Animal protein hydrolysate reduces visceral fat and inhibits insulin resistance and hepatic steatosis in aged mice

  • BACKGROUND/OBJECTIVES: An increasing life expectancy in society has burdened healthcare systems substantially because of the rising prevalence of age-related metabolic diseases. This study compared the effects of animal protein hydrolysate (APH) and casein on metabolic diseases using aged mice. MATERIALS/METHODS: Eight-week-old and 50-week-old C57BL/6J mice were used as the non-aged (YC group) and aged controls (NC group), respectively. The aged mice were divided randomly into 3 groups (NC, low-APH [LP], and high-APH [HP] and fed each experimental diet for 12 weeks. In the LP and HP groups, casein in the AIN-93G diet was substituted with 16 kcal% and 24 kcal% APH, respectively. The mice were sacrificed when they were 63-week-old, and plasma and hepatic lipid, white adipose tissue weight, hepatic glucose, lipid, and antioxidant enzyme activities, immunohistochemistry staining, and mRNA expression related to the glucose metabolism on liver and muscle were analyzed. RESULTS: Supplementation of APH in aging mice resulted in a significant decrease in visceral fat (epididymal, perirenal, retroperitoneal, and mesenteric fat) compared to the negative control (NC) group. The intraperitoneal glucose tolerance test and area under the curve analysis revealed insulin resistance in the NC group, which was alleviated by APH supplementation. APH supplementation reduced hepatic gluconeogenesis and increased glucose utilization in the liver and muscle. Furthermore, APH supplementation improved hepatic steatosis by reducing the hepatic fatty acid and phosphatidate phosphatase activity while increasing the hepatic carnitine palmitoyltransferase activity. Furthermore, in the APH supplementation groups, the red blood cell (RBC) thiobarbituric acid reactive substances and hepatic H2O2 levels decreased, and the RBC glutathione, hepatic catalase, and glutathione peroxidase activities increased. CONCLUSIONS: APH supplementation reduced visceral fat accumulation and alleviated obesity-related metabolic diseases, including insulin resistance and hepatic steatosis, in aged mice. Therefore, high-quality animal protein APH that reduces the molecular weight and enhances the protein digestibility-corrected amino acid score has potential as a dietary supplement for healthy aging.

Effect of Support of Two-Dimensional Pt Nanoparticles/Titania on Catalytic Activity of CO Oxidation

  • Smart catalyst design though novel catalyst preparation methods can improve catalytic activity of transition metals on reducible oxide supports such as titania by enhancement of metal oxide interface effects. In this work, we investigated Pt nanoparticles/titania catalysts under CO oxidation reaction by using novel preparation methods in order to enhance its catalytic activity by optimizing metal oxide interface. Arc plasma deposition (APD) and metal impregnation techniques are employed to achieve Pt metal deposition on titania supports which are prepared by multi-target sputtering and Sol-gel techniques. In order to tailor metal-support interface for catalytic CO oxidation reaction, Pt nanoparticles and thin films are deposited in varying surface coverages on sputtered titania films using APD. To assess the role of oxide support at the interface, APD-Pt is deposited on sputtered and Sol-gel prepared titania films. Lastly, characteristics of APD-Pt process are compared with Pt impregnation technique. Our results show that activity of Pt nanoparticles is improved when supported over Sol-Gel prepared titania than sputtered titania film. It is suggested that this enhanced activity can be partly ascribed to a very rough titania surface with the higher free metal surface area and higher number of sites at the interface between the metal and the support. Also, APD-Pt shows superior catalytic activity under CO oxidation as compared to Pt impregnation on sputtered titania support. XPS results show that bulk oxide is formed on Pt when deposited through impregnation and has higher proportion of oxidized Pt in the form of $Pt^{2+/4+}$ oxidation states than Pt metal. APD-Pt shows, however, mild oxidation with large proportion of active Pt metal. APD-Pt also shows trend of increasing CO oxidation activity with number of shots. The activity continues to increase with surface coverage beyond 100%, thus suggesting a very rough and porous Pt films with higher active surface metal sites due to an increased surface area available for the reactant CO and $O_2$ molecules. The results suggest a novel approach for systematic investigation into metal oxide interface by rational catalysts design which can be extended to other metal-support systems in the future.

