New Records of Two Arcuospathidium Subspecies (Ciliophora: Haptoria: Arcuospathidiidae) from Korea

  • Arcuospathidium is a haptorian ciliate genus composed of 18 species, and only one species has been reported in Korea. Here, we identify two unrecorded Arcuospathidium subspecies by morphological observation of both living and protargol-impregnated specimens with the small subunit ribosomal RNA (18S rRNA) gene sequence. These subspecies, Arcuospathidium cultriforme cultriforme (Penard, 1922) Foissner, 1984 and A. cultriforme scalpriforme (Kahl, 1930) Foissner, 2003, were isolated from various terrestrial habitats in July and August 2013, respectivley. Arcuospathidium cultriforme cultriforme is similar to A. cultriforme scalpriforme by a knife-shaped body, a twisted-shaped macronucleus, number of dorsal brushes, position of dorsal brushes, and shape of macronucleus but former mainly differs from the body length to oral bulge length ratio (27-38% vs. 41-53%), extrusome (one types vs. three types), cyst shape (roughly faceted wall vs. smooth surface and thin wall) and number of somatic kinety rows(18-30 vs. 30-44). Additionally, we analyzed the 18S rRNA gene sequences of two A. cultriforme subspecies and compared them with the sequences from GenBank to confirm their identification at the molecular level. As the results of genetic analysis, the 18S rRNA gene sequence of the Korean A. cultriforme cultriforme population is most similar to that of Austrian population. Also, the sequence of the Korean A. cultriforme scalpriforme population is most similar to that of another population with some nucleotide differences.

Risks for Readmission Among Older Patients With Chronic Obstructive Pulmonary Disease: An Analysis Using Korean National Health Insurance Service - Senior Cohort Data

  • Objectives: The high readmission rate of patients with chronic obstructive pulmonary disease (COPD) has led to the worldwide establishment of proactive measures for identifying and mitigating readmissions. This study aimed to identify factors associated with readmission, as well as groups particularly vulnerable to readmission that require transitional care services. Methods: To apply transitional care services that are compatible with Korea's circumstances, targeted groups that are particularly vulnerable to readmission should be identified. Therefore, using the National Health Insurance Service's Senior Cohort database, we analyzed data from 4874 patients who were first hospitalized with COPD from 2009 to 2019 to define and analyze readmissions within 30 days after discharge. Logistic regression analysis was performed to determine factors correlated with readmission within 30 days. Results: The likelihood of readmission was associated with older age (for individuals in their 80s vs. those in their 50s: odds ratio [OR], 1.59; 95% confidence interval [CI], 1.19 to 2.12), medical insurance type (for workplace subscribers vs. local subscribers: OR, 0.84; 95% CI, 0.72 to 0.99), type of hospital (those with 300 beds or more vs. fewer beds: OR, 0.77; 95% CI, 0.66 to 0.90), and healthcare organization location (provincial areas vs. the capital area: OR, 1.66; 95% CI, 1.14 to 2.41). Conclusions: Older patients, patients holding a local subscriber insurance qualification, individuals admitted to hospitals with fewer than 300 beds, and those admitted to provincial hospitals are suggested to be higher-priority for transitional care services.

Antioxidants in Serum and Induced Sputum of COPD Patients (만성폐쇄성폐질환 환자의 혈청 및 유도객담에서의 항산화제)

  • Background : Although an oxidants and antioxidants imbalance has been considered in the pathogenesis of chronic obstructive pulmonary disease (COPD), there is a paucity of reports focussing on the smoking-induced changes of oxidants and antioxidants in COPD. Method : The concentration of antioxidants (ascorbic acid, uric acid, retinol, and $\alpha$- & $\gamma$-tocopherol) was measured in the serum and induced sputum of 30 healthy controls and 34 stable COPD patients using high performance liquid chromatography (HPLC). The inhibition of lipid peroxidation as an index of antioxidant capacity was measured in the serum by a TBA assay. Results : The serum concentration of ascorbic acid, $\alpha$-tocopherol, and retinol were significantly lower in the patients with COPD than in healthy controls ($484.8{\pm}473.3$ vs $1497.8{\pm}819.2\;pmol/L$, $48.38{\pm}17.34$ vs $73.96{\pm}26.29\;pmol/L$, p<0.001, and $9.51{\pm}8.33$ vs $15.01{\pm}5.88\;pmol/L$, p<0.05, respectively, mean$\pm$SD). However, there were little differences in the ascorbic acid and uric acid concentrations in the induced sputum between the COPD patients and the controls. The induced sputum to serum ratio of ascorbic acid was significantly higher in COPD patients compared with healthy control (0.375 vs 0.085, p<0.05). In the normal controls, the serum ascorbic acid concentration was lower in smokers than in nonsmokers ($1073{\pm}536$ vs $1757{\pm}845\;pmol/L$, p<0.05), but the level was still higher than that of the COPD patients (p<0.05). The serum retinol levels were correlated with $FEV_1$ in COPD patients (r=0.58, p<0.05). The products of lipid peroxidation were increased in normal smokers and COPD compared with norma1 nonsmokers ($115.56{\pm}19.93$ vs $120.02{\pm}24.56$ vs $91.87{\pm}20.71\;{\mu}mol/{\mu}mol$ Pi of liposome, p<0.05). Conclusion : Cigarette smoking may induce the dep1etion of serum antioxidants and this depletion of antioxidants is suggested to play a role in the pathogenesis of COPD.

