Pseudomonas mendocina 에 의한 Polysaccharide 생산 (Cultural Condition of Pseudomonas mendocina for Polysaccharide Production)

  • 토양으로부터 분리 동정된 Pseudomonas mendocina의 다당류 생산조건을 검토하였다. 최적 배지조성은 Sucrose 23.75g/L, $(NH_4)_2SO_4$ 1.57g/L, Yeast extract 0.5g/L, $KH_2PO_4\;2.0g/L,\;MgSO_4.\;7H_2O\;1.0g/L,\;CaCO_3$ 2.5g/L이었으며, 최적 pH 및 배양온도는 6.5 및 $30^{\circ}C$이었다. 이 조건에서 다당류의 생산량은 5.98g/L이며 대당 수율은 25.18%, 비생산속도는 32.83mg/g-cell/h이었다.

인삼열매추출물의 용매분획물이 LPS로 유도된 RAW264.7 세포에 대한 항염활성 (Anti-inflammatory Activity of Solvent Fractions from Ginseng Berry Extract in LPS-Induced RAW264.7 Cells)

  • Anti-inflammatory activity of the extracts of ginseng berry (GBE) was investigated through the evaluation of its inhibitory effect on the production of inflammatory meditator, nitric oxide(NO), tumor necrocis factor-alpha (TNF-${\alpha}$), interleukin-6 (IL-6) in LPS-induced RAW264.7 macrophage cells. GBE was fractionated using n-hexane, chloroform, ethylacetate, buthanol and $H_2O$, sequentially. RAW264.7 cells were induced $100ng/m{\ell}$ of lipopolysaccharide (LPS) and treated with 0, 1.6, 8, 40 and $200{\mu}g/m{\ell}$ of GBE fractions. LPS-induced NO production on all of GBE fractions was inhibited with increasing added concentration of GBE fractions. Chloroform fraction of GBE was the most effective in inhibiting LPS-induced TNF-${\alpha}$ production. Hexane, chloroform and $H_2O$ fractions of GBE exhibit strong inhibition LPS-induced IL-6 production. Especially, $H_2O$ fractions of GBE was the most effective in inhibiting LPD-induced IL-6 production without significant cytotoxicity in RAW264.7 cells, and reduced the activation of mitogen-activated protein kinases (MAPK) and IkB phosphorylation. These results indicate that $H_2O$ fractions of GBE exhibits strong anti-inflammatory effects by inhibition of NF-kB by inhibition of p-38 on MAPK and IkB phosphorylation.

음식물쓰레기의 생물학적 수소생산 및 미생물의 군집특성 (Characteristics of Microbial Community and Bio-hydrogen Production from Food Waste)

  • 탄소원으로 자당을 공급한 합성폐수에 비해 상대적으로 발생수소량은 (7.56 mg $H_2/g$ COD) 적었지만 음식물쓰레기를 기질로 이용한 혐기적 발효공정에서 발생된 가스는 3.47 mg $H_2/g$ COD의 수소생성율을 나타내었다. 자당 합성폐수와 음식물쓰레기의 경우, 각각 8.01, 3.73의 B/A비를 보였으며 수소생성이 많은 경우 주요 유기산 중 Butyric acid가 많이 검출되었다. 동정분석 결과 주요 미생물 군집은 Escherichia 속, Klebsiella 속, Clostridium 속, Bacterium 속, 그리고 Enterobacter 속으로 분석되었다. Clostridium 속은 자당 합성폐수와 음식물 쓰레기 모두에서 관찰되었고 Klebsiella 속은 음식물 쓰레기의 발효과정에서 더욱 활발한 것으로 나타났다. 미생물의 분류학적 관계를 확인한 결과로 60%가 ${\gamma}-proteobacteria$이였으며 Firmicute와 Bacteria가 각각 20%를 차지하였다.

Effect of Phytogenic Feed Additives in Soybean Meal on In vitro Swine Fermentation for Odor Reduction and Bacterial Community Comparison

  • The effect of different phytogenic feed additives on reducing odorous compounds in swine was investigated using in vitro fermentation and analyzed their microbial communities. Soybean meal (1%) added with 0.1% different phytogenic feed additives (FA) were in vitro fermented using swine fecal slurries and anaerobically incubated for 12 and 24 h. The phytogenic FAs used were red ginseng barn powder (Panax ginseng C. A. Meyer, FA1), persimmon leaf powder (Diospyros virginiana L., FA2), ginkgo leaf powder (Ginkgo biloba L., FA3), and oregano lippia seed oil extract (Lippia graveolens Kunth, OL, FA4). Total gas production, pH, ammonianitrogen ($NH_3$-N), hydrogen sulfide ($H_2S$), nitrite-nitrogen ($NO_2{^-}$-N), nitrate-nitrogen ($NO_3{^-}$-N), sulfate (${SO_4}^{--}$), volatile fatty acids (VFA) and other metabolites concentration were determined. Microbial communities were also analyzed using 16S rRNA DGGE. Results showed that the pH values on all treatments increased as incubation time became longer except for FA4 where it decreased. Moreover, FA4 incubated for 12 and 24 h was not detected in $NH_3$-N and $H_2S$. Addition of FAs decreased (p<0.05) propionate production but increased (p<0.05) the total VFA production. Ten 16S rRNA DGGE bands were identified which ranged from 96 to 100% identity which were mostly isolated from the intestine. Similarity index showed three clearly different clusters: I (FA2 and FA3), II (Con and FA1), and III (FA4). Dominant bands which were identified closest to Eubacterium limosum (ATCC 8486T), Uncultured bacterium clone PF6641 and Streptococcus lutetiensis (CIP 106849T) were present only in the FA4 treatment group and were not found in other groups. FA4 had a different bacterial diversity compared to control and other treatments and thus explains having lowest odorous compounds. Addition of FA4 to an enriched protein feed source for growing swine may effectively reduce odorous compounds which are typically associated with swine production.

