카드뮴의 중추신경계 독성유발 기전 (The Mechanism of the Neurotoxicity Induced by Cadmium)

  • Although numerous studies have shown that cadmium disturbs the normal biological processes in central nervous system, the mechanism of toxicity is not well understood. The present study has investigated the effect of cadmium on oxidative stress, Na$^{+}$K$^{+}$ ATPase activity and the aggregation of amyloid beta peptide ($\beta$-amyloid) in neuronal cell line, HT22 cell. LC$_{5}$ and LC$_{50}$ of cadmium for HT22 cell resulted from MTT assay was 4.1 uM and 9.5 uM, respectively. Cadmium (2 to 8 uM) dose-dependently increased the lipid peroxidation and decreased the content of glutathione. Cadmium 4 uM showed a significant decrease in Na$^{+}$/K $^{+}$ ATPase activity as compared with control group. The aggregation of $\beta$-amyloid was accelerated in a dose-dependent manner by the treatment with 2 to 8 uM cadmium. These results suggest that the neurotoxicity of cadmium can be mediated by the increase in oxidative stress and decrease in Na$^{+}$/K$^{+}$ ATPase activity.se activity.

Altered Cardiac $Na^{+}$,$K^{+}$-ATPase Activity in Prehypertensive Spontaneously Hypertensive Rat

  • $Na^{+}$,$K^{+}$-ATPase activity, $Na^{+}$-dependent phosphorylalion, and [$^3$〕ouabain binding in sarcolemma prepared from 4 week old spontaneously hypertensive rat(SHR) ventricles were compared to the same parameters in sarcolemma from age matched normotensive Wistar-Kyoto(WKY) rat ventricles to examine whether the reduced myocardial $Na^{+}$-pump activity in SHR is an inherited enzymatic defect or a second phenomenon due to sustained hypertension. The total body weights, ventricular weights, and blood pressures were the same for SHR and WKY. No significant differences in sarcolemmal protein content and protein recovery were noted between the two pups. Sarcolemma isolated from SHR ventricles showed sigificantly less $Na^{+}$,$K^{+}$-ATPase activity and number of phosphorylation sites when compared to snrcolemma 1mm the WKY ventricles. Equilibrium binding of [$^3H$〕ouabain and the turnovcr number of myocardial $Na^{+}$,$K^{+}$-ATPast however, wee the same for both groups. These results. indicate that the low affinity(${\alpha}$, or ${\alpha}_1$) isoform for ouabain is reduced in SHR compared to WKY but that the high affinity(${\alpha}$+, or ${\alpha}_2$) isoform is the same in ventricles of SHR and WKY. The reduced amount of at isoform of the $Na^{+}$,$K^{+}$-ATPasc in prehypertensive SHR ventricles may play some pole in the development of hypertension.

BC3H-1 분화세포에서의 (Na,K)ATPase ${\alpha}_2$ isoform의 표현증대 (Increased Expression of the ${\alpha}_2$ Isoform of (Na,K)ATPase in the Differentiated Murine Muscle Cell Line BC3H-1)

  • The development of the alpha2 isoform of (Na,K)ATPase which is high affinity ouabain receptors was studied in the differentiating nonfusing muscle cell line BC3H-1. T he differentiation process of BC3H-1 cell line was confirmed by 2-dexy-D-[$^3$H] glucose uptake experiment and the quantity of the expression of ${\alpha}_2$ isoform was measured using a whole cell [$^3$H] ouabain-binding assay. Undifferentiated growing BC3H-1 cells, myoblasts, exhibited low levels of insulin-stimulated glucose uptake and [$^3$H] ouabain-binding sites. In contrast, differentiated BC3H-1 cells, myocytes, had a 5.6-fold increase in insulin-stimulated glucose uptake and 5-fold increase in [$^3$H] ouabain-binding sites. Scatchard analysis showed that myocytes developed more [$^3$H] ouabain-binding sites than myoblasts vath a dissociation constant (kd) of 6${\times}10^{-8}$M and capacity of 6.l${\times}10^{-5}$ sites/cell. Therefore. it seems that myoblasts express low levels of ${\alpha}_2$ subunit and probably the majority of ${\alpha}_1$ subunit, whereas myocytes express high levels of ${\alpha}_2$ isoform. The results indicate that the expression of ${\alpha}_2$ isoform is developmentally regulated during differentiation and that BC3H-1 culture system provides an excellent model for the study of differentiation and mechanism of (Na,K)ATPase action in muscle which requires electrical excitability.

