Macro- and Micro-nutrient Utilization and Milk Production in Crossbred Dairy Cows Fed Finger Millet (Eleucine coracana) and Rice (Oryza sativa) Straw as Dry Roughage Source

  • Finger millet straw and rice straw are the major source of dry roughage in southern India. They distinctly vary in their morphological and nutritional characters. Hence an effort was made to study the nutrient utilization, milk yield and composition in crossbred dairy cows fed either finger millet (group 1) or rice straw (group 2) as a source of dry roughage. The cows in both the groups were fed as per requirement with concentrate, green fodder and straw in the ratio of 30:45:25 parts (DM). At the end of 50 days of preliminary feeding a digestibility trial was conducted for 7 days and pooled samples of feed, fodder, feces, urine and milk were analysed for macro and micro nutrient content. Finger millet straw contained more CP, Ca, P, Mg, Cu, Zn and Co than rice straw and rice straw contained higher ADF, ash and silica. The intake of DM, CP, EE, NDF, ADF and most micronutrients (Ca, P, Mg, Cu, Zn, Fe, Mn and Co) was significantly higher in cows fed finger millet straw. The digestibility of DM, CP, NDF and ADF was significantly higher in cows fed finger millet straw and the gut absorption of Ca, Cu, Mn and Co was significantly higher in cows fed finger millet straw. The dietary requirement of all micronutrients in both the group of cows could be met irrespective of the type of roughage fed except that of Ca, which was low (0.61 and 0.40%) in rice straw fed cows. The average daily milk yield (L/cow) was also higher (7.0 L) in cows fed finger millet straw as compared to cows fed rice straw (6.3 L). The average milk composition also did not differ except that of milk fat which was significantly (4.7 and 4.5%) low in cows fed rice straw. The overall results of this study have indicated that finger millet straw is a better source of dry fodder than rice straw and while feeding rice straw as the sole roughage to dairy cows there is need to supplement additional calcium as this could be one of the limiting nutrients for milk production.

Enhanced Production of Exopolysaccharides by Fed-batch Culture of Ganoderma resinaceum DG-6556

  • The objectives of this study were to optimize submerged culture conditions of a new fungal isolate, Ganorderma resinaceum, and to enhance the production of bioactive mycelial biomass and exopolysaccharides (EPS) by fed-batch culture. The maximum mycelial growth and EPS production in batch culture were achieved in a medium containing 10 g/l glucose, 8 g/l soy peptone, and 5 mM $MnCl_2$ at an initial pH 6.0 and temperature $31^{\circ}C$. After optimization of culture medium and environmental conditions in batch cultures, a fed-batch culture strategy was employed to enhance production of mycelial biomass and EPS. Five different EPS with molecular weights ranging from 53,000 to 5,257,000 g/mole were obtained from either top or bottom fractions of ethanol precipitate of culture filtrate. A fed-batch culture of G. resinaceum led to enhanced production of both mycelial biomass and EPS. The maximum concentrations of mycelial biomass (42.2 g/l) and EPS (4.6 g/l) were obtained when 50 g/l of glucose was fed at day 6 into an initial 10 g/l of glucose medium. It may be worth attempting with other mushroom fermentation processes for enhanced production of mushroom polysaccharides, particularly those with industrial potential.

Bacillus Subtilis W700에서의 Staphylpkinase 대량생산을 위한 배지 최적화 및 배양방법의 비교 (Media Optimization and Comparison of Fermentation Type for Overproduction of Staphylodinase in Bacillus subtilis WB700)

  • Bacillus subtilis WB700에서 P43 프로모터를 사용하여 staphylokinase를 생산하기 위하여 배지 최적화 및 회분식 배양과 유가식 배양 두가지 시스템을 비교하였다. 여러 가지 질소윈 중에서는 tryptone이 가장 좋은 질소원 이었으며, MSR 배지를 사용한 경우 tryptone 15 g/L일 경우에 최적조건임을 알아내었다. MSR 배지에시 포도당을 제한 기질로 사용할 경우는 5 g/L일 때가 SAK의 발현에 최적 조건이었다. MSR 배지를 기본으로 이용하여 포도당 공급을 조절함으로서 발효조 내의 DO를 30%로 유지한 결과 오히려 MSR 배지를 이용하여 회분식 발효를 한 경우보다 좋지 못한 결과를 얻었으며, 이는 B. subtilis 숙주의 영양요구적 특이성과 P43 promoter의 stress 발생시 주 발현되는 특성 등에 기인한 것이라고 사료된다. MSR 배지를 이용하여 회분식 발효를 하였을 때 SAK 활성은 2880 unit이었고, 이때 배지 내로 분비된 SAK 농도는 455 mg/L이었다.

