Expression and regulation of avian beta-defensin 8 protein in immune tissues and cell lines of chickens

  • Objective: Defensins are a large family of antimicrobial peptides and components of the innate immune system that invoke an immediate immune response against harmful pathogens. Defensins are classified into alpha-, beta-, and theta-defensins. Avian species only possess beta-defensins (AvBDs), and approximately 14 AvBDs (AvBD1-AvBD14) have been identified in chickens to date. Although substantial information is available on the conservation and phylogenetics, limited information is available on the expression and regulation of AvBD8 in chicken immune tissues and cells. Methods: We examined AvBD8 protein expression in immune tissues of White Leghorn chickens (WL) by immunohistochemistry and quantitative reverse transcription-polymerase chain reaction (RT-qPCR). In addition, we examined AvBD8 expression in chicken T-, B-, macrophage-, and fibroblast-cell lines and its regulation in these cells after lipopolysaccharide (LPS) treatment by immunocytochemistry and RT-qPCR. Results: Our results showed that chicken AvBD8 protein was strongly expressed in the WL intestine and in macrophages. AvBD8 gene expression was highly upregulated in macrophages treated with different LPS concentrations compared with that in T- and B-cell lines in a time-independent manner. Moreover, chicken AvBD8 strongly interacted with other AvBDs and with other antimicrobial peptides as determined by bioinformatics. Conclusion: Our study provides the expression and regulation of chicken AvBD8 protein in immune tissues and cells, which play crucial role in the innate immunity.

Experimental analysis of vortical structures in a turbulent layer using a dynamic PIV technique (Dynamic PIV를 이용한 난류경계층 내부 와구조 거동의 실험적 분석)

  • The vortical structures in a turbulent boundary layer (TBL) developed over a flat plate have been investigated experimentally. The flow conditions tested in this study were Re$_{\theta}$ = 3700, Re$_{\delta}$ = 11${\times}$105 and the shape factor H = 1.3. Instantaneous velocity fields in the streamwise-wall-normal planes were measured by using a dynamic PIV system. A trip-wire and sandpapers were placed behind the leading edge to promote the turbulent transition. 1000 velocity fields were obtained consecutively with a time interval of 1 millisecond. Streamwise u-velocity components were temporally averaged in the measuring plane. In addition, 2000 velocity fields were obtained randomly and ensemble-averaged to get the fully-developed turbulent characteristics. Profiles of the normalized u-component, turbulent intensities and Reynolds shear stress were evaluated. The structures of spanwise vortices were extracted from the instantaneous velocity fields by determining the swirling strength, ${\lambda}_{ci}$. The wall-normalized locations of vortices were temporally averaged in the measuring plane with respect to their rotational direction. The correlations between the temporally averaged u and the temporally averaged $y^+$ of vortices were evaluated. For the case of positive vortices, the correlation is not significant. However, the negative vortices show a strong negative correlation. The y-location of negative vortices tends to increase, as the averaged u decreases and vice versa. These findings indicate that the number of negative vortices in the outer layer increases during the outward bursting events.

  • The purpose of this research, through kinetic analyses and pilot plant experimentation of biofilm filtration reactor, is to study the theoretical equation of head loss in the Biofilm Reactor. The Head loss in the biofilm reactor has occurred due to the biofilm growth and the adhesion to the media surface and stagnation of upflow air bubble, which have caused the pore spaces to become smaller. Performance Comparison of Pressure Sensitive Paint and Pressure Field Measurement of Oblique Impinging Jet (Pressure Sensitive Paint의 성능비교 및 경사충돌분류의 압력장 측정)

  • The pressure sensitive paint (PSP) has recently received a considerable attention in the fields of aerodynamics and fluid mechanics as a new revolutionary optical technique to measure pressure fields on a body surface. In this study, the feasibility and effectiveness of the PSP pressure field measurement technique have been investigated experimentally. Seven different PSP formulations including two porphyrins(PtOEP and PtTFPP) and four polymers(Polystyrene, cellulous acetate butyrate, GP-197 and Silicon-708) were tested to check the performance and characteristics of each combination. The static calibration of each PSP formulation was carried out in a constant-pressure chamber. The PSP technique was applied to an oblique impinging jet flow to measure variation of pressure field on the impinging plate at on oblique jet angle of ${\theta}=60^{\circ}$. Pressure field images were captured by an 12bit intensified CCD(ICCD, $1K{\times}1K$)camera. As a result, the dynamic response of PSP depends on the oxygen permeability of polymer and the photochemical interaction between luminophore and polymer as well as the reaction of luminophore itself. The reaction of luminophore was changed by employing different polymers. In conclusion, Among 7 PSP formulation tested, the combination of PtTFPP and cellulous acetate butyrate show the best performance. In addition, the detail pressure field of an oblique high-speed impinging jet was measured effectively using the PSP technique.

