Isolation and Identification of Histamine Degrading Bacteria from Kwamegi (과메기에서 histamine 분해능을 나타내는 세균의 분리 동정)

  • To isolate and identify histamine degrading bacteria from Kwamegi, bacteria were screened with restriction media containing histamine. Ten strains were selected through morphological and biochemical identification procedure followed by comparison with DNA sequence of 16 rRNA gene. And also, these strains were confirmed by the histamine degrading assay such as turbidity and enzymatic assay. The results of identification are as followings : Ewingella americana B791, Arthrobacter sp. R45S, Halomonas marisflava, Psychrobacter sp. 9B-7, Bacillus sp. LMC 21002, Psychrohacter cibarius BC-220, Bacillus megaterium KL-197 were identified showing homology of $99\%,\;95\%,\;98\%,\;99\%,\;99\%,\;99\%\;and\;98\%$, respectively. Three strains remain unidentified. Arthrobacter sp. R45S, H. marisflava, Bacillus sp. LMG 21002, B. megaterium KL-197 showed histamine degrading activity, whereas, Psychrobacter sp. 9B-7 only showed weak activity. Three unidentified strains also have histamine degrading activity. In contrast, E. american B791 and p. cibarius JG-220 did not show any significant activity of histamine degradation. The strains isolated from this study showed relatively fast growth rate and histamine degrading rate as compared to those from salted mackerel.

OMICS approaches in cardiovascular diseases: a mini review

  • Ranked in the topmost position among the deadliest diseases in the world, cardiovascular diseases (CVDs) are a global burden with alterations in heart and blood vessels. Early diagnostics and prognostics could be the best possible solution in CVD management. OMICS (genomics, proteomics, transcriptomics, and metabolomics) approaches could be able to tackle the challenges against CVDs. Genome-wide association studies along with next-generation sequencing with various computational biology tools could lead a new sight in early detection and possible therapeutics of CVDs. Human cardiac proteins are also characterized by mass spectrophotometry which could open the scope of proteomics approaches in CVD. Besides this, regulation of gene expression by transcriptomics approaches exhibits a new insight while metabolomics is the endpoint on the downstream of multi-omics approaches to confront CVDs from the early onset. Although a lot of challenges needed to overcome in CVD management, OMICS approaches are certainly a new prospect.

Anti-obesity effect of Auricularia spp. (목이버섯의 항비만 효과)

  • The purpose of this study is to discover a food material having anti-obesity effects and to disseminate information on the effects of the material to people who are interested in anti-obesity. For this study, 11 kinds of Auricularia (wood ear) spp., including 8 strains of Auricularia auricula-polytricha, and 3 strains of A. auricula-judae, were presented by the Jeollanamdo Agricultural Research & Extension Services. 3T3-L1 (preadipocyte cell) was used for identifying the inhibition effect on adipocyte differentiation. As a result, this study found that all the extracts had slightly different degrees of inhibition effects on adipocyte differentiation. Among the A. auricula-polytricha strains, strain 21001 showed the most significant effect (4.58%), and the inhibition effect of strain 21002 (4.43%) was the greatest among A. auricula-judae strains. Overall, the inhibition effect of A. auricula-polytricha strains was greater than that of A. auricula-judae strains. The results of mRNA and protein analysis also demonstrated that the inhibition effect of A. auricula-polytricha 21001 was superior to that of any other strains. An in vivo study using 56 ICR mice (6w, male), was performed for 4 weeks. A. auricula-polytricha 21001, which exhibited the most significant effect in the in vitro study was used to compose six different types of feeds. Daily body weight gain of the high-fat diet containing 0.2% 21001 extract feeding group was $0.22{\pm}0.08g$ (*p < 0.05), and it was 31.25% lower than that of the high-fat diet feeding group ($0.32{\pm}0.06$). Internal organ weight measurement and blood analysis were performed immediately after animal sacrifice. The results proved that treatment with more than 0.1% of A. auricula-polytricha strain 21001 could significantly reduce (more than *p < 0.05) the weight of liver and epididymal fat, and levels of glucose, total cholesterol, AST, and ALT in blood.

