Studies on the Substance of Migration for Retort Pouch Packaging Materials for Various Condition (레토르트 파우치 포장재의 사용조건에 따른 이행물질의 조사)

  • The effect of microwave heat to retort pouches and microwavable packages was examined. 186 products were collected, but packages were consisted of five package materials such as PET, Aluminum foil, Nylon, EVOH, and polypropylene. The results showed that all packages did not exceed the limits of current packaging regulation on consumption weight of $KMnO_4$, TDI(Toluene diisocyanate), and Caprolactam. Safety control on heat resistant packages considered satisfactory, but investigation on new substances should be continued for preventing any possible migration problems of packaged foods.

  • S. cerevisiae mutant(reg1-501, gal1), which cannot use galactose and has alleviated glucose repression level, is used as host for optimizing induction of GAL promoter. The optimum concentration of galactose as inducer for recombinant protein production and the galactose consumption rate have been tested with S. cerevisiae mutant and compared with conventional S. cerevisiae. The extent of glucose repression were investigated for both strain and the degradation pattern of produced foreign protein have been compared in both cases. The effect of pH on foreign protein degradation pattern were studied for both strains. The secetion efficiency of both strains were carried out. Through these experiments, optimum condition of recombinant protein production by GAL promoter using S. cerevisiae mutant (reg1-501, gal1) were found.

Survival and Physiological Response of Hybrid Grouper (Epinephleus bruneus♀ × Epinephleus lanceolatus♂) Exposed to Different Salinity Water (교잡바리(대왕자바리)의 생존과 생리학적 반응에 미치는 염분의 영향)

  • The profer salinity range for aquaculture of hybrid grouper (Epinephleus bruneus♀ × Epinephleus lanceolatus♂) were evaluated in terms of their survival, growth, feed intake and physiological responses after 30 days of exposure to different salinities (0 psu, 3 psu, 5 psu, 10 psu, 15 psu, 20 psu, 25 psu, 30 psu). Hybrid grouper all died at 0 psu salinity on the 4th day of exposure but all hybrid survived at a salinity of 5 psu or higher during the exposure period. Body length and weight increased above 5 psu salinity and the growth rate was the highest at 30 psu. Hybrid exposed to 3 psu did not eat the feed pellet. Feed intake did not show a significant difference between salinity 20 psu, 25 psu and 30 psu. Plasma osmolality of the juvenile hybrids exposed to different salinities was 341~368 mg Osmol/kg above 5 psu. The oxygen consumption rate was the highest at 30 psu with 163.6±22.3 mg O2/kg fish/h. SOD, CAT and GSH-PX were significantly higher at 15 psu. Therefore, the critical salinity for survive of the juvenile hybrid was 5 psu, and the proper salinity for optimal culture was analyzed with 20~30 psu.

Efficacy of Sodium Hypochlorite against E. coli on Various Leafy Green and Stem Vegetables (차아염소산나트륨이 비가열 엽경채류 중 병원성 대장균 사멸에 미치는 영향)

  • This study was conducted to evaluate the efficacy of sodium hypochlorite in eliminating Escherichia coli strains from leafy green and stem vegetables, which are frequently sold at community service centers. A cocktail of non-pathogenic E. coli and enterohaemorrhagic E. coli (E. coli O157:H7) was used to artificially contaminate the vegetables (initial numbers of bacteria 7-8 log CFU/g). The contaminated vegetables were soaked in sodium hypochlorite for 5 min and then washed three times with running water. After the treatment, number of viable bacterial cells on the vegetables was estimated. Sodium hypochlorite treatment reduced the E. coli population by 1-2 log CFU/g on leafy green and stem vegetables, a significant reduction from the initial number. Further, sodium hypochlorite showed better antimicrobial efficacy for leaves with a larger surface area, less roughness, and softness. There was no significant difference in the antimicrobial effect between 100 and 200 mg/kg of sodium hypochlorite. Therefore, it is not necessary to increase sodium hypochlorite concentration than the level suggested in the school meal hygiene management guidelines. However, sodium hypochlorite treatment is not sufficient to achieve a safe level of microorganisms on leafy green and stem vegetables since they generally have a high abundance of microorganisms on their surface. Thus, an alternative cooking method for fresh leafy green and stem vegetables in summer should be developed to ensure they are safe for consumption.

