  • Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and manganese superoxide dismutase (MnSOD) has been hypothesized as a mediator in the activation of multiple pathways implicated in the pathogenesis of diabetic disease. The objective of this study was to understand the mechanism that aerobic exercise activate GAPDH and MnSOD in pancreatic cells. To achieve the purpose of this study, thirty male Sprague-Dawley rats were assigned to control group, diabetic group and diabetic exercise group. 10 rats were forced to exercise according to exercise protocol for 8weeks and 20 rats were untrained for control and diabetic group. Pancreatic tissue were extracted from the each. Expressions of GAPDH and MnSOD in diabetic pancreatic tissues were significantly decreased compare to control group. However, swimming (trained diabetic group) significantly increased expressions of GAPDH and MnSOD compare to diabetic group, respectively. In hyperglycemia, GAPDH and MnSOD in pancreatic cells is activated by aerobic exercise, and this inactivates multiple pathways implicated in the pathogenesis of diabetic disease. In conclusion, these findings suggest that increased activity of GAPDH and MnSOD by exercise have beneficial effects on mitochondrial dysfunction and arresting the progression of diabetic disease.

  • The aim of this study was to discover whether Yukmijihwangtang which was added Plygalae Radix, Acori graminei Rhizoma, has antioxidant effects on aged senescence-accelerated mouse (SAM) P8. The control group used 12 months old SAM P8 and the experimental group was treated with Yukmijihwangtang extracts from 8 to 12 months old. In the liver, the MnSOD of the control group were approximately 11% higher than that of the experimental group treated with herbal extracts, but CuZnSOD activities of the experimental group were approximately 20% higher than that of the control group. The catalase activities of the experimental group was approximately 67% higher than that of the control group. The hepatocytes, in the control group compared with the experimental group, contained the dilated cisternae of rough endoplasmic reticulum and the abnormal mitochondira which had electron-dense matrix and indistinguishable cristae. The inner cavity of the smooth endoplasmic reticulum were slightly dilated, and a number of vacuoles and transitional vesicles occured in of the hepatocytes of the control group. These results suggest that the herbal extract has an antioxidant effects on the liver of SAM P8.

  • From the total methanolic extract of Chrysanthemum coronarium L. var. spatiosum (Compositae), nitrite scavenging ability and superoxide dismutase (SOD)-like activity were analyzed as antioxidative characteristics. After successive partitioning with chloroform, n-butanol, and water, the chloroform fraction showed the most significant nitrite scavenging ability with an $IC_{50}$ value of 39 ppm compared with the values of vitamin C and chlorogenic acid, 15 ppm and 36 ppm, respectively. The active fraction was subjected to silica gel and Sephadex LH-20 column chromatography, and the compound was isolated and identified as ${\beta}-sitosterol-O-{\beta}-D-glucoside$ using $^{1}H-NMR$ and $^{13}C-NMR$ spectral data. The glucoside was further hydrolyzed and confirmed as a glycosylated ${\beta}-sitosterol$. The compound and its aglycone, ${\beta}-sitosterol$, showed different nitrite scavenging and SOD-like activity. The $IC_{50}$ value of nitrite scavenging ability of the compound was 335 ppm at pH 1.5, while that of its aglycone was 41 ppm. As for the SOD-like activity, the $EC_{50}$ values of the sterol and the glucoside were 1,291 ppm and >2,000 ppm, respectively, compared with those of vitamin C and chlorogenic acid, 38 ppm and 449 ppm, respectively.

  • Nitric acid and hydrofluoric acid are used for acid pickling in zirconium alloy tubing manufacturing process. Nitrate and fluoride in the wastewater were treated by chemical coagulation and SOD (Sulfur Oxidation Denitrification) process. This study is investigated the effect of fluoride concentration and the optimal condition for SOD process. The limited fluoride concentration for SOD process was below 20 mg F-/L. The adjusted pH and alkalinity by NaOH and $NaHCO_3$ was shown to be more effective for removal of nitrate compared with using NaOH. Furthermore, the microbial activator mixed trace elements and ingredient for alkalinity did not only supplement with alkalinity but also enhance the growth and proliferation for sulfur-oxidizing bacteria. As a result, the inorganic industrial wastewater was successfully treated by the microbial activator in SOD process without continuous addition of seed sludge. Finally, SOD process was shown to remove nitrate in industrial wastewater and to contribute the microbial activator for activation of sulfur-oxidizing bacteria.

