In the fungal genus Cordyceps, the type species Cordyceps militaris produces bioactive ingredients and exhibits medicinal effects as a Traditional Chinese Medicine (TCM), The fruiting bodies of C.militaris have now been mass-produced artificially and used as functional food and medicine in China. The unstable variation in forming fruiting body is however a key restrictive factor in industrial production. The genetic study on in vitro stromata formation of C. militaris has rarely been carried out. Here, we report the effects of genetic variation including the mating system on perithecial stromata formation of C. militaris. Monoconidial isolates which have both MAT1-1-1 and MAT1-2-1(genotype MAT1-1/2) could produce stromata. While the isolates only have either MAT1-1-1 or MAT1-2-1 (genotype MAT1-1 or MAT1-2) failed to produce stromata. Despite obvious heterothallism, homothallism was occasionally observed in a few isolates of C. militaris. High genetic variation was observed amongst the different monoconidial isolates of C. militaris. The unstable variation or lose of fruiting body formation was found to be caused by the inner-species high genetic variation of C. militaris. These results also indicated that C. militaris sexually behaved as both heterothallic and homothallic and required two mating type compatible in the same culture in order to produce regular clubshaped perithecial stromata.
Cordyceps militaris, known as Dong-Chong-Xia-Cao, produces the most cordycepin among Cordyceps species and can be cultured artificially. For these reasons, C. militaris is widely used as herb or functional food in the East Asia. In this study, we developed a new strain of C. militaris that produces higher cordycepin content than parent strains through mating-based sexual reproduction. Twenty parent strains were collected and identified as C. militaris based on internal trasncrived spacer and rDNA sequences. Seven single spores of MAT 1-1 idiomorph and five single spores of MAT 1-2 idiomorph were isolated from 12 parent strains. When 35 combinations were mated on the brown rice medium with the isolated single spores, eight combinations formed a stroma with a normal perithecia and confirmed mated strains. High pressure liquid chromatography analysis showed that mated strain KSP8 produced the most cordycepin in all the media among all the tested strains. This result showed due to genetic recombination occurring during the sexual reproduction of C. militaris. The development of C. militaris strain with increased cordycepin content by this approach can help not only to generate new C. militaris strains, but also to contribute to the health food or medicine industry.
Cordyceps militaris의 액체 배양시 cordycepin(3'-deoxyadenosine) 생성에 영향을 주는 물리화학적 조건 및 배지 조성의 영향을 조사하였다. 사용된 17종 균주 중에서 C. militaris KCTC 6862, C. militaris KCTC 16932 및 C. militaris DGUM 32003이 우수한 cordycepin생산성을 보여주었다. Cordycepin생산에 적합한 최적 온도와 PH는 각각 $24^{\circ}C$와 pH 6.0-10범위이었다. YM배지에 탄소원으로 glucose를, 질소원으로 tryptone을 각 1% 첨가하였을 매 우수한 cordycepin 생산을 보여주었다. Tryptone을 질소원으로 하는 YMG 액체배지에서 5일간 배양하였을 경우, 39mg/l의 cordycepin의 농도가 측정되었다. 0.1%의 다양한 인산원을 각각 첨가한 결과, cordycepin생산을 저해하였다.