The Inhibitory Effect of L. plantarum Q180 on Adipocyte Differentiation in 3T3-L1 and Reduction of Adipocyte Size in Mice Fed High-fat Diet

  • In this study, we examined the inhibitory effect of L. plantarum Q180 on adipocyte differentiation in 3T3-L1 and reduction of adipocyte size in mice fed high-fat diet. L. plantarum Q180 inhibited the adipocyte differentiation of 3T3-L1 cells ($18.47{\pm}0.32%$) at a concentration of $400{\mu}g/mL$ ($10^8CFU/g$). As a result of western blot analysis, the expression of $C/EBP{\alpha}$ and $PPAR{\gamma}$ in 3T3-L1 adipocyte treated with $400{\mu}g/mL$ of L. plantarum Q180 decreased 35.16% and 40.07%, respectively, compared with the control. To examine the effects, mice were fed three different diets as follows: ND (n=6) was fed ND and orally administered saline solution; HFD (n=6), HFD and orally administered saline solution; and HFD+Q180 (n=6), HFD and orally administered L. plantarum Q180. After six weeks, the rate of increase of body weight was 13.7% lower in the HFD+Q180 group compared to the HFD group. In addition, the epididymal fat weights of the HFD+Q180 group were lower than that of the HFD group. The change of adipocyte size was measured in diet-induced obese mice. Consequently, the number of large-size adipose tissue was less distributed in the ND and HFD+Q180 groups than in the HFD group. L. plantarum Q180 has an effect on the inhibition of 3T3-L1 adipocyte differentiation, fat absorption and reduction of adipocyte size. L. plantarum Q180 could be applied to functional food products that help improve obesity.

고농도 세포배양에서 Bacillus thuringiensis의 세포 성장과 포자 형성 속도 (Growth Kinetics and Sporulation of Bacillus thuringiensis in High Cell Density Culture)

  • 유가식배양과 세라믹 막을 장착한 생물반응기에서 운전인 TCRC 조업을 통해서 Bacillus thuringiensis의 고농도 세포배양을 실시하였다. 유가식 배양에서 B. thuringiensis의 세포 성장은 선형적으로 증가하였고, 이것은 세포성장 모델리의 결과와 잘 맞았다. 낮은 세포 성장속에도 불구하고 유가식 배양동안에 포자형성은 관찰할 수 없었고, 이것은 연속배양의 결과와는 반대였다. 유가식 조업 후에 회분식 배양으로 바꾸면 300 g/L의 포도당 공급 농도를 사용했을 때 2.7$\times$$10^9$ CFU/mL 의 포자농도를 얻었다. 생물반응기내에 세라믹 막을 장착한 TCRC 조업에서 포도당 공급 농도의 영향을 결정하였다. 50 g/L의 포도당 농도를 사용했을 때 TCRC 조업에서 82.5 g-cell/L에 해당하는 최대 세포농도 1.8$\times$$10^{10}$ CFU/mL 를 얻었다. TCRC에서 세포성장은 선형적으로 증가하였고 포도당 농도는 제한되었는데 이것은 세포성장은 모델링의 결과와 잘 일치하였다. 1 g/L의 포도당을 공급한 경우 이외에는 TCRC 조업 동안에 포자형성을 관찰할 수 없었다. 50 g/L의 포도당을 공급한 경우 TCRC조업 후에 회분식 배양으로 전환시키면 1.2$\times$$10^{10}$ CFU/mL 의 포자농도를 얻었고, 이것은 연구된 여러 배양형태 중에 가장 높은 포자농도이다. 이 때 최적의 포도당 공급속도는 0.55 g glucose/h로 공급하였을 때였다.

  • Xylitol의 생산성을 높이기 위해 two-stage fed-batch를 수행하였다. Glucose가 고갈되어 pH가 5.7에서 올라가면 glucose를 공급하는 방법에서 최종세포의 OD 185.0과 최종 ethanol 농도 1.0 g/L를 얻었다. 산소전달에 대한 xylitol의 생성 영향에서는 통기량 1 vvm에서 500 rpm의 교반속도일 때 xylitol 수율 55.2%와 생산성 $2.19\;g-xylitol/L\;{\cdot}\;h$를 얻었다.

  • 본 논문에서는 Inset-fed 안테나의 급전구조를 변형하여 기존의 Inset-fed 안테나 보다 그 성능을 향상시킨 안테나를 설계 및 제작하였다. Patch 안테나에서 gap 결합에 의한 급전 방식을 기초로 하여 변형된 급전 방식을 Inset-fed 안테나에 적용하였다. 설계된 안테나는 FDTD에 의한 simulation을 수행하여 최적치를 얻었으며 제작된 안테나는 공진 주파수에서 511은 -l4dB이고 cross polarization level은 -20dB의 특성을 보였다.