Effect of buchu (Allium tuberosum) on lipid peroxidation and antioxidative defense system in streptozotocin-induced diabetic rats (부추가 Streptozotocin 유발 당뇨쥐의 지질과산화와 항산화방어체계에 미치는 영향)

  • The pathogenic effort of high glucose, possibly in concert with fatty acids, is mediated to vascular complications of diabetes via increased production of reactive oxygen species(ROS), reactive nitrogen species(RNS), and subsequent oxidative stress. This study was carried out to investigate the suppressive effect of buchu(Allium tuberosum) on oxidative stress in streptozotocin(STZ)-induced diabetes in Sprague Dawley male rats. The effect of buchu supplementation (10%) on lipid peroxidation, and antioxidative defense system in blood and liver was compared among normal rats fed basal diet(normal) and diabetic rats fed basal diet(DM-control) or 10% buchu-supplemented diet(DM-buchu). Diabetes was experimentally induced by the femoral muscle injection of 50 mg STZ per kg of body weight. Animals were sacrificed after 4 wks of experimental diets feeding. The induction of diabetes by STZ elevated the level of lipid peroxidation represented by thiobarbituric acid-reactive substances(TBARS) and conjugated dienes in plasma, LDL, liver, and erythrocytes. 10% buchu-supplemented diet significantly reduced the levels of conjugated dienes in erythrocytes(p<0.05) and lowered TBARS in liver and LDL to the levels of control. Induction of diabetes by STZ elevated Mn-superoxide dismutase(Mn-SOD) activity and lowered activities of glutathionine reductase(GSH-red) and glutathionine peroxidase(GSH-px). Catalase activity was not affected by the induction of diabetes by STZ. However, buchu supplementation to diabetic rats significantly elevated catalase activity(p<0.05) and slightly elevated GSH-px and GSH-red activities in liver. GSH levels of blood and liver were lowered or not changed by induction of diabetes by STZ, respectively, while buchu supplementation to diabetic rats significantly elevated hepatic GSH level (p<0.05). In conclusion, it can be concluded that buchu might be a food source to attenuate oxidative stress in diabetic patients by inhibiting lipid peroxidation, by increasing hepatic GSH level, and by inducing anti-oxidative enzyme systems.

