Korean Red Ginseng Extract Inhibits IL-8 Expression via Nrf2 Activation in Helicobacter pylori-Infected Gastric Epithelial Cells

  • Helicobacter pylori (H. pylori) causes gastric diseases by increasing reactive oxygen species (ROS) and interleukin (IL)-8 expression in gastric epithelial cells. ROS and inflammatory responses are regulated by the activation of nuclear factor erythroid-2-related factor 2 (Nrf2) and the expression of Nrf2 target genes, superoxide dismutase (SOD) and heme oxygenase-1 (HO-1). We previously demonstrated that Korean red ginseng extract (RGE) decreases H. pylori-induced increases in ROS and monocyte chemoattractant protein 1 in gastric epithelial cells. We determined whether RGE suppresses the expression of IL-8 via Nrf2 activation and the expression of SOD and HO-1 in H. pylori-infected gastric epithelial AGS cells. H. pylori-infected cells were treated with RGE with or without ML385, an Nrf2 inhibitor, or zinc protoporphyrin (ZnPP), a HO-1 inhibitor. Levels of ROS and IL-8 expression; abundance of Keap1, HO-1, and SOD; levels of total, nuclear, and phosphorylated Nrf2; indices of mitochondrial dysfunction (reduction in mitochondrial membrane potential and ATP level); and SOD activity were determined. As a result, RGE disturbed Nrf2-Keap1 interactions and increased nuclear Nrf2 levels in uninfected cells. H. pylori infection decreased the protein levels of SOD-1 and HO-1, as well as SOD activity, which was reversed by RGE treatment. RGE reduced H. pylori-induced increases in ROS and IL-8 levels as well as mitochondrial dysfunction. ML385 or ZnPP reversed the inhibitory effect of RGE on the alterations caused by H. pylori. In conclusion, RGE suppressed IL-8 expression and mitochondrial dysfunction via Nrf2 activation, induction of SOD-1 and HO-1, and reduction of ROS in H. pylori-infected cells.

Effect of the Acute Phase Response on the Performance and Superoxide Dismutase Activity in Broiler Chicks Fed on Dietary Krill Meal (사료 중 크릴 밀을 급여한 육계의 생산성과 SOD 활성에 미치는 급성기 반응의 영향)

  • Two experiments were conducted to examine the effects of the acute phase response on the performance and superoxide dismutase(SOD) activities in liver and erythrocyte of broiler chicks fed dietary krill meals A and B in experiment 1 and krill meal A in experiment 2. The experimental diets are basal diet based on yellow corn and soybean meal and diets substituted 2.0% of krill meal A or B with soybean meal of the basal diet, respectively. Day-old birds fed on the experimental diets and the acute phase response(immunological stress) was activated in the birds on 8-day of age by alternate day injection i.p. with 3 doses the Salmonella typhymurium lipopolysaccharide(LPS) in saline. The values during the acute phase response were compared with those controls injected with saline. The performance; daily gain, feed intake, and feed efficiency were different between dietary krill meal A and B in birds during the acute phase response and in the control. The acute phase response increased relative liver and spleen weights. Recovery of birds from the immunological stress was different between krill meals. Dietary krill meals increased activities of MnSOD and Cu/ZnSOD in erythrocyte cytosols during the actute phase response. Dietary krill meals did not affect the PHA-p response. The results indicated that the dietary krill meals may accentuate oxidative stress during the acute phase response.

Change of SOD, POD Activity and Stomata Resistance for Ozone on Rice(Oryza sativa L.) (오존 처리에 의한 벼 품종간 SOD, POD 활성과 기공저항성의 변화)

  • This study was carried to select the tolerance in rice varieites for ozone treatment through comparing SOD, POD and stomata resistance. In SOD Acivity, Sangnambatbyeo showed the highest activity on 1 hr after ozone treatment and after 24 hr, SOD activity was decreased. SOD activity of Hangangchalbyeo was not different in observed all times after ozone treatment. In POD activity, Hangangchalbyeo, Sangnambatbyeo and Ilpumbyeo were similar to POD activity but Baeknabyeo was the lowest activity on 1 hr after ozone treatment. Also, all varieties represnted the highest activity (above 90 %) an 24 hr after ozone treatment. In stomata resistance, Sangnambatbyeo was decreased stomata resistance on 1 hr after ozone treatment. Hangangchalbyeo, Sangnambatbyeo and Baekna were decreased stomata resistiance on 24 hr after ozone treatment except Ilpumbyeo.

Salsolinol, a Tetrahydroisoquinoline Catechol Neurotoxin, Induces Human Cu,Zn-superoxidie Dismutase Modificaiton

  • The endogenous neurotoxin, 1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline (salsolinol), has been considered a potential causative factor for the pathogenesis of Parkinson's disease (PD). In the present study, we examined the pattern of human Cu,Zn-superoxide dismutase (SOD) modification elicited by salsolinol. When Cu,Zn-SOD was incubated with salsolinol, some protein fragmentation and some higher molecular weight aggregates were occurred. Salsolinol led to inactivation of Cu,Zn-SOD in a concentration-dependent manner. Free radical scavengers and catalase inhibited the salsolinol-mediated Cu,Zn-SOD modificaiton. Exposure of Cu,Zn-SOD to salsolinol led also to the generation of protein carbonyl compounds. The deoxyribose assay showed that hydroxyl radicals were generated during the oxidation of salsolinol in the presence of Cu,Zn-SOD. Therefore, the results indicate that free radical may play a role in the modification and inactivation of Cu,Zn-SOD by salsolinol.

