Effects of Organic Materials on Soil Organisms in a Korean Ginseng Field (인삼재배지 유기물 시용이 토양미생물과 미소동물에 미치는 영향)

  • The aim of this study is to evaluate the effects of organic materials on soil organisms. Changes in the community structure, and population density of soil organisms (microbes, nematodes, and microarthropods) were studied in a Korean ginseng field. Phospholipid fatty acids analysis showed that the relative abundances of bacteria, fungi, and actinomycetes did not differ significantly. The aerobes/anaerobes ratio was the lowest in soils amended with leaf mold, indicating that the decomposition speed was slow. Further, the addition of leaf mold to the soil enhanced the saturated/monounsaturated fatty acid ratio and cyclopropyl fatty acid/precursor ratio, which indicated an increase in environmental stresses. Application of pig manure compost (PMC) had positive effects on the population density of nematodes, and negative effects on that of oribatid mites. The population densities of nematodes, and microarthropods remained relatively low in the plots that had been treated with leaf mold or pig manure compost. It is suggested that pre-planting soil management directed at enhancing the biological decomposition efficiency should be continued over a long period to increase the soil bioactivity in virgin soils.

Control of the CaCO3 Saturation Index Parameters for Protecting the Corrosion of Waterworks Pipe (상수도관 부식방지를 위한 탄산칼슘 포화지수(LI) 인자 제어에 관한 연구)

  • Calcium Carbonate Saturation Index (LI: Langelier Index), an indicator of $CaCO_3(s)$ saturation, indicates corrosiveness of drinking water and it has been used to monitor drinking water conditions in USA, E.U, and Japan. The objective of this research was to measure LI parameters including water temperature, pH, total alkalinity, calcium ion concentration, and electric conductivity, and to evaluate possibility of using LI in domestic system. Results showed that water temperature varied from 2.0 to $26^{\circ}C$ during 15 months, indicating an average annual temperature of $23.9^{\circ}C$. Total alkalinity was from 20 to 45 mg/L. The concentration difference between total alkalinity and $HCO_3{^-}$ value was hardly observed; the concentration of total alkalinity can be replaced by that of $HCO_3{^-}$. Tap water had a medium corrosiveness since LI values were from 2.0 to 0.5. To reduce the corrosiveness and to increase LI values of drinking water, the results of this study showed that chemicals such as $Ca(OH)_2$, $CaCO_3$, NaOH, or $NaHCO_3$ should be added to water treatment plants.

Impacts of Air Pollution on Forests : A Summary of Current Situations (대기오염이 삼림에 미치는 영향 : 피해현황과 원인을 중심으로)

  • Issues of declining growth and vigor in forests are major concerns in many areas around the world, especially in response to predictions in the 1980s of widespread forest declines. This paper summarizes the current state of knowledge for forests in North America, Europe, and East Asia. Forest declines near point-sources of pollution(such as metal smelters) have been well recognized for a century, but evidence of widespread impacts away from point-sources remains relatively uncommon. In North America, significant forest decline has resulted from high concentrations of ozone near Los Angeles, California, and around Mexico City. Some high-elevation forests of red spruce in the eastern U.S. have declined in the past 20 years ; evidence indicates a role of low-pH fog in reducing they cold-tolerance of spruce. In Europe. most attention has focused on Norway spruce stands that developed yellow foliage, needle loss, and in some cases mortality. This syndrome appears to be related generally to an inadequate supply of magnesium, perhaps coupled with a very high supply of nitrogen. Despite localized areas that show declining trees, overall stand growth and standing biomass in Europe increased from. 1970 to 1990. Much less information is available for East Asia. Many industrialized regions in China have a pH of rain <4.5, and some connections between pollution and forest decline have been suggested. Pollution impacts on forests near cities in Korea include needle chlorosis, reduced needle retention, and declining species diversity. Overall, temperate forests show no widespread declines, and no evidence of substantial effects of pollutants on forest growth or vigor. Small areas showing declining forests may indeed demonstrate pollution impacts, and may provide cause for concerns about future impacts on larger areas.

