가리비, Patinopecten yessoensis 소화맹낭의 계절변화 (Seasonal Variations of Digestive Diverticula in the Scallop, Patinopecten yessoensis)

  • 가리비, Patinopecten yesso두냔의 소화맹낭 및 그 상피세포의 지방에 대한 계절변동과 생식소 발달과의 관계를 알아보기 위하여, 일본 북해도의 오호츠크해에 면한 망주만산 방류양식 개체를 1982년 9월부터 1984년 5월까지 월별로 채집하여 조사한 결과는 다음과 같다. 1. 소화맹낭의 지수는 $5{\~}6$월에 가장 높았으며, 12월 및 유빙에 최저치를 나타냈다. 2. 소화맹낭의 조직단면적 1 $mm^{2}$당 세관수는 $9{\~}10$월에 최소치를 나타냈으나, 3월에 최다치를 보였다. 3. 생식소 지수(X)와 소화맹낭의 지방세관 비율(Y)은 $Y=145.6X^{-0793}$의 역지수함수적 관계를 나타냈다. 4. 소화맹낭의 지방은 생식소의 발달과 함께 생식소로 전이되는 계절변동을 보였다.

참가리비(Patinopectin yessoensis) 종패 생산중 모패의 산란횟수별 유생의 질적특성 및 동해 연안환경 적응력 (Batch-Specific Quality of the Reproductive Outputs and Nursery Acclimation in the Seed Production of Patinopecten yessoensis - case study on Korean coasts of the East Sea)

  • We studied two research items that can be undetermined issues in the artificial seed production of the Japanese scallops, Patinopecten yessoensis, batch-specific quality of the reproductive outputs and nursery acclimation, in the embayed waters of Yangyang, Gangwon, Korea. The first batch of the spawner showed better results in terms of survival, growth, and resistance against parasitic ciliate infection over the second batch that was obtained in 5 days after first batch from the same spawner. The early attached spats directly placed in the farming ground (the unacclimated) were resistant against the water current of the open environment, by showing survival of about 80% for a month, a normal survival compared with other results. However, the spat survival during the nursery acclimation was significantly lowered in comparison with that in the unacclimated condition (P<0.01). We discussed the research results of the two items, particularly focusing on their availability for mass seed production for aquaculture in the Korean coastal waters of the East Sea (Sea of Japan).

Ultrastructure of Germ Cell during Spermatogenesis and the Reproductive Cycle of the Hanging Cultured Male Scallop Patinopecten yessoensis (Pelecypoda:Pectinidae) on the East Coast of Korea

  • Ultrastructure of germ cell differentiation during supermatogenesis and the reproductive cycle in male Patinopecten yessoensis was studied by histological and cytological observations. The gonadosomatic index (GSI) in males rapidly increased and reached a maximum in April when seawater temperature gradually increased. Then the GSI gradually decreased from May through July when spawning occurred. Accordingly, monthly changes in the GSI in males coincided with testicular maturation and spawning periods. The sperm morphology of P. yessoensis belongs to the primitive type and showed general characteristics of external fertilization species. The head of the spermatozoon is approximately $3.50{\mu}m$ in length: the sperm nucleus and acrosome are approximately $2.90{\mu}m\;and\;0.60{\mu}m$ in length, respectively. The nuclear type of the spermatozoon is vase in shape, and the acrosome is cone type. The axoneme of the tail flagellum consists of nine pairs of microtubules at the periphery and a pair of central microtubules in the center The satellite body (which is formed by the centriole) and four mitochondria appear in the middle piece of the spermatozoon. The spawning period was from April through July and the main spawning occurred from May to June when seawater temperatures gradually increased. The reproductive cycle of this species can be classified into five successive stages; early active stage (September to November), late active stage (October to March), ripe stage (February to August), spawning stage (April to July), and spent/inactive stage (July to November).

