber seedlings was 90-95% as PPF was ranged from $25{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ to $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. However, the graft-taking ratio of grafted seedlings healed under PPF of $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ was decreased to 80%. Maximum PPF measured required for smooth joining of rootstock and scion was assumed to be $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. At healing stage of grafted cucumber seedlings, Fv/Fm of scion decreased and at least two days after grafting were required for rooting of grafted seedlings. Chlorophyll fluorescence response of rootstock and scion was linked to light irradiation. Therefore, it was concluded that physical environment including light and humidity during healing process of grafted seedlings should be controlled more precisely to facilitate root formation and to prevent scion from lowering Fv/Fm. Further studies are required to investigate the effects of root development and joining of vascular bundles of grafted seedlings on the chlorophyll content of scion.

Effects of Salt Stress on Dry Matter, Glucose, Minerals Content and Composition in Potato (Solanum tuberosum L.) (염스트레스가 감자(Solanum tuberosum L.)의 건물, 환원당, 무기성분의 함량 및 조성에 미치는 영향)

  • BACKGROUND: Salinity is one of the main environmental stresses deteriorating qualities as well as yields of food crops. This study was conducted to identify the effects of salt stress on dry matter ratio, glucose content, and mineral content and composition in potatoes (Solanum tuberosum L.). METHODS AND RESULTS: Four potato varieties were grown in plastic pots (diameter 20 cm and height 25 cm) with three salinity levels (EC: 1.0, 4.0, and 8.0 dS/m) in a glasshouse. Dry matter ratio, specific gravity, starch, and glucose content in tubers harvested at 90 days after sowing were analyzed. Also, mineral contents (T-N, T-C, $P_2O_5$, $K^+$, $Ca^{2+}$, $Mg^{2+}$, $Na^+$) in stem, leaf, and tuber were investigated and statistically analyzed for analysis of variance (ANOVA). Dry matter ratio, specific gravity, and starch content in tubers were reduced in all varieties as the salt concentration increased. Glucose content tended to decrease according to the salt concentration. In ANOVA analysis of mineral contents, there were significant differences in $K^+$ and $Mg^{2+}$ of leaf and stem, in $Na^+$ of leaf and tuber, and also in $Ca^{2+}$ of leaf by the interactions of variety and salinity. In the case of $K^+/Na^+$ and $Ca^{2+}/Na^+$, the stem was more sensitively influenced by the salt treatment than the leaf or the tuber. The $K^+/Na^+$ and $Ca^{2+}/Na^+$ decreased in leaf, stem, and tuber of four varieties, as the salt concentration became higher. The decreasing level varied according to the varieties. 'Kroda' and 'Duback' maintained relatively higher $K^+/Na^+$ and $Ca^{2+}/Na^+$ than 'Atlantic' or 'Goun' under the salt stress conditions. CONCLUSION: The composition and accumulation of minerals in potato plant as well as dry matter ratio, starch, and glucose contents were significantly influenced by salt stress. The respond patterns were different depending on the varieties and it was related to the salt tolerance among varieties.

The Giant Magnetoresistance Properties of CoFe/Cu/NiFe Pseudo Spin Valve (CoFe/Cu/NiFe Pseudo스핀밸브의 자기저항 특성)

  • The pseudo spin valve with a structure of Tl/CoFe(t $\AA$)/Cu(30 $\AA$)/NiFe(50 $\AA$)/Ta, showing giant magnetoresistance properties by utilizing coercivity difference between only two soft ferromagnetic layers were produced by d.c UHV magnetron sputtering system. In pseudo spin valve Ta/CoFe/Cu/NiFe/Ta, the magnetic and magnetoresistance properties with change of CoFe thickness were investigated. When the thickness of CoFe was 60 $\AA$, a typical MR curve of pseudo spin valve structure was obtained, showing MR ratio of 3.8 cio and the coercivity difference of 27.4 Oe with a sharp change of hard layer switching. When the CoFe thickness was varied from 20 to 100 $\AA$, coercivity difference between two layers was increased to 40 $\AA$. and decreased to 100 $\AA$ gradually. It is thought the change in coercivity of hard layer was due to the crystallinity and magnetostriction of thin CoFe layer. In order to improve the MR property in CoFe/Cu/NiFe trier layer structure, CoFe layer with change of 2-20 $\AA$ thick was inserted between Cu and NiFe. When the thickness of CoFe was 10 $\AA$, MR ratio was 6.7%, showing excellent MR property. This indicates 50 % higher than that of CoFe/Cu/NiFe pseudo spin valve.