Effects of Chamaeneron angustifolia Extract on Lipid Metabolism and Differentiation of 3T3-L1 Preadipocyte (Chamaeneron angustifolia 추출물이 3T3-L1 Preadipocyte의 지방대사 및 분화에 미치는 영향)

  • Objectives : Obesity is an imbalance between energy intake and consumption due to overeating and lack of exercise, and if it persists, it increases non-infectious diseases such as high blood pressure, diabetes, and hyperlipidemia. In this study, we tried to investigate the possibility of using Chamaeneron angustifolia (CA) as a material for anti-obesity by confirming the effect of inhibiting lipid differentiation. Methods : We measured the effects of CA extract on oil-red-o staining, cell cytotoxicity evaluation activity using 3T3-L1 cells. Additionally, we assessed fat decomposition and metabolism-related protein expression through Western blot analysis. Results : In this study, the anti-obesity effects of CA extract were experimentally assessed. Results showed significant inhibition of adipocyte differentiation and accumulation at concentrations of 0.05, 0.1, and 0.2 mg/ml of oil-red-o staining, with reductions of 80% or more. CA notably increased the phosphorylation of AMPK protein expression compared to the control group across all concentrations. Additionally, phosphorylation of ACC significantly increased at a concentration of 0.2 mg/ml compared to the control. PPAR-γ, which regulates adipogenesis, exhibited a significant decrease compared to the control, while protein expression of CPT-1, involved in fatty acid oxidation, showed a concentration-dependent increase across all groups. Therefore, CA extract demonstrates potential as a functional material for anti-obesity by increasing the expression of proteins related to fat decomposition and synthesis while decreasing others. Conclusions : These results suggest that CA may also be useful as an anti-obesity functional substance.

Changes in Blood pH, $Pco_2$ and $Po_2$ During Passive Tilt (체위 변화시의 혈액 pH, $Pco_2$$Po_2$의 변화)

  • To evaluate orthostatic brain hypoxia from the passive tilt, anesthetized dogs were tilted from supine to $90^{\circ}C$ upright and $90^{\circ}C$ head down position. Blood pH, $Pco_2$ and $Po_2$ changes during tilt were measured on 8 dogs. Respiratory rate was decreased in upright position and increased in head down position comparing to that in rte horizontal position. pH in arterial blood was increased in upright position and decreased in venous blood comparing to that in the horizontal position. No significant changes were found in the head down position. $Pco_2$ of arterial and venous blood was decreased both in upright and head down. position A significant decrease was found in the head down position. $Po_2$ in arterial blood was increased both in the upright and head down position. However, it was decreased in the venous blood. The cause of increased $Po_2$ in arterial blood seemed to do due to hyperventilation and the cause of decreased $Po_2$ in venous blood was thought to be due to increased $O_2$ consumption of animals during the tilt.

The Correlation between HRCT Emphysema Score and Exercise Pulmonary Testing Parameters (HRCT Emphysema Scoring과 운동부하 폐기능검사 지표들 간의 상관관계)

  • Background : The correlation between the high resolution computed tomography(HRCT) emphysema score and the physiologic parameters including resting and exercise pulmonary function test was investigated in 14 patients($60.6{\pm}10.3$ years) with pulmonary emphysema. Methods : The patients underwent a HRCT, a resting pulmonary function test, and incremental exercise testing(cycle ergometer, 10 W/min). Computed tomography scans were obtained on a GE highlight at 10 mm intervals using 10 mm collimation, from the apex to the base after a full inspiration. The emphysema scores were determined by a CT program 'Density mask' outlining the areas with attenuation values less than -900 HU, indicating the emphysema areas, and providing an overall percentage of lung involvement by emphysema. Results : Among the resting PFT parameters, only the diffusing capacity(r=-0.75) and $PaO_2$ (r=-0.66) correlated with the emphysema score(p<0.05). Among the exercise test parameters, the emphysema score correlated significantly with the maximum power(r=-0.74), maximum oxygen consumption(r=-0.68), anaerobic threshold(V-slope method: r=-0.69), maximal $O_2$-pulse(r=-0.73), and the physiologic dead space ratio at the maximum workload(r=-0.80)(p<0.01). Conclusion: We could find that exercise testing parameters showed a much better correlation with the HRCT emphysema score, which is known to have a good correlation with the pathologic severity than the resting PIT parameters. Therefore it is suggested that exercise testing is superior to resting PIT for estimating in the estimation of the physiologic disturbance in emphysema patients.

Oxidative Pathway of $C^{14}-glucose$ in Various Human Cancer Tissues (각종 인체 암조직의 당의 산화경로 분석)