큰가리비 (Patinopecten yessoensis) 치패의 양성지역에 따른 성장 특성 연구 (A Study on the Growth of Juvenile Patinopecten yessoensis from Different Aquaculture Regions)

  • 현 시점에서 동해 북부에 한정되어 양식이 이루어지고 있는 큰가리비 양식생산량 확대를 위하여 동부 북부에서 남부까지 양식시험을 통하여 성장 특성을 분석하여 생식생산량 확대를 위한 기초자료를 활용하고자 한다. 조기 종묘의 성장 특성으로 2014년 2월에 채란 및 채묘한 치패의 평균 각장, 각고는 $0.23{\pm}0.002mm$이였다. 고성장군의 경우, 6월에 평균 각장, 각고는 각각 $4.11{\pm}0.08mm$, $4.28{\pm}0.09mm$이였다. 10월에는 $23.63{\pm}3.4mm$, $24.19{\pm}4.5mm$이었다. 저성장군의 경우, 6월에 평균 각장, 각고는 각각 $2.23{\pm}0.7mm$, $1.99{\pm}0.9mm$이였고, 10월에 $1.99{\pm}0.9mm$, $17.59{\pm}4.5mm$로 성장하였다. 일간성장률은 고성장군은 0.037 mm/day, 저성장군은 0.027 mm/day 이었다. 8월에 지역별로 큰가리비의 성장을 보면 양양군, 강릉시, 영덕군, 포항시의 평균각장은 $7.93{\pm}0.2mm$으로 지역별로 큰가리비를 양성 시험한 결과, 포항 지역에서 각장 $26.90{\pm}3.6mm$로 가장 성장이 양호하였다. 이어서 영덕 지역이 $23.56{\pm}3.4mm$으로 나타났다. 이러한 결과로는 포항, 영덕 해수온과 식물플랑크톤이 큰가리비 성장에 적합하게 형성된 것으로 판단된다.

  • 한국산 가리비, 큰가리비(Patinopecten yessoensis)와 주문진가리비(Chlamys swifti), 2종에 대한 28S ribosomal RNA 유전자의 PCR- 산물을 이용 RFLP 및 염기서열을 밝히고, 이미 보고된 2과 3종의 염기서열과 상동성을 비교 분석하였다. 그 결과 28S rRNA유전자를 이용하여 7가지 제한효소를 처리한 PCR-RFLP의 종간 차이에서 Taq I 제한효소에서만 차이를 볼 수 있었다. 한편 두종간에 28S rRNA유전자의 D1 부위의 염기서열에서 231개 부위 중 14군데에서 변이를 보였다.

Growth Responses of the Scallop Patinopecten yessoensis (Pelecypoda: Pectinidae) to Shell Bioerosion and Bottom Sediment Type

  • Data obtained from field observation revealed that the degree of shell bioerosion of the scallop, Patinopecten (Mizuhopecten) yessoensis, by endolithic organisms significantly higher on the muddy sand than on the sand. At the area studied, the polychaete worm, Polydora brevipalpa (=Polydora ciliata brevipalpa, Polydora ciliata Okuda, Not Johnston, Polydora variegata), which is common symbiotic species for the scallop made 95-100% of total scallop shell bioerosion at the area studied. The muddy bottom sediments enriched by organic matter create favourable conditions for development of microphytobenthos and bacteria, which are predominantly consumed by P. brevipalpa. Linear regressions for the degree of shell bioerosion on the scallop shell height, total wet weight and adductor muscle wet weight revealed negative relationships between them for the scallops inhabiting both sand and muddy sand. The influence of polychaetes on scallops is complex. They may be food competitors. Polychaete can directly affect the host through their boreholes. Scallop expends energy for shell regeneration to prevent the polychaete penetration into its interior cavity. It was found that the degree of shell bioerosion increased considerably with scallop age.

한국 주변해역 가리비로부터 분리한 18S rDNA의 염기서열 분석 (Sequence Analysis of the 18S rDNA from Scallops Collected around Korean Sea)

  • Sequences of partial 18S rDNA have been analyzed to elucidate genetic diversity of scallops collected around Korean sea, The scallops used in genetic comparison are Argopecten irradians concentricus, Amusium japonicum japonicum, Chlamys farreri farreri, Chlamys (Swiftopecten) swifti and Patinopecten yessoensis. The 18S rDNA sequences were aligned by Clustalx program. Phylogenetic tree was drawn by Treecon program, The scallops were divided into two groups-the Family Pectinidae containing A. japonicum japonicum and the Family Propeamussiidae containing Argopecten, Chlamys and Patinopecten genera. The Family Propeamussiidae was also divided into the Supergenera Aequipecten containing A. irradians concentricus and Supergenera Chlamys containing C. farreri farreri, C. swifti and P. yessoensis. The species of C. swifti was closer to the P. yessoensis rather than C. farreri farreri in respect to nuclear 18S rDNA sequence.

