Molecular Species of Triglycerides in Sunflower Seed Oil (해바라기씨 기름의 triglyceride분자종에 관한 연구)

  • From the results of triglyceride composition and the fatty acid at ${\beta}-position$ of glycerol, triglyceride molecular species of sunflower seed oil were found to be 26 kinds. The major triglyceride molecular species in sunflower seed oil were identified to PLL; 10.4%, OLL; 22.3%, and characterized that LLL species existed more than 31% of the total triglyceride molecular specie.

Quantitative Changes of Polyphenolic Compounds in Mulberry (Morus alba L.) Leaves in Relation to Varieties, Harvest Period, and Heat Processing

  • Six polyphenolic compounds, such as chlorogenic acid (CA), rutin (RT), isoquercitrin (IQT), quercetin-3-O-(6-O-malonyl)-${\beta}$-D-glucoside (QMG), astragalin (AG), kaempferol-3-O-(6-O-malonyl)-${\beta}$-D-glucoside (KMG), were isolated from mulberry leaves by a series of isolation procedures, such as Diaion HP-20, silica-gel, Sephadex LH-20, and ODS-A column chromatographies. The chemical structures of the phenolic compounds were identified by UV and NMR spectral analyses. Levels of polyphenols in mulberry leaves from six different mulberry cultivars ranged from 1,042.16 to 1,871.97 mg% per dry weight; Guksang cultivar showed the highest levels of polyphenols, whereas Gaeryangdaehwa contained the least polyphenol contents. Generally, levels of polyphenols in mulberry leaves decreased with increasing harvest time, except for Yoolmok, but increased with heat processing time, except QMG and KMG. These results suggest that the heat processed mulberry leaves of Guksang cultivar harvested in early May can be potentially useful sources for production of high quality mulberry leaf teas.

  • Five compounds were isolated from the whole plant of Arabis glabra (Cruciferae) through repeated silica gel and Sephadex LH-20 column chromatography. Their chemical structure were elucidated as salicylic acid, 2,5-dihydroxybenzoic acid, astragalin, rutin, and $quercetin-3,7-O-{\beta}-D-diglucopyranoside$ by spectroscopic analysis.

  • In order to search for anti-Herpes simplex virus type 1(HSV-1) agents, we screened 80 specimens of Korean traditional medicine by SRB assay. The methanol extracts of Thujae orientalis Semen (Cupressaceae) showed strong anti-HSV activity among samples tested. From the butanol fraction of Thujae orientalis Semen anti-HSV-1 agent was isolated by chromatographic separation using Amberlite XAD-4 and Sephadex LH-20. The structure was elucidated by spectroscopic methods, and was identified as ${\beta}-sitosterol$ (compound I). Compound I exhibited anti-HSV-1 activity with $EC_{50}$ of 0.6 mg/ml and $CC_{50}$ of 5.99 mg/ml, respectively.

Phenolic Compounds from Capsiccum annuum Leaves Showing Radical Scavenging Effect (고추 잎에서 라디칼소거활성을 가지는 페놀성 화합물의 분리)

  • In the present study, we investigated the antioxidant activity of methanol extract and its organic fractions (n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$) of Capsiccum annuum leaves against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. As results, methanol extract, and its ethyl acetate and n-butanol fractions showed good scavenging effect at 100 ${\mu}g/ml$. Four compounds were isolated from the ethyl acetate fraction through silica gel and Sephadex LH-20 column chromatographies. Their chemical structures were elucidated as apigenin (1), vanillic acid (2), uracil (3) and apigenin $7-O-{\beta}-D-apiofuranosyl(1{\rightarrow}2)-{\beta}-D-glucopyranoside$ (4) by comparison of spectral data with those in reference. $IC_{50}$ value of compound 2 was 14.7 ${\mu}g/ml$, while compounds 1, 3 and 4 had no effect on the DPPH radical.