  • Tissue homogenates of 12 kinds of human cancer tissues were incubated separately in medium containing $C^{14}-1-glucose$ and $C^{14}-6-glucose$ as a substrate in order to observe the oxidative pathway of glucose in the tumor tissues. At the end of 3 hours incubation in the Dubnuff metabolic shaking incubator, respiratory $CO_2$ samples trapped by alkaling which was placed in the center well of incubation flask were analysed for total $CO_2$ production rates and their radioactivities. The tissue homogenate samples after incubation were analyzed for their concentrations of glucose, lactate and pyruvate. Calculations were made on the glucose consumption rate and accumulation rates of lactate and pyruvate. Fractionation of oxidative pathway of glucose was carried out by calculating $C^{14}O_2 yields from C-1 and C-6 carbon of glucose. The following results were obtained. 1. In 12 kinds of human cancer, total $CO_2$ production rates were less than $8{\mu}M/gm$ except 2 cases. These lower values impressed that oxidative metabolism in the tumor tissues generally inhibited as compared with that in normal tissues. On the other hand, fractions of $CO_2$ derived from glucose to total $CO_2$ production rates (RSA) were less than 10% in every case. These facts showed that oxidation of glucose into $CO_2$ was remarkably inhibited in the tumor tissues. 2. Factions of glucose disappeared into $CO_2\;(RGD_{CO_2})$, lactate $(RGD_L)$, pyruvate $(RGD_P)$ to glucose consumption rates were as follows. $RGD_{CO_2}$ were less than 2% in cases of in this experiment and $RGD_L$ showed more than 5% except in 2 cases. These facts showed that anaerobic degradation of glucose into 3 carbon compounds was easily proceeded but further degradation into $CO_2$ via the TCA cycle was greatly inhibited resulting in accumulation of lactate. There are large variation in values of $RGD_P$ in different kinds of tumor tissue but relatively higher values in $RGD_{CO_2}$ were obtained in the tumor tissues as compared with those of normal tissues. 3. The oxidative pathway of glucose in tumor tissues were analyzed from the values of RSA which were obtained in $C^{14}-1\;and\;C^{14}-6-glucose$ incubation experiments. It was found that 3% of $CO_2$ derived from glucose were oxidized via the principal EMP-TCA cycle and the remainder were via alternate pathway such as HMP in the liver cancer and values in other cancer tissues were as follows; 4% in the tongue cancer, 6% in the colon cancer, 6% in the lung cancer, 9% in the stomach cancer, 11% in the ovarian cancer, 12% in the neck tumor, 22% in the uterine cancer, 22% in the bladder tumor, 32% in the spindle cell sarcoma and 65% in the brain tumor. These values except later 2 cases showed less than 30% which is the lowest value among the normal tissues. Even in the brain tumor in which showed highest value in the tumor group. It is reasonable to suppose that this fraction was remarkably decreased because values in normal brain tissue was more than 90%. From the above data, it was concluded that in tumor tissues, oxidation of glucose via TCA cycle was greatly inhibited but correlation between degree of inhibited oxidation of glucose via TCA cycle and malignancy of tumor were not clarified in this experiments.

Evaluation on Heating Effects of Geothermal Heat Pump System in Farrowing House (지열 난방시스템을 이용한 분만돈사의 난방효과 분석)

  • Geothermal heat pump system (GHPS) is an energy-efficient technology that use the relatively constant and renewable energy stored in the earth to provide heating and cooling. With the aim of using GHPS as a heating source, it's possibilities of application in farrowing house were examined by measuring environmental assessment and sow's performance. A total of 96 sows were assigned to 2 pig housings (GHPS and conventional housing) with 48 for four weeks in winter season. During the experimental period, indoor maximum temperature in GHPS-housing was measured up to $26.7^{\circ}C$, average temperature could maintain $21.2^{\circ}C$. The mean value of dust levels and $CO_2$, $NH_3$ and $H_2S$ gas emissions were decreased in GHPS-housing compare with those of conventional housing. Litter size, birth weight, parity and weaning weight did not differ between housings. However, feed intake of sow in GHPS-housing was lower than that of conventional housing. In energy consumption for heating, electric power consumption increased in GHPS-housing than the conventional housing, a 2,250 kwh increase, whereas there is no fuel usage for heater in GHPS-housing. Amount of ground water circulated for heating in cold weather for earth heat exchanger was 8.4-12.9 ton per day. In conclusion, GHPS may have environmental benefits and effectiveness of heating in farrowing housing and affect the performance in sows.

  • With ever rising concerns about saving of fossil fuel resource, there have been an increasing demand for use of energy more efficiently. The electric motor driven inverters can be a great help to improve energy efficiency. They are also used to control the motor speed to the actual need. Therefore the use of them can lead to reduce energy consumption. In particular, the medium voltage(MV) drive systems used for pumps, fans, steel rolling mills and tractions have widespread applications in the industry. They cover power ratings from 0.4MW to 40MW at the MV level of 2.3kV to 13.8kV. The majority of the installed MV drive systems however, are in the 1MW to 4MW range with voltage rating from 3.3kV to 6.6kV. But they are required to reduce size and weight like other power electronic equipments. In this paper, we studied on the 3.3kV(105A) compact rack type inverter system for improving the cooling efficiency. At first, we confirmed the tendency of temperature with computational simulation using ANSYS ICEPAK and actual experimental tests. And then we researched thermal performance improvement designs in order to reduce temperature of the transformer for the safe operation. It can reduce temperature of transformer that using pipe type flow guide in the system. As a result, we found out more efficient solution by thermal-fluid analysis.
