Fluorescence Sensing Properties of Thiazolobenzo-crown Ether Incorporating Coumarin

  • A new coumarin-thiazolobenzo-crown ether based fluorogenic chemosensor BTC (1) was reported. The ion-selective binding properties of 1 with different alkali, alkaline earth metals and transitional metals were investigated in an ethanol-DMSO system. BTC (1) showed the highest binding constant toward $Hg^{2+}$ over $Ag^+$, $Pb^{2+}$ and $Cu^{2+}$.

Chlorophyll a Fluorescence Response to Mercury Stress in the Freshwater Microalga Chlorella Vulgaris (담수산 클로렐라(Chlorella vulgaris)의 수은 스트레스에 대한 엽록소형광 반응)

  • The response of the freshwater microalga Chlorella vulgaris to mercuric ion ($Hg^{2+}$) stress was examined using chlorophyll a fluorescence image analysis and O-J-I-P analysis as a way to monitor the toxic effects of mercury on water ecosystems. The levels of photosynthetic pigments, such as chlorophyll a and b and carotenoids, decreased with increasing $Hg^{2+}$ concentration. The maximum photochemical efficiency of photosystem II(Fv/Fm) changed remarkably with increasing $Hg^{2+}$ concentration and treatment time. In particular, above $200{\mu}M\;Hg^{2+}$, considerable mercury toxicity was seen within 2 h. The chlorophyll a fluorescence transient O-J-I-P was also remarkably affected by $Hg^{2+}$; the fluorescence emission decreased considerably in steps J, I, and P with an increase in $Hg^{2+}$ concentration when treated for 4 h. Subsequently, the JIP-test parameters (Fm, Fv/Fo, RC/CS, TRo/CS, ETo/CS, ${\Phi}_{PO}$, ${\Psi}_O$ and ${\Phi}_{EO}$) decreased with increasing $Hg^{2+}$ concentration, while N, Sm, ABS/RC, DIo/RC and DIo/CS increased. Therefore, a useful biomarker for investigating mercury stress in water ecosystems, and the parameters Fm, ${\Phi}_{PO}$, ${\Psi}_O$, and RC/CS can be used to monitor the environmental stress in water ecosystems quantitatively.

  • Kinetic studies were carried out for the substitution of $H-2$O for $Cl^-$ ligand of chloropentaamminecobalt(III) in the presence of $Hg^{2+}$ catalyst by UV spectrophotometer. It has been found that $Hg^{2+}$ ion appears first order dependence and the overall reaction is second order. The reaction system can be believed to occur via $S_N2$ reaction, because the calculated rate equation for our presumed $S_N2$ mechanism is in agreement with the observed one.

Spectroscopic, Thermal and Biological Studies of Zn(II), Cd(II) and Hg(II) Complexes Derived from 3-Aminopyridine and Nitrite Ion

  • Microwave assisted syntheses of Zn(II), Cd(II) and Hg(II) complexes with 3-aminopyridine (3AP) and nitrite ($NO_2{^-}$) ions have been reported. The metal complexes were characterized by elemental analyses, molar conductance, IR, Far-IR, electronic, NMR ($^1H$, $^{13}C$), thermal and electron impact mass spectral studies. The spectroscopic studies reveal the composition, the nature of nitrite ligand in the complexes, electronic transitions, chemical environments of C and H atoms thermal degradation of the complexes. On the basis of characterization data, distorted tetrahedral geometry is suggested for Zn(II), Cd(II) and Hg(II) complexes. The organic ligand (3AP) and their metal complexes were screened against gram negative pathogenic bacteria and fungi in vitro. The results are compared with our previous report J. Korean Chem. Soc. 2013, 57, 341 on 4-aminopyridine and nitrite ion complexes of the same metal ions.

  • The dependence of ionic activity and electrode potential upon electrolyte concentration in ethylenediamine includes the effects of ion-pair formation involving all possible combinations of cations and anions represent in the solution and includes the ion-atmosphere effects of dissociated ions. For calculating activity coefficients, Debye-Huckel limiting law, extended Debye-Huckel,equation, and Marshall Grunwald equation were used in comparison of experimental and calculated plots of the electromotive force of the cell versus the logarithum of concentration. The fit of the experimental points to the theoretical curves was improved in case of the Marshall-Grunwald equation.

Expression of Ion Channels in Perivascular Stem Cells derived from Human Umbilical Cords

  • Potassium channels, the largest group of pore proteins, selectively regulate the flow of potassium ($K^+$) ions across cell membranes. The activity and expression of $K^+$ channels are critical for the maintenance of normal functions in vessels and neurons, and for the regulation of cell differentiation and maturation. However, their role and expression in stem cells have been poorly understood. In this study, we isolated perivascular stem cells (PVCs) from human umbilical cords and investigated the expression patterns of big-conductance $Ca^{2+}$-activated $K^+$ ($BK_{Ca}$) and voltage-dependent $K^+$ ($K_v$) channels using the reverse transcription polymerase chain reaction. We also examined the effect of high glucose (HG, 25 mM) on expression levels of $BK_{Ca}$ and $K_v$ channels in PVCs. $K_{Ca}1.1$, $K_{Ca}{\beta}_3$, $K_v1.3$, $K_v3.2$, and $K_v6.1$ were detected in undifferentiated PVCs. In addition, HG treatment increased the amounts of $BK_{Ca}{\beta}_{3a}$, $BK_{Ca}{\beta}_4$, $K_v1.3$, $K_v1.6$, and $K_v6.1$ transcripts. These results suggested that ion channels may have important functions in the growth and differentiation of PVCs, which could be influenced by HG exposure.