Development of Phragmites spp. and Miscanthus spp. Sod Using Natural Fiber Materials for a Vegetational Restoration (천연 섬유를 이용한 식생 복원용 갈대 및 억새속 식물의 뗏장개발)

  • Covering rate, visual rating and sod development were evaluated under three natural fiber materials with Phragmites spp. when over a plastic sheet. The results were as follows. (1) The last covering rate was high on jute net, coir mat and on Miscanthus sacchariflorus, respectively while the early covering rate was high on coir mat and on Miscanthus sinensis+perennial ryegrass. (2) The early growth was good on perennial ryegrass but the covering rate gradually turned poor because of summer drought. (3) Sod was highly developed on Phragmites japonica, Miscanthus sacchariflorus and Miscanthus sinensis compared with other species and mixtures. (4) The covering rate and visual rating were high on natural fiber materials such as coir mat and jute net when compared with on natural fiber materials such as none treatment plots. (5) The natural fibers materials on Phragmites spp. and Miscanthus spp. were effect on sod establishment. Sod coir mat was highly established. (6) The carpet-type sod was best developed on the coir mat.

The Effect of Red Ginseng Extract on Superoxide Dismutase Activity in the Kidney of Gamma-ray Irradiated Mice (홍삼 투여가 방사선에 조사된 생쥐 신장의 Superoxide Dismutase에 미치는 영향)

  • This study was prepared to observe the change of enzyme activities in kidney treated with red ginseng extract in the gamma ray irradiated mice. Determine the activity of SOD, peroxidase, catalase in the kidney a period of 1 day, 2 day, 3 day, 4 day, 5 day after a saline injection or injection of red ginseng extract or gamma ray irradiatied group into four classify. The activity SOD and catalase showed a tendency to increase and recovery at the early state but pay no regard. Wherease, the activity of peroxide restored and increased pay regard. A physiological saline injection group after gamma ray irradiation showed a tendency to diminish after remakable increase of activity of SOD, peroxidase and catalase than control group. Injection group of red ginseng extract after gamma ray irradiation observed rapid recovery on activity of SOD, peroxidase, catalase than a saline injection group. Experimental result suggested that injection of red ginseng extract after irradiation have the recovery effect on the changed of activity of SOD, peroxidase and catalase against radiation injury.

Cu,Zn-Superoxide Dismutase Is an Intracellular Catalyst for the H2O2-dependent Oxidation of Dichlorodihydrofluorescein

  • Dichlorodihydrofluorescein ($DCFH_2$) is a widely used probe for intracellular $H_2O_2$. However, $H_2O_2$ can oxidize $DCFH_2$ only in the presence of a catalyst, whose identity in cells has not been clearly defined. We compared the peroxidase activity of Cu,Zn-superoxide dismutase (CuZnSOD), cytochrome c, horseradish peroxidase (HRP), $Cu^{2+}$, and $Fe^{3+}$ under various conditions to identify an intracellular catalyst. Enormous increase by bicarbonate in the rate of $DCFH_2$ oxidation distinguished CuZnSOD from cytochrome c and HRP. Cyanide inhibited the reaction catalyzed by CuZnSOD but accelerated that by $Cu^{2+}$ and $Fe^{3+}$. Oxidation of $DCFH_2$ by $H_2O_2$ in the presence of a cell lysate was also enhanced by bicarbonate and inhibited by cyanide. Confocal microscopy of $H_2O_2$-treated cells showed enhanced DCF fluorescence in the presence of bicarbonate and attenuated fluorescence for the cells pre-incubated with KCN. Moreover, DCF fluorescence was intensified in CuZnSOD-transfected HaCaT and RAW 264.7 cells. We propose that CuZnSOD is a potential intracellular catalyst for the $H_2O_2$-dependent oxidation of $DCFH_2$.

Effect of 2,4,5-Trichlorobiphenyl (PCB-29) on Oxidative Stress and Activities of Antioxidant Enzymes in Tomato Seedlings

  • Leaves of two-week old seedlings of tomato (Lycopersicon esculentum) were treated with various concentrations (0, 0.2 and 0.4 $\mu$g/1) of 2,4,5-trichlorobiphenyl (PCB-29) and subsequent growth of seedlings, symptoms of oxidative stress and activities of antioxidant enzymes were investigated. Compared with the non-treated control, foliar application of PCB-29 decreased both biomass and superoxide ($O_2$) radical production but increased hydrogen peroxide production and lipid peroxidation such as malondialdehyde (MDA) formation with increased activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and guaiacol peroxidase (GPX). Further studies on the isozymes of SOD, peroxidase (POD) and APX showed that all three isozymes of SOD such as Mn-SOD, Fe-SOD and Cu/Zn-SOD, two among four isozymes of POD and all three isozymes of APX were selectively increased in response to PCB. Therefore, we suggest that a possible cause for the reduction of seedling growth by PCB exposure is the oxidative stress including over production of hydrogen peroxide and the selective expression of specific isozymes of some antioxidant enzymes.

