In our previous study, the first structural gene (GGTI) encoding ${\gamma}-glutamyl$ transpeptidase was cloned and characterized from the fission yeast Schizosaccharomyces pombe, and its transcription, using the GGTI-lacZ fusion gene, containing the 1,085 bp upstream region from the translational initiation point, was found to be enhanced by sodium nitroprusside and L-buthionine-(S,R)-sulfoximine (BSO). In the present work, regulation of the GGTI gene was further elucidated. Non-fermentable carbon sources, such as acetate and ethanol, markedly enhanced the synthesis of ${beta}-galactosidase$ from the GGTI-lacZ fusion gene. However, its induction by non-fermentable carbon sources appeared to be independent of the presence of the Pap1 protein. Nitrogen starvation also gave rise to induction of GGTI gene expression in a Pap1-independent manner. The three additional fusion plasmids, carrying 754, 421 and 156 bp regions, were constructed. The sequence responsible for the induction by non-fermentable carbon sources and nitrogen starvation was identified to exist within a -421 bp region of the GGTI gene. Taken together, the S. pombe GGTI gene is regulated by non-fermentable carbon sources and nitrogen starvation.
Stimulation of epidermal growth factor receptor (EGFR) is essential in signaling pathway of tumor cells. Thus, EGFR has intensely studied as an anticancer target. We developed hologram quantitative structure activity relationship (HQSAR) models for data set which consists of tricyclic azepine derivatives showing inhibitory activities for EGFR. The optimal HQSAR model was generated with fragment size of 6 to 7 while differentiating fragments having different atom and connectivity. The model showed cross-validated $q^2$ value of 0.61 and non-cross-validated $r^2$ value of 0.93. When the model was validated with an external set excluding one outlier, it gave predictive $r^2$ value of 0.43. The contribution maps generated from this model were used to interpret the atomic contribution of each atom to the overall inhibition activity. This can be used to find more efficient EGFR inhibitors.
RI-Biomics field comes into the limelight as a new fusion radiation technology. These rapid development of RI-Biomics cause the necessity of establishment of a new methodical education program model for consistent training of professional manpower in RI-ADME, Biomics field. But domestic current status is not satisfied to training human resource development in RI-Biomics. Actually domestic educational organization related to RI-Biomics just run educational programs oriented basic theory, so practical and fusion education are not existed nowadays for preliminary RI-Biomics expert. Therefore we established a new education program model for educate of the expert in RI-Biomics field to overcome current problem about the route of knowledge that has more monotonous and concentrated tendency and non-professional education. To improve universality and practicality, we conduct education-training model survey about domestic and foreign country. This new human resource development model will contribute to fostering new expert in RI-Biomics field.
The ecotoxicological effects of 4-tert-octylphenol were observed on non-biting midge, Chironomus plumosus collected from Anyang stream in Seoul. The survival rate and adult emergence rate on C. plumosus exposed to octylphenol were not significantly affected. However, 4-tert-octylphenol induced developmental delay and disrupted sex ratio in high concentration of octylphenol. The mouth deformity such as tooth deletion or fusion in mentum, and tooth deletion in mandible were observed exposure to 4-tert-octylphenol. The deformity type of the mentum showed deletion (LT, 6.7%), and fusion (LT, 6.3%). Moreover, tooth deletion of mandible was observed in 4-tert-octylphenol treated groups (6.7%, 3 ppm).

The Korea Aerospace Research Institute plans to launch a lunar module by 2025, and so is carrying out a preliminary study. Landing stability on the lunar surface is a key design factor of a lunar module. In this paper, a 1/6 scale model of a lunar module is investigated, for its landing stability on non-level surfaces. The lunar module has four tripod legs, with aluminum honeycomb shock absorbers in each leg strut. ADAMS$^{TM}$, the most widely used multi-body dynamics and motion analysis software, is used to simulate the module's lunar landing. Three types of dampers in the struts (rigid, viscous, and aluminum honeycomb dampers), and two types of lunar surfaces (rigid and elastic) are considered. The Sforce function is adopted, to model the aluminum honeycomb dampers. Details on the modeling and analysis of the landing stability of the 1/6 scale lunar module and the simulation results are provided in this paper.
$\gamma$-Glutamyl transpeptidase (GGT, EC 2.3.2.2.) catalyzes the transfer of the $\gamma$-glutamyl moiety from $\gamma$-glutamyl containing ompounds, notably glutathione (GSH), to acceptor amino acids and peptides. A second gene (GGTII) encoding GGT was previously isolated and characterized from the fission yeast Schizosaccharomyces pombe. In the present work, the GGTII-lacZ fusion gene was constructed and used to study the transcriptional regulation of the S. pombe GGTII gene. The synthesis of $\beta$-galactosidase from the GGTII-lacZ fusion gene was significantly enhanced by NO-generating SNP and hydrogen peroxide in the wild type yeast cells. The GGTII mRNA level was increased in the wild-type S. pombe cells treated with SNP. However, the induction by SNP was abolished in the Pap1-negative S. pombe cells, implying that the induction by SNP of GGTII is mediated by Pap1. Fermentable carbon sources, such as glucose (at low concentrations), lactose and sucrose, as a sole carbon source, enhanced the synthesis of $\beta$-galactosidase from the GGTII-lacZ fusion gene in wild type KP1 cells but not in Pap1-negative cells. Glycerol, a non-fermentable carbon source, was also able to induce the synthesis of $\beta$-galactosidase from the fusion gene, but other non-fermentable carbon sources such as acetate and ethanol were not. Transcriptional induction of the GGTII gene by fermentable carbon sources was also confirmed by increased GGTII mRNA levels in the yeast cells grown with them. Nitrogen starvation was also able to induce the synthesis of $\beta$-galactosidase from the GGTII-lacZ fusion gene in a Pap1-dependent manner. On the basis of the results, it is concluded that the S. pombe GGTII gene is regulated by oxidative and metabolic stress.