  • Numerical simulations of freely propagating flames burning $H_2/HCl/Air$ Air mixtures were performed at atmospheric pressure in order to understand the effect of hydrogen chloride on flame structures. The chemical and physical effects of hydrogen chloride on flame structures were observed. A chemical kinetic mechanism was developed, which involved 26 gas-phase species and 198 forward elementary reactions. Under several equivalence ratios the flame speeds were calculated and compared with those obtained from the experiments, the results of which were in good agreement. As hydrogen chloride as additive was added into $H_2/Air$ flame, the flame speed, radical concentration and NO production rate were decreased. The chemical effect of hydrogen chloride caused the reduction of radical concentration, and then the decrease of the net rate of NO production. It was found that the influence in the reduction of $EI_{NO}$ with the addition of hydrogen chloride was attributed more due to the chemical effect than the physical effect.

혐기 발효 공정을 통한 음식물류 폐기물 탈리액으로부터 수소 생산 (Dark Fermentative Hydrogen Production using the Wastewater Generated from Food Waste Recycling Facilities)

  • The authors examined the effects of operating parameters on the $H_2$ production by dark fermentation of the wastewater generated from food waste recycling facilities, in short "food waste wastewater (FWW)". Central composite design based response surface methodology was applied to analyze the effect of initial pH (5.5-8.5) and substrate concentration (2-20 g Carbo. COD/L) on $H_2$ production. The experiment was conducted under mesophilic ($35^{\circ}C$) condition and a heat-treated ($90^{\circ}C$ for 20min)anaerobic digester sludge was used as a seeding source. Although there was a little difference in carbohydrate removal, $H_2$ yield was largely affected by the experimental conditions, from 0.38 to 1.77 mol $H_2$/mol $hexose_{added}$. By applying regression analysis, $H_2$ yield was well fitted based on the coded value to a second order polynomial equation (p = 0.0243): Y = $1.78-0.17X_1+0.30X_2+0.37X_1X_2-0.29X_1{^2}-0.35X_2{^2}$, where $X_1$, $X_2$, and Y are pH, substrate concentration (g Carbo. COD/L), and hydrogen yield (mol $H_2$/mol $hexose_{added}$), respectively. The 2-D response surface clearly showed a high inter-dependency between initial pH and substrate concentration, and the role of these two factors was to control the pH during fermentation. According to the statistical optimization, the optimum condition of initial pH and substrate concentration were 7.0 and 13.4 g Carbo. COD/L, respectively, under which predicted $H_2$ yield was 1.84 mol $H_2$/mol $hexose_{added}$. Microbial analysis using 16S rRNA PCR-DGGE showed that $Clostridium$ sp. such as $Clostridium$ $perfringens$, $Clostridium$ $sticklandii$, and $Clostridium$ $bifermentans$ were main $H_2$-producers.

Rumen fermentation, methane production, and microbial composition following in vitro evaluation of red ginseng byproduct as a protein source

  • The main objective of this in vitro study was to evaluate red ginseng byproduct (RGP) as a protein resource and its effects on rumen fermentation characteristics, microflora, CO2, and CH4 production in ruminants. Four treatments for in vitro fermentation using buffered rumen fluid over a 48 h incubation period were used: 1, RGP; 2, corn gluten feed (CGF); 3, wheat gluten (WG); and 4, corn germ meal. In vitro dry matter digestibility (IVDMD), in vitro neutral detergent fiber digestibility (IVNDFD), in vitro crude protein digestibility (IVCPD), volatile fatty acids, pH, and ammonia nitrogen (NH3-N) were estimated after 48 h incubation. Gas production was investigated after 3, 6, 12, 24, 36 and 48 h. The CO2 and CH4 were evaluated after 12, 24, 36, and 48 h. A significant difference in total gas production and CO2 emissions was observed (p < 0.01) at all incubation times. CH4 production in RGP were higher (p < 0.05) than that in other treatments but a higher CH4 portion in the total gas production was observed in WG (p < 0.05) at 48 h incubation. The IVDMD, IVNDFD, and IVCPD of RGP was lower than those of other conventional ingredients (p < 0.01). The RGP had the lowest NH3-N value among the treatments (p < 0.01). The RGP also had the lowest total VFA concentration (p < 0.01), but presented the highest acetate proportion and acetate to propionate ratio among the treatments (both, p < 0.01). The abundance of Prevotella ruminicola was higher in RGP than in WG (p < 0.01), whereas RGP has lower methanogenic archaea (p < 0.01). In conclusion, based on the nutritive value, IVDMD, low NH3-N, and decreased methanogenic archaea, RGP inclusion as a protein source in ruminant diets can be an option in replacing conventional feed sources.

