Antioxidant and QR Inductive Activities of Novel Functional Soybean 'Agakong3'

  • In order to evaluate the bioactivity of 'Agakong3', which was newly bred, quinone reductase (QR) inductive activity and antioxidant activity were both assessed. The methanol extract of 'Agakong3' showed a significantly stronger QR inductive activity than other soybeans. The methanol extract of 'Agakong3' also showed a significantly stronger cytotoxicity on hepa1c1c7 than other soybeans. 'Agakong3' exhibited the most potent antioxidant activity in the Trolox equivalent antioxidant capacity (TEAC) assay whereas it showed significantly weak antioxidant in the DPPH assay. In total phenol and flavonoid contents, 'Agakong3' showed the highest contents regarding phenol and flavonoid compounds. Major isoflavones such as daidzein and genistein were quantitified by high performance liquid chromatography. 'Agakong3' also showed the highest total isoflavone contents. Results of correlation analysis showed that there were high correlation coefficients between the contents of isoflavone and TEAC and the contents of isoflavone and QR inductive activity, respectively. These results suggest that 'Agakong3' will be a promising and functional food material.

  • 고품질의 아가콩 음료개발을 위해 반응표면분석법에 의한 추출조건을 최적화 하였다. 아가콩의 최적 볶음조건은 250 ℃, 30분으로 설정하였다. 추출시간, 추출온도에 따른 아가콩의 품질지표인 pH, 색도 및 이소플라본 함량은 1% 이내에서의 유의적인 영향을 미치는 것으로 나타났다. 추출온도와 추출시간이 길어질수록 이소플라본 함량은 높은 것으로 나타났다. 반응표면분석의 이소플라본 추출 공정 최적화 결과 추출온도는 99.5 ℃, 추출시간은 1.7 h으로 나타났으며, 이 조건에서 이소플라본의 최적 수율은 10.63 ㎍/mL로 예측되었다.

  • This study was conducted to identify the differences in proteomic characteristics of 'Agakong', recombinant inbred line, and its parental genotypes 'Eunhakong' (Glycine max) and 'KLG10084' (G. soja). The isoflavone content of 'Agakong' was 3 times higher than that of its parental lines. A combined high-throughput proteomic approach was employed to determine the expression profile and identity of proteins using 2-dimensional gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry. The overall distribution patterns of proteins are quite similar, but lots of protein spot intensities varied among the genotypes. A total of 41 proteins, representing significant difference in the quantities of protein among the lines, were successfully identified. Among them, more than 50% of the proteins identified were subunits of glycinin and $\beta$-conglycinin, 2 major storage proteins. This study showed that the proteomic analysis could help to define specific changes in protein level and composition, which can occur in the generation of new soybean varieties.

Yield Performance and Nutritional Quality of 'Agakong' Soybean Harvested in Drained-Paddy and Upland Fields

  • The study was conducted to evaluate the performance of the popular isoflavone-rich soybean 'agakong' in upland and in drained-paddy fields. Analysis revealed no significant variation in terms of plant height, number of seeds per pod, number of nodes, and 100-seed weight between the two cropping system. Number of pods was significantly higher in paddy field(234.2kg 10a$^{-1}$) compared to those harvested in the upland field, which was later manifested on the yield where paddy soybeans obtained 278.1kg 10a$^{-1}$ whereas upland only obtained 179.3kg 10a$^{-1}$. This observed difference in yield was attributed to the observed higher amount of N in the paddy soil (0.907%) as compared to the upland soil (0.458%). In terms of nutritional content, protein and phytic acid contents were the only parameters that showed significant differences while oil, sugar, reducing power and fatty acids were all comparable in paddy and filed condition. Protein content was higher in upland soil (47.4%) than that of the paddy (44.9%) soil. On the opposite, phytic acid was higher in paddy (2.90%) than in upland (1.09%). This study showed that the yield of soybean is generally a factor of soil N, drained-paddy field production of soybean is comparable to upland-filed production with the benefit of increasing phytic acid content while maintaining its nutritional value.