본 연구는 동충하초 추출물을 PC12 및 BV2 세포에서 인지능력 개선에 대한 효능을 평가하고자 하였다. 동충하초 추출물을 증류수로 추출하여 PC12 및 BV2 세포로 MTT 분석을 통해 세포 생존율을 확인하고 L-glutamate로 유도한 PC12 세포를 통해 세포 보호 효능과 아세틸콜린 함량 및 아세틸콜린에스테아제 활성을 평가하였다. 또한, LPS로 유도한 BV2 세포를 통해 nitric oxide (NO) 및 prostaglandin E2 (PGE2) 생성량 등의 항염증 효능을 측정하고 western blot 분석을 통해 $NK-{\kappa}B$, p38, JNK, caspase-3 등의 단백질 발현량을 확인하였다. 동충하초 추출물은 $200{\mu}g/m{\ell}$ 농도를 제외하고 1, 10, $100({\mu}g/m{\ell})$ 농도에서 세포 독성이 나타나지 않았다. L-glutamate로 유도한 PC12 세포에서 유의성있게 세포 보호 효능과 아세틸콜린 함량의 증가, 아세틸콜린에스테아제 활성 감소가 나타났다. 또한, 동충하초 추출물은 NO 및 PGE2 생성량과 $NK-{\kappa}B$, p38, JNK, caspase-3 등의 단백질 발현을 억제하였다. 이와 같은 결과는 동충하초 추출물이 인지능력에 대한 예방 및 개선 효능이 있음을 나타낸다. 따라서 동충하초 추출물은 인지능력 개선을 위한 새로운 천연 소재로 활용될 수 있다.
We previously isolated cordycepin, guanosine and tryptophan from Cordyceps militaris as antiadipogenic constituents. For the quality control of C. militaris for anti-adipogenic activity, simultaneous analytical method using high-performance liquid chromatography (HPLC)-diode array detection (DAD) was developed and validated. Quantitation of these compounds in various Cordyceps samples from different sources and various extraction methods were conducted using developed method. Our study shows that natural Cordyceps and host insect possess higher content than cultured ones and fruiting bodies, respectively. The content of cordycepin showed great difference in different C. militaris samples whereas trytophan content was similar in tested samples. Addition of water to extraction solvent greatly increased the yield of guanosine and tryptophan. High temperature and longer extraction time increased yield of guanosine, whereas the content of trytophan was decreased in high temperature during extraction with water. Extraction using ultrasonic apparatus slightly increased extraction efficiency. Cordycepin, however, has little variation in different extraction method tested. Strong anti-adipogenic activity was observed in the samples that contain all the three constituents. Taken together, quantitation of these compounds using developed analytical method might provide basic requirement for the anti-adipogenic activity of C. militaris.
This study was conducted to investigate the effects of the Cordyceps militaris extract on the improvement of the glucide metabolism in serum of streptozotocin (STZ, 50 mg/kg BW, IP injection)-induced diabetic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of creatinine, lipid peroxide and NEFA in sera were significantly higher in the STZ-induced diabetic group (group DMG) and STZ+Cordyceps militaris extract administration group (group DMS) than those in the control group (group BD). However, the concentrations of creatinine, lipid peroxide and NEFA in sera were reduced in the group DMS than those in the group DMG. The activity of creatine phosphokinase (CPK) in serum was lower in the group DMS than in the group DMG. The activity of LCAT in serum was increased in the group DMS (Cordyceps militaris extract administration) than in the group DMG. The results indicate that Cordyceps militaris extract were effective in the improvement of the glucide metabolism in the sera of STZ-induced diabetic rats.

Background: Cordyceps militaris have been reported to modify the immune and inflammatory responses both in vivo and in vitro. Macrophages play important roles in the innate immunity through the phagocytosis of antigens. This study examined the effects of Cordyceps militaris on the activation of murine macrophage RAW 264.7 cells and primary macrophages. Methods: The components contained in culture broth of Cordyceps militaris were purified by propyl alcohol extraction and HP 20 column chromatography to CMDB, CMDBW, CMDB5P, and CMDB25P. The amounts of nitric oxide (NO) were determined by using ELISA, Griess reagent respectively. The amounts of some cytokines were determined by using ELISA, western blot, and RT-PCR The expression levels of cell surface molecules (ICAM-1, B7-1 and B7-2) were measured by flow cytometric analysis. Results: All the components of Cordyceps militaris produced significant amounts of NO. In particular, CMDB produced much more NO in RAW 264.7 cells and primary macrophages than other fractions of Cordyceps militaris. CMDB increased significantly the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-1${\beta}$, and IL-6 dose-dependently in RAW 264.7 cells. Examination of the gene expression level also showed that the enhanced production of cytokines was correlated with the up-regulation of i-NOS expression, cycloxygenase (COX)-2 expression, IL-1${\beta}$ and IL-6 expression, and TNF-${\alpha}$ expression on the expression of mRNAs by semi-quantitative RT-PCR Western blot analysis also confirmed that CMDB enhances the expression level of these cytokines. Conclusion: These results show that CMDB stimulates the production of NO and pro-inflammatory cytokines and can also up-regulate the gene expression levels in macrophages.
