Rhizopus japonicus가 생산하는 인삼 Saponin 전환효소의 효소학적 특성 (Enzymatic Properties of the Convertible Enzyme of Ginseng Saponin Produced from Rhizopus japonicus)

  • 인삼 saponin 중에서 조성 비율이 가장 큰 ginsenoside R $b_1$을 약효면에서 훨씬 우수한 ginsenoside-Rd로 선택적 전환할 수 있는 전환효소를 Rhizopus japonicus 배양물로부터 순수하게 정제한 후 그 효소학적 특성을 조사하였다. 본 효소는 pH 4.8~5.0, 45$^{\circ}C$에서 그 활성도가 가장 높았으며 pH4~9의 범위에서 안정하였고, pH5.0에서 6$0^{\circ}C$ 열처리할 경우 50% 실활하는데 2시간이 소요되었다. M $n^{++}$, F $e^{++}$에 의해 효소활성이 촉진되었으며 C $u^{++}$, A $g^+$에 의해서는 저해되었다. 효소 저해제중에는 EDTA, o-phenanthroline 등에 의해 저해되기도 했다. 본 효소는 당류중에는 gentiobiose, cellulose만을, glycoside 중에서는 amygdalin, purnasin, salicin 등을 분해할 수 있었으며 ginsenoside R $b_1$, 이외의 다른 ginsenoside는 전혀 분해할 수 없었다. 본 효소의 Km치는 total saponin 기질 및 ginsenoside Rb group saponin 기질은 ginsenoside R $b_1$으로서 5.0mM 이었으며, gentiobiose는 4.8mM, amygdalin은 3.7mM 이었다.

인지능 개선 효과 증진을 위한 백삼 추출물 조제 연구 (Study on White Ginseng Extract Preparation for Cognition Improvement)

  • Background: The study was conducted to elucidate the extraction conditions under which white ginseng has cognition-improving efficacy. Methods and Results: Extracts from white ginseng under different solvent and temperature conditions were analyzed for ginsenoside content and inhibitory effect on N-methyl-D-aspartate (NMDA) receptor and acetylcholinesterase. The total ginsenoside contents and amounts of ginsenoside Rb1 plus ginsenoside Rg1 from the 1st extracts (prepared with EtOH/$H_2O$ as solvent) were higher than those from the 2nd extracts (extracted with $H_2O$ after the 1st EtOH/$H_2O$ extraction). The contents in the 1st and 2nd extracts produced at $80^{\circ}C$ were also higher than those obtained at $50^{\circ}C$. Samples from the 1st extraction at $80^{\circ}C$ indicated higher inhibitory activities on NMDA receptors-whose excessive activation is thought to mediate the calcium-dependent neurotoxicity associated with several neurodegenerative diseases-than those from the 2nd extraction. Among the samples prepared at varying temperatures, the extract prepared at $50^{\circ}C$ showed the highest suppression activity on NMDA receptors. Note, however, that the extracts from the 2nd extraction at $50^{\circ}C$ inhibited acetylcholinesterase-whose inhibition could be a therapeutic strategy for neurodegenerative diseases with cognitive deficits and memory malfunction-more effectively than those from the 1st extraction. Conclusions: To enhance the cognition-improving activity of white ginseng extract, it is suggested that the extracts be utilized after being combined the 1st extracts (made with EtOH/$H_2O$ solvent) and the 2nd extracts (prepared with $H_2O$) at low temperature.