Studies on the Quick Ripening Process for Soysauce (속양(速釀)간장 제조(製造)에 관(關)한 연구(硏究))

  • This experiment was conducted as a preliminary test to invent the quick ripening process for soysauces. The bran koji extract of 2 kinds of mold (Asp. niger and Asp. sojae) was used as enzyme sources in order to improve the rate of enzymatic conversion of raw materials and shorten the ripening period of soysauces. Soybean koji (inoculated Asp. sojae) and steamed soybean were enzymatically decomposed for 8 hours at $40^{\circ}C$, pH 4-6 with stirring at a rate of 1500 rpm. After the enzymatic treatment, soysauces were fermented by 4 kinds of processes with the above-mentioned salt tolerant microorganisms inoculated. After 4 weeks, sensory evolution for these soysauces products and analysis of chemical components were carried out. The results obtained were summarized as follows. 1. Soysauces prepared from decomposed koji by Asp. niger enzyme complex showed higher extractables, such as total nitrogen, amino nitrogen, pure extract and reducing sugar, than any other procedure. Extracted amounts of each components of soysauces prepared from this process during 1 month were similar to those of soysauces made from general procedure (control) for 5 to 6 months. 2. According to sensory evaluation, soysauce prepared from decomposed koji also showed best taste and were similar to that of soysauces prepared by general method for 6 months.

Phospholipids Isolation from Squid Viscera Residues After Supercritical Carbon Dioxide Extraction (오징어 내장의 초임계 이산화탄소 추출 잔류물로부터 인지질의 분리)

  • Phospholipids were recovered from squid viscera residues by ethanol extraction after supercritical carbon dioxide($SCO_2$) extraction and from squid viscera was not processed $SCO_2$ by various organic solvent extraction. $SCO_2$ extraction were performed at $45^{\circ}C$ and 20 MPa for removal of non polar lipid molecules from freeze dried squid viscera sample. Phospholipids were extracted from freeze dried squid viscera sample by chloroform, hexane, methanol, and ethanol and from $SCO_2$extracted squid viscera sample by ethanol. The pH was fixed at 5.7 for all phospholipids extraction conditions. Phospholipid classes were analyzed by HPLC equipped with evaporative light scattering detector (ELSD). Phosphatidyl choline(PC) extracted by ethanol from $SCO_2$ extracted residues was higher than that of extracted by ethanol from squid viscera. But phosphatidyl ethanolamine(PE) and phosphatidic acid(PA) were extracted higher percentage in raw squid viscera. The fatty acid compositions in phospholipids extract by ethanol extract from $SCO_2$ extracted residues were analyzed by gas chromatography(GC). Docosahexanoic acid(DHA) was found in highest percentage in phospholipid extract.

Cloning, Expression, and Characterization of a Novel GH-16 β-Agarase from Agarivorans sp. JA-1 (Agarivorans sp. JA-1 유래 신규 GH-16 β-agarase의 클로닝, 발현 및 특성)

  • Authors report the glycoside hydrolase (GH) family 16 ${\beta}$-agarase from the strain of Agarivorans sp. JA-1, which authors previously stated as recombinant expression and characterization of GH-50 and GH-118 ${\beta}$-agarase. It comprised an open reading frame of 1,362 base pairs, which encodes a protein of 49,830 daltons consisting of 453 amino acid residues. Valuation of the total sequence showed that the enzyme has 98% nucleotide and 99% amino acid sequence similarities to those of GH-16 ${\beta}$-agarase from Pseudoalteromonas sp. CY24. The gene corresponding to a mature protein of 429 amino acids was recombinantly expressed in Escherichia coli, and the enzyme was purified to homogeneity by affinity chromatography. It showed maximal activity at $40^{\circ}C$ and pH 5.0, representing 67.6 units/mg. Thin layer chromatography revealed that mainly neoagarohexaose and neoagarotetraose were produced from agarose. The enzyme would be valuable for the industrial production of functional neoagarooligosaccharides.

Kinetics for the Transformation of Outer Charge Transfer Complex to Inner Complex (Outer Charge Transfer Complex가 Inner Complex로의 변환에 따른 속도론적 연구)

