In vitro Degradation of Saikosaponin-a in Physiological Condition

  • In vitro degradation of saikosaponin-a in physiological condition was conducted. Saikosaponin-a in 0.1 N-HCl of 25% 1,4-dioxane was incubated at $37^{\circ}$ and the products were analyzed and the time course of degradation was observed. Saikosaponin-a(Sa) was transformed to $saikosaponin-b_1(Sb_1)$ and saikosaponin-g(Sg) in the course of time. Sa was decreased drastically and not detected after 6 hours. $Sb_1$0 and Sg was increased with the time. After 6 hours the ratio of $Sb_1$ and Sg was about 4:1 and was maintained for 24 hours.

  • The optimal condition for the determination of saikosaponin a and d, the major pharmacologically active saponins of the roots of Bupleurum falcatum, was studied with the conversion of these saponins into diene saponins $(saikosaponin\;b_1\;and\;b_2)$. The complete separation and quantitative analysis of these saponins were performed by the method of high performance liquid chromatography using $NH_2$ column. The conversion of saikosaponin a and d into diene saponins under gastric pH was calculated. Thirty-three percent of saikosaponin a was converted to saikosaponin $b_1$ and 63 percent of saikosaponin d was converted to saikosaponin $b_2$.

  • Saikosaponin derivatives such as saikosaponins A, B1, B2, B3, B4, C, and D present in Bupleurum falcatum were analyzed by a high performance liquid chromatograph equipped with an evaporative light scattering detector, using different extraction solvents (water and 70% ethanol). The samples were injected into a YMC Pack Pro C18 column and separated using a gradient elution system with a mobile phase composed of acetonitrile and water at a flow rate of 1.1 mL/min. The content of saikosaponin derivatives was higher in 70% ethanol extract than in water extract. This study provides an efficient analytical method for determining the optimal conditions for extraction of saikosaponin derivatives, which can be used as a basis for development of functional foods and pharmaceutical products from B. falcatum.

  • A HPLC-ELSD method was developed to determine saikosaponin derivatives from Bupleuri Radix. Eight saikosaponins, saikosaponin c, i, h, a, $b_2$, g, $b_1$ and d, were analyzed under optimized HPLC conditions [column: Eclipse XDB $C_{18}$ ($150{\times}4.6mm$ i.d., $5{\mu}m$; mobile phase: $H_2O$ with 0.1% $CH_3$COOH (v/v) for solvent A and AcCN with 0.1% $CH_3$COOH (v/v) for solvent B, gradient elution; flow rate: 1mL/min; injection volume: $20{\mu}L$]. Good linearity was achieved in the range from 62.5 to $250{\mu}g/mL$ for each compound, and intra-day precision and accuracy at each concentration level varied between 0.05 and 5.45% and between 93.9 and 109.6%, respectively, whereas those for inter-day variations were between 0.91 to 2.73% and 94.3 to 106.1%. This HPLC-ELSD method was applied for the determination of sakosaponins from Bupleuri Radix samples, and saikosaponin a $(0.79{\pm}0.20mg/g)$, c $(0.33{\pm}0.06mg/g)$ and d $(0.48{\pm}0.15mg/g)$ were observed as major compounds. The other saikosaponins were shown under limit of quantification level thus couldn't be quantified. The present study suggested that the introduced HPLC-ELSD method is selective and reliable, and not only saikosaponin a, but also saikosaponin c and d should be employed as the standard markers for Bupleuri Radix.

Saikosaponin Contents and Growth Characteristics on Cutting and Flower Removal in Bupleurum falcatum L. (예취 및 적화처리에 따른 시호의 생육특성 및 saikosaponin 함량)