백서의 DMBA 유도 악하선종양에서의 $Na^+,K^+$-ATPase에 대한 연구 (A STUDY OF $Na^+,K^+$-ATPase IN THE RAT SUBMANDIBULAR GLAND TUMOR INDUCED BY DMBA)

  • 체중 100g 내외의 Sprague Dawley계 정상백서 암, 수 각 50마리씩을 실험동물로 하여 분말형태의 순수 DMBA를 소구(pellet)로 압착제작하여 우측 악하선에 매식후 즉시, 1,3,5,7,9,11,13,15,17주째 10마리씩 희생시켜 좌, 우측악하선을 적출, 무게를 측정하고, 육안적, 조직학적, 생화학적으로 실험기간에 따라 비교분석하여 다음과 같은 결론을 얻었다. 1. 매식후 11 주째부터 우측 경부 악하선부위의 종창을 관찰할 수 있었으며, 결절덩어리를 촉진할 수 있었다. 2. 우측악하선의 무게의 크기는 실험기간의 경과에 따라 괄목할만한 증가를 보였다(매식후 17주째는 좌측에 비해 약 5.5배의 무게를 나타냈다). 3. 매식후 7주와 11주째부터 조직학적으로 상피이형성과 침윤성 유표피암의 소견을 각각 관찰할 수 있었다. 4. 우측 악하선에서의 유표피암의 발생율은 매식후 17주째 76%이었다. 5. DMBA 는 현저히 억제된 $Na^+,K^+$-ATPase 활성도를 야기하며 실험기간의 경과의 따라 활성도 비율도 현저히 저하시킨다.

  • Background The stenosis of the coronary artery may decrease myocardial oxygen supply and occur myocardial ischemia or infarction. Soojeomsan, one of analgesics is generally regarded to have the effect of vitalizing blood, expelling blood stasis and alleviation cardiac pain. Methods The purpose of this experimental study is to find the influence of Soojeomsan on cardiac enzyme (CPK, Na-K ATPase) of ischemic and reperfused rat hearts which are isolated under the Langendorff apparatus. Ischemia was induced In isolated hearts of Sprague-Dawley rats by ceasing the perfusion for 20 minutes. The experiments were divided into a normal saline orally administered group(control group), a Soojeomsan orally 20ml administered group(sample A) and a Soojeomsan orally 30ml administered group(sample B). The CPK (creatinine phosphokinase) and Na-K ATPase activity of this three group were measured and compared in order to assess the influence of Soojeomsan on protection of isolated rat hearts from ischemia. Results 1. CPK was significantly reduced in Sample A group and Sample B group in comparison with control group in reperfusion(P<0.01), and there were no significant difference between Sample A and B. 2. Na-K ATPase activity was significantly increased in Sample A group and Sample B group in comparison with control group in ischemia(P<0.001), and the activity was significantly higher in Sample B then in Sample A.(P<0.01) 3. There were no significant difference in Na-K ATPase activity of the three groups after reperfusion. Conclusion Soojeomsan has effects to decrease CPK activity and activate Na pump. This result in protection of the myocardium of isolated rat hearts from ischemia.

인삼이 토끼 적혈구막의 $Na^{+}-K^{+}-ATPase$의 활성도에 미치는 영향 (Effect of Ginseng on Sodium-Potassium activated ATPase in Rabbit Red Cell Membrane)

  • The effect of ginseng on the ATPase activity of rabbit ref cell membrane has been investigated. The experiments were also designed to determine whether the components of ginseng could be attributed to the effect on ATPase activity which dependent upon sodium plus potassium and is sensitive to ouabain. The following results were observed. 1. The activity of the $Na^{+}-K^{+}-ATPase$ from red cell membrane is stimulated by ginseng, and the concentration of ginseng for half-maximal activity is about 15 mg%. The pH optimum for the ginseng sensitive component is 7.6. 2. The portion of the enzyme activity stimulated by ginseng is completely abolished by ouabain. 3. The activating effect of ginseng on the ATPase, with a given concentration of sodium in the medium, is increased by raising the potassium concentration but activity ratio is decreased. 4. The activating effect of ginseng on the ATPase, with a given concentration of potassium in the medium, is increased by raising the sodium concentration but the activity ratio is decreased. 5. The ATPase activity is increased by small amounts of calcium but inhibited by larger amounts and the rate of activity by ginseng is constant. 6. The action of ginseng on the ATPase activity was not related to the sulfhydryl group of cysteine, the amino group of lysine, the imidazole group of histidine, the quanidinium group of arginine, the carboxyl group of aspartic acid, or the hydroxyl group of threonine. 7. The activating effect of ginseng on the ATPase activity may be not due to a saponin which is contained in ginseng.

