Gastroprotective Activities of Sennoside A and Sennoside B via the Up-Regulation of Prostaglandin E2 and the Inhibition of H+/K+-ATPase

  • Sennoside A (erythro) and sennoside B (threo) are dianthrone glycosides and diastereomers. We investigated their abilities to prevent the gastric lesions associated with diseases, such as, gastritis and gastric ulcer. To elucidate their gastroprotective effects, the inhibitions of $HCl{\cdot}tOH$-induced gastritis and indomethacin-induced gastric ulcers were assessed in rats. It was observed that both sennoside A and sennoside B increased prostaglandin $E_2$ ($PGE_2$) levels and inhibited $H^+/K^+$-ATPase (proton pump). In a rat model, both compounds reduced gastric juice, total acidity and increased pH, indicating that proton pump inhibition reduces gastric acid secretion. Furthermore, sennoside A and B increased $PGE_2$ in a concentration-dependent manner. In a gastric emptying and intestinal transporting rate experiment, both sennoside A and sennoside B accelerated motility. Our results thus suggest that sennoside A and sennoside B possess significant gastroprotective activities and they might be useful for the treatment of gastric disease.

Comparison of Biological Activities of Korean Halophytes

  • Halophytes are expected to possess abundant secondary metabolites and various biological activities because of habitat in extreme environments. In this study, we collected 14 halophytes (Asparagus oligoclonos, Calystegia soldanella, Carex pumila, Chenopodium glaucum, Elymus mollis, Glehnia littoralis, Limonium tetragonum, Messerschmidia sibirica, Rosa rugosa, Salsola komarovii, Spergularia marina, Suaeda glauca, Suaeda maritima, and Vitex rotundifolia) native to Korea and compared their total polyphenol contents, antioxidant and anti-inflammatory activities. The total polyphenol contents of R. rugosa (27.28%) and L. tetragonum (13.17%) were significantly higher than those of the other 12 halophytes and L. tetragonum, R. rugosa, and M. sibirica showed significantly greater antioxidant activities than the other 11 halophytes, as determined by DPPH (2,2-diphenyl-1-picrylhydrazyl). A. oligoclonos, E. mollis, and C. pumila showed significantly greater anti-inflammatory activities than the other 11, as determined by NO (Nitric oxide) and $PGE_2$ (Prostaglandin $E_2$) levels. In contrast, these three extracts had normal and low total polyphenol contents among the 14 halophytes. Consequently, the total polyphenol content in the 14 studied halophytes appeared to be related to antioxidant, but not anti-inflammatory activity levels.

The Ameliorative Effect of Rubi Fructus on DSS-induced Colitis in Mice

  • Ulcerative colitis (UC) is an inflammatory bowel disease and a chronic gastrointestinal disorder. Rubi Fructus (RF), the fruit of Rubus coreanus Miquel, is known to exert several pharmacological effects including anti-oxidative, anti-obesity and anti-inflammatory properties. However, the improving effect and mechanism of RF on intestinal inflammation is not been fully understood. The purpose of this study was to investigate the regulatory effect of RF on dextran sulfate sodium (DSS)-induced colitis in mice. We evaluated the effects of RF on DSS-induced clinical signs by analyzing weight loss and colon length. The inhibitory effects of RF on inflammatory mediators such as prostaglandin E2 (PGE2), cyclooxygenase (COX)-2, as well as the activation of nuclear factor-κB (NF-κB), were determined in colitis tissue. Our data indicated that mice treated with DSS showed clinical symptoms of colitis, including weight loss, colon length decrease and diarrhea. However, we observed that RF treatment significantly improved these clinical symptoms of weight loss, colon length decrease and diarrhea induced by DSS. RF inhibited the enhanced levels of COX-2 and PGE2 caused by DSS. We also showed that the anti-inflammatory mechanism of RF by suppressing the activation of NF-kB in DSS-treated colon tissues. Collectively, the findings of this study indicate the prospect of developing new drugs from RF for UC treatment.

