Effects of Nutrition Education on Diet Control Program S-28 (다이어트프로그램 S-28에 대한 영양교육의 효과)

  • This study was carried out to investigate effects of nutrition education on diet program S-28 for a per-iod of 5 months. The participants in this diet program were 40 Chungkang college female students with over 30% body fat. During the first 2 weeks they were provided 800kcal a day and 1,200kcal a day for the lat 2 weeks. Since then weight maintenance program was continued. Exercise and nutrition e여-cation program for nutrition knowledge attitude and food habit were conducted during the entire test period, Results during the test period were as follows : 1) Weight loww was statistically significant from 69.5$\pm$4.9kg at the beginning to65.6$\pm$3.0kg(P<0.01) in 3 months to 63.2$\pm$3.5kg(P<0.01) in the 5th month. 2) Obesity rate was significantly decreased from 23.9$\pm$12.1% to 21.3$\pm$9.2%(P<0.05) in 3 mon-ths to 19.7$\pm$8.5% in the 5th month. 3) BMI was significantly declined from 27.3$\pm$1.5kg/m2 to 26.1$\pm$1. 2kg/m2(P<0.01) in 3 months to 25.7$\pm$0.9kg/m2(P<0.05) in 3 months to 28.2$\pm$1.8%(p<0.05) in the 5th month. 5) The mean scores of food habit were 33.4$\pm$4.5 at the beginning 35.2$\pm$3.7 in 3 mon-손 37.4$\pm$4.0 in the 5th month and each of nutrition attitude was 53.8$\pm$5.3, 58.0$\pm$6.2. There-fore these data showed that effects of nutrition education on diet program S-28 were feflected remark-ably not only in the antropmetric measurements but also in the mean scores of food habit and nu-trition attitude.

Marginal and internal discrepancy of 3-unit fixed dental prostheses fabricated by subtractive and additive manufacturing (절삭 및 적층 가공법으로 제작된 3본 고정성 국소의치의 변연 및 내면 적합도에 관한 연구)

  • Purpose: This study was to evaluate marginal and internal discrepancy of 3-unit fixed dental prostheses (FDP) fabricated by subtractive manufacturing and additive manufacturing. Materials and methods: 3-unit bridge abutments without the maxillary left second premolar were prepared (reference model) and the reference model scan data was obtained using an intraoral scanner. 3-unit fixed dental prostheses were fabricated in the following three ways: Milled 3-unit FDP (MIL), digital light processing (DLP) 3D printed 3-unit FDP (D3P), stereolithography apparatus (SLA) 3D printed 3-unit FDP (S3P). To evaluate the marginal/internal discrepancy and precision of the prosthesis, scan data were superimposed by the triple-scan protocol and the combinations calculator, respectively. Quantitative and qualitative analysis was performed using root mean square (RMS) value and color difference map in 3D analysis program (Geomagic control X). Statistical analysis was performed using the Kruskal-Wallis test (α=.05), MannWhitney U test and Bonferroni correction (α=.05/3=.017). Results: The marginal discrepancy of S3P group was superior to MIL and D3P groups, and MIL and D3P groups were similar. The D3P and S3P groups showed better internal discrepancy than the MIL group, and there was no significant difference between the D3P and S3P groups. The precision was excellent in the order of MIL, S3P, and D3P groups. Conclusion: Within the limitation of this study, the 3-unit fixed dental prostheses fabricated by additive manufacturing showed better marginal and internal discrepancy than the those of fabricated by subtractive manufacturing, but the precision was poor.

Effect of Sphingosine-1-Phosphate on Intracellular Free Ca2+ in Cat Esophageal Smooth Muscle Cells

  • A comprehensive collection of proteins senses local changes in intracellular $Ca^{2+}$ concentrations ($[Ca^{2+}]_i$) and transduces these signals into responses to agonists. In the present study, we examined the effect of sphingosine-1-phosphate (S1P) on modulation of intracellular $Ca^{2+}$ concentrations in cat esophageal smooth muscle cells. To measure $[Ca^{2+}]_i$ levels in cat esophageal smooth muscle cells, we used a fluorescence microscopy with the Fura-2 loading method. S1P produced a concentration-dependent increase in $[Ca^{2+}]_i$ in the cells. Pretreatment with EGTA, an extracellular $Ca^{2+}$ chelator, decreased the S1P-induced increase in $[Ca^{2+}]_i$, and an L-type $Ca^{2+}$-channel blocker, nimodipine, decreased the effect of S1P. This indicates that $Ca^{2+}$ influx may be required for muscle contraction by S1P. When stimulated with thapsigargin, an intracellular calcium chelator, or 2-Aminoethoxydiphenyl borate (2-APB), an $InsP_3$ receptor blocker, the S1P-evoked increase in $[Ca^{2+}]_i$ was significantly decreased. Treatment with pertussis toxin (PTX), an inhibitor of $G_i$-protein, suppressed the increase in $[Ca^{2+}]_i$ evoked by S1P. These results suggest that the S1P-induced increase in $[Ca^{2+}]_i$ in cat esophageal smooth muscle cells occurs upon the activation of phospholipase C and subsequent release of $Ca^{2+}$ from the $InsP_3$-sensitive $Ca^{2+}$ pool in the sarcoplasmic reticulum. These results suggest that S1P utilized extracellular $Ca^{2+}$ via the L type $Ca^{2+}$ channel, which was dependent on activation of the $S1P_4$ receptor coupled to PTX-sensitive $G_i$ protein, via phospholipase C-mediated $Ca^{2+}$ release from the $InsP_3$-sensitive $Ca^{2+}$ pool in cat esophageal smooth muscle cells.

