Production of Xylooligo-Saccharides and Purification of Extracellular Xylanase from Streptomyces chibaensis J-59 (방선균 Streptomyces chibaensis J-59 Xylanase의 정제 및 자일로 올리고당(Xylooligo-Saccharides)의 생산)

  • S. chibaensis J-59 produced an extracellular xylanase in a CSL medium composed of 1.5% com steep liquor, 0.1% $MgSO_4{\cdot}7H_2O$, 0.012% $CoCl_2{\cdot}6H_2O$, and 0.15% glucose containing xylan. but it did not produce in the culture medium containing xylose. The production of enzyme reached to a maximum level (0.83 uints/ml) when bacteria were cultured in 2.5 l jar fermentor for 48hrs at $30^{\circ}C$ and pH 7.0. Furthermore, S. chibaensis J-59 produced an intracellular glucose isomerase in a medium containing xylan and/or xylose. Xylanase was purified 29-fold over the culture supernatants of S. chibaensis J-59 by ammonium sulfate fractionation, chromatography on DEAE-Sephadex A-50, and gel filtration on Sephadex G-200. The purified enzyme is a monomeric enzyme with a native molecular mass of 25 kDa and a subunit molecular mass of 25 kDa. The purified enzyme requires $Mg^{2+}$ for activity, $Ca^{2+}$, $Co^{2+}$ is not an inhibitor but inhibit by $Fe^{3+}$, $Hg^{2+}$, and $Cu^{2+}$, sodium dodecyl sulfate, N-bromosuccinide. Pattern of hydrolysis demonstrated that the xylanase was an endo-splitting enzyme able to break down birchwood xylan at random giving xylobiose, xylotriose and xylotetrose as the main end products.

Studies on the Heavy Metals in Paddy Rice and Soils in Jang-hang Smelter (제연소인근지역(製鍊所隣近地域)의 토양(土壤) 및 수도체중(水稻體中) 중금속함량(重金屬含量)에 관(關)한 조사연구(調査硏究))

  • Soils and rice plants subject to smoke from the Jang-hang Smelter were samples at two depths to assess the nature and extend of Cd, Cu, Pb, and Zn contamination, particularly with respect to distance from pollution source. Soils at east site within 1km of the center of the smelter were highly contaminated with Cd, Cu, Pb, and Zn. Metal levels in the smoke-polluted area were higher in surface soil than in subsurface soil, and decreased rapidly with distance reaching nearly background levels at 5km from pollution source. A position correlation was found between contents of total and 0.1 N HCl-extractable, or 1.0N $CH_3COONH_4$- extractable heavy metals in surface soils of pollution. Contents of heavy metals in soils were positively correlated with soil organic matter and cation exchange capacity. The contents of heavy metals in brown rice collected from the paddy field around the Jang-hang Smelter were in the range of 0.23-1.33 ppm for Cd, 2.39-6.25 for Cu, 0.95-8.32 ppm for Pb and 14.60-27.31 for Zn. The contents of Cd, Cu, and Zn in rice straw were positively correlated with those in the brown rice. Negative correlation was found between the contents of Cd, Cu, Pb, and Zn in brown rice and the distance from the source.

Nitrogen Removal Characteristics in DynaFlow Biofilter System Using Sewage Wastewater of Low C/N Ratio (낮은 C/N비에서 운영되는 유로변경식 생물여과 공정의 질소 제거 특성)

  • In this study, a 3-stage biological aerated filter (BAF) system was proposed to enhance nitrogen removal in the treatment of low carbon to nitrogen ratio (C/N ratio) municipal wastewater. Laboratory experiments were conducted to evaluate the effects of dynamic-flow at the HRT of 6 h. Results of the long-term operation of 3-stage BAF systems showed that the dynamic-flow enabled the total nitrogen removal (T-N) removal efficiency of the system to be about 7 % higher than that of non-dynamic-flow system in treating domestic wastewater due to the more efficient use of organic substrates. The overall $NH_4$-N removal performance was stable during the operational period due to the unique system configuration where independent nitrification occurred. It was concluded that the 3-stage BAF system proposed in this study provided excellent performance in the removal of nitrogen by employing dynamic-flow and three columns functioning as sorption, denitrification and nitrification, respectively.