콩 뿌리조직에서의 $Ca^{2+}$에 의한 마이크로솜 이온펌프 활성저해 ($Ca^{2+}-induced$ Inhibition of Microsomal ATPases in Soybean Roots)

  • 콩 뿌리조직의 이온 흡수와 관련된 생리활성을 조사하기 위하여 뿌리조직으로부터 마이크로솜을 분리하였고, 마이크로솜 ATPase (이온점프) 활성을 분광학적 방법인 enzyme-coupled 분석방법에 따라 측정하였다. 마이크로솜 ATPase의 활성에 미치는 여러 가지 이온의 효과 또는 ATPase의 이온선택성을 조사하기 위하여 $10mM\;Na^+$$120mM\;K^+$을 포함하는 대조용액에서의 평균활성을 측정한 결과 190 nmol/min/mg protein으로 나타났다. 대조활성에 비하여 $Na^+$을 포함하지 않은 $130mM\;K^+$ 용액에서는 활성이 150%로 증가하였고, $K^+$을 포함하지 않은 $130mM\;Na^+$ 용액에서는 활성이 63%로 감소되었다. 반응용액의 $K^+$ 농도에 따른 활성변화를 측정한 결과, ATPase의 활성은 외부용액의 $K^+$ 농도 증가에 따라 활성이 증가됨을 알 수 있었다. 또한 마이크로솜 ATPase 활성은 반응용액의 pH 감소에 따라 증가되어 $pH\;6{\sim}7$에서는 비교적 높은 활성을 보였으나, pH 8 이상에서는 급격히 활성이 감소되었고, pH 9에서는 80%이상의 활성이 저해되었다. $Ca^{2+}$에 의한 이온펌프의 활성조절 여부를 평가하기 위해서 마이크로솜 내부 및 외부의 $Ca^{2+}$에 의한 ATPase 활성변화를 측정하였다. 마이크로솜 ATPase의 활성은 반응액의 $Ca^{2+}$ 농도가 낮아질수록 증가하여 $10^{-9}M$ 이하에서 최대활성이 관측되었고, $Ca^{2+}$ 농도가 증가할수록 활성은 감소하여 $500\;{\mu}M$ 전후에서 50%의 활성이 감소하였다. 또한 ATPase의 활성은 마이크로솜 내부의 $Ca^{2+}$ 농도증가에 의해서 저해되어, $Ca^{2+}\;ionophore\;A23187$처리에 의한 외부의 $Ca^{2+}$ 유입에 의해서 약30%의 활성감소를 보였으며, EGTA 처리에 의한 $Ca^{2+}\;chelation$에 의해서 마이크로솜 내부의 $Ca^{2+}$ 농도가 감소되었을 때, ATPase 활성은 증가하였다. 위의 조건에서 실제 마이크로솜 내부로의 $Ca^{2+}$ 유입 여부는 $‘Ca^{2+}’$를 이용하여 확인하였다. 이상의 결과는 마이크로솜 막에 위치한 ATPase의 내부 및 외부에 $Ca^{2+}$에 의한 효소활성 조절부위가 각각존재함을 시사한다.

가토 신피질 절편에서의 K-pNPPase활성에 대한 Ouabain 및 Vanadate의 영향 (Effects of Ouabain and Vanadate on K-pNPPase Activity in Rabbit Renal Cortical Slices)