An Experimental Study on the Optimum Design of Sirocco Fan by Using Taguchi Method (다구찌 방법을 이용한 시로코 홴의 최적설계에 관한 실험적 연구)

  • This paper is studied to find the optimum condition of double-inlet Sirocco fan installed in an indoor PAC for low noise operation by the Taguchi method. The goal of this study is to obtain the best combination of each control factor which results in a desired flowrate of Sirocco fan with minimum variability. In this study, the parameter design of the Taguchi method is adopted for robust design by the dynamic characteristic analysis using orthogonal arrays and S/N ratios. The flowrate measurements are conducted by using a multiple-nozzle-type fan tester according to the orthogonal array L9($3^4$). The results of this study can be summarized as follows ; (i) The optimum condition of control factor is a set of where A is an inner to outer diameter ratio($D_1/D_2$), B is a width to outer diameter ratio($L/D_2$), C is a blade attachment angle(${\theta}$) and D is a number of blade(Z), (ii) The flowrate under the optimum condition satisfies the equation $y=0.0384{\cdot}M$ where M is a signal factor, namely number of revolution. The flowrate performance improves about 7.3% more largely as compared with the current condition, which results in about 35RPM reduction of number of revolution for the target flowrate $18.5m^3/min$, and (iii) The sensitivity analysis shows that the major factors in contribution to flowrate performance are A, B, and D ; the percentage contributions of each control factor are 44.01%(Z), 26.77%($D_1/D_2$) and 20.42%($L/D_2$).

Distribution of Glutathione S-Transferase Omega Gene Polymorphism with Different Stages of HBV Infection Including Hepatocellular Carcinoma in the Egyptian Population

  • Background: Infection with hepatitis B virus (HBV) is a major global public health problem, with a wide spectrum of clinical manifestations. Human cytosolic glutathione-S-transferases (GSTs) include several classes such as alpha (A), mu (M), pi (P), sigma (S), zeta (Z), omega (O) and theta (T). The present study aimed to investigate the role of GST omega genes (GSTO1 and GSTO2) in different groups of patients infected with HBV. Materials and Methods: HBV groups were classified according to clinical history, serological tests and histological analysis into normal carriers (N), acute (A), chronic (CH), cirrhosis (CI) and hepatocellular carcinoma (HCC) cases. The study focused on determination of the genotypes of GST omega genes (GSTO1 and GSTO2) and GST activity and liver function tests. Results: The results showed that GSTO1 (A/A) was decreased in N, A, CH, CI and HCC groups compared to the C-group, while, GSTO1 (C/A) and GSTO1(C/C) genotypes were increased significantly in N, A, CH, CI and HCC groups. GSTO2 (A/A) was decreased in all studied groups as compared to the C-group but GSTO2(A/G) and GSTO2(G/G) genotypes were increased significantly. In addition, GST activities, albumin and TP levels were decreased in all studied groups compared to the C-group, while the activities of transaminases were increased to differing degrees. Conclusions: The results indicate that GSTO genetic polymorphisms may be considered as biomarkers for determining and predicting the progression of HBV infection.

The Study on Cu2ZnSnSe4 Thin Films without Annealed Grown by Pulsed Laser Deposition for Solar Cells

  • The $Cu_2ZnSnSe_4$ (CZTSe) thin films solar cell is one of the next generation candidates for photovoltaic materials as the absorber of thin film solar cells because it has optimal bandgap (Eg=1.0eV) and high absorption coefficient of $10^4cm^{-1}$ in the visible length region. More importantly, CZTSe consists of abundant and non-toxic elements, so researches on CZTSe thin film solar cells have been increasing significantly in recent years. CZTSe thin film has very similar structure and properties with the CIGS thin film by substituting In with Zn and Ga with Sn. In this study, As-deposited CZTSe thin films have been deposited onto soda lime glass (SLG) substrates at different deposition condition using Pulsed Laser Deposition (PLD) technique without post-annealing process. The effects of deposition conditions (deposition time, deposition temperature) onto the structural, compositional and optical properties of CZTSe thin films have been investigated, without experiencing selenization process. The XRD pattern shows that quaternary CZTSe films with a stannite single phase. The existence of (112), (204), (312), (008), (316) peaks indicates all films grew and crystallized as a stannite-type structure, which is in a good agreement with the diffraction pattern of CZTSe single crystal. All the films were observed to be polycrystalline in nature with a high (112) predominant orientation at $2{\theta}{\sim}26.8^{\circ}$. The carrier concentration, mobility, resistivity and optical band gap of CZTSe thin films depending on the deposition conditions. Average energy band gap of the CZTSe thin films is about 1.3 eV.