Effects of Rice Straw Particle Size on Chewing Activity, Feed Intake, Rumen Fermentation and Digestion in Goats

  • Effects of particle size and physical effective fibre (peNDF) of rice straw in diets on chewing activities, feed intake, flow, site and extent of digestion and rumen fermentation in goats were investigated. A 4${\times}$4 Latin square design was employed using 4 mature Liuyang black goats fitted with permanent ruminal, duodenal, and terminal ileal fistulae. During each of the 4 periods, goats were offered 1 of 4 diets that were similar in nutritional content but varied in particle sizes and peNDF through alteration of the theoretical cut length of rice straw (10, 20, 40, and 80 mm, respectively). Dietary peNDF contents were determined using a sieve for particle separation above 8 mm, and were 17.4, 20.9, 22.5 and 25.4%, respectively. Results showed that increasing the particle size and peNDF significantly (p<0.05) increased the time spent on rumination and chewing activities, duodenal starch digestibility and ruminal pH, and decreased ruminal starch digestibility and $NH_{3}$-N concentration. Intake and total tract digestibility of nutrients (i.e. dry matter, organic matter, and starch) and ruminal fermentation were not affected by the dietary particle size and peNDF. Increased particle size and peNDF did not affect ruminal fibre digestibility, but had a great impact on the intestinal and total tract fibre digestibility. The study suggested that rice straw particle size or dietary peNDF was the important influential factor for chewing activity, intestinal fibre and starch digestibility, and ruminal pH, but had minimal impact on feed intake, duodenal and ileal flow, ruminal and total tract digestibility, and ruminal fermentation.

Characterization of Plant Growth-Promoting Traits of Free-Living Diazotrophic Bacteria and Their Inoculation Effects on Growth and Nitrogen Uptake of Crop Plants

  • The search for diverse plant growth-promoting (PGP) diazotrophic bacteria is gaining momentum as efforts are made to exploit them as biofertilizers for various economically important crops. In the present study, 17 diazotrophic strains belonging to eight different genera isolated from rice paddy fields were screened for multiple PGP traits and evaluated for their inoculation effects on canola and rice plants. All of the strains tested positive for 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity and production of indole 3-acetic acid (IAA) and ammonia ($NH_3$). Additionally, four of the strains were able to solubilize phosphorus (P), five tested positive for zinc (Zn) solubilization and sulfur (S) oxidation, and eight strains produced siderophores. Based on the presence of multiple PGP traits, 10 strains were selected for inoculation studies. Treatment with Herbaspirillum sp. RFNB26 resulted in maximum root length (54.3%), seedling vigor, and dry biomass in canola, whereas Paenibacillus sp. RFNB4 exhibited the lowest activity under gnotobiotic conditions. However, under pot culture conditions, Paenibacillus sp. RFNB4 significantly increased plant height and dry biomass production by 42.3% and 29.5%, respectively. Canola plants and rhizosphere soils inoculated with Bacillus sp. RFNB6 exhibited significantly higher nitrogenase activity. In greenhouse experiments, Serratia sp. RFNB18 increased rice plant height by 35.1%, Xanthomonas sp. RFNB24 enhanced biomass production by 84.6%, and rice rhizosphere soils inoculated with Herbaspirillum sp. RFNB26 exhibited the highest nitrogenase activity. Our findings indicate that most of the selected strains possess multiple PGP properties that significantly improve the growth parameters of the two plants when tested under controlled conditions.