  • Na-K-ATPase의 일부로 알려져 있으며 intact cell에서도 쉽게 활성의 측정이 가능한 K-pNPPase활성을 신피질 절편에서 측정하여 ouabain에 영향을 받는 산소 소모량 및 세포내 전해질 함량, 그리고 microsome에서 측정한 Na-K-ATPase활성에 대한 ouabain 및 vanadate의 효과를 비교 관찰하여 다음과 같은 결과를 얻었다. 1) 신피질 절편에서 p-NPPase활성은 60분간 시간에 직선적으로 비례하여 증가하였고 이중 1 mM ouabain에 의해 억제되는 K-pNPPase활성은 악 55% 정도로서 전 incubation기간 중 일정한 값을 보였다. 2) K-pNPPase활성을 50% 억제하는 ouabain 및 vanadate의 농도는 각각 $7.9{\times}10^{-6}M$$1.3{\times}10^{-5}M$이었다. 3) 신피질 절편에서 측정한 전체 산소 소모량 중 1 mM ouabain에 의해 억제되는 부분(ouabain-sensitive fraction)에 대한 ouabain과 vanadate의 50% 억제 농도는 각각 $6.3{\times}10^{-6}M$$2.5{\times}10^{-5}M$로서 K-pNPPase에서의 값과 유사하였다. 4) Ouabain 및 vanadate에 의한 세포내 $Na^+$$K^+$함량의 변화 양상은 절편에서 측정한 K-pNPPase활성의 변화 양상과 유사하였다. 5) Microsome에서 측정한 Na-K-ATPase활성은 $10^{-3}M$의 ouabain이 나 vanadate에 의해 완전 억제되었으며 50% 억제 농도는 각각 $1.2{\times}10^{-6}M$$1.6{\times}10^{-6}M$로 1 mM ouabain에 영향을 받는 K-pNPPase 활성 및 산소 소모량을 50% 억제하는 농도보다 ouabain은 약 5배 vanadate는 약 15배 정도 낮았다. 이상의 결과로 미루어 가토 신피질 절편에서 측정한 K-pNPPase활성은 $Na^+-K^+$교환 점프의 활성을 나타내는 좋은 지표가 될 수 있으며 특히 세포막을 통한 물질 이동이나 세포의 기능에 대한 영향과 Na-K-ATPase활성에 대한 영향을 동시에 보고자 할 때는 microsome에서 측정한 Na-K-ATPase활성보다 더 이상적이라고 사료된다.

Aloe vera의 연 유도성 신경계 독성 저감 기전에 관한 연구 (A study on the mechanism for reduction of lead-induced toxicity in nervous system by aloe vera)

  • Effects of water extract of aloe vera on lead-induced neurotoxicity were investigated in sciatic nerve isolated from rat. The mechanism on toxicity reduction by measuring activities of axonal enzymes, metabolism of myo-inositol in nerve, lead concentration in several organs and so on were further examimed. In the lead-treated rats, the transport rate of axonal enzyme, such as acetyl cholinesterase and choline acetyltransferase, was reduced by from 50% to 30% respectively. Reduction in myo-inositol concentration and $Na^+/K^+$ ATPase activity were also observed in sciatic nerve from lead-treated rat. However, the aloe extract administration significantly eliminated the impairment and maintained myo-inositol concentration to about 85% of normal level. Also aloe extract promoted the excretion rate of lead which is accumulated in blood, sciatic nerve and kidney. These results suggest that lead-induced neurotoxicity was significantly reduced by administration of aloe extract and the mechanism might be partly increase in kidney excretion rate of lead and parity normalization of $Na^+/K^+$ ATPase activity which is critical factor in order to keep nerve maintaining normal myo-inositol level.

식이 $\omega$ 3 지방산이 쥐의 신장 인지질과 Na-K-ATPase 활성에 미치는 영향 (Effect of Dietary $\omega$3 Fatty Acid on Kidney Phospholipid and Na-K-ATPase)

  • 식이 $\omega$3 지방산이 신장의 지질구성 및 기능에 미치는 영향을 조사하기 위해 젖뗀 55-60 g의 흰쥐에게 고등어유, 들깨유, 콩기름을식이의 10% (W/W)포함하는 먹이를 조제하여 24일간 섭취시켰다. 고등어유군의 신장 인지질 지방산에 C20~C22 $\omega$ 3 지방산의 함량이 월등히 많은 반면 들깨유군은 콩기름군에 비해 약간 높을 뿐이어서 C18 : 3 ($\omega$3) 지방산으로 단기간에 쉽게 전환되지 않음을 보였다. C18 : 2($\omega$6) 지방산은 고등어군만 낮았고 C20 : 4($\omega$6)는 들깨유군에서는 낮았지만 고등어유군은 콩기름군과 같은 수준이었다. 또한 고등어유군에서 C18 : 1($\omega$9)을 포함한 단일 불포화 지방산이 높고 포화지방산 함량이 낮은 것도 특징이었다 . 인지질 종류별 분석결과에서 고등어유군이 sphingomyelin 이 높고 phosphatidylethanol-amine이 낮았다. 반면 신장 Cholest대기 함량은 고등어유군이 높았으며, 이러한 지질구성 변화는 신장 Na-K-ATPase 활성감소시킨 것이다.