  • 자연하천의 안정은 하상경사, 유사이동, 원지반의 상태, 저수로의 깊이, 하도 및 하안의 구조와 하안식생 등에 달려있다. 이 중 하상경사는 가장 중요한 요소로써 하도의 안정을 위해서는 무엇보다도 이를 적절하게 유지하는 방안을 모색하여야 한다(김혜주 등, 2003). 이러한 자연하천의 안정을 위하여 주로 하상유지시설을 설치하는 것이 보편적이다. 이러한 낙차공의 설치는 하천설계기준을 근거로 하상의 안정성 확보라는 설치 목적에 맞게 설계 및 시공이 이루어져 하도의 안정에는 나름대로 효과적이지만 하천환경의 연속성 차단 등 다양한 생태·환경적 폐해가 나타나고 있는 것으로 알려져 있다(김혜주 등, 2003). 이러한 단점을 보완하기 위해 낙차공과 같은 횡단구조물의 본체에 어도를 설치하거나 계단형 또는 경사형 본체를 갖는 구조물이 제안되고 있으나 현재까지 제안된 보완적인 방법들로는 근본적인 생태단절을 해소하기에는 무리가 있다. 따라서 낙차공 등 하천 수공구조물 설치로 인해 야기되는 생태 환경적 문제를 해결하기 위해서는 대상하천에 서식하는 생물에 대한 이해가 우선 이루어져야 하며 이를 바탕으로 설계 및 시공하는 것이 가장 효과적이다. 이를 위해서는 구조물 설치에 따라 변화되는 수리현상에 대한 정확한 분석이 필요하다. 이에 본 연구에서는 국내외에서 제안되고 있는 낙차공 중 통나무를 이용한 낙차공의 평면형태에 따른 하류부 세굴현상을 수리실험을 통해 분석하였다. 본 연구에서는 평면형태(horizontal)에 따른 세굴영향을 파악하고자 평면각을 $70^{\circ}$에서 최대 $180^{\circ}$로 변화시켜 세굴 실험을 수행하였다. 상향 V형만을 고려한 이유는 하향 V형의 경우($\theta$>$180^{\circ}$)에는 낙차공을 월류하는 흐름으로 인해 구조물 하류부 좌 우안측에 세굴이 발생하기 쉽기 때문에 자연하천의 경우 호안측 침식이 발생한 수 있고 또한 실험수로 벽면영향으로 인해 세굴형태가 왜곡될 우려가 있어 하향 형태의 낙차공은 실험대상에서 제외하였다. 낙차공의 평면 형태가 세굴심에 미치는 영향을 검토한 결과, 유입 Froude 수( )에 대해 높은 상관이 있으며 또한 평면각도에 따라 변하는 낙차공의 횡단길이가 낙차공 하류부의 세굴심에 큰 영향을 미치고 있음을 확인할 수 있었고, 세굴길이의 경우, 평면형태에 따른 세굴길이와 세굴심의 비는 낙차공의 평면각도의 증가에 따라 세굴 길이가 짧아짐을 확인할 수 있었다.

  • 자연하천의 유량값은 일반적으로 횡단면의 면적과 이를 통과하는 유속의 곱으로 산정된다. 유량측정 방법은 하천의 형태와 저 평수기 및 홍수기의 수위에 따라 도섭법, 교량법, 부자법 및 보트를 이용한 ADVM 측정법 등 다양한 방법으로 실시된다. 그러나 현장 여건에 따라 흐름에 직각이 아닌 사교에서 측정이 이루어지는 경우에는 단면적의 오차를 포함할 가능성이 크기 때문에 횡단면의 측선 각도에 따라 각보정을 실시해야 한다. 현재 사교에서 유량 측정을 실시하는 경우, 흐름의 직각을 기준으로 처짐각을 측량하여 각 측선에 $cos{\theta}$를 적용하여 단면적을 보정하고 있는데, 이 처짐의 정도가 유량의 참값에서 어느 정도의 영향을 미치는지에 대한 검토가 요구된다. 본 연구에서는 한강수계 왕숙천에 위치한 퇴계원 지점에서 실시간 수위에 따른 유속을 측정하였으며, 횡단면에 직각인 측선을 기준값으로 제시하고, 처짐각의 정도를 $10^{\circ}$, $20^{\circ}$, $30^{\circ}$, $40^{\circ}$, $50^{\circ}$까지 늘려 산정된 유량값을 기준값과 비교 분석하였다. 본 연구에 쓰인 측정기기는 Price AA 유속계이고, 측정방법으로는 유량의 흐름 방향을 기준하여 직각으로 수면에서 0.6d 지점의 유량측정방법(1점법)을 적용하였다. 그 결과 유량의 흐름 방향을 기준하여 직각인 경우 $1.39m^3/s$의 유량에서 보정 전 각 $10^{\circ}$의 유량 $1.36m^3/s$, $30^{\circ}$의 유량 $1.49m^3/s$, $50^{\circ}$의 유량 $2.25m^3/s$로 각이 클수록 단면적이 크게 나타나며 유량 역시도 과대 산정됨을 알 수 있었다. 따라서 도섭법을 이용한 유량측정이나 사교에서의 교량법 등을 적용하여 유량측정을 실시할 경우 유량의 흐름방향을 기준으로 직각으로 유량측정을 실시하여 유량을 산정하되 부득이한 경우로 사교에서의 측정이 이루어 졌을 시 흐름 방향을 기준으로 각도를 측정하여 크게 나타나는 단면적에 처짐각을 보정하여 유량을 산정함이 오차를 줄일 수 있으며, 신뢰성 있는 유량자료 생산의 방법이 라 할 수 있겠다.