A Comparative Study on the Physiological Activities of Auricularia spp. (목이버섯 품종간 생리활성 비교 연구)

  • This study investigated the physiological effects of three species of Auricularia known as Auricularia polytricha (JNM21001), Auricularia auricula-judae black (JNM21002), and Auricularia auricula-judae brown (JNM21012). In the ORAC assay, Auricularia spp. showed antioxidant activities in the order of JNM21001>JNM21012>JNM21002. All Auricularia spp. strongly inhibited the action of ${\alpha}$-amyloglucosidase up to 60%. In order to further test in vivo anti-obesity effects, high fat diet induced ICR obese mice fed a diet containing 20% fat were used. All Auricularia spp. supplementation during high fat diet feeding significantly reduced body weight gain, epididymal fat pads weight, and lowered the food efficiency ratio compared to the high fat control (HFC) group. In particular, the group fed with JNM21012 had a lower average daily body weight gain of 0.45 g/day, demonstrating similar levels to the normal diet fed group. The group fed with JNM21012 significantly reduced lowered serum triglycerides (42%), total cholesterol (81%), and LDL-cholesterol level (66%) compared with the HFC group.

Influencing factors of using Korean Medicine services - focusing on the 2017 Korean Medicine Utilization Survey (한방의료이용 선택 요인에 관한 연구 - 2017 한방의료이용실태조사를 중심으로)

  • Objectives: The aim of this study was to investigate influencing factors of using Korean medicine services (KMS) using the 2017 Korean Medicine Utilization Survey (KMUS). Methods: Demographic statistics of the survey were summarized and influencing factors of the KMS experience and the intention to visit KMS were analyzed using logistic regression model with complex sample design. Influencing factors were specified based on Andersen's behavioral model of health care utilization and factors associated with individual recognitions of KMS. Additionally, using the ordinary logistic regression model without complex sample design, the survey data were analyzed to compare the results. Results: In the logistic regression analysis, sex, age, health condition, presence of chronic disease, a degree of knowledge about Korean Medicine, and a view about herbal medicine safety were statistically significant both in the KMS experience, and the intention to visit KMS. Marital status was statistically significant in the KMS experience, while family income, a view about the cost of KMS were statistically significant in the intention to visit KMS. Conclusion: Individual recognitions of KMS and enabling components should be considered when establishing KMS policies. In addition, future studies analyzing KMUS need to take into account the complex sample design features of the survey to avoid statistically misleading results.

A Comparison of Typhoon Wind Models with Observed Winds (해상풍 관측자료에 근거한 태풍 해상풍 모형간의 상호비교)

  • The sea-surface winds during the passage of 64 typhoons for 1979-1999 were simulated using two different typhoon wind models, ie, typhoon parametric model(TPM) and primitive vortex model(PVM). The model hindcast winds were compared with the winds observed at JMA ocean buoys(22001 and 21002) and Kyushu ocean observation tower. The analysis of ms and relative errors between hindcast and observed winds was made to find the accuracy and sensitivity of the typhoon wind prediction models. Both hindcast winds of TPM and PVM underestimate the observed typhoon winds, but PVM winds are more closer to the observations with less rms and relative errors. Relative errors of two model winds were small within 200km from typhoon center, but TPM's relative errors increase up to 70% as the radial distance from typhoon center increases beyond > 200km although PVM's relative errors remain in 20% with less sensitive to the distance from typhoon centers.

Characterization of a new cultivar of Auricularia auricula-judae 'Yong-A' (목이 신품종 '용아'의 특성)

  • 'Yong-A' was bred by Jeollanamdo Agricultural Research and Extension Services in 2015. It was primarily selected through monospore crossing between JNM21008 and JNM21013 in 2009. The strain is named as JNM-Mi-194 and 'Yong-A', showing major characteristics with good production yield. The cultivation characteristics, yield potential, and fruiting body characteristics of 'Yong-A' are as follows; The wrinkle of the fruiting body was of branching type its shape was wave type. MCM, YM, Malt, and PDA media were suitable for growth of this cultivar. The number of effective stipes was 13 ea/0.9 kg. The minor axis of pileus was 5 cm and the major axis was 9 cm. The yield was 291 g per plastic bag (0.9 kg). Yong-A required 60 days for mycelial running at $20^{\circ}C$. The growth and primordial period required 24 days, which was shorter than that of JNM-Mi-21002 (the conventional cultivar). Somatic incompatibility was observed between parental lines and Yong-A. Analysis of genetic diversity in the new variety 'Yong-A' showed a different profile compared to that of the parent strain when Random Amplified Polymorphic DNA primers were used.
