A Study of Surface Electromyography Measurement of Wrist Exercise (Extension, Flexion, Grasping) in Healthy People (정상인의 완관절 신전, 굴곡, 악력(握力) 시 표면근전도 측정 연구)

  • Purpose : This study aimed to prove that surface electromyography(SEMG) can be used to identify the degree of symptoms of diseases in the upper extremities; it also aimed to confirm various potential therapeutic effects through an inquiry into the value measured by the SEMG on certain acupuncture points in the upper extremities. Methods : Fifty healthy people received instructions for the method of exercise: wrist flexion, extension and hand grasping. Disposable electrodes were attached to acupuncture points of Susamni($LI_{10}$), Naegwan($PC_6$), Oegwan($TE_5$) and below Sohae($HT_3$) two cun on both sides in flexion, extension and grasping to measure the SEMG values. The research results were analyzed using SPSS statistics Ver. 22.0(IBM, USA). Results : The average value was highest on Naegwan($PC_6$) in grasping, and the average SEMG value was higher in the order of grasping, extension and flexion. The asymmetry index of each point was Susamni($LI_{10}$) $16.26{\pm}13.59%$, Oegwan($TE_5$) $20.38{\pm}15.59%$, below Sohae($HT_3$) two cun $20.89{\pm}16.77%$, Naegwan($PC_6$) $22.49{\pm}14.91%$ in wrist extension, Susamni($LI_{10}$) $25.99{\pm}21.44%$, Oegwan($TE_5$) $21.15{\pm}15.94%$, below Sohae($HT_3$) two cun $19.62{\pm}15.46%$, Naegwan($PC_6$) $19.93{\pm}16.85%$ in wrist flexion, Susamni($LI_{10}$) $16.60{\pm}12.21%$, Oegwan($TE_5$) $10.94{\pm}8.29%$, below Sohae($HT_3$) two cun $15.20{\pm}12.60%$, Naegwan($PC_6$) $11.68{\pm}7.77%$ in grasping. Conclusions : In this study, to identify the degree of symptoms of diseases in the upper extremities and confirm therapeutic effects, it is necessary to analyze the calculated percentage and compare the SEMG measurement of special points with those of other points, and with the asymmetry index.

Suppression of Cyclooxygenase-2 Expression in Colonic Epithelial Cells by Ilekudinol B Isolated from Weigela subsessilis

  • Ilekudinol B is one of the flavonoids isolated from Weigela subsessilis (Caprifoliaceae). In the present study, the suppression effect of ilekudinol B on tumor necrosis factor $(TNF)-{\alpha}-induced$ cyclooxygenase-2 (COX-2) expression was investigated in human colon epithelial cell line HT-29. Interleukin-8 (IL-8) production and prostaglandin $E_2\;(PGE_2)$ secretion was measured by enzyme-linked immunosorbent assay (ELISA). COX-2 and nuclear factor $(NF)-{\kappa}B$ expression were determined by Western blot analysis. Ilekudinol B significantly inhibited $TNF-{\alpha}-induced$ secretion of IL-8 and prostaglandin $E_2\;(PGE_2)$ from the human colon epithelial cell line HT-29 in a concentration-dependent manner. In addition, ilekudinol B remarkably diminished $TNF-{\alpha}-induced$ COX-2 expression and $NF-{\kappa}B$ p65 subunit translocation to the nucleus. In conclusion, our results indicate that ilekudinol B may have anti-inflammatory activity on $TNF-{\alpha}-dependent$ colonic inflammation.

Cytoprotective Effects of Phaeophyta Extracts from the Coast of Jeju Island in HT-22 Mouse Neuronal Cells (제주 연안 갈조류 추출물의 신경세포 보호효과)

  • Marine algae have long been recognized as a health and beauty food, based on its anti-tumor, anti-inflammatory and anti-obesity activities. In this study, methanol extracts were prepared from 10 different phaeophyta, after which DPPH radical scavenging and cytoprotective activities of HT-22 cells against ${\beta}$-amyloid protein ($A{\beta}$), which has neurotoxic effects, were investigated. In DPPH experiments, Ecklonia cava and Ishige okamurai showed strong ROS scavenging activities, whereas eight other phaeophyta including Petalonia binghamiae (P. bin) showed weak ROS scavenging activities. To validate the cytoprotective effects of 10 different phaeophyta in $A{\beta}$-induced HT-22 cells, protein expression levels of APP, BACE1, iNOS, phosphorylated ERK1/2, phosphorylated p38 and phosphorylated JNK1/2 were determined along with MTT assay. In the MTT assay, P. bin showed the best effective cytoprotective activity at a concentrations of $25{\mu}g/mL$, whereas Sargassum confusum, Colpomenia sinuosa, Myelophycus simplex, and Sargassum hemiphyllum showed potential. Determination of protein expression levels related to $A{\beta}$-induced neurotoxicity in the five selected phaeophyta showed that P. bin inhibited BACE1 and iNOS expression in $A{\beta}$-induced HT-22 cells. These results indicate that the cytoprotective effects of P. bin are mediated by suppressing the pathways involving $A{\beta}$-induced ERK and p38 activation.