  • Ginseng(Panax ginseng C.A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng callus and hairy roots grow vigorously and may Produce the same or more biologically active compounds for human health than natural ginseng roots. Therefore, ginseng callus and hairy roots can be used for commercial purposes. Polyacetylene, one of anti-cancer compounds in ginseng, was not detected in the callus cultured on the medium containing 2, 4-B, but cells derived from the callus growth was excellent, The ginseng calli cultured on the medium containing 2mg11 CPA and 0.05mg/1 BA was grown vigorously and produced panaxydol, one of ginseng polyacetylene. The biosynthesis of polyacetylene in callus was not affected by addition of NAA and sucrose in media. The SH medium was better than the MS medium for ginseng callus growth and biosynthesis of panaxydol. Another ginseng anti-cancer compounds, ginsenoside-Rg$_3$, Rh$_1$and Rh$_2$ were detected in ginseng hairy roots by heat treatment. Those of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes Rl000 $A_4$T in dark condition after one month of culture. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark(22$^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5 ι Erlenmeyer flasks, 1ι roller drums, 10ι jar-fermenters, and especially in 20ι air-lift .culture vessels. All heat treatments had remarkably different ginsenoside contents. Eleven ginsenosides were determined in heat treatment, eight in freeze dried hairy roots. Contents of ginsenoside-Rbl , Rb2, Rc, Rd. Re, Rf, and Rg$_1$tested in all heat treatments were less than those of freeze dried hairy roots. Contents of glnsenoside-Rg$_2$ in heat treatment for 1 hour at 105$^{\circ}C$ was 4.92mg/g dry wt, 3.9 times higher than 1.27 mg/g dry wt of freeze dried hairy roots. The optimum condition of heat treatment for the production of ginsenoside-Rg$_3$and Rhl was 2 hours at 105$^{\circ}C$, and ginsenoside content was 2.58mg/g dry wt and 3.62mg/g dry wt, respectively. The production of ginsenoside-Rh2 was the highest in heat treatment for 2 hours at 105$^{\circ}C$ among treatments examined, and ginsenoside-Rh$_2$content was 1.08mg/g dry wt.

Perioperative stress prolong post-surgical pain via miR-339-5p targeting oprm1 in the amygdala

  • Background: The decreased expression of mu-opioid receptors (MOR) in the amygdala may be a key molecular in chronic post-surgical pain (CPSP). It is known that miR-339-5p expression in the amygdala of a stressed rat model was increased. Analyzed by RNAhybrid, miR-339-5p could target opioid receptor mu 1 (oprm1) which codes MOR directly. So, the authors hypothesized that miR-339-5p could regulate the expression of MOR via targeting oprm1 and cause the effects to CPSP. Methods: To simulate perioperative short-term stress, a perioperative stress prolongs incision-induced pain hypersensitivity without changing basal pain perception rat model was built. A pmiR-RB-REPORTTM dual luciferase assay was taken to verify whether miR-339-5p could act on oprm1 as a target. The serum glucocorticoid level of rats was test. Differential expressions of MOR, GFAP, and pERK1/2 in each group of the rats' amygdala were tested, and the expressions of miR-339-5p in each group of rats' amygdalas were also measured. Results: Perioperative stress prolonged the recovery time of incision pain. The expression of MOR was down-regulated in the amygdala of rats in stress + incision (S + IN) group significantly compared with other groups (P < 0.050). miR-339-5p was up-regulated in the amygdala of rats in group S + IN significantly compared with other groups (P < 0.050). miR-339-5p acts on oprm1 3'UTR and take MOR mRNA as a target. Conclusions: Perioperative stress could increase the expression of miR-339-5p, and miR-339-5p could cause the expression of MOR to decrease via targeting oprm1. This regulatory pathway maybe an important molecular mechanism of CPSP.

가공방법에 따른 몇가지 백삼 제품의 사포닌 및 유리당 조성 변화 (Composition of Saponin and Free Sugar of Some White Ginsengs with Processing Conditions)