  • Ganoderma lucidum이 생산하는 polygalacturonase의 유용 활용 방안을 위한 최적 생산조건을 검토한 결과는 다음과 같다. Polygalacturonase 생산을 위한 최적 배양조건은 $30^{\circ}C$, pH5.5에서 14일이었다. Polygalacturonase 생산을 위한 최적 영양조건은 pectin 10 g, soluble starch 10 g, yeast extract 1 g, peptone 2 g, phenylalanine 1 g, $KH_2PO_4$ 2 g, $MgSO_4$ 0.2 g, $CaCI_2$ 50 mg, $thiamin{\cdot}HCI\;100{\mu}g$, D.W. 1000ml이었다.

Pyunkang-hwan (Pyunkang-tang) Regulates Hypersecretion of Pulmonary Mucin from Rats with Sulfur Dioxide-Induced Bronchitis and Production and Gene Expression of MUC5AC Mucin from Human Airway Epithelial Cells

  • Pyunkang-hwan (Pyunkang-tang) extract (PGT) is a traditional folk medicine for controlling diverse pulmonary diseases including bronchitis, tonsiltis and pneumonitis. We investigated whether PGT significantly affects secretion, production and gene expression of airway mucin using in vivo and in vitro experimental models reflecting the hypersecretion and/or hyperproduction of mucus observed in inflammatory pulmonary diseases. For in vivo experiment, effect of PGT was checked on hypersecretion of pulmonary mucin in sulfur dioxide-induced bronchitis in rats. For in vitro experiment, confluent NCI-H292 cells were pretreated with PGT for 30 min and then stimulated with EGF (epidermal growth factor), PMA (phorbol 12-myristate 13-acetate) or TNF-${\alpha}$ (tumor necrosis factor-${\alpha}$) for 24 h. The MUC5AC mucin gene expression and mucin protein production were measured by RT-PCR and ELISA. The results were as follows: (1) PGT inhibited the expression of MUC5AC mucin gene induced by EGF, PMA or TNF-${\alpha}$ from NCI-H292 cells, respectively; (2) PGT also inhibited the production of MUC5AC mucin protein induced by the same inducers from NCI-H292 cells, respectively; (3) PGT inhibited secretion of mucin in sulfur dioxide-induced bronchitis rat model. This result suggests that PGT can regulate secretion, production and gene expression of airway mucin.

CSTR의 장기운전을 통한 포도당으로부터의 고온 수소생산 (Thermophilic Biohydrogen Production from Glucose with a Long-term Operation of CSTR)

  • 1. $H_2$ 생산속도와 $H_2$ 수율의 안정화를 근거로 판단컨대 start-up 기간은 30일 이내로 나타나 중온 CSTR에 비해 짧은 편이었다. 2. 고온 CSTR의 최고 $H_2$ 수율은 2.4 mol $H_2/mol$ glucose로 나타나 보고 된 중온의 것에 비해 우수한 편에 속하였다. 3. 운전 초기에 $CH_4$이 발생하였으나 14일 이후부터는 pH를 5.0 이하로 유지하면 거의 검출되지 않는 것으로 봐서 메탄생성균이 식종균에 남아 있더라도 반응기 운전 조건을 통해 $CH_4$ 발생을 억제할 수 있었다. 4. 고온 CSTR은 초기 운전 후에 적용한 운전조건의 변화(유입 포도당 농도, pH, 및 온도)에 민감한 것으로 나타났다. 특히 pH 및 온도변화에 대해 $H_2$ 생산속도와 $H_2$ 수율, 포도당 제거율 면에서 반응기 성능의 감소 및 불안정이 나타나, 운전 조건 변화 후에 나타난 고온 CSTR의 성능회복이 쉽지 않음을 알 수 있었다. 5. 문헌에 보고 된 중온 CSTR과는 달리 고온 CSTR는 일정한 조건에서도 불안정 한 성능을 나타내기도 하였다. 6. 불안정한 반응기 성능은 lactate 농도 증가와 더불어 n-butyrate와 acetate 농도 감소를 동반하였다. 생산된 n-butyrate와 acetate의 농도는 lactate의 농도변화와 반대의 경향을 나타내었다. 7. 비교적 긴 HRT와 침전조를 이용한 biomass의 재순환에도 불구하고, 유입 포도당의 농도가 낮아 biomass 농도는 다른 중온 반응기에서 보고된 것에 비해 낮은 편이었다. 8. T. thermosaccharolyticum와 계통발생학적으로 관련된 개체군이 반응기 운전 후 약 40일부터 우점으로 나타나 반응기 성능과 상관없이 그 이후로 계속 우세한 것으로 나타났다.