  • The expression vector (pWHM3-TR1R2) for sprT gene encoding Streptomyces griseus trypsin (SGT) followed by two regulatory genes, sgtR1 and sgtR2, was introduced into Streptomyces lividans TK24 and Streptomyces griseus IFO 13350. Various media with different compositions were used to maximize the productivity of SGT in the recombinant trains. he SGT productivity was best when the transformant of S. lividans TK24 was cultivated in R2YE medium (0.74 unit/mL) at 5 days of cultivation. C5/L (0.66 unit/mL) medium also gave a good productivity, but Livid (0.08 unit/mL) and NDSK (0.06 unit/mL) yielded poor productivities. S. griseus IFO 13350/pWHM3-TR1R2 produced SGT by 1.518 unit/mL (C5/L), 1.284unit/mL (R2YE),0.932 unit/mL (NDSK), and 0.295 unit/mL (Livid) at 7 days of cultivation, which was much higher than those from S. lividans TK24/TR1R2. The SGT protein was purified from the culture broth of S. griseus IFO 13350/pWHM3-TR1R2 in C5/L to homogeneity via ammonium sulfate fractionation, and CM-sepharose and SP-sepharose column chromatographies. The specific activity of purified SGT was 69,252 unit/mg, and the final purification fold and recovery yield were 6.5 and 1.4%, respectively.

  • Biogeochemistry of a 34-year-old red pine plantation on a Typic Udipsamment was investigated in the central sand plains of Wisconsin Biomass and net primary production were $106Mg\;ha^{-1}$ and $11.0Mg\;ha^{-1}y^{-1}$ respectively. Concentrations of elements in the vegetation were greatest in the 2- to 3-year-old needles, followed by current needles, current twigs, bole bark, live branches, dead branches, bolewood production, and bolewood. For a given tree component (excluding the current twigs), elemental concentrations were greatest in the upper one- third and least in the lower one- third of the tree. Total elements in the vegetation are ranked: N($222kg\;ha^{-1}$), Ca(217), K (85), Mg(38), S(31), and p. The soil contained from 60 to 89% of the readily available P, K, Ca, Mg, and S in the ecosystem. Whereas precipitation was the major source of N and S, weathering supplied the bulk of the Ca, Mg, P, and K. Output of a given element by leaching was less than $7kg\;ha^{-1}y^{-1}$ except for S which was $12kg\;ha^{-1}y^{-1}$ Whereas throughfall returned the greatest amounts of N, K, and S to the forest floor, litterfall accounted for the greatest returns of Ca, Mg, and P. Because of foliar leaching, net loading of all elements studied in the throughfall exceeded that of the precipitation. Ammonium in net precipitation was reduced by a factor of three within the upper 7.5cm due to uptake by fine roots.

Effect of Different pH Processes on Branched β-1,3-Glucan Production from Submerged Culture of Ganoderma lucidum (영지 (G. lucidum)의 액체배양에 의한 β-1,3-Glucan 생산에 미치는 서로 다른 pH Process의 영향)

  • A submerged cultivation of Ganoderma lucidum was carried out in an air-lift fermenter system, and the effects of different pH processes on extracellular branched ${\beta}$-1,3-glucan(EPS) production and mycelial growth(MDW) were investigated. The controlled pH process improved the production of branched ${\beta}$-1,3-glucan and biomass in comparison to the uncontrolled pH process. However, the maximum production of branched ${\beta}$-1,3-glucan were obtained by the bi-staged pH process. From these results, we confirmed that the bi-staged pH process was the most effective for improving the production of branched ${\beta}$-1,3-glucan from submerged culture of G. lucidum.

Comparison between the Balance of Skilled and Less-Skilled Players during Successful and Failed Front Kick and Turning Side Kick Motions (태권도 품새 우수·비 우수선수 간 앞차고 몸돌아 옆차기의 성·패에 따른 균형성 비교)

  • The aim of this study is to identify the appropriate movement for maintaining postural balance during Front Kick and Turning Side Kick motions. To do so, ten Taekwondo athletes: five skilled players(S, body mass: $65.0{\pm}5.8kg$, height: $172.3{\pm}3.7cm$, age: $20.0{\pm}1.2yrs$, career: $9.0{\pm}1.9yrs$) and five less-skilled players(LS, body mass: $67.1{\pm}5.5kg$, height: $173.2{\pm}5.1cm$, age: $19.4{\pm}1.7yrs$, career: $9.6{\pm}1.7yrs$) participated in this study. A three-dimensional motion analysis was performed on the participants using eight infrared cameras and two force plate(sampling frequency of 200 Hz and 2000 Hz for S and LS players, respectively). The participants' motions were divided into: a front-kick phase(P1) and a turning-side-kick phase(P2). For P2(p<.05), the range and root mean square(RMS) of the ground reaction torque and the M-L mean velocity of COP were greater for LS than for S; similarly, for P2(p<.05), the M-L range, A-P range, and velocity of the COP were greater for LS than for S. Further, the M-L range and maximum velocity of the COP was greater for failure than for success(p<.05). The femoral biceps muscle for bending the knee joint was significantly stronger in S than in LS(p<.05). It is expected that these results will be useful in developing a training program for improving the balance and stability of Taekwondo poomsae athletes and improve their front-kick and turning-side-kick motions.