  • 수삼으로 제조된 백삼 제품들의 가공방법의 차이에 따른 성분 변화에 대한 조사로서 원료삼인 수삼과 전통적인 방법으로 제조한 백삼, 태극삼 A, 새로운 방법으로 제조된 태극삼 B의 일반성분, 추출수율, 사포닌 함량 및 조성, 유리당 조성을 비교 분석 하였다. 4가지 시료의 조단백질은 13.34-15.33%, 조지방은 1.04-2.05%, 조회분은 3.13-5.25%, 총 식이섬유는 9.72-15.46%, 당질은 64.75-73.48% 범위의 함량이었다. 추출수율은 열수 추출이 80% 메탄올 추출보다 전체적으로 약 2배 이상 높게 나타났으며, 가공하지 않은 수삼의 열수 추출 수율이 56.4%로 가장 높았다. 사포닌 함량은 열수 추출시 수삼의 총 사포닌이 2.40%로 가장 많이 추출되었으며, 태극삼 A는 태극삼 B보다 비율로 볼 때 약 20% 이상 적은 1.45%의 총 사포닌 함량을 나타내었다. Ginsenoside 조성은 열수 추출시 $ginsenoside-Rg_1$이 수삼은 36.52%이지만 태극삼 A와 B는 68.66, 67.89%고 약 2배의 함량을 나타내었다. $Ginsenoside-Rb_1$은 태극삼 A차 B에 18.41, 17.43%만이 함유되어 있었다. 한편 태극삼 A와 태극삼 B의 총 사포닌 양에는 큰 차이가 있었지만 ginsenosides의 조성비에 있어서는 서로 큰 차이가 없는 것으로 나타났다. 80% 메탄올 추출의 경우에도 가공방법이 다르게 제조된 백삼 제품별로 ginsenoside조성에 큰 차이를 나타내었다. 수삼과 백삼에서는 열수 추출과 80% 메탄올 추출 모두에서 함량비는 다를지라도 fructose, glucose, sucrose, maltose만이 함유되어 있었지만 열 처리된 태극삼 A와 B에는 rhamnose도 각각 2.56, 4.01%씩 함유되어 있었다.

Changes in ginsenoside compositions and antioxidant activities of hydroponic-cultured ginseng roots and leaves with heating temperature

  • Background: This study evaluated changes in ginsenoside compositions and antioxidant activities in hydroponic-cultured ginseng roots (HGR) and leaves (HGL) with heating temperature. Methods: Heat treatment was performed at temperatures of $90^{\circ}C$, $110^{\circ}C$, $130^{\circ}C$, and $150^{\circ}C$ for 2 hours Results: The ginsenoside content varied significantly with heating temperature. The levels of ginsenosides Rg1 and Re in HGR decreased with increasing heating temperature. Ginsenosides F2, F4, Rk3, Rh4, Rg3 (S form), Rg3 (R form), Rk1, and Rg5, which were absent in the raw ginseng, were formed after heat treatment. The levels of ginsenosides Rg1, Re, Rf, and Rb1 in HGL decreased with increasing heating temperature. Conversely, ginsenosides Rk3, Rh4, Rg3 (R form), Rk1, and Rg5 increased with increasing heating temperature. In addition, ginsenoside contents of heated HGL were slightly higher than those of HGR. The highest extraction yield was 14.39% at $130^{\circ}C$, whereas the lowest value was 10.30% at $150^{\circ}C$ After heating, polyphenol contents of HGR and HGL increased from 0.43 mg gallic acid equivalent/g (mg GAE eq/g) and 0.74 mg GAE eq/g to 6.16 mg GAE eq/g and 2.86 mg GAE eq/g, respectively. Conclusion: Antioxidant activities of HGR and HGL, measured by 1,1-diphenyl-2-picrylhydrazyl and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging ability, increased with increasing heating temperature. These results may aid in improving the biological activity and quality of ginseng subjected to heat treatments.

