Paraquat 유도독성에 대한 금은화 엑스의 효과(IV) (Scavenging Effects of Lonicera Japonica Extracts on Paraquat Induced Toxicity(IV))

  • Scavenging effects on paraquat induced toxicity were investigated by using methanol (MeOH) and ethylacetate (EtoAC) extracts of Lonicera japonica. The results are summerized as follows: 1. To Fe(III)-ADP-NADPH induced microsomal lipid peroixdation, MeOH and EtoAC extracts showed antioxidative activiies and inhibition ratio at 100 $\mu\textrm{g}$/$m\ell$ 44.4% and 73.8% respectively 2. To microsomal NADPH dependent cytochrome p -450 reductase in rat liver, MeOH and EtoAC extracts inhibited the enzyme activiies and inhibition ratio were 26.3% and 44.8% respectively. 3. Administration (30 mg/kg, iv) of paraquat to rats caused the marked elevation of GOT, GPT, LDH, ALP in the serum and lipid peroxides in the microsome as compared to the control group. Serum GTP, LDH, ALP and liver microsomal LPO were reduced significantaly by administration of MeOH extract. (1,000 mg/kg), EtoAC extract (40 mg/kg) and Silymarin (150 mg/kg) as compared to the paraquat group. From the results, MeOH and EtoAc exuacts. of Lonicera japonica showed the useful scavenger and reducer on the paraquat induced hepatotoxicty.

챠넬메기 간에서 Hydroxylated Polychlorinated Biphenyl의 Glucuronidation (Glucuronidation of Hydroxylated Polychlorinated Biphenyl by Channel Catfish Liver)

  • Hydroxylated polychlorinated biphenyl (OH-PCBs)는 PCB의 CYP-dependent oxidation의 대사물로서 잠재적 독성이 강하고 지질친화성을 가지며 생물군에 지속적인 축적성을 나타낸다. OH-PCBs의 해독화 효능을 조사하기 위하여 channel catfish 간에서 glucuronidation을 통한 해독작용 가능성을 평가하고 biphenyl 구조에 다양한 염소치환의 구조적 차이점에 따른 영향을 비교 분석하여 보았다. Kinetic parameters에서 $K_m$$V_{max}$$192{\sim}871{\mu}M$, $869{\sim}1774$ pmo1/min/mg으로써 4'-OH-PCB35와 4'-OH-PCB69이 가장 높은 속도의 glucuronidation을 나타냈으며, 구조적 차이점에서 phenolic group에 한 개의 염소치환이 존재할 경우보다 두 개의 염소치환이 존재할 경우에 OH-PCBs(p<0.001)의 glucuronidation에 대한 $V_{max}$를 현저하게 낮추는 결과를 보였다.

인진쑥 추출물의 항산화 및 항암 활성 (Antioxidant and Anticancer Activities of Extract from Artemisia capillaries)

  • Artemisia capillaries is a major important food and medicinal resource in Korea. In order to confirm the biological activities of Artemisia capillaries, we investigated antioxidant and anticancer activities from in vitro assays. The Artemisia capillaries methanol (MeOH) extracts was used for the evaluation of DPPH scavenging, total phenolic content, total flavonoid content, hydroxyl radical (${\bullet}OH$) scavenging, reducing power assay as antioxidant activity, as well as anticancer activities as MTT assay. As a result, the Artemisia capillaries MeOH extracts showed potent antioxidative activity and anticancer activity in vitro. These results suggest that the Artemisia capillaries MeOH extracts have a potential alleviated oxidation process, cell motility activity, and tumorigenesis.

  • As the continued studies for Korean cultivated rhubarb, MeOH extract of the leaves was fractionated with ether, ethylacetate, and n-butanol. From the ethyl acetate fraction of MeOH extract, one flavone glycoside, apigenin-8-${\beta}$-D-glucopyranoside(vitexin, $C_{21}H_{20}O_{10}$) and from the n-BuOH fraction of MeOH extract, two flavonol glycosids, kaempferol-3-O-(2,6-di-O-rhamnopyranosyl)-${\beta}$-D-galactopyranoside$(C_{33}H_{40}O_{19})$and quercetin-3-O-rutinoside(rutin, $C_{27}H_{30}O_{16}$) were isolated and identified through the physico-chemical properties and spectroscopic evidences(UV, IR, NMR, Mass) respectively.