콩나물의 asparagine 함량에 미치는 요인 구명 (Study on the factors affecting asparagine content in soy-sprout)

  • 알코올의 숙취해독에 뛰어난 효과가 있는 것으로 알려진 asparagine은 콩나물의 뿌리에 함량이 가장 많은 것으로 알려져 있다. 본 연구논문에서는 콩나물 뿌리 발생에 미치는 재배실의 온도, 일당 관수횟수, 재배일수 및 동일용기에 사용한 콩 량에 대한 영향을 조사하여 얻어진 연구결과를 요약하면 다음과 같다. 1. 아가콩과 풍산나물콩의 수율은 재배일수에 따라 경시적으로 증가하였으며, 재배실 온도가 $20^{\circ}C$에서 보다 $30^{\circ}C$에서 높았으며, 콩나물의 수율은 5회 관수, 8일 재배시 가장 높았다. 2. 아가콩과 풍산나물콩의 콩나물 부위별 건물중의 비율은 자엽이 가장 많았고 뿌리가 가장 낮았으며, 재배실 온도가 $20^{\circ}C$에서 보다 $30^{\circ}C$에서 비율이 더 높았다. 3. 아가콩의 asparagine 함량은 재배실 온도 $30^{\circ}C$에서 5회 관수 8일 재배한 하배축에서 18.9%로 가장 높게 나타났다. 4. 풍산나물콩의 asparagine 함량은 재배일수가 길수록, 재배온도가 높은 경우 높았으며, 하배축에서 가장 높았고, $30^{\circ}C$에서 2회 관수 8일 재배시 하배축에서 18.8%로 가장 높게 나타났다. 5. 아가콩은 300g, 풍산나물콩은 500g 치상시 수율이 가장 높았으며, 잔뿌리의 수는 아가콩과 풍산나물콩 모두 치상량이 증가함에 따라 증가하는 경향이었다.

The Characteristics of Cheonggukjang, a Fermented Soybean Product, by the Degree of Germination of Raw Soybeans

  • This study was conducted to observe the quality characteristics of cheonggukjang fermented with new small-seed soybeans ('Agakong') according to the degree of germination of the raw soybeans. The sprouting rate was $8.6{\pm}5.6%$ after 12 hr of germination, but at 24 hr it increased rapidly to $85.4{\pm}4.7%$. We confirmed that the total isoflavone content immediately after soaking was 273.9 mg%, which was at least 3 times greater than for common soybeans; content increased at the start of germination, and increased to 338.4 mg% by 24 hr of germination, but then decreased. The quantity of viscous substance of the cheonggukjang increased in proportion to the degree of germination of the raw soybeans. The levels of amino acids in the cheonggukjang made from non-germinated soybeans, and soybeans germinated 48 hr, were 12.45 and 10.06%, respectively. The isoflavone levels in the cheonggukjang were determined by the degree of germination of the raw soybeans. There were no significant differences between the odor, sweet taste, savory taste, bitter taste, and overall acceptability of the cheonggukjang with different germination times of 0 to 36 hr.

싹튼 콩으로 제조한 청국장의 발효기간에 따른 품질 변화 (Changes in Quality Characteristics of Cheonggukjang Made with Germinated Soybean)

  • 본 연구에서는 24시간 동안 발아시킨 콩을 이용하여 제조한 청국장의 발효기간에 따른 품질변화를 확인하고자 하였다. 24시간 발아시킨 콩의 pH는 6.7이었으며, 발효가 진행됨에 따라 pH가 점차 증가하여 48시간 후 발효를 종료할 때 pH 7.4를 나타내었다. 발아된 아가콩으로 만든 청국장의 점질물은 발효 48시간 동안 지속적인 증가를 보여 발효 종료 시에는 5.89%로 36시간 발효 이후는 점질물의 함량의 증가는 다소 미비하였다. 유리당은 raffinose, sucrose, glucose 및 fructose가 검출되었으며, 발효가 진행됨에 따라 급격히 감소하였다. 24시간 발아된 아가콩의 유리아미노산의 함량은 391.0 mg%로 나타났으며, 48시간 발아된 아가 콩의 유리아미노산은 10,994.8 mg%이었다. 발효 전기간 동안 glutamic acid가 가장 많았으며, aspartic acid, lysine, leucine, proline의 순이었다. 필수아미노산이 차지하는 비율은 전체 유리아미노산의 43%의 수준을 유지하였다. 발아 아가콩으로 만든 청국장의 관능검사 결과, 24시간 발아시킨 아가콩으로 제조한 청국장은 24-36시간 동안 발효시킨 것이 가장 우수하였다.
