  • A fucoidanase, which was screened from the heptopancreas of Patinopectin yessoensis, was fractionated according to by ammounium sulfate precipitation and anion exchange chromatography. The crude fucoidanase activity was hydrolyzed a small amounts of fucoidan agar and not arrageenan. The crude enzyme hydrolyzed fucoidan to produce oligosaccha-rides of fucose, glucose, maltose maltotriose and maltoteraose as the reaction products.

  • 우리나라 동해안에서 주요 양식대상종으로 각광을 받고 있는 가리비의 산란유발은 종묘생산시 생산성을 높이는 가장 중요한 요소라고 할 수 있다. 따라서 본 연구는 자극방법에 따른 가리비의 산란유발 효과와 유생사육에 관하여 조사한 결과, 가리비의 생식소 지수는 4월에 암수 각각 32.4와 33.2로 최고치를 보였고, 정자현탁액 첨가구에서 개체당 백만개 이상과 80%이상으로 가장 좋았다.

  • The genetic relationship was examined with PCR-RAPD markers among three scallop species, Chlamys farreri farreri, Patinopecten yessoensis, and Agropecten irradians. Six primers were selected from 60 primers used to compare PCR-RAPD profiles among species. All primers showed distinct RAPD band patterns between the three species. In Chiamys farreri farreri, the morphological characteristics such as shell size and color were considerably different between the two geographical populations. RAPD profile, however, showed that no significant genetic differences were found between the two geographical populations. Polymorphic alleles were observed within a population of each species. Thus, PCR-RAPD markers are useful in identifying scallop species and in understanding scallop population genetic structure.

  • Unexpected mass mortality has been one of the drawbacks in front of the stable production of Japanese scallop (Patinopecten yessoensis) on the Gangwon coasts of the East Sea. The preliminary data from our routine observation revealed that the mortality appeared to be related to variation of water temperature in the farming site and the degree of the mortality was dependent on scallop strain. The present study performed to verify the preliminary findings exhibited that the mortality was closely related to daily temperature variation rather than monthly variation. Daily temperature variation was particularly damageable to the scallop during the temperature elevation period. Scallops from hatchery seeds (Chinese strain) were more tolerant against the temperature variation over those from wild seeds. The hatchery scallop gain of the temperature tolerance was probably due to their larval experience to higher temperature in the hatchery as well as their maternal genetic acclimation to upper temperature extreme of the Chinese environment which was recently found.

  • Toxicity and toxin compositions of wild and cultured scallops (Patinopecten yessoensis), collected from coastal waters near Kangnung of East Sea, were examined from January to June, 1997. By mouse bioassay methods, the toxicity was detected with low toxicity of $2 MU\;g^{-1}$, and paralytic shellfish poisoning (PSP) toxin was detected in the specimens from 30 April to 15 May by HPLC. GTXs and PXs were identified as the major toxin components.

  • Few studies on parasitic scutica-like ciliates (SLCs) influencing larval growth and survival during the seed production of the Japanese scallop, Patinopecten yessoensis have been performed in spite of relatively high magnitude of their infectious damage to the larvae. In the mass seed production of the scallop, SLCs were never infectious during D-larval stage which lasted 6 to 7 days. Indeed, the infection first occurred in 2 to 4 days after first umbo-staged larvae. A regular selection of active larvae in swimming behavior kept larval SLC infection low throughout the culture compared to unselected control (P<0.05) in which all the larvae alive were contained. Higher infection frequencies of the unselected control resulted in lower larval survivals and wider range of larval size distribution that drove larval attachment rate lower. To the worse, the nursery spats from the higher SLC infection exhibited a retarded growth and elevated occurrence of abnormal scallop.

