Anticariogenic activity of acanthoic acid isolated from Acanthopanax koreanum $N_{AKAI}$ against oral pathogens (섬오가피로부터 분리된 acanthoic acid의 항치아우식효과)

  • This research was aimed to search for natural antimicrobial agents that are sefe for humans and specific for oral pathogens. Acanthoic was isolated from the chloroform fraction of methanol extract of Acanthopanax koreanum $N_{AKAI}$ and its structure were elucidated by 13-NMR, 1H-NNR and ESIMS. Antibacterial activity of acanthoic acid was investigated by the minimum inhibitory (MIC) and minimun bactericidal (MBC) concentration. MIC/MBC of acanthoic acid against Streptococcus mutans $N_{AKAI}$ causing dental caries was determined to be $2/4\;{\mu}g/mL$, which was much lower than these of other natural antimicrobial agents such as $8/16\;{\mu}g/mL$ of sangurinarine and $250/500\;{\mu}/mL$ of green tea extract, $500/600\;{\mu}g/mL$ of thymol and borneol. Acanthoic and significantly inhibited the growth of other cariogenic bacteria such as Streptococcus sobrinus $N_{AKAI}$ and Streptococcus sanguis $N_{AKAI}$, and Streptococcus gordonii $N_{AKAI}$ in the MIC range of $4{\sim}32\;{\mu}g/mL$. My finding suggests that acanthoic acid could be employed as a potential antibacterial agent for preventing dental caries.

Determination of new antifouling agents in seacoasts in Korea by gas chromatography-mass spectrometry (GC/MS를 이용한 한국연안의 새로운 방오제 분석)

  • Antifouling agents including tributyltin (TBT) compound and its derivatives have been used for many years, but the usage of TBT in Korea was legally restricted in 2003, due to its significant environmental impact. Following this, many new alternative antifouling agents have been used. In this experiment, four major antifouling agents were selectively analyzed to study their release in seawater and tidal flats on the Korean Peninsula. These new antifouling agents were extracted from the seawater and tidal flats using a liquid-liquid extraction method and microwave extraction, respectively. The measured concentrations of Irgarol 1051, Sea-Nine 211, Dichlofluanid and Chlorothalonil ranged from N.D.$-23.80ng/{\ell}$, N.D.$-15.30ng/{\ell}$, N.D.$-61.69ng/{\ell}$ and N.D.$-4.19ng/{\ell}$ in the seawater samples and from N.D.-159.45 ng/g, N.D.-476.57 ng/g, N.D.-59.79 ng/g and N.D.-21.27 ng/g in the tidal flat samples, respectively. Interestingly, these new antifouling agents were not detected in any area in the tidal flats at Pusan, whereas a certain amount of them was found in the seawater.

Effect of Crop Rotation on the Growth of Sesame(Sesamum indicum L.) and Soil Properties (윤작이 참깨의 생육과 토양의 이화학성에 미치는 영향)

  • The objective of this study was to investigate the effect of sesame monoculture and sesame in rotation (SR) with maize. soybean. barley or rye on reduction of injury by continous cropping in sesame. Field studies were conducted for 3-years (1995~1997) at the experimental field of the Crop Experiment Station. Compared to CS (continuous sesame). SR treatments turned out to keep more organic matter and higher exchangeable cation concentrations in soils. BS (barley and sesame in a 1-year rotation) and RS (rye and sesame in a 1-yea r rotation) treatments had significantly greater available $P_2O_5$ contents in soils than CS, MS (maize and sesame in a 3-year rotation) and SbS (soybean a nd sesame in a 3-year rotation). The pH of the soils under different treatments were not significantly different. SR treatments exhibited significantly lower bulk density and higher pore space than CS. Soil microbial biomass C (SMBC) and N (SMBN) were determined by the chloroform fumigation-extraction method. SMBC and SMBN were significantly higher in soils under BS and RS than those under CS, but only during the 1 year of monitoring. MS and SbS treatments resulted in higher SMBC and SMBN than CS. The occurrence of injury by disease of sesame is the important primary factor of injury by continous cropping, but the disease occurrence with rotation did not decrease in th is experiment. Under CS treatment, the growth and grain of sesame was significantly lower than those under other treatments. Compared to CS, the increments of grain yield of sesame were 68, 63, 57 and 51% for MS, RS, SbS and BS, respectively in the first harvest. In the second harvest, they were 24% for MS, 28% for RS, 20% for SbS and 19% for BS. The average increase ratios during the two years were 41, 41, 34, and 33% for MS, RS, SbS and BS, respectively.