Determination of Selenium and Tellurium by HG-AAS in Foods

  • A method has been investigated for the determination of selenium and tellurium in Ginseng, Ganoderma and Garlic using hydride generation atomic absorption spectrometry(HG-AAS). The concentration effects of hydrochloric acid and sodium tetrahydroborate on the hydride generation for the determination of selenium and tellurium were investigated. The method of sample decomposition was also investigated using various mineral acids, such as nitric, perchloric and sulfuric acid in the closed system, and foreign ion effects containing in the samples were studied. The calibration curves of selenium and tellurium were obtained in the range of 0~40 ppb. The detection limits(S/N=2) of selenium and tellurium are 0.1 and 0.2 ppb. Analytical data of selenium and tellurium in Garlic, Ganoderma and Ginseng are 289, 296 and 198 ng/g for selenium and 146, 127 and 110 ng/g for tellurium, respectively.

Selective Trace Analysis of Mercury (II) Ions in Aqueous Media Using SERS-Based Aptamer Sensor

  • We report a highly sensitive surface-enhanced Raman scattering (SERS) platform for the selective trace analysis of mercury (II) ions in drinkable water using aptamer-conjugated silver nanoparticles. Here, an aptamer designed to specifically bind to $Hg^{2+}$ ions in aqueous solution was labelled with a TAMRA moiety at the 5' end and used as a Raman reporter. Polyamine spermine tetrahydrochloride (spermine) was used to promote surface adsorption of the aptamer probes onto the silver nanoparticles. When $Hg^{2+}$ ions are added to the system, binding of $Hg^{2+}$ with T-T pairs results in a conformational rearrangement of the aptamer to form a hairpin structure. As a result of the reduced of electrostatic repulsion between silver nanoparticles, aggregation of silver nanoparticles occurs, and the SERS signal is significantly increased upon the addition of $Hg^{2+}$ ions. Under optimized assay conditions, the concentration limit of detection was estimated to be 5 nM, and this satisfies a limit of detection below the EPA defined limit of 10 nM in drinkable water.

Effects of Fineness of Chitosan Fiber on the Adsorption Characteristics of Metal Ion (금속이온흡착 특성에 미치는 키토산 섬유 굵기의 영향)

  • This article describes the metal ions adsorption of chitosan fibers. The chitosan fibers were manufactured by wet spinning using 2% acetic acid as solvent, 10% aqueous sodium hydroxide as non solvent, and 4%chitosan solution as a solvent. The adsorption characteristics of chitosan fibers towards 100ppm solutions of various metal ions such as Cu(II), Cd(II), Cr(III), Hg(II) were examined at different pH value by ICP-Atomic Emission Spectrometer. The adhesiveness of metallic ions to the chitosan fiber were increased with the increase of pH and the decrease of denier. On the other hand, from pH4, chitosan fiber that is immersed in metal ion aqueous solution of Cu(II) and Cd(II) became homogeneous solution because is dissolved. The adhesiveness of metallic ions to chitosan fiber were found to increased in a sequence of Hg(II)> Cr(III)> Cu(II)> Cd(II). The antimicrobial characteristics of the chitosan fiber by adhered metal ions, virgin chitosan fiber, and cotton fiber were evaluated. The antimicrobial activity of the fibers were increased with the decrease of denier.

A highly effective route for removal of Hg2+ from the waste water using 3-nitrobenzelidenemalononitrile as a modifier of Fe3O4@SiO2 nanoparticles

  • SiO2-coated magnetic nanoparticles (Fe3O4@SiO2 NPs) were modified by 3-nitrobenzelidenmalononitrile and used as green linkages for removal of Hg2+ form the wastewater. In this research, it has been attempted to refer to the harmful effects of mercury ions for living things and how to remove such ions using very easy and practical technique. This study shows that by optimizing the test conditions, the efficiency of the removal of harmful ions such as mercury from the water contaminated with these ions can be increased. Conditions such as temperature, speed of agitation, pH of solution were tested for removal of mercury ions. The advantages of this method over other methods listed in the article are the rapid and easy nanocry synthesis. The generated and modified Fe3O4@SiO2 nanoparticles were characterized by X-ray diffraction, fourier transform infrared and scanning electron microscopy spectroscopy. The results show that the synthesized magnetic nanoparticles have the excellent performance for the removal of mercury(II) ion from the waste water.