고온고압증기멸균이 우황청심원에 미치는 영향 (The Effects of High Temperature High Pressure Steam Sterilization on Woohwangchungsimwon)

  • Objectives To check marker content for appropriate quality control of Woohwangchungsimwon sterilized to ensure microbiological safety and to observe antioxidant activity for any changes in efficacy. Methods To measure any effects of sterilization on the effective compounds, 8 ingredients of Woohwangchungsimwon were screened for any changes in marker content using HPLC-DAD. Using the colorimetric method on the microplate reader any changes in total phenolic compound and flavonoid levels were observed. Antioxidant activity was measured using the DPPH, ABTS, and FRAP. Results Of the ingredients of Woohwangchungsimwon, 8 were subject to quantitative analysis before and after sterilization. 21.6 mg and 1.93 mg of Glycyrrhizin was found in Glycyrrhiza uralensis Fischer pre and post sterilization, respectively. Decursin found in Angelica gigas Nakai increased from 0.16 mg to 0.29 mg after sterilization. Bilirubin found in Gallstone of Bostaurusvar. domesticus increased from 0.24 mg to 0.33 mg. Cinnamic acid found in Cinnamomum cassia Blume increased from 0.02 mg to 0.05 mg. Ginsenoside Rb1 found in Panax ginseng C. A. Meyer decreased from 0.02 mg to 0.14 mg. Paeoniflorin found in Paeonia lactiflora Pallas increased from 1.05 mg to 1.13 mg. Amygdalin found in Armeniacae Amarum Semen increased from 2.68 mg to 2.83 mg. L-muscone found in Musk increased from 0.63 mg to 0.76 mg. As for total phenolic compound and total flavonoid content, there was a 1.22 and 4.15-fold increase. DPPH and ABTS increased by 20.45% and 20.69%, respectively. FRAP activity was 2.78 times more active post stabilization. Conclusions This study confirmed that high temperature high pressure steam sterilization, a method used to ensure microbiological safety of Woohwangchungsimwon, does not affect marker content; in other words, does not affect quality of the Woohwangchungsimwon. It could also be seen that total phenolic compound and flavonoid content increased after sterilization. An antioxidant activity test showed that there was significantly increased activity of antioxidants.

전염성 조혈기 괴사 바이러스(IHNV)의 항원 유도 단백질 특성 (Characterization of Immunogens of Infectious Hematopoietic Necrosis Virus Isolated in Korea)

  • 우리나라에 존재하며 외국에서 분리된 것과 다른 특성을 보이는 IHNV를 대상으로 하여 이 바이러스의 면역유도단백질을 확인하고자 하였다. 먼저 우리나라에서 분리된 4종류의 IHNV를 미국에서 분리된 3종류의 IHNV(OSV, SRCV 및 RB-76)와 SDS-PAGE상에서 구조단백질의 크기와 혈청학적 특성 등을 비교하였다. 그 결과 우리나라에서 분리된 4종류의 IHNV중 2종류(PRT, MRT)의 IHNV는 미국에서 분리된 IHNV와 차이가 있었으며 (Park et al., 1993), IHNV-PRT를 대상으로 면역유도단백질을 확인하기 위해 IHNV-PRT에 대한 monoclonal antibodies(MAbs)를 만들었다. 이들 중 4종류의 hybridoma를 선택하여 hybridoma cell들이 분비하는 MAbs가 어떤 class인지를 ELISA 실험을 통하여 확인한 결과 4종류 모두 IgG class에 속하는 것으로 확인되었다. 이와같이 만들어진 4종류의 MAbs가 IHNV-PRT 구조단백질들 중 어떤 것에 대한 것인지를 western blotting 실험을 통해 확인한 결과, 2종류의 MAbs는 G단백질과 특이성이 있는 것들이었고, 나머지 2종류는 G보다 조금 큰 size의 단백질에 대한 것들이었다. 다음은 IHNV-PRT에 감염된 무지개송어의 혈청을 뽑아 여기에 존재하는 IHNV-PRT에 대한 항체를 western blotting 방법으로 분석을 한 결과 G, $M_1$, $M_2$ 및 G 보다 조금 큰 size의 단백질에 대한 항체가 존재하는 것으로 나타났다. 이상의 결과로부터 IHNV-PRT의 구조단백질들 중 G, $M_1$, $M_2$ 및 G 보다 조금 큰 size의 단백질들이 면역 유도 특성이 있음을 확인할 수 있었다.

