심근허혈검출을 위한 심박변이도의 시간과 주파수 영역에서의 특징 비교 (Comparison of HRV Time and Frequency Domain Features for Myocardial Ischemia Detection)

  • 심박 변이도 (HRV) 분석은 심근허혈 (MI)를 평가하기 위한 편리한 도구이다. HRV에 대한 분석법은 시간 영역과 주파수 영역 분석으로 나눠질 수 있다. 본 논문은 단기간의 HRV 분석에 있어서 웨이블릿 변환을 주파수 영역 분석과 시간 영역 분석 비교하기 위하여 사용하였다. ST-T와 정상 에피소드는 각각 European ST-T 데이터베이스와 MIT-BIH Normal Sinus Rhythm 데이터베이스에서 각각 수집되었다. 한 에피소드는 32개 연속하는 RR 간격으로 나눠질 수 있다. 18개 HRV 특징은 시간과 주파수 영역 분석을 통하여 추출된다. 가종 퍼지소속함수 신경망 (NEWFM)은 추출된 18개의 특징을 이용하여 심근허혈을 진단하였다. 결과는 보여주는 평균 정확도로부터 시간영역과 주파수영역의 특징은 각각 75.29%와 80.93%이다.

Non-canonical targets play an important role in microRNA stability control mechanisms

  • MicroRNAs (miRNAs) regulate gene expression by guiding the Argonaute (Ago)-containing RNA-induced silencing complex (RISC) to specific target mRNA molecules. It is well established that miRNAs are stabilized by Ago proteins, but the molecular features that trigger miRNA destabilization from Ago proteins remain largely unknown. To explore the molecular mechanisms of how targets affect the stability of miRNAs in human Ago (hAgo) proteins, we employed an in vitro system that consisted of a minimal hAgo2-RISC in HEK293T cell lysates. Surprisingly, we found that miRNAs are drastically destabilized by binding to seedless, non-canonical targets. We showed that miRNAs are destabilized at their 3' ends during this process, which is largely attributed to the conformational flexibility of the L1-PAZ domain. Based on these results, we propose that non-canonical targets may play an important regulatory role in controlling the stability of miRNAs, instead of being regulated by miRNAs.

IL-6-miR-210 Suppresses Regulatory T Cell Function and Promotes Atrial Fibrosis by Targeting Foxp3

  • The aim of this study was to explore the role of IL-6-miR-210 in the regulation of Tregs function and atrial fibrosis in atrial fibrillation (AF). The levels of interleukin (IL)-6 and IL-10 in AF patients were detected by using ELISA. Proportions of Treg cells were detected by fluorescence activated cell sorting analysis in AF patients. The expression of Foxp3, α-SMA, collagen I and collagen III were determined by western blot. The atrial mechanocytes were authenticated by vimentin immunostaining. The expression of miR-210 was performed by quantitative real-time polymerase chain reaction (qRT-PCR). TargetScan was used to predict potential targets of miR-210. The cardiomyocyte transverse sections in AF model group were observed by H&E staining. The myocardial filaments were observed by masson staining. The level of IL-6 was highly increased while the level of IL-10 (Tregs) was significantly decreased in AF patients as compared to normal control subjects, and IL-6 suppressed Tregs function and promoted the expression of α-SMA, collagen I and collagen III. Furthermore, miR-210 regulated Tregs function by targeting Foxp3, and IL-6 promoted expression of miR-210 via regulating hypoxia inducible factor-1α (HIF-1α). IL-6-miR-210 suppresses regulatory T cell function and promotes atrial fibrosis by targeting Foxp3.

Molecular insights into the role of genetic determinants of congenital hypothyroidism