Isolations of Flavonoids and a Higher Alcohol from the Aerial Parts of Brassica juncea (갓 지상부에서 플라보노이드와 고급 알콜 화합물의 분리)

  • The aerial parts of Brassica juncea (Cruciferae) called by brown or oriental mustard have been widely used as a spice in food and also traditional folk medicine as stimulant, diuretic and expectorant agent. And Gatkimchi made of the aerial parts of this plant are very popular in Korea. The aerial parts of this plant was refluxed with MeOH and then fractionationed with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$, successively. One higher alcohol compound and three flavonoids were isolated from the EtOAc fraction through silica gel and Sephadex LH-20 column chromatographies. Their structures were elucidated as n-hexacosanol(1), kaempferol(2), isorhamnetin $3-O-{\beta}-D-glucopyranoside(3)$ and isorhamnetin $3-O-(6'-O-acetyl)-{\beta}-D-glucopyranoside(4)$ by comparison of spectral data with those in references. And compounds 1 and 4 were firstly isolated from this plant.

  • As a series of the studies on the specific and indigenous plants of Mt. Halla, the constituents from stem of Rhamnus taquetii (Rhamnaceae) were investigated. From the water fraction of the MeOH extract, a new flavonol glycoside, kaempferide-3-O-${\alpha}$-L-rhamnopyranosyl(1->3)-${\alpha}$-L-rhamnopyranosyl(1-6)-${\beta}$-D-galactopyranoside, was isolated with three known compounds, quercetin, quercetin-3-methylether, kaempferol, by column chromatographic separation using Amberlite XAD-2, ODS-gel and Sephadex LH-20 and elucidated physico-chemical evidences($^1H-NMR,\;^{13}C-NMR$,IR, EI-Mass, FAB-Mass, and G.C.),respectively.

Hydrolysable Tannins from Cercidiphyllum japonicum Bark

  • The EtOAc and $H_2O$ soluble fractions of Katsura tree (Cercidiphyllum japonicum Sieb. Et Zucc) bark extracts were chromatographed on a Sephadex LH-20 column with various aqueous MeOH. Gallic acid (1), methyl galate (2), kurigalin (3), 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose (4) and 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucose (5) were isolated from EtOAc fraction. Isocorilagin (6) and methyl galate (2) were separated from $H_2O$ fraction. The structure determination was done by $^1H$ and $^{13}C$ NMR. Of these isolated compounds, methyl galate (2), kurigalin (3) and isocorilagin (6) were isolated, for the first time, from the bark extracts of Cercidiphyllum japonicum.

Optimization of Spray Drying Process for Manufacturing Dried Vinegar using Response surface methodology (분말식초제조를 위한 분무건조공정의 최적화)

  • This study was conducted to develop processing method for vinegar powder from natural vinegar for encapsulation applications. To optimize the spray drying process, experiment was designed by central composition method to find optimal conditions for manufacturing vinegar powder. The acidity, water absorption, solid content and heat stability of vinegar powder were selected as response variables. The optimal concentration of inclusion complex which was made with vinegar and ${\beta}$-cyclodextrin was determined on 30。bx. On increasing the concentration of ${\beta}$-cyclodextrin as a wall material, the quality of the vinegar was decreased. The optimum conditions of spray drying process for manufacturing vinegar powder were 188∼192$^{\circ}C$ and 500∼600 Lh$\^$ -1/ as inlet temperature and flow rate, respectively.

Chlorogenic Acid, an Antioxidant Principle from the Aerial Parts of Artemisia iwayomogi that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Artemisia iwayomogi was determined by measuring the radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract of A. iwayomogi showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate > n-butanol > water > chloroform > n-hexane fraction. The ethyl acetate and n-butanol soluble fractions exhibiting strong antioxidant activity were further purified by repeated sitica get and Sephadex LH-20 column chromatography. Antioxidant chlorogenic acid was isolated as one of the active principles from the n-butanol fraction, together with the inactive components, 1octacosanol, scopoletin, scopolin, apigenin $7, 4^{I}$-di-O-methylether, luteolin $6, 3^{I}$-di-O-methylether (jaceosidin), apigenin methylether (genkwanin), 2, 4-dihydroxy-6-methoxyacetophenone $4-O-{\beta}-$D-glucopyranoside and quebrachitol. The antioxidant activity of chlorogenic acid was comparable to that of L-ascorbic acid, which is a well known antioxidant.