The Anti-inflammatory Mechanism of Protaetia brevitarsis Lewis via Suppression the Activation of NF-κB and Caspase-1 in LPS-stimulated RAW264.7 Cells

  • The larva of Protaetia brevitarsis Lewis (P. brevitarsis), edible insect, is traditionally consumed as alternative source of nutrients and has various health benefits. However, the exact pharmaceutical effects of P. brevitarsis on inflammatory response are still not well understood. Thus, we investigated the anti-inflammatory effects and mechanisms of P. brevitarsis in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. We investigated the effects of P. brevitarsis on the expression levels of inflammatory-related genes, including inflammatory cytokines, prostaglandin E2 (PGE2), cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS) in LPS-stimulated RAW264.7 cells. To understand the anti-inflammatory mechanism of P. brevitarsis, we explored the regulatory effect of P. brevitarsis on nuclear factor (NF)-κB and caspase-1 activation. The findings of this study demonstrated that P. brevitarsis inhibits the LPS-induced inflammatory cytokine and PGE2 levels, as well as COX-2 and iNOS expression. Moreover, we confirmed that the anti-inflammatory effect of P. brevitarsis occurs via suppression of the activation of NF-κB and caspase-1. Conclusively, these findings provide experimental evidence that P. brevitarsis may be useful candidate for the treatment of inflammatory-related diseases.

Relationship between Antioxidant Nutrient Intakes, Blood Oxidative Stress and Inflammatory Markers of Overweight and Obese Adults in Yeongdong Area (영동지역 과체중 및 비만 성인에서 항산화 영양소섭취상태와 혈중 산화 및 염증관련 지표와의 관련성)

  • This study was designed to investigate the relationship between the nutrient intakes, antioxidants, and inflammatory markers of overweight and obese adults (46 females and 16 males) in Yeongdong area. The study was conducted through questionnaires, anthropometric checkups, 2-day 24 hr recalls and blood biomarker analyses. Body fat was significantly higher in women compared to men. Diastolic blood pressure (DBP) and systolic blood pressure (SBP) were significantly higher in men compared to women. There were no significant differences in height, weight, body mass index, and waist size among the two groups. The intake of nutrients was significantly higher in men compared to women. Ferric reducing ability plasma (FRAP) was significantly higher in men compared to women. Body fat was positively associated with blood IL-6 and IL-8 levels. DBP was positively associated with FRAP. The intake of protein was negatively associated with IL-6 levels. The intake of carbohydrates was negatively associated with total antioxidant capacity (TAC). Prostaglandin E2 (PGE2) levels were negatively associated with TAC. These results suggest that antioxidant and inflammatory markers may be related to the body fat percentage and dietary intake in overweight and obese adults.

Anti-inflammatory Effect of Indirubin-3'-Monoxime-5-Sulphonic Acid on Lipopolysaccharide-stimulated Murine Macrophage

  • Indirubin is the active ingredient of Danggui Longhui Wan, a mixture of plants that is used in traditional Chinese medicine to treat chronic diseases. In this study we investigated the anti-inflammatory effects of an indirubin derivative, indirubin-3’-monoxime-5-sulphonic acid (I3M-5S, $C_{16}H_{11}N_3O_5S$). We found that I3M-5S inhibits the production of various inflammatory mediators such as nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) as well as inflammatory cytokines, tumor necrosis factor-${\alpha}$ and interleukin-6 in lipopolysaccharide (LPS) stimulated murine macrophage, RAW264.7 cells. In addition, the expression of inducible nitric oxide synthase and cyclooxygenase-2, which are essential enzymes to produce NO and $PGE_2$, respectively, was blocked by I3M-5S treatment in LPS-stimulated RAW264.7 cells. Present data suggest that I3M-5S exhibits potent anti-inflammatory activity in cultured macrophages and merit further study as potential therapeutic agents for inflammatory disorders.

Study on the Anti-inflammatory Effects of the Remedy Prescripted with Lonicerae Flos and Scutellariae Radix in U937 cells (금은화(金銀花)와 황금(黃芩)이 배오(配伍)된 처방제(處方劑)의 항염증(抗炎症) 효과(效果) 연구(硏究))

  • Inflammatory reaction is characterized by over-production of inflammatory mediators due to an up-regulation of inflammatory pathways, which produce pro-inflammatory mediators, such as interleukin-1beta (IL-$1{\beta}$), IL-6, tumour necrosis factor alpha (TNF-$\alpha$), prostaglantin $E_2$ ($PGE_2$), and nitric oxide (NO) in U937 cells. We investigate the anti-inflammatory effects of water extracts from Lonicerae Flos and Scutellariae Radix in lipopolysaccharide (LPS)-stimulated U937 cells. Each extract suppressed the production of inflammatory mediators (NO, IL-$1{\beta}$, TNF-$\alpha$, and $PGE_2$) and the expression of inducible NO synthase and cyclooxygenase-2 in LPS- stimulated U937 cells in a dose-dependent manner. These suppressive effects were synergistically increased by their combination. Their combination extract also inhibited NF-${\kappa}B$-DNA complex of NF-${\kappa}B$ binding activity and translocation of NF-${\kappa}B$ from cytosol to nucleus. These results suggest that the combination of water-extractable components of Lonicerae Flos and Scutellariae Radix may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

