Determination of blood concentration of higenamine by high pressure liquid chromatography

  • A procedure utilizing high pressure liquid chroatography coupled with UV detection is described for the determination of blood concentration of higenamine. Deproteinized serum was pretreated with$C_{18}$(Sep-pak $C_{18}$ cartridge) and the 70% EtOH eluent was applied onto a reversed-phase column ($\mu$ Bondapak $C_{18}]$) with a 15% acetonitrile in 0.05 N $NaH_2$$PO_4$-trichloroacetic acid mixed buffer (pH 2.8) as a mobile phase. With the UV detection at 232 nm, the retention times of higenamine and 1, 2, 3, 4-tetrahydropapaveroline, an internal standard, were 5.2 min and 3.9 min respectively. The blood concentration of higenamine was meausred at regular intervals after i. v. injection of higenamine to rabbit. A drastic decrease in higenamine concentration to 30% of the maximum value obtained immediately after the injection, was observed during the first 1-2 min period and thereafter the rate of decrease was slowed down. The analytical result seemed to coincide with the pharmacological effect of higenamine exerting the maximum chronotropic and hypotensive effect at the completion of the injections which were progressively recovered.

  • A microorganism capable of producing high level of extracelluar cyclomaltodextrin glucanotransferase(EC 2.4.1.19; CGTase) was isolated ’rom soil. The isolated strain No. 239 was identified as Bacillusstearothermophilus. The maximal CGTase production (about 7.0 unitslml) was observed in medium containing2% soluble starch, 0.5% defatted soybean meal, 0.1% NaH_2PO_4.2H_2O$ and 0.015% $ CaC_l2 $ with initial pH 7.0. The strain was cultured at $55^{\circ}C$ for 48 hr with reciprocal shaking. At 0.83% substrated concentration potato starch was the optimum substrate with 50.1% conversion to cyciodextrin (CD)after the reaction at $65^{\circ}C$ for 24 hr (CGTase 10 unitlg starch). Using soluble starch as substrate (5% substrate concentration, CGTase 10 unitlg starch), the maximum conversion of 40% was obtained at11 hr reaction, and the ratio of $\alpha-, \beta-$ and $\gamma$-CD production at this time were 1.0:1.3:0.4, respectively., respectively.

  • The strain producing biosurfactant was isolated from soil smples. The isolated strain was identified as the genus Nocardia through its morphological, cultural and physiolgical characteristics. A high concentration of the biosurfactant by Nocardia sp. L-417 was obtained after 4 days of cultivation in the culture medium containing 3% n-hexadecane, 0.1% $NaNO_3$, 0.02% $K_2HOP_4$, 0.01% $H_2PO_4$, 0.01% $MgSO_4$.$7H_2O$, 0.01% $CaCl_2$, 0.02% yeast extract, and 0.02% tryptone. The optimum pH and temperature for biosurfactant production were pH 6.0 and $30^{\circ}C$, respectively. Furthermore, most biosurfactans were produced during the exponential growth phase, and this fact indicated that the biosurfactans production was growth-associated. The biosurfactant showed the good emulsification activities on various emulsifying substrates such as bunker A, paraffin, corn oil which are used widely in industries.

  • Graft copolymerization of methlymethacrylate(MMA) onto sodium alginate(SA) aqueous solution by sodium metaperiodate$(NaIO_4)$ as an initiator was carried out with the variation of the reaction time, the reaction temperature and the concentrations of initiator and monomer. The results obtained were as follows: 1. SA was easily separated from the reaction mixture of homopolymer, graft polymer and sodium alginate ungrafted by the treatment of concentrated boiling $Na_3PO_4$, solution with the small amount of $Na_2SO_3$ to the mixture and then isolated as the acid form by acidifying the salts solution containing SA. 2. The amount of graft polymer was larger than that of homopolymer below $70^{\circ}C$ whereas above $70^{\circ}C$ the amount of homopolymer was larger. 3. The sum of each amount of graft polymer ana homopolymer was always increased with increase of the reaction time, the reaction temperature and the concentrations of initiator and monomer. 4. With increase of the concentration of initiator, the graft efficiency was increased below $70^{\circ}C$ while decreased above $70^{\circ}C$. 5. Graft copolymerization of MMA onto SA in aqueous solution was carried out without initiator.

Preparation and Characteristics of Konjac Noodle-added Mugwort (쑥을 첨가한 곤약국수의 제조 및 특성)

  • Konjac gel is known as a low calorie food, because the main component is water. Mugwort has been used for food and medicine for a long time due to itsfunctional property, which contains a lot of vitamins and minerals. Therefore, konjac noodle-added mugwort was prepared. The konjac gel was made by from a centrifuge after adding water, alkali and mugwort power in konjac flour. The centrifuge was used to remove air in the gel. After centrifuging by heating the gel in boiling water for one hour, konjac noodle was made by pressing using a flat heating press. The results are as follows. In the drying process of mugwort freeze drying after blanching in 1% $NaHCO_3$ solution is better than other drying methods. The physical properties (hardness, elasticity, cohesiveness, gumminess) of konjac gel were measured with a Rheometer. The optimum processing conditions were decided by these measured values. The optimum condition of making konjac gel is 9% concentration of konjac flour, a ph of 12.0, $Na_3PO_4$ coagulant and 1.5% concentration of mugwort power in konjac flour.