  • 시간 상관 단일 광자 계수법을 사용하여 PVP/C/sub 2/H/sub 5/OH와 PEG/H/sub 2/O 혼합 용매에 위치한 rhodemine 6G 분자의 회전 재배열 시간을 측정하였다. 주어진 온도에서 혼합비를 변화시키면서 측정한 결과, PEG/H/sub 2/O와 PVP/C/sub2/H/sub 5/OH에서 각각 회전 이완 시간이 혼합액의 점성도에 선형적으로 비례하지 않는 것으로 관측되었다. 고분자 주의에 cluster가 형성된다는 관점에서 혼합비에 따른 회전 이완 시간의 변화를 설명하였다.

Charges of TIP4P water model for mixed quantum/classical calculations of OH stretching frequency in liquid water

  • The potential curves of OH bonds of liquid water are inhomogeneous because of a variety of interactions with other molecules and this leads to a wide distribution of vibrational frequency which hampers our understanding of the structure and dynamics of water molecules. Mixed quantum/classical (QM/CM) calculation methods are powerful theoretical techniques to help us analyze experimental data of various vibrational spectroscopies to study such inhomogeneous systems. In a type of those approaches, the interaction energy between OH bonds and other molecules is approximately represented by the interaction between the charges located at the appropriate interaction sites of water molecules. For this purpose, we re-calculated the values of charges by comparing the approximate interaction energies with quantum chemical interaction energies. We determined a set of charges at the TIP4P charge sites which better represents the quantum mechanical potential curve of OH bonds of liquid water.

근관치료에서 Ca(OH)₂의 사용

  • Calcium Hydoxide는 치수복(pulp capping), 생활치수절단술(vital pulpotomy)등에서 bactericidal effect 때문에 많은 사용을 하고 있으며, 최근에는 무수치(pulpless teeth)와 관련된 치근단병소가 있는 경우, 미완성치근단인 경우, 치근흡수, 횡주치근파절(Transverse root fracture)등의 경우에서도 Ca(OH)₂paste를 사용하여 성공적인 근관치료를 할 수 있다. 근관의 instrumentation 과 irrigation후 근과 dressing 은 약 1주일간 행하고 Ca(OH)₂paste를 건조된 근관내에 넣는다. 이때 치근단(root apex)으로부터 약 3~5mm 되게 저속으로 아주 가볍고 부드러운 압력을 가하여 Ca(OH)₂paste를 cotton pellet로 밀어넣고 Sealing(가봉)한다. 이 Temporary sealing은 1개월이상 견딜수 있도록 Cement 또는 Amalgam filling 함이 좋다. Ca(OH)₂paste의 제법은 normal irrigating agents 와 minimal filing으로 시행한다.

산성 광산 배수의 처리를 위한 철(Fe) 성분의 플럭 형성 특성 (The Characteristics of Iron(Fe) Floc Formation for Treatment of Acid Mine Drainage)

  • The characteristics of floc formation of the iron(Fe) ions was studied for developing the process treating the acid mine drainage. The metal ions in aqueous solution oxidized with oxygen in air, which generated hydrogen ion and lowered the pH of the aqueous solution. The iron(Fe) ions were formed into flocs by the acid-base reaction with the added $Ca(OH)_2$ for the neutralizing the solution. There were several variables affecting the formation, size and color of floc; whether air was present or not, air feeding rate, oxidizing time, concentration of $Ca(OH)_2$, the acid-base reaction time of the $iron(Fe)-Ca(OH)_2$. For proper formation of the $iron(Fe)-Ca(OH)_2$ flocs and developing the floc treating system, the control variables mentioned above should be considered.

  • Thermal and electrical conductivities of the natural, artificial, and expandable graphites were analyzed after treatment in NaOH aqueous solution. In order to investigate the elimination of the oxidized groups and impurities on the graphite surfaces after NaOH treatment, the graphite samples were structurally characterized by using XRD, XPS, Raman, FE-SEM. The thermal and electrical conductivities of the graphite samples were significantly improved after NaOH treatment. These results were caused by the structural rehabiliation.

애기거머리말의 항산화 활성 (Antioxidant Activity of the Seagrass Zostera japonica)

  • In this study, crude extract of the seagrass Zostera japonica, and its solvent-partitioned fractions were evaluated for their antioxidant activity. The crude extract was successively fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water fractions by liquid-liquid partition. These include DPPH radical scavenging, hydroxyl radical scavenging in HT-1080 cells, peroxynitrite scavenging, and protective effect on DNA damage caused by hydroxyl radicals generated. In all assays, except for DPPH radical, 85% aq.MeOH and n-BuOH fraction showed the strong antioxidant activity. These results suggest that Z. japonica may be used as a potential source of natural antioxidants for the development of cosmetic product or functional food in the future.