  • Myostatin (GDF8) is a growth factor that limits muscle tissue growth and development in vertebrates. We isolated a myostatin-like gene (Py-MSTN) from the marine invertebrate, the scallop Patinopecten yessoensis. Py-MSTN was highly expressed in the adductor muscle and in the gill unexpectedly. Amino acid analysis showed that Py-MSTN has 49% amino acid sequence identity and 64% similarity to human myostatin (Hs-MSTN), and 42% identity and 61% similarity to myoglianin, the only invertebrate homolog. These results indicated that Py-MSTN may be functionally similar to the vertebrate MSTN than the invertebrate homolog. Phylogenetic analysis suggested that Py-MSTN is an ancestral form of vertebrate MSTN and GDF11 and does not belong to other $TGF-{\beta}$ family members. Molecular modeling showed that Py-MSTN exhibits a similar tertiary structure to mammalian BMP7, a member of $TGF-{\beta}$ family. In addition, the amino acid residues which contact extracellular domain of the receptor were relavively conserved. Given these results, we propose that Py-MSTN is a functionally active member of the $TGF-{\beta}$ family and is involved In muscle growth and regulation.

Genetic Diversity and Population Structure of the Scallop Patinopecten yessoensis in Korea, China, and Japan by Random Amplified Polymorphic DNA Markers (RAPD 마커에 의한 한국, 중국, 일본 참가리비의 유전적 다양성과 집단 구조)

  • Sixty individuals of the scallop $Patinopecten$ $yessoensis$ (Genus Pecten) were sampled to examine the genetic diversity and population structure of this species. Random amplified polymorphic DNA (RAPD) identified 109 genotypes and produced 79 polymorphic loci (72.8%). Total genetic diversity values ($H_T$) and interlocus variation in the within-population genetic diversity ($H_S$) were 0.254 and 0.178, respectively. On a per-locus basis, the proportion of total genetic variation due to differences among populations ($G_{ST}$) was 0.299. This indicated that about 70.1% of the total variation was within populations. The unique loci and bands of $P.$ $yessoensis$ were shown in only one population among the three countries. RAPD markers were very effective in classifying the natural population levels of $P.$ $yessoensis$ in Korea, China, and Japan. In addition, insights into the relative gene diversity among and within populations of $P.$ $yessoensis$ would be useful in breeding and for the development of strategies for animal genetic resources.

Egg Development and Lana Growth of the Scallop, Patinopecten yessoensis (가리비, Patinopecten yessoensis 난 발생과 유생의 성장)

  • In order to obtain the basic information for seedling production of the scallop, Patinopecten yessoensis, the egg development and larva growth were investigated at different conditions such as water temperature, salinity and phytoplankton. Eggs were demersal isolated eggs, which averaged 77.3${\pm}$2.7$\mu\textrm{m}$ in diameter after spawning. The fertilized eggs developed to D-shaped larva of shell length 117.5${\pm}$3.8$\mu\textrm{m}$ after 60 hours at 15$^{\circ}C$. The range of water temperature and salinity during egg development were 10-20$^{\circ}C$ and 28-34 ppt, respectively. The time of egg development was shorter with higher water temperature. After 10 days of spawning, D-shaped larva reached 160 $\mu\textrm{m}$ in shell length, and after 25 days became full-grown larva 250 $\mu\textrm{m}$ in shell length, in which could be observed eye spots. The relative growth formula between shell length (SL) and shell height (SH) was SH = 1.0425SL-27.731 (r$^2$= 0.9749) during the entire larva period. In regard to water temperature, growth and survival rates of larvae were good at 16$^{\circ}C$. Lower growth and survival rates were observed at 12$^{\circ}C$ and 20$^{\circ}C$ than that at 16$^{\circ}C$. When larvae were fed mixed phytoplankters, such as isochrysis galbana, Pavlova lutheri and Chaetoceros calcitrans, their growth and survival rates were the highest among groups.

  • Our previous finding summarizing that larval sieving process is inevitable but triggers the outbreak of scutica-like ciliates (SLCs) in the seed production of Patinopecten yessoensis urged further study to determine best suggestable sieving interval in an agreeable range of water quality. In the mass seed production of the scallop, SLC outbreak was closely related to the larval sieving in which larvae were drained on the basis of every 3-day (5T), 5-day (3T), 7-day (2T), or 9-day (1T) from culture tanks onto a mesh screen and placed back into new water in cleaned tanks. The larval performance of growth and survival was clearly dependent on the sieving intervals. It was in order of 3T, 5T, 2T, and 1T for both of growth and survival and in reverse order for SLC infection frequency, confirming that larval sieving is necessary but damageable if it overwhelms the larval resistance. Interestingly, the larval damages by the sieving persisted to their ensuing spat life in terms of nursery growth, survival, and abnormality.