  • Validamycin A is an aminoglycoside fungicide produced by Streptomyces hygroscopicus that inhibits trehalase. The purpose of this study was to develop a method for detecting validamycin A in agricultural samples to establish MRL values for use in Korea. The validamycin A residues in samples were extracted using methanol/water (50/50, v/v) and purified with a hydrophilic-lipophilic balance (HLB) cartridges. The analyte was quantified and confirmed by liquid chromatograph-tandem mass spectrometer (LC-MS/MS) in positive ion mode using multiple reaction monitoring (MRM). Matrix-matched calibration curves were linear over the calibration ranges (0.005~0.5 ng) into a blank extract with $R^2$ > 0.99. The limits of detection and quantification were 0.005 and 0.01 mg/kg, respectively. For validation validamycin A, recovery studies were carried out three different concentration levels (LOQ, $LOQ{\times}10$, $LOQ{\times}50$, n = 5) with five replicates at each level. The average recovery range was from 72.5~118.3%, with relative standard deviation (RSD) less than 10.3%. All values were consistent with the criteria ranges requested in the Codex guidelines (CAC/GL 40-1993, 2003) and the NIFDS (National Institute of Food and Drug Safety) guideline (2016). Therefore, the proposed analytical method is accurate, effective and sensitive for validamycin A determination in agricultural commodities.

  • This study was to test whether in vitro matured Hanwoo oocytes can be successfully cryopreserved by a new vitrification procedure using MVC method. For the vitrification, oocytes were pretreated in 10% ethylene glycol (EG10) for 5~10 min, exposed in EG30 for 30 sec, each oocyte was individually put on the inner wall of 0.25 $m\ell$ straw, and then straws were directly plunged into L$N_2$. Thawing was taken by 4-step procedures 〔1.0 M sucrose (MS), 0.5 MS, 0.25 MS, and 0.125 MS〕 at 37$^{\circ}C$. In vitro developmental capacity (survival, cleavage ($\geq$2-cell) and blastocyst rates) in vitrified group was no significant difference compared to that in other treatment groups (exposed; 100.0, 74.4, 32.3% and control; 100.0, 78.3, 36.3%): high mean percentage of oocytes (91.2%) was survived, 69.4% of them were cleaved and 27.9% of cleaved embryos were developed to blastocyst. Especially, after transfer of in vitro developed embryos in vitrified group, four of six recipient animals were pregnant and three of them were ongoing-pregnant by manual palpation at 250 days after transfer. This result demonstrates that MVC method is very appropriate freezing method for the Hanwoo in vitro matured oocytes and that ovum bank can be maintained efficiently by MVC cryopreservation method.

Isolation of Antifungal Compound and Biocontrol Potential of Lysobacter antibioticus HS124 against Fusarium Crown Rot of Wheat

  • Fusarium graminearum is the main cause of substantial economic loss in wheat production. The aim of this study is to investigate biocontrol potential of Lysobacter antibioticus HS124 against F. graminearum and to purify an antifungal compound. In preliminary study, n-butanol crude extract revealed destructive alterations in the hyphal morphology of F. graminearum and almost degraded with $1,000{\mu}g\;mL^{-1}$ concentration. For further study, the antifungal compound extracted from the n-butanol crude extract of L. antibioticus HS124 was identified as N-Butyl-tetrahydro-5-oxofuran-2-carboxamide ($C_9H_{16}NO_3$) using NMR ($^1H-NMR$, $^{13}C-NMR$, $^1H-^1H\;COSY$, HMBC, and HMQC), and HR-ESI-MS analysis. To our knowledge, N-Butyl-tetrahydro-5-oxofuran-2-carboxamide may be a novel compound with molecular weight of 186.1130. The minimum inhibitory concentration value of antifungal compound was $62.5{\mu}g\;mL^{-1}$ against F. graminearum. In an in vivo pot experiment, crown rot disease from F. graminearum was inhibited when wheat seeds were treated with both HS124 culture and F. graminearum. Growth of wheat seedling was enhanced by treatment of HS124 compared to control. Our results suggest that L. antibioticus HS124 characterized in this study could be successfully used to control F. graminearum and could be used as an alternative to chemical fungicides in modern agriculture.

Identification of Flavor Components in Korean Ordinary Kochujang Inoculated with Bacillus sp. and Saccharomyces sp. (Saccharomyces 속(屬) 및 Bacillus 속(屬)을 접종(接種)한 한국재래식(韓國在來式)고추장의 향기성분동정(香氣成分同定))

  • Kochujang (Red pepper paste) is one of a traditional fermented seasoning foods having peculia. flavor. In present paper, the effects of microorganisms on the formation of volatile components of Kochujang during fermentation were observed. The volatiles of Kochujang were extracted by a steam-distillation under the reduced pressure and determined by gas chromatography and gas chromatography-mass spectrometry (CC-MS), respectively. During fermentation of Kochujang, Saccharomyces sp. and three strains of Bacillus sp. were isolated as main microorganisms participating in the formation of volatile components. According to the analytical results, tetrahydro geroniol, furfuryl alcohol, methoxy acecoptenone and myrtanal in Kochujang inoculated with Saccharomyces spl, 2-methyl-propan-2-ol, furfuryl alcohol an furfuryl-n-butyrate in Kochujang inoculated with Bacillus sp. B-1, 2-methyl-propan-2-ol, furfuryl alcohol and furfuryl-n-butyrate in Kochujang inoculated with Bacillus sp. B-2, and 2-methyl-propan-2-ol, furfuryl alcohol and n-propylbenzene in Kochujang inculated with Bacillus sp. B-3, were identified as major components, respectively.