  • The twelve-protostane analogues were synthesized from alisol B 23-acetate and assessed for their in vitro antitumor activity against six different human and murine tumor cell lines. Of the compounds synthesized, 23S-acetoxy-24R(25)-epoxy-11$\beta$,23S-dihydroxyprotost-13(17)-en-3-hy-droxyimine (12) exhibited significant cytotoxic activities against A549, SK-OV3, B16-F10, and HT1080 tumor cells with $ED_{50}/$ values of 10.0, 8.7 ,5.2, and 3.1 ${\mu}g$/ml, respectively. Furthermore, 23S-acetoxy-13(17),24R(25)-diepoxy-11$\beta$-hydroxyprotost-3-one (5), 13(17),24R(25)-diepoxy-11$\beta$, 23S-dihydroxyprotostan-3-one (6), 24R,25-epoxy-11$\beta$,23S-dihydroxyprotost-13(17)-en-3-one (7), and 11$\beta$,23S,24R,25-tetrahydroxyprotost-13(17)-en-3-one (9) showed moderate cytotoxic activities against 816-F10 and HT1080 tumor cells. These results mean that a hydroxyimino group at C-3 position in the protostane-type terpene enhances cytotoxic activity.

Physiological Responses of Starry Flounder Platichthys stellatus during Freshwater Acclimation with Different Speeds in Salinity Change (염분변화 속도를 달리한 담수순화 과정에서 강도다리 Platichthys stellatus의 생리학적 반응)

  • Physiological responses (hematological factors, cortisol, glucose, osmolality, $Na^+$, $K^+$ and $Cl^-$) in starry flounder Platichthys stellatus were investigated during freshwater acclimation in the conditions of different speeds in salinity change with acute-decrease (AD) or stepwise-decrease (SD I and II). In AD of acute-decrease salinity, hematocrit (Ht), red blood cell (RBC) and hemoglobin (Hb) were rapidly increased more than SD I of stepwise-decrease salinity. But in case of SD II, Ht, RBC and Hb were no significant difference from beginning to end of this experiments. In AD, cortisol level significantly increased from $2.1{\pm}1.0{\mu}g/mL$ at the beginning to $13.7{\pm}0.2{\mu}g/mL$ at 6 hours and recovered to the basal levels ($3.1{\mu}g/mL$) at 10 days. In SD I, cortisol level was significantly increased from $2.1{\pm}1.0{\mu}g/mL$ at the beginning to $13.6{\pm}0.6{\mu}g/mL$ at 6 hours and recovered to the basal levels ($3.1{\pm}0.4{\mu}g/mL$) at 10 days. In SD II, cortisol level was a little increased from $2.1{\pm}1.0{\mu}g/mL$ at the beginning to $10.5{\pm}2.5$, $10.8{\pm}5.6{\mu}g/mL$ at 6, 12 hours and recovered to the basal level at 48 hours. Glucose level of AD, SD I, II were no significant difference from beginning to end of this experiments. Osmolality was $286.8{\pm}3.3\;mOsm/kg$ at the beginning. In SD II of stepwise-decrease, osmolality was no significant difference during rearing in freshwater (FW). But AD of stepwise-decrease and SD I of stepwise-decrease, osmolality was a little decreased end of this experiments. In AD of acute-decrease, only $Cl^-$ level was showed no significant difference from beginning to end of experiment and $Na^+$, $K^+$ levels were decreased. In case of SD I, $Cl^-$ level was showed no significant difference from beginning to end of experiment and $Na^+$, $K^+$ levels were decreased.

In vitro evaluation of fracture strength of zirconia restoration veneered with various ceramic materials

  • PURPOSE. Fracture of the veneering material of zirconia restorations frequently occurs in clinical situations. The purpose of this in vitro study was to compare the fracture strengths of zirconia crowns veneered with various ceramic materials by various techniques. MATERIALS AND METHODS. A 1.2 mm, $360^{\circ}$ chamfer preparation and occlusal reduction of 2 mm were performed on a first mandibular molar, and 45 model dies were fabricated in a titanium alloy by CAD/CAM system. Forty-five zirconia copings were fabricated and divided into three groups. In the first group (LT) zirconia copings were veneered with feldspathic porcelain by the layering technique. In the second group (HT) the glass ceramic was heat-pressed on the zirconia coping, and for the third group (ST) a CAD/CAM-fabricated high-strength anatomically shaped veneering cap was sintered onto the zirconia coping. All crowns were cemented onto their titanium dies with Rely $X^{TM}$ Unicem (3M ESPE) and loaded with a universal testing machine (Instron 5583) until failure. The mean fracture values were compared by an one-way ANOVA and a multiple comparison post-hoc test (${\alpha}$= 0.05). Scanning electron microscope was used to investigate the fractured interface. RESULTS. Mean fracture load and standard deviation was $4263.8{\pm}1110.8$ N for Group LT, $5070.8{\pm}1016.4$ for Group HT and $6242.0{\pm}1759.5$ N for Group ST. The values of Group ST were significantly higher than those of the other groups. CONCLUSION. Zirconia crowns veneered with CAD/CAM generated glass ceramics by the sintering technique are superior to those veneered with feldspathic porcelain by the layering technique or veneered with glass ceramics by the heat-pressing technique in terms of fracture strength.