The Process Efficiency Evaluation of the Food Supernatant Using A/G (Acid/Gas) Phased Anaerobic Digestion (산/가스 분리 혐기소화공정을 이용한 음식물 탈리액의 처리효율 평가)

  • Several acidogenesis batch tests, and BMP (Biochemical Methane Potential) with food waste leachate was tested at various organic loading rates (OLRs) on the mesophilic ($35^{\circ}C$) and thermophilic ($55^{\circ}C$) conditions. In acidogenesis batch test, VS removal efficiencies were 27.3% and 30.6% at $35^{\circ}C$ and $55^{\circ}C$, respectively. Removal efficiency of VS at $55^{\circ}C$ was higher than that at $35^{\circ}C$. With decrease in VS, SCOD increased as reaction time increased. Solubilization efficiency of VS were 27.4% and 33.4% at each reaction temperature within 4 days acid fermentation. Methane yield were 461 and 413 $mLCH_4/gVS$ at mesophilic and thermophilic BMP test, respectively. SCOD solubilizations in the themophilic acid fermenter showed 8~17% higher than those in the mesophilic fermenter. COD removal efficiency showed higher in the mesophilic acid fermenter at low organic loading rate. While at high organic loading rate, it was higher in the thermophilic acid fermenter. VS removal efficiency was higher at the mesophilic temperature, however, it decreased at OLR higher than 6 kg $COD/m^3{\cdot}day$. On the contrary, VS removal efficiency did not decrease but maintain at thermophilic temperature. The amount of methane gas generated from mesophilic methanogenesis digester was 12.6, 21.6, 27.4 L/day at OLR of 4, 5, 6 $COD/m^3{\cdot}day$, respectively. The amount of methane gas generated from themophilic methanogenesis digester was 14.3, 20.6, 25.2 L/day at each OLR, respectively, which is about 15~20 L/day lower than those generated at mesophilic digester.

Determination of VOC in aqueous samples by the combination of headspace (HS) and solid-phase microextraction (SPME) (HS-SPME 방식에 기초한 물 중 VOC 성분의 분석기법에 대한 연구: 3가지 실험 조건의 변화와 분석감도의 관계)

  • The application of solid phase microextraction (SPME) is generally conducted by directly immersing the fiber into the liquid sample or by exposing the fiber in the head space (HS). The extraction temperature, the time of incubation, and application of stirring are often designated to be the most important parameters for achieving the best extraction efficiencies of HS-SPME analysis. In this study, relative importance of these three analytical parameters involved in the HS-SPME method is evaluated using a polydimethylsiloxane/carboxen (PDMS/CAR) fiber. To optimize its operation conditions the competing relationships between different parameters were investigated by comparing the extraction efficiency based on the combination of three parameters and two contracting conditions: (1) heating the sample at 30 vs. 50 C, (2) exposing samples at two durations of 10 vs. 30 min, and (3) application of stirring vs. no stirring. According to our analysis among 8 combination types of HS-SPME method, an extraction condition termed as S50-30 condition ((1) 1200 rpm stirring, (2) $50^{\circ}C$ exposure temp, and (3) 30 min exposure duration) showed maximum recovery rate of 45.5~68.5% relative to an arbitrary reference of direct GC injection. According to this study, the employment of stirring is the most crucial factor to improve extraction efficiency in the application of HS-SPME.

Effect of Thermal Stress on Sexual Behaviour of Superovulated Bharat Merino Ewes

  • The present study was undertaken to study the effect of thermal stress on sexual behaviour of superovulated ewes. Fourteen adult Bharat Merino ewes with an average body weight of 29.4${\pm}$2.34 kg were randomly allocated into two groups of 7 each. All the animals were grazed on natural pasture in the morning and evening hours and housed in shed during night. Animals of Group-1 were housed in shed from 10:00 to 16:00 h while the animals of Group-2 were exposed to thermal stress in a hot chamber ($40^{\circ}C$ /6 h/day). All the animals were offered drinking water once a day at 16:30 h. Meteorological observations i.e. dry bulb, wet bulb, minimum and maximum temperature were recorded daily inside the shed as well as in hot chamber throughout the experimental period. For superovulation of animals, standard protocol developed at the Institute, using FSH (Ovagen 5.4 mg in eight injections) and PMSG (200 IU) was followed. Various sexual behaviour parameters (circling, tail fanning, head turning, standing and approaching to ram) and estrus incidence (onset of estrus and estrus duration) were observed in both the groups. The different estrus symptoms were graded subjectively on arbitrary scale of 0-5 where 0 representing no sexual behaviour (0%) and 5 representing maximum intensity in sexual behaviour (100%). Estrus was detected with the help of a marked aproned ram of proven vigor at six hourly intervals. The average percent values for sexual behaviour parameters recorded in Group-1 and Group-2 animals were 53.7${\pm}$3.76 vs. 41.1${\pm}$2.18 for circling, 71.8${\pm}$5.42 vs. 49.0${\pm}$4.41 for tail fanning, 64.7${\pm}$3.30 vs. 44.5${\pm}$4.34 for head turning, 90.1${\pm}$3.16 vs. 75.8${\pm}$4.02 for standing and 63.8${\pm}$4.8 vs. 41.9${\pm}$4.58 for approaching to ram. Animals exposed to thermal stress had significantly lower values of these sexual behaviour parameters. The animals kept in shed exhibited estrus earlier (25.4${\pm}$1.12 h) and duration was higher (37.7${\pm}$1.59 h) as compared to animals exposed to thermal stress i.e. 30.6${\pm}$1.16 h and 31.7${\pm}$3.57 h, respectively. The findings of the study indicate that thermal stress reduces the intensity of sexual behaviour in ewes and may result in failure of the animal to mate and conceive.