MeV Si 자기 이온주입된 단결정 Silicon내의 결함 거동 (Defect Formatìon and Annealìng Behavìor in MeV Si Self-Implanted Silicon)

  • 본 연구에서는 MeV Si 자기 이온주업을 실시하여 주업원자와 모재 원자와의 화학적 영향이 배제된 결함 형성 거동을 관찰하였다. 자기 이온주업을 위하여 Tandem Accelerator가 사용되었고 1~3 MeV의 에너지 범위의 이온주입이 실시되었다. MeV 이온주입된 시편의 격자결함은 표면으로부터 고립된 $R_p$ 근처에 집중된 것이 관찰되었다. 주입에너지 변화에 따른 격자결함 생성 거동을 관찰하기 위하여 조사량을 $1{\times}10^{15}/cm^2$으로 고정하고 주입에너지를 1~3 MeV로 증가하였다. RBS 분석 결과 격자결함의 형성층 깊이는 에너지 증가에 따라 증가하였고 표면층에는 에너지 증가시 더욱 좋은 결정성을 유지하였다. 또한 주입에너지가 일정한 경우 조사량 증가시 $R_p$ 부근에 집중된 결함층의 농도는 증가하였으나 표면부근의 결함농도는 임계조사량 이상에서 포화되는 것이 관찰되었다. XTEM 분석 결과는 RBS의 결과와 잘 일치하였다. XTEM 관찰 결과 이온주업 상태의 결함층은 dark band의 형태로 관찰되었고 열처리시 이차결함은 이곳으로부터 생성되었다. 2MeV $Si^+$ 자기 이온주입시 이차결함이 형성되는 임계조사량은 $3{\times}10^{14}{\sim}5{\times}10^{14}/cm^2$ 사이로 관찰되었다. 열처리시 dark band의 하단부의 위치는 변화하지 않고 상단부만이 제거되었다. 실험을 통하여 얻은 결과들은 Monte-Carlo technique을 이용한 TRIM-code를 사용하여 해석하였다. SIMS 분석을 통하여 이차결함은 모재내에 존재하는 oxygen 불순물을 gettering함을 관찰하였다.

논토양에서 비닐하우스를 이용한 재배방법이 인삼의 수량 및 품질에 미치는 영향 (Effect of Cultivation Using Plastic-Film House on Yield and Quality of Ginseng in Paddy Field)

  • The average and maximum temperature were $29.5^{\circ}C$ and $33.2^{\circ}C$ at 2:00 p.m. respectively, in the plastic-film house covered with shade net, and both of temperature were lower $0.6^{\circ}C$ and $1.3^{\circ}C$ than those of conventional shade. Light transmittance was 14% in the plastic-film house, while 9.9% in conventional shade during growing season from May to October. Withering time of aboveground part was on October 3rd in conventional shade with 60% of withering leaf, while it was on November 10th with 3.7% of withering leaf in the plastic-film house, about 40 days longer survival. The main disease incidence were 15% of anthracnose, 17% of leaf spot, 5% of phytophthora blight and 3% of gray mold in the conventional shade, while 0 ~ 0.1% disease incidence and 95% of emergence rate in the plastic-film house. The growth in the aboveground and underground part of ginseng was totally better, particularly characteristics affecting yield such as root length, main root length and diameter in the plastic-film house. The fresh weight was increased by 128% compared to the conventional shade and harvested roots per $3.3m^2$ were 36 roots in the conventional shade and 58 roots in the plastic-film house and futhermore yield per $3.3m^2$ was increased by 216% compared to the conventional shade. As covering materials, the rice straw in the plastic-film house was excellent. The ginsenoside contents affecting the quality of ginseng were higher in the plastic-film house indicating 0.333% of Rg1, 0.672% of Rb1, 0.730% of Rc and rate of red rusty root was less than 4.0 ~ 6.1%. Above the results, the quality of ginseng grown in the plastic-film house covered with shade net was improved than that of the conventional shade.