Gelatinized Carbohydrates in the Diet of Catla catla Fingerlings: Effect of Levels and Sources on Nutrient Utilization, Body Composition and Tissue Enzyme Activities

  • A Feeding trial was conducted to study the effects of three different sources and two levels of dietary gelatinized carbohydrate (GC) on nutrient utilization, growth, tissue composition and tissue enzyme activities of fingerlings of Catla catla (15.1-15.3 g). Six isocaloric (17.1-17.5 kJ/g) semi-purified diets were prepared either with rice, corn or tapioca at 40 or 50% GC each. The crude protein (CP) level used in the diet was 35% and 25% for 40% and 50% GC level, respectively to study the protein sparing effect of GC. The degree of gelatinization was higher for corn and tapioca than rice under similar cooking conditions. After a 60-d feeding trial, dry matter, carbohydrate, protein and lipid digestibility were higher in tapioca fed groups at both the levels of GC. However, the highest specific growth rate (SGR) and protein efficiency ratio (PER) were observed in the corn fed groups at 50% GC level indicating better utilization of nutrients from gelatinized corn. Feed conversion ratio (FCR) was almost similar in corn and tapioca fed groups between two levels of GC but in rice fed groups, FCR was lower in 40% GC than 50% GC level. The results indicated higher protein-sparing effect in corn and tapioca fed groups than rice fed groups. The order of gelatinized carbohydrate utilization in Catla catla fingerlings at 50% GC level was corn>tapioca>rice. At 40% GC level, corn and tapioca were comparable and more efficiently utilized than rice. In the corn fed groups, 50% GC was comparable with 40% GC level, whereas in rice and tapioca fed groups the 40% GC was better in terms of nutrient utilization. Liver glycogen content and hepatosomatic index were significantly (p<0.05) higher in those groups fed high GC (50%) irrespective of carbohydrate sources. Higher intestinal amylase and glucose-6-phosphate dehydrogenase activities were observed in higher GC fed groups than the lower GC groups. No mortality was found in any groups at any levels of GC.

Effects of Feeding Probiotics on the Odor-Generating Substances and Development of maggots(Fly's larvae) in Pig's Feces (육성비육돈에 대한 생균제의 첨가급여가 분 악취 및 파리유충 발생에 미치는 영향)

  • This research was conducted to investigate the effects on the amount of nitrogen and phosphorus and the number of maggots in the feces from the experimental pigs fed probiotics additives, and the variance of NH3-concentration(ppm) generated in the experimental pig-pens. The experiment was performed twice in the W-farm(under the condition of sufficient energy and protein in Spring) and B-farm(insufficient energy and protein in Summer). The experiment was designed with 5 treatments (control and 4 treatments according to the kinds of additives used). In the W-farm experiment, 75 three-way crossbred pigs weighing average 38kg were randomized with 3 replications for 102 days. In the B-farm experiment, 105 three-way crossbred pigs weighing average 33kg were randomized with 3 replications for 130 days. The results obtained are as follows: 1. The amount of dry matter, organic matter, nitrogen and phosphorus in the feces was not significantly different among the treatments. But the amount of OM, N and P shows a tendency to be somewhat lowered in the treatment A. On the otherhand, the amount of dry matter was somewhat lowered in the treatment B. The amount of such matters in the W-farm was somewhat lowered than those in B-farm. But there was not significant difference between W-farm and B-farm. 2. On the variance of the number of maggots developed in the pig's feces according to each treatment, the number of maggots in the treatments added probiotics were markedly decreased in comparison with those of the control(T), and it was significantly different(p<0.01). Especially the decrease in the treatment A and B was much remarkable. 3. The addition of probiotics to the pigs' feeds made the NH3-concentration generated in the pig-pens decrease markedly. especially the NH3-concentration in the treatment A had the highest decreasing ratio among all the treatments. The decreasing ratio of NH3-concentration of the treatments added probiotics as compared to control(T) was 59.1∼80.0%(average 70.3%) in the treatment A, 56.8∼80.0%(ave. 68.5%) in the treatment B and 24.4∼46.0%(ave. 35.0%) in the treatment C and D. On the variance of the NH3-concentration according to the experimental times)seasonal conditions), the NH3-concentration generated in the pig-pens was gradually increased from April to August, on the other hand it was decreased in the September. By the result of this study, it was appeared that the probiotics added to the pigs' feeds made the NH3-concentration decrease in the pig-pens to the extent of safety degree(25 ppm and less) permitted to swine especially in the treatment A and B.

Cracking Behavior of RC Tension Members Reinforced with Amorphous Steel Fibers (비정질 강섬유로 보강된 철근콘크리트 인장부재의 균열거동)

  • This paper presents the experimental results concentrically reinforced concrete tension members and compares cracking behavior of amorphous steel fiber and normal steel fiber reinforced concrete members. Two kind of steel fibers were included as a major experimental parameter together with the six cover thickness to bar diameter ratio ($c/d_b$). The presence of amorphous steel fibers effectively controlled the splitting cracks initation and propagation. In the amorphous steel fiber reinforced specimens, no splitting cracks were observed that becomes higher with cover thickness to bar diameter ratio is 2.0. Crack spacing of the each specimens reinforced with amorphous steel fibers and normal steel fibers becomes larger with the increase in cover thickness, and also measured maximum and average crack spacing are significantly smaller than current design code provision. Based on the measured crack spacings, a relationships for predicting the crack spacing is proposed using the measured average crack spacing in amorphous steel fiber reinforced concrete tension members.