Comparative study on response of thiocyanate shock load on continuous and fed batch anaerobic-anoxic-aerobic sequential moving bed reactors

  • A comparative study on response of a toxic compound thiocyanate ($SCN^-$) was carried out in continuous and fed batch moving bed reactor systems. Both systems had three sequential anaerobic, anoxic and aerobic reactors and operated at same hydraulic retention time. Feed $SCN^-$ was first increased from 600 mg/L to 1,000 mg/L for 3 days (shock 1) and then from 600 to 1,200 mg/L for 3 days (shock 2). In anaerobic continuous reactor, increase of effluent COD (chemical oxygen demand) due to shock load was only 2%, whereas in fed batch reactor it was 14%. In anoxic fed batch reactor recovery was partial in terms of $SCN^-$, phenol, COD and $NO{_3}{^-}$-N and $NO{_2}{^-}$-N removals and in continuous reactor complete recovery was possible. In both systems, inhibition was more significant on aerobic reactors than anaerobic and anoxic reactors. In aerobic reactors ammonia removal efficiency deteriorated and damage was irreversible. Present study showed that fed batch reactors showed higher substrate removal efficiency than continuous reactors during regular operation, but are more susceptible to toxic feed shock load and in nitrifying reactor damage was irreversible.

Comparative study on growth of leafy vegetables grown in a hybrid BFT-aquaponics using Japanese eel, Anguilla japonica and hydroponics

  • Aquaponics is a cultivation system that combines aquaculture and agricultural hydroponics. This study investigated the productivity of leafy vegetables cultivated in the hybrid biofloc technology-aquaponics (HBFT-AP) using Japanese eel fed two kinds of diets (Mash, commercial powdered feed and extruded pellet [EP]) and hydroponics (HP).The mash was fed to the fish in a type of a dough mixed with water for 6 weeks (Exp1) and switched to the EP containing 2.7% monobasic potassium phosphate (MKP) for subsequent 6 weeks (Exp2). Leafy vegetables of 8 cultivar were employed in the experiment and water quality [dissolved oxygen (DO, mg/L), pH, water temperature (℃), electrical conductivity (EC; ㎲/cm), turbidity (NTU), TAN (NH3 + NH4+) (mg /L), NO2-N (mg/L), NO3-N (mg/L) and PO4-P (mg/L)] was measured 6 times a week. Leafy vegetable productivity (HBFT-AP vs HP) was compared in respective experiment, which was similar or somewhat higher in HBFT-AP. During the 12-week feeding trial, concentrations of nitrite (NO2-N) and phosphorus (PO4-P) were kept low in variability while total ammonia nitrogen (TAN) and NO3-N levels increased with time in HBFT-AP. At the end of two feeding trials, values of weight gain (WG, %), feed efficiency (FE, %), specific growth rate (SGR, %) and protein efficiency ratio (PER) were higher in Exp2 than in Exp1. As well, higher values in hematocrit (PCV, %), plasma K (mEq/L) and inorganic phosphorus (mg/dL) were found (p < 0.05) in Exp2 where fish were fed the EP (EP: 38.60%, 2.80 mEq/L and 7.04 g/dL; Mash: 33.20%, 1.95 mEq/L and 5.50 g/dL). Leafy vegetables in HBFT-AP using Japanese eel fed the EP with MKP 2.7% had a productivity similar (4 kinds of cultivar) or somewhat higher (4 kinds of cultivar) compared to those in HP. Also, Japanese eel fed the EP showed higher values of Pi (m/dL) and K (mEq/L) in plasma compared to those fed commercial powder diet.

Rhamnolipid Production in Batch and Fed-batch Fermentation Using Pseudomonas aeruginosa BYK-2 KCTC 18012P

  • The optimization of culture conditions for the bacterium Pseudomonas aeruginosa BYK-2 KCTC 18012P, was performed to increase its rhamnolipid production. The optimum level for carbon, nitrogen sources, temperature and pH, for rhamnolipid production in a flask, were identified as 25 g/L fish oil, 0.01% (w/v) urea, 25 and pH 7.0, respectively. Optimum conditions for batch culture, using a 7-L jar fermentor, were 200 rpm of agitation speed and a 2.0 L/min aeration rate. Under the optimum conditions, on fish oil for 216 h, the final cell and rhamnolipid concentrations were 5.3 g/L and 17.0 g/L respectively. Fed-batch fermentation, with different feeding conditions, was carried out in order to increase, cell growth and rhamnolipid production by the Pseudomonas aeruginosa, BYK-2 KCTC 18012P. When 2.5 g of fish oil and 100 mL basal salts medium, containing 0.01 % (w/v) urea, were fed intermittently during the fermentation, the final cell and rhamnolipid concentrations at 264 h, were 6.1 and 22.7 g/L respectively. The fed-batch culture resulted in a 1.2-fold increase in the dry cell mass and a 1.3-fold increase in rhamnolipid production, compared to the production of the batch culture. The rhamnolipid production-substrate conversion factor (0.75 g/g) was higher than that of the batch culture (0.68 g/g).