증착온도에 의한 전기적 착색 니켈 산화물 박막의 특성 분석

  • 니켈 산화물 박막을 전자비임 증착법으로 기판온도는 RT~25$0^{\circ}C$의 범위에서 제작하였다. 제작시 초기 베이스 압력은 2$\times$10-6mbar로 하고 산소주입후 작업진공도를 3$\times$10-4mbar로 유지하여 증착하였다. 제작시 기판온도에 따라 제작된 시료들은 각각 X선회절장치(XRD)로 막의 구조과 그림과 같이 입방체 구조 또는 팔면체구조를 갖음을 알 수 있었으며 막의 표면형상은 SEM을 이용하여 분석하였다. 각각의 여러 기판온도에 따라 제작된 니켈 산화물 박막의 전기 화학적인 특성을 분석하기 위해 순환전압전류법을 이용하였다. 또한, 전기적인 광학소자로써의 특성을 분석하기 위해 UV-Vis 광분광기를 사용하여 투과율을 측정하여 그 특성을 알아보았다. 순환전압전류법에 의한 각 시료에 대한 박막의 전기화학적 특성은 0.5M KOH 전해질 수용액에서 기판온도가 150~20$0^{\circ}C$로 제작된 니켈 산화물 박막이 다른 온도에서 제작된 시료들보다 높은 전기화학적 안정성을 보임을 알 수 있었다. 마찬가지로 광학적 특성에서 착색과 탈색의 순환과정시 분광광도계에서 나타나는 광투과율을 비교해 보면 100~20$0^{\circ}C$에서 제작된 니켈 산화물 박막이 가역적인 착탈색의 색변화가 현저하게 나타남을 알 수 있었다. 결과적으로 광학적 특성 및 전기화학적 안정성 분석으로 인해 막의수명과 전기적착색 물질의 특성면에서 증착시 기판온도가 150~20$0^{\circ}C$에서 제작된 시료가 가장 내구성면에서 막의 이온 누적이 적고 활성적인 광투과율의 성질을 갖는다는 것이다. 이와같이 니켈산화물 박막제작시 기판온도가 전기적착색물질의 특성과 내구성에 큰 영향을 미침을 분석할 수 있었다.electron Microscopy)과 AFM(Atomim Force microscopy)으로 증착박 표면의 topology와 roughness를 관찰하였다. grain의 크기는 10nm에서 150nm이었고 증착막의 roughness는 4.2nm이었다. 그리고 이 산화막에 전극을 형성하여 유전 상수와 손실률 등을 측정하였다. 이와 같이 plasma를 이용한 3-beam에 의한 증착은 금속의 산화막을 얻는데 유용한 기술로 광학 재료 및 유전 재료의 개발 및 연구에 많이 사용될 것으로 기대된다.소분압 조건에서 RuO2의 형성을 관찰하였으며, 이것은 열역학적인 계산을 통해서 잘 설명할 수 있었다.0$\mu\textrm{m}$, 코일간의 간격은 100$\mu\textrm{m}$였다. 제조된 박막 인덕터는 5MHz에서 1.0$\mu$H의 인덕턴스를 나타내었으며 dc current dervability는 100mA까지 유지되었다. CeO2 박막과 Si 사이의 결함때문이라고 사료된다.phology 관찰결과 Ge 함량이 높은 박막의 입계가 다결정 Si의 입계에 비해 훨씬 큰 것으로 나타났으며 근 값도 증가하는 것으로 나타났다. 포유동물 세포에 유전자 발현벡터로써 사용할 수 있음으로 post-genomics시대에 다양한 종류의 단백질 기능연구에 맡은 도움이 되리라 기대한다.다양한 기능을 가진 신소재 제조에 있다. 또한 경제적인 측면에서도 고부가 가치의 제품 개발에 따른 새로운 수요 창출과 수익률 향상, 기존의 기능성 안료를 나노(nano)화하여 나노 입자를 제조, 기존의 기능성 안료에 대한 비용 절감 효과등을 유도 할 수 있다. 역시 기술적인 측면에서도 특수소재 개발에 있어 최적의 나노 입자 제어기술 개발 및 나노입자를 기능성 소재로 사용하여 새로운 제품의 제조와 고압 기상 분사기술의 최적

Effect of Supplemental Corn Dried Distillers Grains with Solubles Fed to Beef Steers Grazing Native Rangeland during the Forage Dormant Season

  • Two experiments were conducted to evaluate the effects of the level of corn dry distillers grains with solubles (CDDGS) supplementation on growing performance, blood metabolites, digestion characteristics and ruminal fermentation patterns in steers grazing dormant forage. In Exp. 1, of growth performance, 120 steers ($204{\pm}5kg$ initial body weight [BW]) were distributed randomly into 3 groups (each of 40 steers), which were provided with the following levels of CDDGS supplement: 0%, 0.25%, or 0.50% BW. All groups of steers were grazed for 30 days in each of 3 grazing periods (March, April, and May). Approximately 1,000 ha of the land was divided with electric fencing into 3 equally sized pastures (333 ha in size). Blood samples were collected monthly from 20 steers in each grazing group for analysis of glucose (G), urea-nitrogen (UN) and non-esterified fatty acids. Final BW, average daily gain (ADG) and supplement conversion (CDDGS-C) increased with increasing levels of CDDGS supplementation (p<0.05).The CDDGS supplementation also increased the plasma G and UN concentrations (p<0.05). In Exp. 2, of digestive metabolism, 9 ruminally cannulated steers ($BW=350{\pm}3kg$) were distributed, following a completely randomized design, into groups of three in each pasture. The ruminally cannulated steers were provided the same levels of CDDGS supplementation as in the growing performance study (0%, 0.25%, and 0.50% BW), and they grazed along with the other 40 steers throughout the grazing periods. The dry matter intake, crude protein intake, neutral detergent fiber intake (NDFI), apparent digestibility of dry matter (ADDM), crude protein (ADCP) and neutral detergent fiber (ADNDF) increased with increasing levels of CDDGS supplementation (p<0.05). The ruminal degradation rates of CP (kdCP), NDF (kdNDF) and passage rate (kp) also increased with increasing levels of CDDGS supplementation (p<0.05). Ruminal ammonia nitrogen ($NH_3$-N) and propionate concentrations also increased with increasing levels of CDDGS supplementation (p<0.05). However, acetate concentrations decreased with increasing levels of CDDGS supplementation (p<0.05). Liquid dilution rate increased with increasing levels of CDDGS supplementation but ruminal liquid volume decreased (p<0.05). On the basis of these findings, we can conclude that CDDGS supplementation enhanced the productive performance of cattle grazing native rangeland without negatively affecting forage intake, glucose and urea-nitrogen blood concentrations, ruminal degradation and ruminal fermentation patterns.