Combination of an Enzymatically Hydrolyzed Yeast and Yeast Culture with a Direct-fed Microbial in the Feeds of Broiler Chickens

  • A balance trial experiment was carried out to evaluate the potential relationship between an enzymatically hydrolyzed yeast (EHY) and yeast culture combined with a live Bacillus subtilis (Bs) on the productive parameters, ileal digestibility, retention of nutrient and energy and villus morphology in broilers. Seventy two 28 d old, Ross B308 male broilers were assigned to a factorial combination of 2 levels of EHY (0 and 1 kg/ton of feed) and 2 levels of Bs (0 and 125 g/ton of feed). The experiment lasted 2 weeks. Several treatment interactions were observed. EHY-fed broilers showed the lowest feed intake and feed conversion ratio whereas Bs-fed broilers showed the highest feed intake and intermediate feed conversion ratio (EHY and BS interaction, p<0.05). Also, EHY-fed broilers had greater ileal digestibility of dry matter (EHY and BS interaction, p<0.01) and energy (EHY and BS interaction, p<0.05) but these responses were counterbalanced by the combination of EHY and Bs. The thickness of the mucosa was similar between the control and EHY-fed broilers, but was lowest when Bs was added alone (EHY and BS interaction, p<0.01). The thickness of the villus was greater in EHY plus Bs-fed broilers, intermediate for the control and lower for Bs or EHY-fed broilers (EHY and BS interaction, p<0.05). The area of the villus was greater in the control and EHY plus Bs-fed broilers (EHY and BS interaction, p<0.05). In addition, EHY-fed broilers showed greater breast yield and nitrogen retention (p<0.01) and ashes digestibility (p<0.05). On the other hand, Bs-fed broilers had greater carcass and breast weight, nitrogen retention, energy excretion and villus height (p<0.05). In summary, EHY and Bs enhanced some growth, carcass and nutrient retention responses, but did not show any synergic relationship in these responses. Opposite to this, the results suggest that the positive effect of EHY on the feed conversion and digestibility of nutrients were counterbalanced by the addition of Bs.

Influence of Feeding Processed Cottonseed Meal on Meat and Wool Production of Lambs

  • In order to assess the effect of feeding raw or processed cotton (Gossypium) seed meal (CSM) on meat and wool production, 30 male crossbred lambs (3-4 months) of uniform body weight were assigned equally to five dietary treatments in a completely randomised design. The CSM was processed by three different methods i.e., cooking the meal at $100^{\circ}C$ for 45 minutes, treatment with 1% calcium hydroxide ($Ca(OH)_2$) for 24 h and iron treatment in the ratio of 1 part free gossypol (FG) to 0.3 parts of iron for 30 minutes. The lambs were fed isonitrogenous and isocaloric concentrate mixtures, containing 30% deoiled peanut meal (reference diet) and 40% of either raw, cooked, $Ca(OH)_2$ or iron treated CSM for 180 days. The raw and variously processed CSM replaced about 50% nitrogen of reference concentrate mixture. The concentrate mixtures were fed to meet 80% of the protein requirements (NRC, 1985) along with ad libitum chopped maize (Zea mays) hay. The slaughter weight, empty body weight and carcass weight was higher ($p{\leq}0.01$) in lambs fed cooked CSM incorporated diets, compared to diets containing deoiled peanut meal (DPNM). These parameters were not influenced by feeding diets containing either raw, $Ca(OH)_2$ or iron treated CSM in comparison DPNM diets. The carcass length, loin eye area and edible and inedible portion of carcass and the meat: bone ratio in whole carcass were also not affected by feeding CSM based diets. Among various primal cuts, the yield of legs was lower ($p{\leq}0.05$) from raw CSM fed lambs in comparison to DPNM fed lambs. The fat content in the Longissimus dorsi muscle was reduced ($p{\leq}0.05$) in lambs fed processed CSM based diets compared to those fed DPNM diet. Replacing DPNM with either raw or processed CSM based diets did not influence the sensory attributes and overall acceptability of meat. The wool yield was higher ($p{\leq}0.05$) in iron treated CSM fed lambs. The fibre length and fibre diameter were comparable among lambs on various dietary regimes. Among lambs fed variously processed CSM diets, the feed cost per kg of edible meat production was lower ($p{\leq}0.05$) on $Ca(OH)_2$ treated CSM, followed by cooked CSM diet and then on raw CSM based diets compared to DPNM diet. The CSM after 1% $Ca(OH)_2$ treatment or cooking for 45 minutes appears to be a satisfactory protein supplement in lamb diets for meat and wool production to replace at least 50% nitrogen of scarce and costly peanut meal.
