Synthesis and Characterization of New Thermotropic Liquid Crystalline Polyurethanes (새로운 형태의 액정폴리우레탄의 합성 및 특성)

  • A series of thermotropic polyurethanes containing biphenyl units was synthesized by polyaddition reaction of diisocyanates such as 2,6-tolylene diisocyanate, 2,5-tolylene diisocyanate, 2,4-tolylene diisocyanate, 1,4-phenylene diisocyanate, and hexamethylene diisocyanate with 4,4'-Bis(9-hydroxynonoxy)biphenyl (BP9). 4,4'-bis(9-hydroxynonoxy)biphenyl exhibited a smectic type mesophase. Mesophase was found for all synthesized liquid crystalline polyurethanes except 1,4-PDI/BP9 based polyurethane. Structures of the monomer and the corresponding polymers were identified using FT-IR and $^1H-NMR$ spectroscopies. Their phase transition temperatures and thermal stability were also investigated by differential scanning calorimetry and optical polarizing microscopy.

Chemical Components and Pulverization conditions of the Pollens of Pinus rigida and Pinus koraiensis (리기다소나무와 잣나무 화분의 화학적 성분과 파쇄 조건에 관한 연구)

  • This study was to analyze the chamical components of the pollens of Pinus rigida and P. koraensis and to investigate the optimal conditions for pollen pulverization. The results are as follow: (1) The contents of moisture, crude ash, crude protein, crude fat, crude filber and carbohydrate in the pollen of P. rigida were 9.9%, 2.5%, 13.1% 2.9% 7.5% 64.1% respectively, while those of P. koraiensis were 9.8%, 2.1%, 11.1% 2.8% 8.1% 66.1% respectively. All the contents of P. rigida and P. koraiensis were much higher than those of general crop grains. (2) Eighteen different amino acids were detected in the pollen of P. rigida and P. koraiensis. Among them, ten essential amino acids were indentified, which showed high nutritions values. (3) The contents of vitamin A, B1, B2, C, and E in the pollen of P. rigida were 8.7 mg, 10.1 mg, 15.9 mg, 32.9 mg, 1.9 mg, respectively, while those of in the pollen of P. koraiensis were 7.9 mg, 9.7 mg, 14.5 mg, 34.2 mg, and 2.5 mg respectively. Vitamin C among them was abundant. (4)When the pollen grains of P. prigida was pulverized for 50minutes at 5,000rpm and P. koraiensis for 40 minutes at 5,000rpm by Overhead stirrer, the 100% of pollen was pulverized.

  • Melt copolymerization reactions of several bis(diethylamino)silane derivatives, bis(diethylamino)methylphenylsilane, bis(diethylamino)methyloctylsilane, 1,2-bis(diethylamino)tetramethyldisilane, and 1,3-bis(diethylamino) tetramethyldisiloxane, with 2,7-dihydroxyfluoren-9-one were carried out to yield poly[oxy(2,7-fluoren- 9-onenylene)oxy(diorganosilylene)]s bearing the fluoren-9-one fluorescent aromatic group in the polymer main chain: poly[oxy(2,7-fluoren-9-onenylene)oxy(methylphenylsilylene)], poly[oxy(2,7-fluoren-9-onenylene) oxy(methyloctylsilylene)], poly[oxy(2,7-fluoren-9-onenylene)oxy(tetramethyldisilylene)], and poly[oxy- (2,7-fluoren-9-onenylene)oxy(tetramethyldisiloxanylene)]. These polymeric materials are soluble in common organic solvents such as $CHCl_3$ and THF. FTIR spectra of all the materials reveal characteristic Si-O-C stretching frequencies at 1012-1018 $cm^{-1}$. In the THF solution, the prepared materials show strong maximum absorption peaks at 258-270 nm, strong maximum excitation peaks at 260-280 nm, and strong maximum fluorescence emission bands at 310-420 nm. TGA thermograms suggest that most of the polymers are essentially stable to $200^{\circ}C$ without any weight loss and up to $300^{\circ}C$ with only a weight loss of less than 5% in nitrogen.

  • For the examination of the role of monocerin(1) on the biological activity, (8S, 9S, 11R)-(-)-monocerin(20) and (9S, 11R)-(+)-fusarentin 4, 5-dimethyl ether(19) were synthesized by a condensation of the benzylic anion of ethyl 2, 3, 4-trimethoxy-6-methylbenzoate(16) with modifyed (R)-ethyl 3-hydroxyhexanoate (9). In a key step, bioreduction with active dried baker's yeast in organic solvent system was employed to get a chiral aldehyde 12. Their phytotoxic activities were tested on rice seedlings and lettuce seeds.

  • Calbindin-$D_{9k}$ (CaBP-9k), a cytosolic calcium-binding protein, is expressed in a variety of tissues, i.e., the duodenum, uterus, placenta, kidney and pituitary gland. Duodenal CaBP-9k is involved in intestinal calcium absorption, and is regulated at transcriptional and post-transcriptional levels by 1,25-dihydroxyvitamin D3, the hormonal form of vitamin D, and glucocorticoids (GCs). Uterine CaBP-9k has been implicated in the regulation of myometrial action(s) through modulation of intracellular calcium, and steroid hormones appear to be the main regulators in its uterine and placental regulation. Because phenotypes of CaBP-9k-null mice appear to be normal, other calcium-transporter genes may compensate for its gene deletion and physiological function in knockout mice. Previous studies indicate that CaBP-9k may be controlled in a tissue-specific fashion. In this review, we summarize the current information on calcium homeostasis related to CaBP-9k gene regulation by GCs, vitamin D and its receptors, and its molecular regulatory mechanism. In addition, we present related data from our current research.