Microstructure and Biocompatibility of Porous BCP(HA/β-TCP) Biomaterials Consolidated by SPS Using Space Holder

  • $HA(hydroxyapatite)/{\beta}-TCP$ (tricalcium phosphate) biomaterial (BCP; biphasic calcium phosphate) is widely used as bone cement or scaffolds material due to its superior biocompatibility. Furthermore, $NH_4HCO_3$ as a space holder (SH) has been used to evaluate feasibility assessment of porous structured BCP as bone scaffolds. In this study, using a spark plasma sintering (SPS) process at 393K and 1373K under 20MPa load, porous $HA/{\beta}-TCP$ biomaterials were successfully fabricated using $HA/{\beta}-TCP$ powders with 10~30 wt% SH, TiH2 as a foaming agent, and MgO powder as a binder. The effect of SH content on the pore size and distribution of the BCP biomaterial was observed by scanning electron microscopy (SEM) and a microfocus X-ray computer tomography system (SMX-225CT). The microstructure observations revealed that the volume fraction of the pores increased with increasing SH content and that rough pores were successfully fabricated by adding SH. Accordingly, the cell viabilities of BCP biomaterials were improved with increasing SH content. And, good biological properties were shown after assessment using Hanks balanced salt solution (HBSS).

STUDIES ON THE UTILIZATION OF RICE STRAW BY SHEEP III. EFFECT OF SOYBEAN MEAL AND BARLEY SUPPLEMENTATION ON VOLUNTARY INTAKE, DIGESTIBILITY AND RUMINAL FERMENTATION

  • The effect of soybean meal and barley supplementation of the utilization of rice straw was investigated. Balance trials were conducted with three Japanese Corriedale wethers fed rice straw supplemented with soybean meal and barley at three different levels of protein: low (40 g CP/d, LCP), medium (67 g/d, MCP) and high (94 g/d, HCP). In addition, all the supplements were formulated to contain the same amount of TDN (275 g/d). Voluntary intake of rice straw was not affected by any supplementation, while digestibility of organic matter in sheep given HCO diet was significantly higher (p<0.05) than those on LCP diet. Crude protein, neutral detergent fiber (NDF) and acid detergent fiber (ADF) digestibilities of MCP and HCP diets were significantly improved (p<0.05) over the LCP diet. Average daily gain of the animals under MCP and HCP diets were significantly higher (p<0.05) than those under LCP diet. Differences of rumen pH among the treatments were not significant, while concentration of rumen $NH_3-N$ was significantly higher (p<0.05) for HCP diet than for LCP and MCP diets. Total volatile fatty acids ($VFA_s$) and blood urea nitrogen (BUN) concentrations were significantly higher (p<0.05) in sheep fed MCP and HCP diets than those fed LCP diet, while plasma total protein concentration was not affected by any supplementation. Sheep fed MCP diet had a higher nitrogen retention than those fed LCP and HCP diets. It was concluded that rice straw was utilized better by sheep when SBM and barley were supplemented at the medium level of protein.