  • Formation of charge transfer complex between iodine and substituted aniline [aniline, N,N-dimethylaniline(N,N-DMA), 2,6-dimethylaniline(2,6-DMA), 2,4,6-trimethylaniline(2,4,6-TMA)] in CHCl$_3$, CH$_2$Cl$_2$ : CHCl$_3$ (1 : 1), and CH$_2$Cl$_2$ have been studied kinetically by using conductivity method. In the transformation of initially formed outer charge transfer complex to inner complex, the effects of substituted aniline as electron donor and polar medium on the reaction were investigated. The rate of transformation depend on the dielectric contribution of medium and pK$_a$ value of substituted aniline. The order of rate increasing is 2,4,6-TMA, 2,6-DMA, aniline, and N,N-DMA. The activation enthalpy ${\Delta}H^{\neq}$ for 2.5 M-substituted aniline in CHCl$_3$ at 25$^{\circ}C$ is respectively N,N-DMA, 3.47 kcal/mol; aniline, 4.25 kcal/mol; 2,6-DMA, 7.79 kcal/mol and 2,4,6-TMA, 7.96 kcal/mol; and activation entropy ${\Delta}S^{\neq}$ is large and negative value of -41 ~ -55 cal/mol${\cdot}$K.

CLINICAL APPLICATION OF MTA(MINERAL TRIOXIDE AGGREGATE) FOR APEXIFICATION (치근단 형성술(Apexification)에 있어서 MTA(Mineral Trioxide Aggregate)의 적용)

  • Traumatic injuries in young patients can result in the interruption of the development of the incompletely formed roots. In teeth with incomplete root-end formation and necrotic pulps, the root canals must be completely debrided. Because of a lack of an apical stop and the presence of thin and fragile walls in these teeth, it is imperative to perform apexification to obtain an adequate apical seal. Calcium hydroxide has become the material of choice for apexification. Despite its popularity for the apexification procedure, calcium hydroxide therapy has some inherent disadvantages that include variablility of treatment time, unpredictability of apical closure, difficulty in patient follow-up, and delayed treatment. An alternative treatment to long-term apexification procedure is the use of an artificial apical barrier that allows immediate obturation of the canal. MTA(Mineral Trioxide Aggregate) is a powder consisting of fine hydrophilic particles of tricalcium silicate, tricalcium aluminate, tricalcium oxide and silicate oxide. MTA has a pH of 12.5 after setting, similar to calcium hydroxide. This may impart some antimicrobial properties. MTA has low solubility and a radiopacity slightly eater than that of dentin. Also, MTA leaked significantly less than other materials and induced hard-tissue formation more than other materials.

Rapid Processing of the Fish Sauce and Its Quality Evaluation (속성어간장 제조 및 품질 평가)

  • Changes in chemical characteristic, microflora, and sensory evaluation of fish sauce extracted at an interval of one week from fermented solution were investigated. pH was reduced from 6.0 to 4.5, and trimethylamine oxide from 132.5 to 87.2 mg/100g during fermenting periods. Trimethylamine increased from 5.6 to 50.2 mg/100g, and volatile basic nitrogen from 48.3 to 232.5 mg/100g. Bacterial flora isolated from the fish sauce were 70% Lactobacillus sp. and 13% Bacillus sp. Among the free amino acids, alanine, glutamic acid, valine, and methionine contents constitute 40% of the total free amino acids. Major non-volatile organic acid of the fish sauce was lactic acid (76%). Sensory evaluation results of the fish sauce were higher than the traditional soybean sauce after 28 days of fermentation.

A Study on the Tannin Weighting of Silk (견의 탄닌가공에 관한 연구)

  • The studies were performed to investigate the optimum conditions for tannic processing of silk by use Chinese Gallotannin and synthesized tannic acid, which are aimed at weighting, dyeing and physical properties of tannin treated silk. 1. It was reasonable that the concentration of tannin solution is 30 grams per liter of Chinese Gallotannin, 15 grams per liter of tannic acid for the efficient weighting of processed silk. The temperature and time for tannin treatment was optimum at 80$^{\circ}C$, 60 minutes and the acidity of tannin solution at pH 2 to 3. 2. In dyeing the tannintreated silk by Acid dye Orange II, the temperature and time was reasonable at 60$^{\circ}C$, 90 minutes to control the desorption of tannin components weighted onto silk. 3. The colour differences ($\Delta$E) of dyed silk fabric by soaping could be remarkably narrowed by tannin treatment, resulting in improving the washing fastness of tannin treated silk by two grades more than that of untreated one. 4. The light fastness of tannin treated silk could be drastically improved by reducing the dye-loss of dyed silk fabric which was coused from the Ultra-violet ray irrdiation. 5. The rubbing fastness and water repellency of tannin treated silk was at the same level with that of untreated one. However, the Drape coefficient of tannin treated silk was decreased more than that of untreated one, which is closely related with fabric softness and dressing appearence.