  • In our previous studies, we have demonstrated the association of certain variants of the thyroid-stimulating hormone receptor (TSHR), thyroid peroxidase (TPO), and thyroglobulin (TG) genes with congenital hypothyroidism. Herein, we explored the mechanistic basis for this association using different in silico tools. The mRNA 3'-untranslated region (3'-UTR) plays key roles in gene expression at the post-transcriptional level. In TSHR variants (rs2268477, rs7144481, and rs17630128), the binding affinity of microRNAs (miRs) (hsa-miR-154-5p, hsa-miR-376a-2-5p, hsa-miR-3935, hsa-miR-4280, and hsa-miR-6858-3p) to the 3'-UTR is disrupted, affecting post-transcriptional gene regulation. TPO and TG are the two key proteins necessary for the biosynthesis of thyroid hormones in the presence of iodide and H2O2. Reduced stability of these proteins leads to aberrant biosynthesis of thyroid hormones. Compared to the wild-type TPO protein, the p.S398T variant was found to exhibit less stability and significant rearrangements of intra-atomic bonds affecting the stoichiometry and substrate binding (binding energies, ΔG of wild-type vs. mutant: -15 vs. -13.8 kcal/mol; and dissociation constant, Kd of wild-type vs. mutant: 7.2E-12 vs. 7.0E-11 M). The missense mutations p.G653D and p.R1999W on the TG protein showed altered ΔG(0.24 kcal/mol and 0.79 kcal/mol, respectively). In conclusion, an in silico analysis of TSHR genetic variants in the 3'-UTR showed that they alter the binding affinities of different miRs. The TPO protein structure and mutant protein complex (p.S398T) are less stable, with potentially deleterious effects. A structural and energy analysis showed that TG mutations (p.G653D and p.R1999W) reduce the stability of the TG protein and affect its structure-functional relationship.

한국한의학연구원 논문의 통계적 오류에 관한 연구 (An Assessment of Statistical Validity of Articles Published in "Korean Journal of Oriental Medicine"-from 1995 to 2007)

  • Background and Purpose: The purpose of this study was investigate statistical validities of previously reported articles that used various statistical techniques such as t-test and analysis of variance. Methods: To analyze the statistical procedures, 66 original articles using those statistical methods were selected from "Korean Journal of Oriental Medicine(KJOM)" published from 1995 to 2007. Results: Twenty-one articles(32%) did not report correct p-values, 33 articles(50%) used mean${\pm}$standard error(mean${\pm}$SE) and 11 articles(l7%) used mean${\pm}$standard deviation(mean${\pm}$SD). Fifty-two articles(95%) of 55 ones which were tested for normal distribution made an error in describing normal distribution. Seventeen articles misused t-test and 12 articles did not carry out the multiple comparison. Conclusions: The training of researchers with clinical statistics or the participation of statisticians in research design will reduce the significant errors in statistical interpretation of the results.

Engineered T Cell Receptor for Cancer Immunotherapy

  • Among the therapeutic strategies in cancer immunotherapy-such as immune-modulating antibodies, cancer vaccines, or adoptive T cell transfer-T cells have been an attractive target due to their cytotoxicity toward tumor cells and the tumor antigen-specific binding of their receptors. Leveraging the unique properties of T cells, chimeric antigen receptor-T cells and T cell receptor (TCR)-T cells were developed through genetic modification of their receptors, enhancing the specificity and effectiveness of T cell therapy. Adoptive cell transfer of chimeric antigen receptor-T cells has been successful for the treatment of hematological malignancies. To expand T cell therapy to solid tumors, T cells are modified to express defined TCR targeting tumor associated antigen, which is called TCR-T therapy. This review discusses anti-tumor T cell therapies, with a focus on engineered TCR-T cell therapy. We outline the characteristics of TCR-T cell therapy and its clinical application to non-hematological malignancies.

항암화학요법에 관한 가족참여 개별교육이 암환자의 자가간호 지식, 자가간호 수행, 가족지지 및 불안에 미치는 효과 (Effects of Individual Education for Cancer Patients and Their Family about Chemotherapy on the Self­-Care Knowledge, Self­-Care Performance, Family Support, and Anxiety)

  • Purpose: The study was to examine the effects of the individual education about chemotherapy for cancer patients as well as their family who are receiving the first chemotherapy on self-care knowledge, self-care performance, family support, and anxiety level. Methods: A non-equivalent control group was used in this interventional study. A total of 50 patients with cancer aged above 19 in the K university hospital, Seoul, Korea, participated in the study (Experimental group: n=25, Control group: n=25). The education about chemotherapy was offered one week after hospital discharge, which was followed by telephone counseling Data were analyzed by the SPSS/WIN 20.0 program using percentage, frequency, mean, standard deviation, $x^2$ test, and independent t-test. Results: The experimental group showed a significantly better self-care knowledge (t=4.094, p<.001), self-care performance (t=3.532, p=001), family support (t=2.170, p=.035), and anxiety control (t=4.780, p<.001) than the control group. Conclusion: The findings suggest that the individual education should be considered for cancer patients and their family in the clinical fields.