Effect of Space Holder Content on Pore Size and Distribution in HA/β-TCP Composites Consolidated by SPS (SPS로 제조된 HA/β-TCP 복합재의 기공의 크기와 분포에 미치는 지지체 량의 영향)

  • Ceramics biomaterials are useful as implant materials in orthopedic surgery. In this study, porous HA(hydroxyapatite)/${\beta}$-TCP(tricalcium phosphate) composite biomaterials were successfully fabricated using HA/${\beta}$-TCP powders with 10-30 wt% $NH_4HCO_3$ as a space holder(SH) and $TiH_2$ as a foaming agent, and MgO powder as a binder. The HA/${\beta}$-TCP powders were consolidated by spark plasma sintering(SPS) process at $1000^{\circ}C$ under 20 MPa conditions. The effect of SH content on the pore size and distribution of the HA/${\beta}$-TCP composite was observed by scanning electron microscopy(SEM) and a microfocus X-ray computer tomography system(SMX-225CT). These microstructure observations revealed that the volume fraction of the pores increased with increasing SH content. The pore size of the HA/${\beta}$-TCP composites is about $400-500{\mu}m$. The relative density of the porous HA/${\beta}$-TCP composite increased with decreasing SH content. The porous HA/${\beta}$-TCP composite fabricated with 30%SH exhibited an elastic modulus similar to that of cortical bone; however, the compression strength of this composite is higher than that of cortical bone.

The Effect of Vitamin {TEX}$B_{6}${/TEX} Deficiency on the Utilization and Recuperation of Stored Fuel in Highly Intense Exercised Rats

  • The purpose of this study was to investigate the effect of vitamin {TEX}$B_{6}${/TEX} deficiency on the utilization and recuperation of stored fuel in exercising rats. Thirty six rats were fed either a vitamin {TEX}$B_{6}${/TEX} deficient diet(-{TEX}$B_{6}${/TEX}) or a control diet(+{TEX}$B_{6}${/TEX}) for 5 weeks, then subdivided into 3 group: non-exercise group(NE), exercise group(EX), exercise and recuperation group(EX). EX group were exercised in treadmill({TEX}$10^{o}${/TEX}, 0.5~0.8km/h)for 2 hours and ER group were recuperated three days with the respective diet after exercise. Glucose(GLU), glycogen(GLY), protein(PRO), trigyceride(TG) and free fatty acid(FFA) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. Compared to +{TEX}$B_{6}${/TEX} rats, in NE group, the level of L-GLY of -{TEX}$B_{6}${/TEX} rats was higher, M-TG, L-PRO of -{TEX}$B_{6}${/TEX} rats were lower and there were no differences in P-PRO, P-FFA, P-TG, M-GLY, M-PRO and L-TG. In EX group, the levels of P-FFA, L-PRO of -{TEX}$B_{6}${/TEX} rats were higher. P-TG, L-TG of -{TEX}$B_{6}${/TEX} rats were lower and there was no difference in L-GLY. In EG group, the levels of P-GLU, P-PRO, P-RG, L-PRO of -{TEX}$B_{6}${/TEX} rats were lower and there were no differences in L-GLY, L-TG, M-TG and M-GLY. these results suggest that a lowered intake of vitamin {TEX}$B_{6}${/TEX} may impair the recuperation of aminals after exercise related to exercise fuel stores although there is a compensation among stored fuel utilization during exercise.

Do platelet concentrates accelerate orthodontic tooth movement?: a systematic review

  • Purpose: Surgical techniques in orthodontics have received widespread attention in recent years. Meanwhile, biomaterials with high molecular content have been introduced, such as platelet concentrates (PCs), which may accelerate orthodontic tooth movement (OTM) and reduce periodontal damage. The present systematic review aimed to answer the following PICO question: "In patients in whom orthodontic surgical techniques are performed (P), what is the effectiveness of using PCs over the surgical site (I) when compared to not placing PCs (C) to achieve faster tooth movement (O)?" Methods: A search was performed in 6 databases. The criteria employed were those described in the Preferred Reporting Items for Systematic Reviews and Meta-Analyses declaration. The present review included studies with a control group that provided information about the influence of PCs on the rate of OTM. Results: The electronic search identified 10 studies that met the established criteria. Conclusions: The included studies were very diverse, making it difficult to draw convincing conclusions. However, a tendency was observed for OTM to be accelerated when PCs were used as an adjuvant for canine distalization after premolar extraction when distalization was started in the same session. Likewise, studies seem to indicate an association between PC injection and the amount of canine retraction. However, it is not possible to affirm that the use of PCs in corticotomy shortens the overall treatment time, as this question has not been studied adequately.