Processing of Low Salt Fermented Sauce of Shellfish with Citric Acid Pretreatment (구연산 전처리에 의한 개량조개의 저염젓갈가공)

  • In order to develop low salt fermented hen clam sauce with high stability, the effects of citric acid pretreatment and fermentation temperature were examined. The VBN contents of surf clam sauce treated with citric acid (SCA) were lower than those of control and the same sauces added alcohol at two temperature conditions ($5^{\circ}C$ and room temperature) during fermentation. The maximum $NH_2-N$ contents of control and SCA during fermentation at $5^{\circ}C$ were 501.3 and $618.4{\sim}691.6\;mg/100g$, respectively, and the pH of those showed $5.61{\sim}6.24$ and $2.43{\sim}3.21$. The total creatine contents of control and SCA, respectively, were $36.8{\sim}27.6\;mg/100g$ ranges. As the time of treatment with citric acid was longer, the degradation of ATP, ADP and AMP in the SCA was faster. In the control, the Ala content was $19.6{\sim}23.02%$ and was highest level among all free amino acids. As fermenting term was longer, among the free amino acids, Ala and Gly were large amounts in control and SCA, respectively, and Gly was slowly increased in SCA during fermentation. As the results of organoleptic test, the tastes of SCA showed good score than control.

  • In the fermentation for preparation of petroprotein by Rhodotorula sp. in $C_{12}{\sim}C_{14}$n-alkane mixture it was investigated how the effects due to the difference of chemical form of the nitrogen sources are, that is, how the inorganic nitrogen sources such as nitrates and ammonium salts and organic nitrogen source such as urea effect on the view of fermentation time and yield and how the ratios of nitrogen to carbon with $NaNo_3$ effect. Then following results were obtained: the time required to maximum growth, when $NaNo_3$ or $(NH_4)_2SO_4$ was used as nitrogen source, was 40 hrs. and 45 hrs., respectively, but when urea was used, it was 66 hrs. much longer than above nitrogen sources. On the view of yield, however, in use of the both inorganic sources, when the yield is represented as consumption of 0.1 N-NaOH, it was 0.36 and 0.38 ml, respectively, but, in the case of urea, it amounted to 0.78 ml. In the effect of the ratios of nitrogen to carbon in medium, when n-alkane mixture was added in 1 % (vol.) and N/C with $NaNo_3$ was 0.2 the best results were obtained and generally the higher the value of the ratio the better growth effects were shown.

  • Okamoto cavity was modified to generate high power (2.45 GHz, 2 kW) He, N2 and Ar plasmas with WR-340 waveguide. Many factors which influence to the plasma generation were optimized and investigated for the spectroscopic properties of the He plasma generated. Some of the important factors are the diameter of the inner conductor, the distance between the inner and outer conductors and the distance between the tip of the inner conductor and the torch. After optimization for the He, two torches (a commercial mini torch for ICP and a tangential flow torch made locally) were compared and showed similar results for the helium plasma gas flow of 25 L/min~30 L/min. A tall torch (extended) was used to block the air in-flow and reduced the background intensity at 340 nm region (NH band). Emission intensity was measured for determination of halogen element in the aqueous solution with power and carrier gas flow rate. Electron number density and the excitation temperature were on the order of $3.67{\times}10^{11}/cm^3$ and 4,350 K, respectively. These values are similar or a bit smaller than other microwave plasmas. It has been possible to analyze aqueous samples. The detection limit for Cl (479.45 nm) was obtained to be 116 mg/L and needs analytical optimization for the better performance.

Case Study of the Heavy Asian Dust Observed in Late February 2015 (2015년 2월 관측된 고농도 황사 사례 연구)

  • Asian dust is a seasonal meteorological phenomenon influencing most East Asia, irregularly occurring during spring. Unusual heavy Asian dust event in winter was observed in Seoul, Korea, with up to $1,044{\mu}g\;m^{-3}$ of hourly mean $PM_{10}$, in 22~23 February 2015. Causes of such infrequent event has been studied using both ground based and spaceborne observations, as well as numerical simulations including ECMWF ERA Interim reanalysis, NOAA HYSPLIT backward trajectory analysis, and ADAM2-Haze simulation. Analysis showed that southern Mongolia and northern China, one of the areas for dust origins, had been warm and dry condition, i.e. no snow depth, soil temperature of ${\sim}0^{\circ}C$, and cumulative rainfall of 1 mm in February, along with strong surface winds higher than critical wind speed of $6{\sim}7.5m\;s^{-1}$ during 20~21 February. While Jurihe, China, ($42^{\circ}23^{\prime}56^{{\prime}{\prime}}N$, $112^{\circ}53^{\prime}58^{{\prime}{\prime}}E$) experienced $9,308{\mu}g\;m^{-3}$ of hourly mean surface $PM_{10}$ during the period, the Asian dust had affected the Korean Peninsula within 24 hours traveling through strong north-westerly wind at ~2 km altitude. KMA issued Asian dust alert from 1100 KST on 22nd to 2200 KST on 23rd since above $400{\mu}g\;m^{-3}$ of hourly mean surface $PM_{10}$. It is also important to note that, previously to arrival of the Asian dust, the Korean Peninsula was affected by anthropogenic air pollutants ($NO_3^-$, $SO_4^{2-}$, and $NH_4^+$) originated from the megacities and large industrial areas in northeast China. In addition, this study suggests using various data sets from modeling and observations as well as improving predictability of the ADAM2-Haze model itself, in order to more accurately predict the occurrence and impacts of the Asian dust over the Korean peninsula.