This study aimed to verify the whitening effect of Cordyceps militaris, which is distributed in several countries worldwide, including Korea, Japan, and China, and has various medical effects. To screen the efficacy of C. militaris, the inhibitory activity of tyrosinase, which was 66% at a concentration of 1 mg/mL, was measured. Thereafter, the survival rate of melanoma cells was measured, and cell experiments were conducted at a concentration of 90% or more in which C. militaris was not toxic to cells. After measuring the inhibitory effect of TRP-1, TRP-2, tyrosinase protein, and mRNA expression, which are factors influencing melanin synthesis, C. militaris was found to decrease in all factors, with an expression level that was significantly lower compared to quercetin. This confirmed that C. militaris stimulated with LED has excellent whitening activity and can be used as a functional whitening cosmetics material.
1999년 5월 중순부터 10월까지 완주군 일대의 연석산, 모악산, 대아리 수목원의 해발 약 400m의 그늘지고 습한곳에서 약 20여종의 동충하초를 채집하였다. 이 중 2속 5종을 확인하였고 이것들에서 10균주를 분리하여 9균주를 자실체유도 실험에 이용하였다. 그결과 Cordyceps militaris (99-0007) 1균주, Isaria japonica (99-0001, 99-0002, 99-0003) 3균주, Cordvceps soborifera (99-0008) 1균주, C.kanzashiana (99-0009) 1균주는 이용가치가 충분히 있다고 사료되어 우량균주로 선발하였다.
본 실험은 동충하초 분말과 8가지 균주로 각각 발효한 동충하초 분말의 혈전용해 활성, 항산화 활성, tyrosinase 저해 활성 그리고 총 폴리페놀 함량, 플라보노이드 함량, 미네랄 함량을 비교 검토하였다. 발효한 결과 CAo군의 총 폴리페놀과 플라보노이드 함량이 발효 전에 비해 가장 증가하였고, 주요 미네랄인 K, Mg, Ca, Zn 등의 함량 역시 증가하였다. 또 native-PAGE를 이용하여 발효 전과 발효 후 단백질을 비교하였을 때, CAo군에서 새로운 단백질이 관찰되었다. DPPH 라디칼 소거능 측정에서는 CMp군이 다른 군에 비해 경미하게 높은 활성을 나타냈지만, 모든 군들이 합성항산화제인 butylated hydroxytoluene (BHT)에 비해 낮은 활성을 나타내었다. tyrosinase 저해 활성을 보았을 때 CAo군이 C군 보다 높은 활성을 나타내었다. 발효 후 번데기 동충하초의 항혈전을 비교한 결과, CAk군과 CBs군에서 큰 항혈전능을 나타내었다. 두 균주를 1%(v/w), 5%(v/w)로 접종하여 발효 시간과 발효 온도를 조정하면서 각각의 항혈전 최적 조건을 분석한 결과, CAk군과 CBs군 모두 5%(v/w)로 접종하여 $40^{\circ}C$에서 24시간 발효하였을 때 최대 항혈전 활성을 나타내었다. 따라서, 번데기 동충하초는 발효 균주에 따라 건강 기능성식품의 원료 또는 혈전용해제의 원료로써 가능성을 보이며 이에 대한 심층적인 연구가 필요할 것으로 사료된다.
