Fusarium spp. were isolated from the postharvest fruit rot of oriental melon fruits at commercial fruit markets in Korea during 2001 to 2003. The decayed fruits were covered with the fungal mycelia and eventually soft rotted. The disease started at the fruit stalk area, the calyx end of the fruit and skin of fruit. As the disease advanced, white to pinkish mycelia covered with the surface of decayed fruit. The cultural and morphological characteristic of Fusarium spp. were compared with descriptions of those reported previously, and identified as Fusarium equiseti, F. graminearum, F. moniliforme, F. proliferatum, F. sambucinum, and F. semitectum. Pathogenicity of the isolates was proved by artificial wound and unwound inoculation onto the healthy fruits. Two days after inoculation, aerial mycelia were noticed on the wound inocultion region of the fruit and developed soft rot symptoms. Although Fusarium spp. causing fruit rot disease in oriental melon have been reported in Korea, identification of the those species was not described. Therefore, this is the first report of Fusarium spp. causing postharvest fruit rot on oriental melon in Korea.
This study examined the effects of pearling, simple heating, baking and extrusion in processing of barley contaminated with deoxynivalenol (DON) on the reduction or destruction of DON. The DON contamination level of barley used in the experiment was 2.08 ppm. The DON-forming strain isolated from the barley was Fusarium graminearum, and the DON formation potential of which was 6.3 ppm on the average in Czapek-Dox broth medium. The DON removal efficiency of pearling was 56.5%, simple heating 3.8-32.7%, extrusion 53-59%, and baking 10.4%. As a large quantity of DON still remains after processing, it tis necessary to develop a new method of removing DON comletely.
Bacillus licheniformis KMU-3 shown a strong antifungal activity was isolated from Swedish forest soils. B. licheniformis KMU-3 produced a maximum level of antifungal substance under incubation aerobically at $24^{\circ}C$ for 24 hours in LB broth containing $1.0\%$ sodium acetate, $1.0\%$ ammonium sulfate at 180 rpm and initial pH adjusted to 8.0. Chloroform extraction of culture broth was confirmed inhibitory zone by plate assay and Rf value 0.49 substance by thin layer chromatography (TLC) represented high antifungal activity against Rhizoctonia solani AG-1. This substance also exhibited against Rhizoctonia solani AG-4, Colletotrichum orbiculare, Colletotrichum gloeosporioides, Cladosporium cucumerinum, Fusarium oxysporum, and Fusarium graminearum.
Mycotoxins are toxic secondary metabolites produced by fungi. They can be present in where agricultural-based commodities are contaminated with toxigenic fungi. These mycotoxins cause various toxicoses in human and livestock when consumed. Small grains including corn, barley, rice or wheat are frequently contaminated with mycotoxins due to infection mainly by toxigenic Fusarium species and/or under environment favorable to fungal growth. One of the most well-known Fusarium toxin groups in cereals is trichothecenes consisting of many toxic compounds. Deoxynivalenol (DON), nivalenol (NIV), T-2 toxin, and various derivatives belong to this group. Zearalenone and fumonisin (FB) are also frequently produced by many species of the same genus. In order to monitor Korean cereals for contamination with Fusarium and other mycotoxigenic fungal species as well, barley, corn, maize, rice grains, and soybean were collected from fields at harvest or during storage for several years. The fungal colonies outgrown from the grain samples were identified based on morphological and molecular characteristics. Trichothecene chemotypes of Fusarium species or presence of FB biosynthetic gene were determined using respective diagnostic PCR to predict possible toxin production. Heavy grain contamination with fungi was detected in barley, rice and wheat. Predominant fungal genus of barley and wheat was Alternaria (up to 90%) while that of rice was Fusarium (~40%). Epicoccum also appeared frequently in barley, rice and wheat. While frequency of Fusarium species in barley and wheat was less than 20%, the genus mainly consisted of Fusarium graminearum species complex (FGSC) which known to be head blight pathogen and mycotoxin producer. Fusarium composition of rice was more diverse as FGSC, Fusarium incarnatum-equiseti species complex (FIESC), and Fusarium fujikuroi species complex (FFSC) appeared all at considerable frequencies. Prevalent fungal species of corn was FFSC (~50%), followed by FGSC (<30%). Most of FFSC isolates of corn tested appeared to be FB producer. In corn, Fusarium graminearum and DON chemotype dominate within FGSC, which was different from other cereals. Soybeans were contaminated with fungi less than other crops and Cercospora, Cladosporium, Alternaria, Fusarium etc. were detected at low frequencies (up to 14%). Other toxigenic species such as Aspergillus and Penicillium were irregularly detected at very low frequencies. Multi-year survey of small grains revealed dominant fungal species of Korea (barley, rice and wheat) is Fusarium asiaticum having NIV chemotype.
Fusarium graminearum species complex (FGSC) causes Fusarium head blight in small grain cereals. To date, four species (F. graminearum, F. asiaticum, F. boothii, and F. meridionale ) belonging to FGSC frequently occur in Korean cereals. In addition, we first reported the occurrence of additional species (F. vorosii ) within FGSC, which was isolated from barley, corn, and rice in Korea. Phylogenetic analysis of the Fusarium isolates of this group using combined multigene sequences confirmed species identification. Moreover, the macroconidia produced by these isolates were morphologically similar to those of the F. vorosii holotype. Chemical analysis indicated that the F. vorosii isolates produced various trichothecenes such as nivalenol and deoxynivalenol with their acetyl derivatives along with zearalenone. Pathogenicity tests demonstrated that all of the F. vorosii isolates examined were pathogenic on barley, corn, and rice with variation in aggressiveness. This study is the first report of F. vorosii in Korean cereals, their pathogenicity towards barley and corn, and their ability to produce trichothecenes and zearalenone.