The Effect of Yellow Soil on Mortality of Korean Scallops, Patinopecten yessoensis at Indoor Tank

  • In other to understand the effect of yellow soil to mortality of Korean scallops, P. yessoensis, We investigated its mortality at indoor tanks. The environmental conditions such as water temperature, Salinity, Do and pH were continued constantly during the experimental periods. The 100% of survival rate showed in two experiments groups such as 0.1% and 0.4% of concentration of yellow soil and the other groups as 0.05% and 0.2% of concentration of yellow soil was appeared one dead scallop at each group for 8 days of the experiment periods. the gills of scallop in high concentration of yellow soil (0.2% and 0.4% groups) were covered by yellow soil particles so that this group's scallop should be got a high stress from yellow soil. I think this situation will be more continued for long time the scallop will become to dead. The results of bacteriological analysis did not isolated from haemolymph of scallops and no Perkinsus infectious disease in scallops and the scallops showed necrosis and degeneration on digestive grand and gills of scallop.

  • Recent years have seen steadily increasing concerns about human exposure to heavy metals through seafood consumption. In Korea, the diverse dietary patterns of different bivalve shellfish necessitate tissue-specific evaluation of heavy metal contents for risk assessment. We analyzed the heavy metal contents of various tissues in scallops (Patinopecten yessoensis) and pen shells (Atrina pectinate). In both species, we found higher levels of cadmium (Cd) in the mantle than in the adductive muscle, while there was no significant difference between the two tissue types in total mercury (THg) and lead (Pb) (P<0.05). Levels of THg and Pb were acceptable by Korean standards. However, the level of Cd in pen shell mantle tissue exceeded the standard by 40%. The rate of heavy metal exposure from consumption of these species was shown to be 0.003-0.010% of the provisional tolerable weekly intake of THg and 0.312-0.600% of the provisional tolerable monthly intake (PTWI) of Cd as recommended by the Joint FAO/WHO Expert Committee on Food Additives. These findings on the tissue-specific heavy metal contents of these species will be useful in assessing the health risks of Korean dietary habits.

Identification and Characterization of Three Differentially Expressed Ovarian Genes Associated with Ovarian Maturation in Yesso Scallop, Patinopecten yessoensis

  • Despite great commercial interest, relatively little has been described about molecular mechanism of bivalve reproduction. We investigated genes involved in ovarian maturation of the Yesso scallop, Patinopecten yessoensis. GSI index and histological analysis revealed that maturation of ovary begin in February and spawning period is from April to June which is similar to the previous study in the East Sea. As result of combination analysis of differential display RTPCR (DDRT-PCR) and histological examination, vitellogenin (Vg), ferritin (Ft) and ADT/ATP carrier protein (ACC) were identified as differently expressed genes in maturating ovary. Endpoint RT-PCR results showed that Vg is ovary-specific genes whereas Ft and ACC are expressed ubiquitously suggesting that Vg can be good molecular markers for ovarian development and sex determination in bivalves. Quantitative PCR results revealed that Vg were expressed highest during growth stage and appears to play a major role in oocyte maturation. On the contrary, expression of Ft was highest after spawning stage, which suggests that up-regulation may be involved in spawning and inactive stages in which the scallops recover from spawning. In addition, high level of the mitochondrial gene, ACC, may play a role in energy metabolism in maturating oocytes. Isolation and molecular studies of these key genes will expand our knowledge of the physiological changes from various exogenous factors including temperature, salinity, pH, even or numerous endocrine disrupting chemicals (EDCs) during reproductive cycle. In addition, further study of these genes implicates various industrial applications including the stable seed production, increased food quality, or economic aquaculture system.