Effects of Workplace Nutrition Education Program Tailored for the Individual Chronic Disease Risks (직장인의 만성질환 위험 요소별로 구성된 영양 교육의 효과 비교 연구)

  • The incidence of chronic disease is continuously increasing in Korea. Especially, office workers have higher risk of chronic disease because of their dietary habit and lifestyle. The study aimed to investigate the effect of tailored nutrition counseling on improving chronic disease risk factors. Ninety-nine male workers (age $46.9{\pm}7.0$ yrs) volunteered for 12 weeks of nutrition program containing dietary intake and physical activity adjustment. Five individualized programs were performed with the main theme of weight loss (WL, n = 16), blood pressure lowering (BL, n = 34), normalizing blood glucose (GL, n = 21), lipid lowering (LL, n = 13) and reducing MS risk factors (ML, n = 15). Anthropometric data, blood-pressure, self-reported questionnaire, blood profiles were measured before and after 12weeks of nutrition education. The education program included 5 times of 1:1 interview. Compared to 0 week, anthropometric data (weight, BMI, fat, visceral fat, waist, SBP, DBP) were significantly decreased after 12 weeks (p < 0.001). Fasting blood glucose and total cholesterol were decreased (p < 0.05). The primary outcomes with individually tailored programs showed to be more effective than one general nutrition program.

RIE에서 $C_3F_6$ 가스를 이용한 $Si_3N_4$ 식각공정 개발

  • $SF_6$ gas는 반도체 및 디스플레이 제조공정 중 Dry etch과정에서 널리 사용되는 gas로 자연적으로 존재하는 것이 아닌 사용 목적에 맞춰 인위적으로 제조된 gas이다. 디스플레이 산업에서 $SF_6$ gas가 사용되는 Dry etch 공정은 주로 ${\alpha}$-Si, $Si_3N_4$ 등 Si계열의 박막을 etch하는데 사용된다. 이러한 Si 계열의 박막을 식각하기 위해서는 fluorine, Chlorine 등이 사용된다. fluorine계열의 gas로는 $SF_6$ gas가 대표적이다. 하지만 $SF_6$ gas는 대표적인 온실가스로 지구 온난화의 주범으로 주목받고 있다. 세계적으로 온실가스의 규제에 대한 움직임이 활발하고, 대한민국은 2020년까지 온실가스 감축목표를 '배출전망치(BAU)대비 30% 감축으로' 발표하였다. 따라서 디스플레이 및 반도체 공정에는 GWP (Global warming Potential)에 적용 가능한 대체 가스의 연구가 필요한 상황이다. 온실가스인 $SF_6$를 대체하기 위한 방법으로 GWP가 낮은 $C_3F_6$가스를 이용하여 $Si_3N_4$를 Dry etching 방법인 RIE (Reactive Ion Etching)공정을 한 후 배출되는 가스를 측정하였다. 4인치 P-type 웨이퍼 위에 PECVD (Plasma Enhanced Chemical Vapor Deposition)장비를 이용하여 $Si_3N_4$를 200 nm 증착하였고, Photolithography공정을 통해 Patterning을 한 후 RIE공정을 수행하였다. RIE는 Power : 300 W, Flow rate : 30 sccm, Time : 15 min, Temperature : $15^{\circ}C$, Pressure : Open과 같은 조건으로 공정을 수행하였다. 그리고 SEM (Scanning Electron Microscope)장비를 이용하여 Etching된 단면을 관찰하여 단차를 확인하였다. 또한 Etching 전후 배출가스를 포집하여 GC-MS (Gas Chromatograph-Mass Spectrophotometry)를 측정 및 비교하였다. Etching 전의 경우에는 $N_2$, $O_2$ 등의 가스가 검출되었고, $C_3F_6$ 가스를 이용해 etching 한 후의 경우에는 $C_3F_6$ 계열의 가스가 검출되었다.

Design and Fabrication of 25 W Ka-Band SSPA Based on GaN HPA MMICs (GaN HPA MMIC 기반 Ka 대역 25 W SSPA 설계 및 제작)

  • We designed and manufactured Ka-band SSPA include drive amplifier and high power amplifier MMICs by $0.15{\mu}m$ GaN commercial process. Also, we fabricated main components micro-strip line to WR28 waveguide transition and WR28 wave guide power combiner for Ka-band SSPA. This Ka-band SSPA shows saturated output power 44.2 dBm, power added efficiency 16.6 % and power gain 39.2 dB at 29~31 GHz frequency band.