  • This study was conducted to investigate the effect of cultural methods(cutting, flower removal) on growth and quality of B. falcatum L. Jeongseon local cultivar collected in Korea and Mishima local cultivar introduced from Japan were used. Some of the results obtained are as follows; Jeongseon local cultivar showed less stem branches and shoot weight compared to Mishima. However, Jeongseon local cultivar showed tall plant height, high root fresh and dry weight, and high levels of saikosaponin-a and total-saikosaponin, but low saikosaponin-c content than that of Mishima. Both cultivars seeded on March 20 had longer main root, bigger stem diameter, few stem branch, and high saikosaponin-c content compared with those of late seeded one, April 30. Growth characteristics such as plant height, stem diameter, stem branch number, shoot weight, root diameter, root fresh and dry weight, and root branch number were increased in a low planting $density(30{\times}15\;cm)$, but the content of saikosaponin was not affected by planting density. Jeongseon and Mishima cultivars seeded on April 10 with $30{\times}15\;cm$ planting density and April 30 with $30{\times}10\;cm$ planting density contained highest total saikosaponin levels, respectively. However, average root dry weight was not affected by planting time or density in both Bupleurum cultivars. Both cutting and flower removal increased the length of main root, fresh and root weight, and saikosaponin-a, saikosaponin-d and total-saikosaponin in Jeongseon cultivar than that of Mishima. As the harvesting time was delayed, plant height, stem diameter, shoot weight, length of main root, root fresh and dry weight were increased, but content of saikosaponin-a, saikosaponin-d and total-saikosaponin were decreased.

  • Seven genetic lines of Bupleurum falcatum L. from different geographical regions were analysed for saikosaponin contents and chromosomal numbers. The somatic chromosome numbers of B. falcatum originated from Euisong, Iri, Milyang, Sangnam, Taejon, and Youngchon were 2n=20 while Mishimasaiko showed 2n=26. However, chromosome features were different in plants grown in different geographical regions. Generally, Korean lines had higher saikosaponin contents than Mishimasaiko which is Japanese and Sangnam lines had highest saikosaponin contents compared to other tested lines.

Saikosaponin Contents in Somaclones Derived from Different Aged Calli of Bupleurum falcatum L. (배양 기간이 다른 캘러스로부터 재분화된 시호 (Bupleurum falcatum L.) 체세포클론의 사이코사포닌 함량 변이)

  • The contents of saikosaponin a, c and d in the root of somaclones and wild plants of B. falcatum L. were determined. The saikosaponin contents of wild plants collected from Chongson-gun, Kangwon Province and Yongdok-gun, Kyongbuk Province were higher than those of the somaclones derived from 3-month and 2-year aged calli. The mean values of saikosaponin content in 2 year-old somaclones derived from the callus cultured three months were higher than those in 1 year-old somaclones from 2-year aged callus. Variation of saikosaponin content depending on the culture periods was not found. Four plants among 1-year old somaclones showed saikosaponin content twice as much as wild plants had. This result implies the possible use of somaclonal variation for crop improvement.

Effect of Organic Fertilizers Application in Root Yield and Saikosaponin Contents in Bupleurum falcatum L. (宥機質 肥料 施用이 柴胡의 根收量과 Saikosaponin 含量에 미치는 影響)

  • This study was carried out to investigate the effect of several organic fertilizers on the growth characteristics, root yield and the levels of the efficacious constitutents saikosaponins in Bupleurum falcatum L. in the field of Chonnam RDA. Naju, 1994. Five organic fertilizers were used ; rice straw manure(RSM), rice straw(byproduct of mushroom)manure[RS(BM)M], pig-dropping sawdust manure(PSM),poultry manure(PM)and sawdust(byproduct of mushroom) manure[S(BM)M]. The results were summarized as follows ; The chemical properties, such as organic matter, available phosphate, $K_2O$, CaO, and MgO of soil were increased on all plots with the addition of organic fertilizers, specifically in PM+RS(BM)M application. PM+RS(BM)M treatment was higher in emergence rate and better in the growth charaters of shoot, root parts of plants than those of conventional fertilization and the highest dried root yield resulted from using 39% PM+RS(BM)M treatment. Thus, the organic constituents of the plant leaves were affected in the same ways by all treatments, but the T-N, $P_2O_5$, $K_2O$, CaO and MgO contents of the roots were highly increased in PM+RS(BM)M treatment, also the total saikosaponin levels in the roots increased with different organic fertilizers in the following order : 1.70% in PM+RS(BM) treatment, 1.66% in RS(BM) treatment, and 1.57% in RSM+N-P-K treatment. There were significant positive correlation observed between the growth characters of shoot, root parts of plants, yield and contents of T-N, $P_2O_5$, $K_2O$, total-saikosaponin in B. falcatum roots.