  • The maximum breathing capacity (MBC) and the maximum mid-expiratory flow rate (MMF) are widely used in evaluation of the ventilatory function, among various parameters of pulmonary function. The MBC volume is the amount of gas which can be exchanged per unit time during maximal voluntary hyperventilation. Performance of this test, unlike that of single breath maneuvers, is affected by the integrity of the respiratory bellows as a whole including such factors are respiratory muscle blood supply, fatigue, and progressive trapping of air. Because of this, the MBC and its relation to ventilatory requirement correlates more closely with subjective dyspnea than does any other test. The MMF is the average flow rate during expiration of the middle 50% of the vital capacity. The MMF is a measurement of a fast vital capacity related to the time required for the maneuver and the MMF relates much better to other dynamic tests of ventilatory function and to dyspnea than total vital capacity, because the MMF reflects the effective volume, or gas per unit of time. Therefore, it is important to have a prediction formula with one can compute the normal value for the subject and the compare with the measured value. However, the formulas for prediction of both MBC and MMF of the Korean children and adolescents are not yet available in the present. Hence, present investigation was attempt to derive the formulas for prediction of both MBC and MMF of the Korean children and adolescents. MBC and MMF were measured in 1,037 healthy Korean children and adolescents (1,035 male and 1,002 female) whose ages ranged from 8 to 18 years. A spirometer (9L, Collins) was used for the measurement of MBC and MMF. Both MBC and MMF were measured 3times in a standing position and the highest values were used. For measurement, the $CO_2$ absorber and sadd valve were removed from the spirometer in order to reduce the resistance in the breathing circuit and the subject was asked to breathe as fast and deeply as possible for 12 seconds in MBC and to exhale completely as fast as possible after maximum inspiration for MMF. During the measurement, investigator stood by the subject to give a constant encouragement. All the measured values were subsequently converted to values at BTPS. The formulas for MBC and MMF were derived by a manner similar to those for Baldwin et al (1949) and Im (1965) as function of age and BSA or age and height. Inhibitory Effect of Ginseng Saponins and Polysaccharides on Infection and Vacuolation of Helicobacter pylori

  • Ginsenosides and polysaccharides were isolated from Panax ginseng C.A. Meyer (Family Araliaceae) by treating at low ($60^{\circ}C$, LT), mild ($100^{\circ}C$, MT), and high ($120^{\circ}C$, HT) temperatures, and their inhibitory effects on growth, infection, and VacA vacuolation of Helicobacter pylori (HP) were investigated. The molecular weights of polysaccharides decreased as the processing temperature increased. Ginseng polysaccharides inhibited the HP infection into KATO III cells, but did not inhibit growth of HP and VacA vacuolation of HeLa cells. HT polysaccharides showed the most potent inhibition with $IC_50$ value of 6.8 mg/ml. Ginseng saponins did not inhibit the infection of HP into KATO cells. However, 20(s)-protopanaxadiol showed the most potent inhibition of HP growth and vacuolation of HeLa by VacA toxin with $IC_50$ values of 0.05 and 0.067 mg/ml, respectively.

The Antimutagenic Activity and the Growth Inhibition Effect of Cancer Cells on Methanol Extracts from Small Water Dropwort (돌미나리 메탄올 추출물의 항돌연변이 작용과 암세포증식 억제효과)

  • The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.

In vitro Cytotoxic Effect of Extracts from Styela plicata (오만둥이(Styela plicata) 추출물의 in vitro 세포독성 효과)

  • The present study describes the preliminary evaluation of the anticancer activity of Styela plicata. Freeze-dried S. plicata was extracted with methanol, ethanol, acetone, and water, and then anticancer effect of the extracts was measured by the MTT reduction assay and phase-contrast microscopy on the HT-29 human colon carcinoma cells. Among the extracts, acetone extract showed the highest anticancer activity. The cell proliferation rates markedly decreased by 94.0% at the concentration of 500 ${\mu}g/mL$ of acetone extract compared with control cells. The acetone extract was further fractionated with hexane, diethyl ether, ethyl acetate, and water layer according to the degree of polarity. The HT-29 cells with hexane layer extract (250 ${\mu}g/mL$) decreased the cell viability to 5.1% of untreated control. The growth of SW620, HeLa, and MCF-7 cells was decreased to about 10%, by the treatment of hexane layer extract 250 ${\mu}g/mL$. Theses results suggest extracts from S. plicata as possible natural cancer therapeutic material.