CO2 Decomposition Characteristics of Activated(Fe1-xMnx)3O4-δ and (Fe1-xCox)3O4-δ (활성화된(Fe1-xMnx)3O4-δ과 (Fe1-xCox)3O4-δ의 이산화탄소 분해 특성)

  • Activated magnetite ($Fe_3O_{4-{\delta}}$) has the capability of decomposing $CO_2$ proportional to the ${\delta}$-value at comparatively low temperature of $300^{\circ}C$. To enhance the $CO_2$ decomposition capability of $Fe_3O_{4-{\delta}}$, $(Fe_{1-x}Co_x)_3O_{4-{\delta}}$ and $(Fe_{1-x}Mn_x)_3O_{4-{\delta}}$ were synthesized and then reacted with $CO_2$. $Fe_{1-x}Co_xC_2O_4{\cdot}2H_2O$ powders having Fe to Co mixing ratios of 9:1, 8:2, 7:3, 6:4, and 5:5 were synthesized by co-precipitation of $FeSO_4{\cdot}7H_2O$ and $CoSO_4{\cdot}7H_2O$ solutions with a $(NH_4)_2C_2O_4{\cdot}H_2O$ solution. The same method was used to synthesize $Fe_{1-x}Mn_xC_2O_4{\cdot}2H_2O$ powders having Fe to Mn mixing ratios of 9:1, 8:2, 7:3, 6:4, 5:5 with a $MnSO_4{\cdot}4H_2O$ solution. The thermal decomposition of synthesized $Fe_{1-x}Co_xC_2O_4{\cdot}2H_2O$ and $Fe_{1-x}Mn_xC_2O_4{\cdot}2H_2O$ was analyzed in an Ar atmosphere with TG/DTA. The synthesized powders were heat-treated for 3 hours in an Ar atmosphere at $450^{\circ}C$ to produce activated powders of $(Fe_{1-x}Co_x)_3O_{4-{\delta}}$ and $(Fe_{1-x}Mn_x)_3O_{4-{\delta}}$. The activated powders were reacted with a mixed gas (Ar : 85 %, $CO_2$ : 15 %) at $300^{\circ}C$ for 12 hours. The exhaust gas was analyzed for $CO_2$ with a $CO_2$ gas analyzer. The decomposition of $CO_2$ was estimated by measuring $CO_2$ content in the exhaust gas after the reaction with $CO_2$. For $(Fe_{1-x}Mn_x)_3O_{4-{\delta}}$, the amount of $Mn^{2+}$ oxidized to $Mn^{3+}$ increased as x increased. The ${\delta}$ value and $CO_2$ decomposition efficiency decreased as x increased. When the ${\delta}$ value was below 0.641, $CO_2$ was not decomposed. For $(Fe_{1-x}Co_x)_3O_{4-{\delta}}$, the ${\delta}$ value and $CO_2$ decomposition efficiency increased as x increased. At a ${\delta}$ value of 0.857, an active state was maintained even after 12 hours of reaction and the amount of decomposed $CO_2$ was $52.844cm^3$ per 1 g of $(Fe_{0.5}Co_{0.5})_3O_{4-{\delta}}$.