  • The purpose of this study was to investigate gender-specific changes of plasma MDA, SOD, and lymphocyte DNA damage during high intensity exercise. In this study, 17 healthy male and 18 healthy female college students ran on a treadmill at 85%$VO_{2max}$ until the point of all-out. Blood-collecting was carried out five times (Rest, Ex-Exha, R0.5h, R4h and R24h), and with the collected blood, plasma malondialdehyde (MDA), superoxide dismutase (SOD), and lymphocyte DNA damage were analyzed. Plasma MDA and SOD concentration increased significantly at the Ex-Exha (p<0.05), and there were no significant differences in gender. For the degree of lymphocyte DNA damage, all %DNA in the tail, tail length and tail moment increased significantly at the Ex-Exha (p<0.05), and %DNA in the tail and tail length were significantly higher in the male group than in the female group (p<0.05). These results suggest that acute high intensity exercise not only causes oxidative stress but also brings about lymphocyte DNA damage. In addition, it was found that males showed higher DNA damage than females in terms of oxidative stress subject to high intensity exercise. Nevertheless, further subsequent studies are required in order to better understand the mechanism behind DNA damage varying with gender, in a way that takes into consideration physical fitness, hormonal level, exercise intensity and duration - additional factors which might affect DNA damage.

  • This study was investigated to analyze the effect of extracts from the Lepista nuda, on the antioxidant activity to form a part of studies on the functional materials of L. nuda. Antioxidant activity of L. nuda extracts was evaluated by measuring the electron-donating ability (EDA), the superoxide dismutase (SOD)-like activity, and the nitrite-scavenging ability. The EDAs of water and ethanol extracts at the concentration of 1,000 ppm by the rotary heating method from L. nuda were $60.47\%$ and $60.13\%$, respectively and those of water and ethanol extracts by microwave-assisted method were $87.73\%$ and $84.84\%$, respectively. The measurements of SOD-like activity were in the range of $24.58\%\~42.03\%$ at 1,000 ppm. EDA and SOD were increased with the concentrations of extracts. The nitrite-scavenging ability at the concentration of 1,000 ppm was the highest and $29.77\%$ at pH 1.2, and was decreased with an increment of pH value. These results indicated that microwaveassisted water extract from L. nuda showed the highest activities on the EDA and nitrate-scavenging ability, while the rotary heating ethanol extract had the highest effect on the SOD-like activity.

  • This study was conducted to compare the physiological and biochemical responses of P. orientalis and L. tulipifera in response to ozone. One-year-old seedlings of P. orientalis and L. tulipifera were exposed to 100 ppb ozone concentration for 2, 4, or 8 hr/day for 1 week. Photosynthesis, stomatal conductance and ozone uptake rate were measured daily, and chlorophyll fluorescence, carboxylation efficiency, chlorophyll content, and SOD activity were measured after 1 week. In P. orientalis, photosynthesis and stomatal conductance were not decreased in the 2h/day ozone treatment, but the L. tulipifera response was more sensitive even in the 2h/day ozone treatment. Increased treatment time decreased photosynthesis and stomatal conductance. Chlorophyll fluorescence was not significantly different among treatment times. However, carboxylation efficiency decreased with increased treatment time, and L. tulipifera was more sensitive than P. orientalis. Chlorophyll content did not differ with species or treatment time. SOD activity response was greater in L. tulipifera than in P. orientalis, increasing to $131\%$ of pretreatment observations. Therefore it was concluded that L. tulipifera was more responsive and had lower ozone tolerance than P. orientalis.

  • Analysis of chlorophyll (Chl) fluorescence implicated treatment of 40 mM NaCl decreased maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm), actual quantum yield of PSII (${\Phi}_{PSII}$), and photochemical quenching (qP) in rice, but increased non-photochemical quenching (NPQ). Decreases in Fv/Fm, ${\Phi}_{PSII}$, and qP were significantly alleviated by $30\;{\mu}M$ jasmonic acid (JA), while NPQ increase was enhanced. Transcription levels of antioxidant isoenzyme genes were differentially modulated by NaCl treatment. Expression of cCuZn-SOD2 gene increased, while those of cAPXb, CATb, and CATc genes decreased. JA prevented salt-induced decrease of pCuZn-SOD gene expression, but caused greater decrease in mRNA levels of cAPXa and Chl_tAPX genes. Investigation of vacuolar $Na^+/H^+$ exchanger (NHX2) and 1-pyrroline-5-carboxylate synthetase (P5CS) gene expressions revealed transcription level of NHX2 gene was increased by JA, regardless of NaCl presence, while that of P5CS gene slightly increased only in co-presence of JA and NaCl. Unlike JA, ${\gamma}$-radiation rarely affected expressions of antioxidant isoenzyme, NHX2, and P5CS genes, except for increase in mRNA level of Chl_tAPX and decrease in that of pCuZn-SOD. These results demonstrate enhanced salt-tolerance in JA-treated rice seedlings may be partly due to high transcription levels of pCuZn-SOD, NHX2, and P5CS genes under salt stress.