Fusarium head blight (FHB) is one of the most serious diseases in barley and wheat, as it is usually accompanied by the production of harmful mycotoxins in the grains. To identify FHB-resistant breeding resources, we evaluated 60 elite germplasm accessions of barley (24) and wheat (36) for FHB and mycotoxin accumulation. Assessments were performed in a greenhouse and five heads per accession were inoculated with both Fusarium asiaticum (Fa73, nivalenol producer) and F. graminearum (Fg39, deoxynivalenol producer) strains. While the accessions varied in disease severity and mycotoxin production, four wheat and one barley showed <20% FHB severity repeatedly by both strains. Mycotoxin levels in these accessions ranged up to 3.9 mg/kg. FHB severity was generally higher in barley than in wheat, and Fa73 was more aggressive in both crops than Fg39. Fg39 itself, however, was more aggressive toward wheat and produced more mycotoxin in wheat than in barley. FHB severity by Fa73 and Fg39 were moderately correlated in both crops (r = 0.57/0.60 in barley and 0.42/0.58 in wheat). FHB severity and toxin production were also correlated in both crops, with a stronger correlation for Fa73 (r = 0.42/0.82 in barley, 0.70 in wheat) than for Fg39.
In this study, five endophytic fungi belonging to the Aspergillus and Alternaria genera were isolated from Lagopsis supina. The antimicrobial activity of all fungal fermented extracts against Staphylococcus and Fusarium graminearum was tested using the cup-plate method. Among them, Aspergillus ochraceus XZC-1 showed the best activity and was subsequently selected for large-scale fermentation and bioactivity-directed separation of the secondary metabolites. Four compounds, including 2-methoxy-6-methyl-1,4-benzoquinone (1), 3,5-dihydroxytoluene (2), oleic acid (3), and penicillic acid (4) were discovered. Here, compounds 1 and 4 displayed anti-fungal activity against F. graminearum, F. oxysporum, F. moniliforme, F. stratum, Botrytis cinerea, Magnaporthe oryzae, and Verticillium dahlia with diverse MIC values (128-512 ㎍/ml), which were close to that of the positive control antifungal, actidione (64-128 ㎍/ml). Additionally, compounds 1 and 4 also exhibited moderate antibacterial activity against S. aureus, Listeria monocytogenes, Escherichia coli, and Salmonella enterica, with low MIC values (8-64 ㎍/ml). Moreover, compounds 1 and 4 displayed selective cytotoxicity against cancer cell lines as compared with the normal fibroblast cells. Therefore, this study proposes that the endophytic fungi from L. supina can potentially produce bioactive molecules to be used as lead compounds in drugs or agricultural antibiotics.
This study aimed to assess the incidence and distribution of toxigenic fungi in Korean oat. Toxigenic fungi were isolated from oat samples collected from 12 oat fields from heading to harvest in 2017 and 2018. A total of 745 fungal colonies were isolated based on morphology and identified using marker genes. About 92% of the fungal isolates were Fusarium spp. and others were Penicillium (5.9%) and Aspergillus (2.1%). Fusarium isolates comprised mostly of F. asiaticum (83.1%), followed by F. incarnatum (5.4%), F. proliferatum (3.5%), F. fujikuroi (2.8%), F. tricinctum species complex (FTSC) 11 (1.5%) and F. graminearum (1.0%). About 97% of F. asiaticum was nivalenol type, and 3-acetyl deoxynivalenol (3.2%) and 15-acetyl deoxynivalenol (0.4%) types also were found. Pathogenicity test of the selected Fusarium isolates revealed that F. asiaticum isolates have a wide range of virulence depending on the tested plants. F. graminearum and FTSC 11 isolates from blighted spikelets were the most virulent in naked oat. All Fusarium isolates (n=18) except one (FTSC 11) produced nivalenol (0.2-7.6 ㎍/g), deoxynivalenol (0.03-6.1 ㎍/g), and zearalenone (0.1-27.0 ㎍/g) on rice medium. This study is first report that F. asiaticum causes Fusarium head blight disease of oat in Korea. These findings demonstrate the dominance of F. asiaticum in oat agroecosystems as in rice, wheat and barley in Korea.
Five cereal grains(wheat, barley, rice, maize and sorghum) were collected from three Egyptian provinces known to be grain producers(Daqahlia, Gharbia and Kafer el-Sheikh). Two species of Alternaria(A. raphani and A. tenusinae); two species of Aspergillus(A. flavus and A. niger); one species of Cunninghamella(C. elegans); one Dreschslera species(D. myakt); three Fusarium species(F. graminearum, F. moniliform and F. solani); one Rhizopus species(R. stolonifer) and two species of Penicillium(P. digitatum and P. notatum) were isolated from the grains. The densities of these fungi and their frequencies of occurrence have been investigated. All the fungal isolates were tested for the production of toxic metabolites in culture media and the percentages of toxigenic isolates were calculated. The biological assay of the toxigenic fungal isolates showed significant variations in toxigenic activity. Thin layer chromatography revealed that the most active isolate produces moniliformin in culture media. The effect of culture conditions on the production of moniliformin was studied.
