Influence of Glycyrrhizic Acid, Menthol and Their Supramolecular Compounds on the Functional Activity of Rat Mitochondria in in-vitro Experiments

  • Menthol (M) is a cyclic monoterpenode and is a major component of essential oils. Menthol, along with menthol, isomenton, etc., gives taste and odor of the mint plant, and when it comes to menthol in general, L- or (-) -menthol is usually used. Included in pharmaceuticals, cosmetics and pesticides. It has antimicrobial, antibacterial, antioxidant properties. It is also known that the licorice plant (Glycyrrhiza Glabra L.) differs from other types of plants by its medicinal properties. For many years it has been used in folk medicine. Extraction of licorice root revealed up to 25% glycyrrhizinic acid (GA). Its aglycone - glycyrrhizic acid is notable for its structural similarity to the adrenal cortex hormones. Currently, GA and glycyrrhizic acid are widely used in medicine as a remedy for colds, allergies, viral diseases, tumors. The biological activity of menthol and GA-based supramolecular compounds has been poorly studied, and their effect on the functional parameters of rat liver mitochondria has been studied little. For this purpose, in our experiments, the effect of menthol (M), glycyrrhizinic acid (GA) and their supramolecular complexes obtained in different proportions on in vitro and in vivo studies on rat liver mitochondria was studied.

Dose-Dependent Cytotoxic Effects of Menthol on Human Malignant Melanoma A-375 Cells: Correlation with TRPM8 Transcript Expression

  • Background: Transient receptor potential melastatin 8 (TRPM8), a principle membrane receptor involved in calcium ion influx and cell signal transduction, has been found to be up-regulated in some cancer types, including melanomas. Efficiency of menthol, an agonist of TRPM8, in killing melanoma cancer cells has been reported previously, but the mechanisms remain unclear. We here determined whether in vitro cytotoxic effects of menthol on A-375 human malignant melanoma cells might be related to TRPM8 transcript expression. Materials and Methods: The $PrestoBlue^{(R)}$ cell viability assay was used to assess the in vitro cytotoxic effect of menthol after 24h of treatment. RT-PCR was used to quantify TRPM8 transcript expression levels in normal and menthol-treated cells. Cell morphology was observed under inverted phase contrast light microscopy. Results: TRPM8 transcript expression was found at low levels in A-375 cells and down-regulated in a potentially dose-dependent manner by menthol. Menthol exerted in vitro cytotoxic effects on A-375 cells with an $IC_{50}$ value of 11.8 ${\mu}M$, which was at least as effective as 5-fluorouracil ($IC_{50}=120{\mu}M$), a commonly applied chemotherapeutic drug. Menthol showed no dose-dependent cytotoxicity on HeLa cells, a TRPM8 non-expressing cell line. Conclusions: The cytotoxic effects on A-375 cells caused by menthol might be related to reduction of the TRPM8 transcript level. This suggests that menthol might activate TRPM8 to increase cytosolic $Ca^{2+}$ levels, which leads to cytosolic $Ca^{2+}$ imbalance and triggers cell death.

  • A study was made of the puff-by-puff delivery of menthol and tar for several commercial brands. The purpose of this study is for the brands constantly to maintain fresh aroma of menthol till the last puff in smoking. The results from this study could be used for development and evaluation of a mentholated cigarette. Prior to the purpose, a straightforward method from extraction through quantitative analysis of 9-menthol contained in cigarettes has been developed.

A STUDY ON THE FIXAT10N OF MENTHOL BY CLINOPTILOLITES (Clinoptilolite의 Menthol보유성에 관한 연구)

  • In this study, clinoptilolites were subjected to an array of experimental conditions, such as variations in heating temperature, reaction time, and molarity of acid and bass, and their responses measured. The activation levels were measured by adsorption indexes of toluene and ethyl alcohol and the X-ray spectra of clinoptilolites treated with acid and base. The effect of clinoptilolites on the menthol fuation was estimated by the amount of menthol produced after a given time of storage. The results were as follows: The optimum conditions for the clinoptilolites treatment were found to be IM HCI, IM NaOH, IMHCl-lM NaOH, and IM NaOH-lM HCl at $100^{\circ}C$ for 2 hours. 2. It was noted that clinoptilolites, untreated or treated with HCl, could not be used for the menthol fixation since the catalytic crating of menthol occurred with them. 3. The results from the residual menthol measurement in clinoptilolites after a specific period of storage showed that the best menthol fixation occurred in those treated with IM NaOH. 4. The menthol fuation by cigarettes with mentholated clinoptilolites was far better than that obtained by the use of mentholated tobacco column. 5. Most components in cigrettes smoke were generally reduced by the adsorption to clinoptilolites treated with IM NaOH.