주름버섯목(目) 느타리버섯과 민주름버섯목(目) 잔나비걸상버섯과의 이목간(異目間) 원형질체(原形質體)를 polyethylene glycol로 유도하여 융합주(融合株) heterokaryon을 선발하였다. 버섯최소배지에서 극히 균사생장이 느렸으며 버섯완전배지에서 3번 계대배양되면서 다소 생장이 빨라졌다. 융합주 36균주의 75%는 양친의 균사가 혼합된 균총형태였으며 16.7%는 새로운 형태, 8.3%는 느타리버섯 형태이었다. 이들 중 양친의 균총(菌叢)이 혼합된 형태는 3번 계대배양 후 모두 느타리버섯 형태로 변하였다. 균사(菌絲)에는 클램프연결체가 없었고 원기(原基)도 형성하지 않았다. 융합주(融合株)를 전기영동법으로 esterase, malate dehydrogenase, peroxidase의 동위효소(同位酵素) 분석(分析)으로 비교하였는데 잔나비걸상버섯 효소는 뚜렷하지 않았으나 새로운 밴드의 형성으로 보아 두 양친 genome간의 상호작용이 존재하였다.
야생형(野生形)인 만가닥버섯 Lyophyllum ulmarium(=Hypsizigus marmoreus) ASI 8007에서 분리한 분열자(分裂子)와 잔나비걸상버섯 영양요구주 Ganoderma applanatum ASI 7-18 (cys met)의 원형질체(原形質體)를 polyethylene glycol로 융합(融合)하여 융합주를 얻었다. 융합주는 GCM(Ganoderma complete modium)에서 양친균총이 혼합된 형태였으나 3회 계대 배양되면서 만가닥버섯 균총으로 변하여 균사에는 클램프연결체와 분열자를 지녔고, 만가닥과 거의 유사한 자실체를 형성하였다. 균사체로 esterase 동위효소(同位酵素)를 분석하였는데 만가닥이 1개 밴드, 잔나비걸상이 2개의 밴드를 가진데 비하여 융합주는 양친의 3개와 새로운 2개의 밴드를 나타내었다. 이들 융합주 상호간에는 균총형태, 생장속도, esterase 동위효소, 자실체 특성에 있어서 거의 차이가 없어 아주 안정되게 양친의 유전물질을 보유하였다.
소나무재선충(pine wood nematode, PWN) 감염 소나무를 현장에서 신속하게 진단을 할 수 있는 방법은 현재 없다. 본 연구에서는 PWN특이적인 항원으로 알려진 PWN-GaLectin을 baculovirus발현체계로 발현시켜서 총 1,464개의 fusion hybridoma 세포 주 라이브러리를 제작 하는 항원으로 이용하였다. 총 1,464개의 fusion hybridoma 세포 주 중, PWN-GaLectin에 대한 높은 반응을 보이는 62개의 fusion hybridoma 세포 주를 선별했다. 이들 중, 표준 PWN 감염소나무 PBS추출물과 PWN 단백질 추출물에 강한 반응을 보이는 세포 주 12개를 선별하여 단클론 항체(monoclonal antibody, Mab) 분비세포 주 확립을 위한 limited dilution을 실시하였다. Mab분비세포 주 확립을 위해서 표준 PWN 감염소나무 추출물에 대한 반응이 표준 정상 소나무 추출물 보다 높은 세포 주들을 선별했다. 그리고 추가로 PWN 단백질 추출물에 대해서 3종의 비 병원성 선충 단백질 추출물 보다 높은 반응을 보이는 세포 주들도 선별, 확립했다. 본 연구에서 확립된 Mab들을 우리는 현장과 실험실에서 사용할 수 있는 신속진단키트의 개발에 이용할 수 있을 것이다.
Glutaredoxins (Grxs) are thioloxidoreductases which are required for maintaining thiol/disulfide equilibrium in living cells. The Grx3 gene, which encodes one of the three monothiol Grxs in the fission yeast Schizosaccharomyces pombe, was characterized, and its transcriptional regulation studied. Genomic DNA encoding Grx3 was isolated by PCR, and a plasmid pTT3 carrying this DNA was produced. The DNA sequence has 1,267 bp, which would encode a monothiol Grx of 166 amino acids with a molecular mass of 18.3 kDa. The putative protein has 27% homology with Grx5, and contains many hydrophobic amino acid residues in its N-terminal region. S. pombe cells harboring pTT3 had increased Grx activity and enhanced survival on minimal medium plates containing aluminum (5 mM), BSO (0.05 mM), menadione (0.01 mM) or cadmium (0.2 mM). The 568 bp upstream region of Grx3 was fused into the promoterless b-galactosidase gene of the shuttle vector YEp367R to generate fusion plasmid pMJS10. Potassium chloride (KCl) and metals including aluminum and cadmium enhanced the synthesis of ${\beta}$-galactosidase from the fusion gene. The synthesis of ${\beta}$-galactosidase was also enhanced, in a Pap1-dependent manner, by fermentable carbon sources such as glucose (at low concentrations) and sucrose, but not by non-fermentable carbon sources such as ethanol and acetate. Grx3 mRNA increased in response to treatment with BSO. These observations indicate that S. pombe Grx3 is involved in the response to stress, and is regulated by stress.