  • Atherosclerosis is the deposition of plaque in the main arteries. It is an inflammatory condition involving the accumulation of macrophages and various lipids (low-density lipoprotein [LDL] cholesterol, ceramide, S1P). Moreover, endothelial cells, macrophages, leukocytes, and smooth muscle cells are the major players in the atherogenic process. Sphingolipids are now emerging as important regulators in various pathophysiological processes, including the atherogenic process. Various sphingolipids exist, such as the ceramides, ceramide-1-phosphate, sphingosine, sphinganine, sphingosine-1-phosphate (S1P), sphingomyelin, and hundreds of glycosphingolipids. Among these, ceramides, glycosphingolipids, and S1P play important roles in the atherogenic processes. The atherosclerotic plaque consists of higher amounts of ceramide, glycosphingolipids, and sphingomyelin. The inhibition of the de novo ceramide biosynthesis reduces the development of atherosclerosis. S1P regulates atherogenesis via binding to the S1P receptor (S1PR). Among the five S1PRs (S1PR1-5), S1PR1 and S1PR3 mainly exert anti-atherosclerotic properties. This review mainly focuses on the effects of ceramide and S1P via the S1PR in the development of atherosclerosis. Moreover, it discusses the recent findings and potential therapeutic implications in atherosclerosis.

Overproduction and High Level Secretion of Glucose Oxidase in Saccharomyces cerevisiae (Glucose Oxidase의 Saccharomyces cerevisiae에서의 대량생산 및 고효율 분비)

  • The overproduction and high level secretion of Glucose Oxidase (GOD) from A. niger in S. cerevisiae was carried out by cloning GOD gene. For this purpose, using two different strong promoters (ADH1 promoter, GAL10 promoter) and signal sequences (${alpha}$-MF signal sequence of S. cerevisiae and ${alpha}$-amylase signal sequence of A. oryzae) and GAL7- and GOD terminator, four expression vectors were constructed. All the expression vectors were transformed in S. cerevisiae 2805 using auxotroph method. By the flask culture, transformants of pGAL expression vector series containing GAL 10 promotor showed much higher GOD productivity than transformants of pADH expression vector series containing ADH1 promoter Transformants of pGALGO2 containing GAL10 promotor and ${alpha}$-amylase signal sequence has shown the best productivity of GOD ($GOD_{total}$: 10.3 unit/mL, $GOD_{ex}$: 8.7 unit/mL) at 115 hr. This value was three fold higher than that of pGALGO1 containing GAL 10 promotor and ${alpha}$-MF signal sequence, even if the same promotor was involved. Through the ${alpha}$-amylase signal sequence of A. oryzae, GOD was secreted much more than the case of ${alpha}$-MF signal sequence from S. cerevisiae. These results suggest that signal sequence may play a important roles in not only the secretion but also the overproduction of foreign protein. Secretion rate of GOD in pGALGO1 and pGALGO2 was 89% and 84%, respectively, Because of the overglycosylation in S. cerevisiae the molecular weight of recombinant GOD in S. cerevisiae was much larger (250 kDa) than that of nature GOD in A. niger (170 kDa).

Isolation of the Alcohol-Tolerant Lactic Acid Bacteria Pediococcus acidilactici K3 and S1 and their Physiological Characterization (알코올 내성 젖산균 Pediococcus acidilactici K3와 S1의 분리 및 생리적 특성)

  • Lactic acid bacteria (LAB) are a representative group of probiotics and used in many fermented foods and beverages. Several recent studies have shown that LAB are present in makgeolli which is a traditional Korean alcoholic beverage. However, most LAB are intolerant of more than 6% (v/v) alcohol concentrations. For this reason, alcohol-tolerant LAB are isolated from kimchi, makgeolli and nuruk using alcohol containing selective media. After being cultured in MRS broth containing 13% (v/v) alcohol, the two strains which showed the highest increasing O.D values, were finally selected. As results of 16S rRNA gene sequencing and biochemical characterization using an API kit, the two species were identified as Pediococcus acidilactici K3 and S1. In addition, the identified two strains produced bacteriocins against Staphylococcus aureus. When compared with the P. acidilactici type strain, the two selected strains possessed two to three time higher growth on 12-13% (v/v) alcohol containing MRS broth. The viability of P. acidilactici K3 and S1 when inoculated in makgeolli and stored at $10^{\circ}C$ did not decrease through a period of one month indicating that the selected strains can be used for LAB containing makgeolli.