  • In this study, it is intended to provide basic research data on hair loss and problematic scalp that is expanding to lower age groups and women. When using Company B's hair ampoule containing L-Menthol, Salicylic acid, and Dexpanthenol daily for women in their 20s with problematic scalp, the effect on the scalp and hair after 4 weeks was investigated. As a result of this study, when using Company B's ampoule containing L-Menthol, Salicylic acid, and Dexpanthenol, the subjects showed improvement effects such as relieving inflammation, reducing scalp fever and erythema, reducing itching and hair loss, and increasing hair thickness. In addition, it was confirmed that the scalp self-diagnosis of the subjects showed improvement effects on scalp satisfaction, scalp vitality and elasticity, heat generation, scalp pulling after cleaning, and itching. Through these research results, it is believed that products containing L-Menthol, Salicylic acid, and Dexpanthenol will help improve problematic scalp and hair.

  • Here we present how a supramolecular complex of Glycyrrhizic acid (GA) with Menthol (Mt) affects the blood glucose levels and glycogen synthesis in the liver of rats in in-vivo experiments with diabetes caused by alloxan. We have shown that Menthol, Glycyrrhizic acid and GA:Mt supramolecular complexes can restore functional dysfunction of the liver mitochondria in alloxan diabetes, i.e., inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular complex GA:Mt (4:1) in alloxan diabetes were more pronounced than those of menthol, GA and its GA:Mt (2:1) and GA: Mt (9:1) supramolecular complexes.

  • The metabolic capability of the cultured cells of peppermint was tested with whole intact cells by feeding appropriate exogenous substrates to the suspension cultures. Conversion of (-)-limonene into any other monoterpenes was not observed with the suspension cultures. (+)-Pulegone was converted into (+)-isomenthone and (-)-menthone, and (-)-menthone into (-)-menthol. The experiments confirmed that the suspension retained most of the menthol biosynthesis pathway in the cell except for a few loci. (-)-Isopiperitenone was transformed into (+)-pulegone, piperitenone, (-)-7-hydroxyisopiperitenone and two unidentified products.

Effect of L-Menthol on the Percutaneous Absorption of Ketorolac Tromethamine Across Human Cadaver Skin (사람 카다베르 피부를 통한 케토롤락 트로메타민의 경피 흡수에 L-menthol이 미치는 영향)

  • Transdermal delivery of ketorolac tromethamine, a potent non-narcotic analgesic, through human cadaver skin was investigated in vitro. A mixture of ethanol/water (40/60) containing 0, 1, 3, 5, and 8 (w/v)% L-menthol were used as a vehicle and penetration enhancer respectively. The permeation of ketorolac through human cadaver skin from saturated drug solution was evaluated at $37^{\circ}C$ with modified Franz diffusion cell. The in vitro skin flux and lag time were $1.23\;{\pm}\;0.11\;{\mu}g/cm^2{\cdot}hr$ and $5.56\;{\pm}\;0.34\;hr$, respectively. The cumulative amount of penetrated ketorolac containing L-menthol in ethanol/water (40/60) binary system was increased by the following order; 3%, 5%, 8%, 1%, 0%, and the lag time was decresed by the following order; 3%, 5%, 8%, 0%, 1%. The results suggested that a potential use of 3% L-methol is an effective penetration enhancer of ketorolac tromethamine through the human cadaver skin.

  • Nowadays a huge variety of products that aim to assist to quit smoking or reduce addictive symptoms are developed and manufactured with safety evaluation, but the safety of the most recent products of interest which do not contain tobacco and nicotine, and shape cigarettes is not evaluated and guaranteed relatively. This study was carried out to evaluate the single and repeated dose inhalation toxicity and genotoxicity of H menthol (Nicotine free-tobacco free) herbal cigarettes provided by Cigastop Ltd. in ICR mice. In this study, doses which we determined to expose to mice were 40 cigarettes for 6 hours a day to mice in single dose and 20 (high dose), 10 (middle dose) and 5 cigarettes (low dose) a day for 28 days in repeated dose inhalation toxicity, in vivo chromosome aberration test and micronucleus test. The particulate substances from H menthol herbal cigarettes also were gathered and used in the Salmonella typhimurium/microsome assay (Salmonella test; Ames test). We could find neither significant changes between control and treatment groups nor dose-response effects of test material at all except serum Ca level of female middle dose treatment group in repeated dose inhalation toxicity test. In conclusion, H menthol herbal cigarettes, when applied clinically intended dose we used, might not show any toxic and/or mutagenic effect.

  • This paper presents the effect of the supramolecular complex of GA (Glycyrrhizic acid) and Mt(menthol) and GA: Mt (4: 1) obtained on their basis can restore functional dysfunction of the liver mitochondria in alloxan diabetes, ie, inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular compound GA: Mt (4: 1) in alloxan diabetes were more pronounced than those of menthol, GA and its GK: Mt (2: 1) and GA: Mt (9: 1) compounds. According to the results obtained, the concentration of GA did not affect the peroxidation of lipid membranes of the liver mitochondria. However, a concentration of 15 μM of GA was found to reduce LPO (lipid peroxidation) formed by the effect of Fe2+ / ascorbate on the mitochondrial membrane by 58.0 ± 5.0% relative to control.