Preparation and Nutritional Evaluation of Hatchery Waste Meal for Broilers

  • Hatchery waste including infertile eggs, dead embryos in shell, dead or low grade chicks was cooked at $100^{\circ}C$ for 15 minutes and then oven dried and ground. Hatchery waste meal (HWM) thus prepared contained 44.25% protein, 4,573 kcal/kg gross energy, 3,600 kcal/kg metabolizable energy, 30% ether extract, 1.9% fibre, 14% ash, 9.8% nitrogen free extract, 7.26% Ca, 0.84%P, 1.86% lysine and 0.66% methionine with no Salmonella and E. coli. In biological evaluation trial, significantly higher weight gain was observed in ration containing 12% HWM compared with that containing similar amount of fish meal. Protein efficiency ratio on the two rations was 3.96 vs 2.85; protein digestibility, 86.02 vs 71.9; net protein utilization, 64.9 vs 42.37 and biological value, 75.37 vs 58.84, respectively, indicating better balance of amino acids in HWM compared with fish meal. Growth performance trial on broiler chicks also revealed better weight gain and feed efficiency on ration containing 12% HWM than that containing similar level of fish meal.

Mixed double-embryo transfer: A promising approach for patients with repeated implantation failure

  • Objective: The purpose of this study was to evaluate the efficacy of frozen mixed double-embryo transfer (MDET; the simultaneous transfer of day 3 and day 5 embryos) in comparison with frozen blastocyst double-embryo transfer (BDET; transfer of two day 5 blastocysts) in patients with repeated implantation failure (RIF). Methods: A total of 104 women with RIF who underwent frozen MDET (n = 48) or BDET (n = 56) with excellent-quality embryos were included in this retrospective analysis. All frozen embryo transfers were performed in natural cycles. The main outcome measures were the implantation rate, clinical pregnancy rate, multiple pregnancy rate, and miscarriage rate. These measures were compared between the patients who underwent MDET or BDET using the chi-square test or the Fisher exact test, as appropriate. Results: The implantation and clinical pregnancy rates were significantly higher in patients who underwent MDET than in those who underwent BDET (60.4% vs. 39.3%, p=0.03 and 52.1% vs. 30.4%, p=0.05, respectively). A significantly lower miscarriage rate was observed in the MDET group (6.9% vs. 10.7%, p=0.05). In addition, the multiple pregnancy rate was slightly, but not significantly, higher in the MDET group (27.1% vs. 25.0%). Conclusion: MDET was found to be significantly superior to double blastocyst transfer. It could be regarded as an appropriate approach to improve in vitro fertilization success rates in RIF patients.

Effect of Sleep Disturbance on Fatigue, Sleepiness, and Near-Miss among Nurses in Intensive Care Units (중환자실 간호사의 수면장애가 피로, 졸음과 근접오류에 미치는 영향)

  • Purpose : This study aims to investigate the differences in fatigue, sleepiness, and near-miss according to sleep disturbance among shift nurses in intensive care units (ICUs). Methods : A cross-sectional study in a tertiary hospital was performed. A total of 122 shift nurses working in the six ICUs were recruited. They completed self-reported questionnaires about sleep disturbance, fatigue, sleepiness, and near-miss in the past two weeks. Results : The prevalence of reported sleep disturbance was 30.3% (37 out of 122 subjects). Compared to the non-sleep disturbance group, the sleep disturbance group reported significantly more sleepiness (11.46 vs. 8.86) and higher fatigue (82.62 vs. 69.39). The sleep disturbance group showed higher rates of near-miss (78.4 vs. 57.6%) and a higher frequency of them (4.49 vs. 2.11/2weeks) compared to the non-sleep disturbance group. Medication error was the most common type of near-miss. Conclusions : This study suggests that sleep disturbances could increase fatigue, sleepiness, and near-miss among ICU shift nurses. Personal and organizational programs should be developed to support the sleep of ICU nurses.