Synthesis Conditions and Rheological Characteristics of Aluminum Phosphate (인산 알루미늄의 합성조건과 유동학적 특성)

  • Aluminum phosphate gel was synthesized by reacting aluminum sulfate as a soluble aluminum salt to tribasic sodium phosphate in this study. The optimal synthesis conditions based on the yield of product were investigated by applying Box-Wilson experimental design. It was found that optimal synthesis conditions were as follows: Reaction temperature; $61~71^{\circ}C$, concentration of two reactants; 12.27~13.83%, concentration ratio of two reactants; [AI$_{2}$(SO$_{4}$)$_{3}$]/[Na$_{3}$PO$_{4}$]= 0.5, reaction time; 10.9~12.1 minutes, drying temperature of product; $60~72^{\circ}C$. Aluminum phosphate gel prepared by the optimal synthesis conditions was suspended with four types of natural and synthetic gums at the concentration of 0.375~1.5wv%. Their Theological properties of aluminum phosphate gels were examined with Haake-Rotovisco RV 20 rotational viscometer. It showed that the higher concentration of suspending agents and lower temperature, the higher viscosity. Aluminum phosphate gel suspended by pectin and agar showed plastic flow with rheopexy, and their gels suspended by sodium alginate and sod. CMC showed plastic flow with thixotropy.

  • Various kinds of native herbage grasses like-Miscanthus sinensis, Arundinella hirta, Cymbopogen geirngii, Themeda japonica etc. are widely distributed in every Korean mountain. So we investigated the availability of native grass, Miscanthus sinensis as a substrate for the production of S. C. P. The results were obtained as follows. 1) At the alkali treatment, NaOH was the most effective with the exclusion of lignin, and pretreated Miscanthus with NaOH appeared to be a good substrate for the microbial growth. 2) When Miscanthus was treated with one to 10% NaOH, the microbial growth increased in proportion to the increased alkali concentration. Beyond 4% NaOH, a slight increase was observed. 3) Phosphoric acid, as a neutralizer, was the most effective in cell production after alkali treatment. 4) On the effect of incubation time, the productivity was best found at 60 hours, and the cell weight was 9.23 mg per 1 ml, and the microbial digestibility of substrate was 75.2%.

Impact of urbanization and industrialization on irrigation water quality of a canal - a case study of Tongi canal, Bangladesh

  • The Dhaka city, the capital of Bangladesh is one of the most densely populated cities in the world. Tongi canal is situated on the north of the city, which connected the Turag river to the west and the Balu river to the east. A total of 26 water samples were collected from the canal to measure irrigation water quality on the basis of their trace metal and major ionic constituents. Trace metals concentrations in water samples were determined using an Atomic Absorption Spectrophotometer. The amount of Fe, Mn, Zn and Pb in water samples ranged from 0.01-0.80, trace-1.02, trace-0.054 and $0.43-0.64{\mu}g\;mL^{-1}$, respectively. The mean concentration of Ca, Mg, Na, K, $HCO_3{^-}$, $Cl^-$, $BO{_3}^{3-}$, $PO{_4}^{3-}$ and $SO{_4}^{2-}$ in water samples were 45.32, 15.33, 151.65, 11.98, 516.06, 94.69, 0.33, 14.02 and $56.21{\mu}g\;mL^{-1}$, respectively. In respect of $HCO_3{^-}$, $SO{_4}^{2-}$, $PO{_4}^{3-}$ and K contents, most of the water samples were found problematic for irrigation. In context of RSC and hardness, 96 and 92% of water samples were graded as unsuitable and hard class, respectively. The study concluded that Pb content in canal water was comparatively high, so it is desirable to take necessary initiative to minimize the contamination level and to monitor its concentration in water routinely.

Optimization of Medium for $\beta$-Mannanase Production by Aspergillus oryzae (Aspergillus oryzae에 의한 $\beta$-Mannanase 생산배지의 최적화)

  • Medium optimization for ${\beta}$-mannanase production by Aspergillus oryzae ATCC 2114 was performed. Effect of carbon source (locust bean gum) concentration on ${\beta}$-mannanase production was investigated. Above 20 g/L locust bean gum, a lag time for ${\beta}$-mannanase production was appeared because high concentration of locust bean gum caused high viscosity which made the mixing of medium poor. As the locust bean gum concentration in the medium increased, ${\beta}$-mannanase activity and cell growth increased proportionally. Effect of various nitrogen sources on ${\beta}$-mannanase production was also studied. (NH4)2SO4 and malt extract were the most effective for ${\beta}$-mannanase production among the inorganic nitrogenous compounds and organic nitrogen nutrients. Inorganic compounds such as KH2SO4, NaCl, Na2CO3, and MgSO4, on ${\beta}$-mannanase production were optimized for ${\beta}$-mannanase production. Locust bean gum of 10 g/L, malt extract of 3 g/L, (NH4)2SO4 of 2 g/L, KH2SO4, of 10 g/L were selected as the optimal medium. Culture in a fermentor by using the optimal medium was carried out. Lag time of ${\beta}$-mannanase production was shorter due to the better mixing of the fermentor. The maximum ${\beta}$- mannanase activity of 9.7 unit/mL and specific ${\beta}$-mannanase activity of 1.9 unit/mg-cell could be obtained at 27 hours and the productivity of ${\beta}$-mannanase was 0.36 unit/mL$.$h.

  • A validated UV determination of loratadine in human plasma was developed and the pharmacokinetic profiles of single dose of loratadine were determined in 8 healthy volunteers. Human serum samples (1.0 mL) spiked with known concentration of loratadine and 50 ng diazepam as an internal standard were alkalinized with 500 ${\mu}\ell$ ${\mu}\ell$ of 10% $Na_{2}CO_{3}$ and extracted with 7 mL of mixture of isopentane and hexane (2 : 1. v/v) for 5 min. Extracts were centrifuged and 6 mL of organic layer was back-extracted with 150 ${\mu}\ell$ of 12.5% $Na_{2}PO_{4}$ for 1 min. (omitted)