Antioxidant effect of Aspalatone on KA-induced neurodegeneration in rat. (카인산 유발 신경세포 변성 모델에 대한 Aspalatone의 항산화 작용.)

  • 뇌신경 변성 / 퇴행과 관련된 중요한 병인론 중의 하나는 변성 과정에서 형성된 유리기(free radical)로 인한 항산화계의 평형 소실로 알려져 있다. Aspalatone (APT)의 예상되는 항산화 효능을 검정하기 위하여 본 실험에서는 Kainic acid (KA) 유발 뇌변성 모델을 적용하였다. KA 모델은 변연계의 간질성 경련과 신경세포 변성에 대하여 재현성 있는 병변 모델을 제공해 주며, 이와 같은 신경세포의 병독 기전에 산소 유리기가 관여함이 강력히 시사되고 있기 때문이다. KA 투여로 인하여 지속적이고도 전형적인 간질성 경련이 관찰되고 1일 이내에 높은 치사율을 보였으나 APT으로 인하여 그 간질성 경련 행위와 비율이 억제되고 KA 유발 치사율도 억제되었다. 최종 KA 투여 3일 후에 얻어진 흰쥐 해마 및 대뇌 피질에서 항산화 효소인 Superoxide dismutase (SOD), Catalase (Cat.), Glutathione peroxidase (GSH-PX) 및 과산화지질의 지표인 Malondialdehyde (MDA)를 검정하였다. 대조군에 비하여 KA는 뇌조직의 SOD-1을 유도하였으나, Cat.와 GSH-PX의 활성은 현저히 유도되지 않았고, 반면에 MDA 치는 현저히 증가하였다. 즉, Cat., GSH-PX와 같은 $H_2O$$_2$중화제가 동반 유도하지 않는 SOD의 유도는 세포내 축적되는 $H_2O$$_2$로 인하여 Fenton/Haber-Weiss 반응을 가속화하여 과산화지질화를 촉진함을 시사한다. APT 병용 투여로 SOD는 현저히 유도되지 않았으나 특히 Cat.가 현저히 유도되어지고 MDA는 억제되었다. 이와 같은 생화학적인 결과는 다음의 형태학적인 소견과 일치한다. Fos 관련 항원 (FRA)와 SOD-1을 면역세포화학 (Immunocytochemistry)적 방법으로 이중 표식 (double-labelling) 하였다. FRA는 KA로 인한 신경세포의 자극에 대한 지표로 응용하였고, SOD-1은 퇴행성 뇌질환에서 산화적 손상의 지표로 사용하였다. KA 투여로 해마의 dentate gyrus (DG) 내에 강한 면역환성 (immunoreactivity)이 나타났고 pyramidal cell layer (PCL)와 glia에 SOD-1이 강하게 염색되었다. APT 병용 투여로 상당수의 경련이 일어나지 않은 흰쥐는 해마의 DG에 FRA가 경미하게 염색되었고, PCL에 SOD-1도 경미하게 나타났으나, 경련이 나타난 쥐에서는 KA만을 투여한 흰쥐와 구별되지 않았다. 이상의 APT의 항산화 효과는 KA로 인한 뇌세포 변성 개선에 중요한 인자로 작용할 것으로 사료되나, 보다 명확한 APT의 기전을 검색하고 직접 임상에 응응하기 위하여는 보다 다양한 실험 조건이 보완되어야 찰 것으로 생각된다.

Manganese Superoxide Dismutase (MnSOD Val-9Ala) Gene Polymorphism and Susceptibility to Gastric Cancer

  • Background: Oxidative stress caused by the generation of reactive oxygen species plays an important role in human carcinogenesis. Manganese superoxide dismutase (MnSOD) Val-9Ala in the mitochondrial target sequence is the best known polymorphism of this enzyme. The purpose of the current research was to assess the association of MnSOD Val-9Ala genotypes with the risk of gastric cancer. Materials and Methods: This case-control study covered 54 gastric cancer patients compared to 100 cancer free subjects as controls. Extraction of DNA was performed on bioptic samples and genotypes were identified with a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: The frequencies of MnSOD Ala/Ala, Ala/Val and Val/Val genotypes in healthy individuals were 24.3, 66.7 and 9%, respectively. However, in gastric cancer patients, Ala/Ala, Ala/Val and Val/Val were observed in 24.0, 48.0 and 28.0% (p=0.01). In patients the frequency of MnSOD Val allele was higher (52%) compared to that in controls (42%). Conclusions: The results of this study show a positive association between MnSOD Val-9Ala gene polymorphism and risk of gastric cancer disease in Iranian population.