Suppressive effects of pinosylvin on prostaglandin E$_2$and nitric oxide production in lipopolysaccharide-stimulated RAW 264.7 cells

  • The inhibitors of prostaglandin biosynthesis and nitric oxide production by corresponding inducible isozyme have been considered as potential anti-inflammatory and cancer chemopreventive agents. In our continuous search for cancer chemopreventive agents from natural products, we have evaluated the inhibitory potential of PGE$_2$and NO production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells. As a result, pinosylvin (3,5-dihydroxy-trans-stilbene), a stilbenoid, mainly found from the heartwood and leaves of the Pinus sylvestris, showed potential inhibitory activity of LPS-induced PGE$_2$and NO production in a dose-dependent manner. Pinosylvin also suppressed the LPS-induced iNOS protein expression. Further study revealed that pinosylvin exhibited antioxidant activity by the DPPH free radical scavenging potential and inhibitory effect of xanthine oxidase activity. In addition, pinosylvin inhibited COX -2 overexpressed human colon cancer cell (HT-29) growth in a time- and dose-dependent manner. These findings suggest that pinosylvin might be a promising candidate for developing cancer chemopreventive agent.

Effect of Chrysanthemum zawadskii var. latilobum on the release of inflammatory mediators from LPS-stimulated mouse macrophages (구절초(九折草)가 LPS로 염증유도(炎症誘導)된 대식세포주(大食細胞柱)에 미치는 영향(影響))

  • 목적 : 구절초는 국화과에 속하는 다년생 초본으로 가을에 줄기와 잎을 가을에 채취한 것을 한약재로 사용하여 대한, 월경불순 등 각종 여성질환과 함께 위냉증, 소화불량, 감고, 폐렴, 기관지염, 배뇨장애 및 신경퇴행성 질환에 활용되고 있다. 본 연구에서는 LPS로 염증유도된 대식세 포주를 활용하여 구절초의 항염증효능을 확인하고자 하였다. 방법 : 구절초 추출물의 항염증효과를 관찰하기 위하여 RAW264.7 대식세포주에서 MTT cytotoxic assay, iNOS 및 COX-2 발현, NO와 PGE2 생성 및 $NF-{\kappa}B$ 활성실험을 수행하였다. 결과 : 세포독성실험을 통하여 구절초 추출물의 안전성이 확인되었으며 LPS로 염증유도된 RAW264.7 대식세포주에서 증가된 iNOS의 발현이 감소되었음을 확인하였다. 또한 NO 및 PGE2의 생성을 억제하였다. 이러한 결과는 구절초가 염증매개물질의 생성을 억제하는 효과가 있음을 확인할 수 있었으며, $NF-{\kappa}B$ 활성도를 감소시킴을 관찰하였다. 결론 : 이러한 결과는 구절초가 $NF-{\kappa}B$ pathway를 조절해 줌으로써 항염증효과를 나타내는 것으로 사료된다.

Experimental study on the Anti-inflammatory and wound healing effect of Hyelgalsan (혈갈산(血竭散)이 항염작용(抗炎作用)에 미치는 영향(影響))

  • Hyelgalsan(HGS) is important prescriptions that have been used in oriental medicine for stomatitis and wound healing. The study was done to evaluate the inhibitory effects of cytotoxicity, formation of superoxide on the macrophage and neutrophil, prostaglandins($PGE_2$), interleukins($IL-1{\beta}$), collagenase activity and synthesis of collagen and DNA. The results were obtained as follows: 1. HGS was not showed the proliferation difference of human fibroblast and monocyte in all concentrations to be experimented and in result, it was concluded that they have no cytotoxicity. 2. HGS inhibited the formation of superoxide to 48% at the concentration of 0.01% in the mouse monocyte. 3. HGS was not showed the proliferation difference of human monocyte in all concentrations to be experimented and in result, it was concluded that they inhibited the formation of superoxide. 4. HGS was not showed the proliferation difference of human neutrophil in all concentrations to be experimented and in result, it was concluded that they inhibited the formation of superoxide. 5. The concentration of inhibiting the production of prostaglandins($PGE_2$) to slight in the human monocyte stimulated with E. coli were 0.01% of HGS. 6. The concentration of inhibiting the production of interleukins($IL-1{\beta}$) to slight in the human monocyte stimulated with E. coli were 0.001% and 0.0001% of HGS. 7. HGS didn't influence on collagen synthesis and total protein in fibroblasts. 8. HGS inhibited the collagenase activity to 22% at 0.1%, 45% at 0.2%, 57% at 0.5% respectively.