A Novel Antifungal Analog Peptide Derived from Protaetiamycine

  • Previously, the 9-mer analog peptides, 9Pbw2 and 9Pbw4, were designed based on a defensin-like peptide, protaetiamycine isolated from Protaetia brevitarsis. In this study, antifungal effects of the analog peptides were investigated. The antifungal susceptibility testing exhibited that 9Pbw4 contained more potent antifungal activities than 9Pbw2. A PI influx assay confirmed the effects of the analog peptides and demonstrated that the peptides exerted their activity by a membrane-active mechanism, in an energy-independent manner. As the noteworthy potency of 9Pbw4, the mechanism(s) of 9Pbw4 were further investigated. The membrane studies, using rhodamine-labeled giant unilamellar vesicle (GUV) and fluorescein isothiocyanate (FITC)-dextran loaded liposome, suggested that the membrane-active mechanism of 9Pbw4 could have originated from the pore-forming action and the radii of pores was presumed to be anywhere from 1.8 nm to 3.3 nm. These results were confirmed by 3D-flow cytometric contour-plot analysis. The present study suggests a potential of 9Pbw4 as a novel antifungal peptide.

Tetrabromobisphenol A Induces MMP-9 Expression via NADPH Oxidase and the activation of ROS, MAPK, and Akt Pathways in Human Breast Cancer MCF-7 Cells

  • Tetrabromobisphenol A (TBBPA), the most common industrial brominated flame retardant, acts as a cytotoxic, neurotoxic, and immunotoxicant, causing inflammation and tumors. However, the mechanism of TBBPA-induced matrix metalloproteinase-9 (MMP-9) expression in human breast cancer cells is not clear. In human breast cancer MCF-7 cells, treatment with TBBPA significantly induced the expression and promoter activity of MMP-9. Transient transfection with MMP-9 mutation promoter constructs verified that $NF-{\kappa}B$ and AP-1 response elements are responsible for the effects of TBBPA. Furthermore, TBBPA-induced MMP-9 expression was mediated by $NF-{\kappa}B$ and AP-1 transcription activation as a result of the phosphorylation of the Akt and MAPK signaling pathways. Moreover, TBBPA-induced activation of Akt/MAPK pathways and MMP-9 expression were attenuated by a specific NADPH oxidase inhibitor, and the ROS scavenger. These results suggest that TBBPA can induce cancer cell metastasis by releasing MMP-9 via ROS-dependent MAPK, and Akt pathways in MCF-7 cells.

Agglutination Activity of Fasciola gigantica DM9-1, a Mannose-Binding Lectin

  • The DM9 domain is a protein unit of 60-75 amino acids that has been first detected in the fruit fly Drosophila as a repeated motif of unknown function. Recent research on proteins carrying DM9 domains in the mosquito Anopheles gambiae and the oyster Crassostrea gigas indicated an association with the uptake of microbial organisms. Likewise, in the trematode Fasciola gigantica DM9-1 showed intracellular relocalization following microbial, heat and drug stress. In the present research, we show that FgDM9-1 is a lectin with a novel mannose-binding site that has been recently described for the protein CGL1 of Crassostrea gigas. This property allowed FgDM9-1 to agglutinate gram-positive and -negative bacteria with appropriate cell surface glycosylation patterns. Furthermore, FgDM9-1 caused hemagglutination across all ABO blood group phenotypes. It is speculated that the parenchymal located FgDM9-1 has a role in cellular processes that involve the transport of mannose-carrying molecules in the parenchymal cells of the parasite.

  • One flavonoid, lignan glycosides and two diarylheptanoids were isolated from the sapwood of Betula maximowicziana. The structures were determined as : catechin 7-O-𝛽-D-xylopyranoside, lyoniresinol 9'-O-𝛽-D-glucopyranoside, 11-oxo-3, 8, 12, 17-tetrahydroxy-9-ene[7, 0]-metacyclophane and 11-oxo-3, 8, 9, 10, 12, 17-hexahydroxy[7, 0]-metacyclophane, respectively, on the basis of spectrosopic data and chemical correlations.

Investigation of the effect of SRSF9 overexpression on HIV-1 production

  • Serine-arginine-rich splicing factors (SRSFs) are members of RNA processing proteins in the serine-arginine-rich (SR) family that could regulate the alternative splicing of the human immunodeficiency virus-1 (HIV-1). Whether SRSF9 has any effect on HIV-1 regulation requires elucidation. Here, we report for the first time the effects and mechanisms of SRSF9 on HIV-1 regulation. The overexpression of SRSF9 inhibits viral production and infectivity in both HEK293T and MT-4 cells. Deletion analysis of SRSF9 determined that the RNA regulation motif domain of SRSF9 is important for anti-HIV-1 effects. Furthermore, overexpression of SRSF9 increases multiple spliced forms of viral mRNA, such as Vpr mRNA. These data suggest that SRSF9 overexpression inhibits HIV-1 production by inducing the imbalanced HIV-1 mRNA splicing that could be exploited further for a novel HIV-1 therapeutic molecule.
