Cloning, Expression and Genomic Organization of Genes Encoding Major Royal Jelly Protein 1 and 2 of the Honey Bee (Apis cerana)

  • Major Royal Jelly Protein cDNAs of Apis cerana (AcMRJP) were cloned and characterized. The open reading frames (ORFs) of the AcMRJP1 and AcMRJP2 genes were 1302 and 1392 nucleotides, encoding 433 and 463 amino acid residues, respectively. The sequence divergences between AcMRJP1 and AcMRJP2 and their corresponding protein families in A. mellifera were 0.0618 and 0.0934 at the nucleotide level and 0.0912 and 0.1438 at the protein level, respectively. Phylogenetic analysis supports the orthologous similarity between these proteins. The deduced amino acids indicated high essential amino acid contents of AcMRJP1 and AcMRJP2 (47.5 and 44.8%, respectively). The genomic organization of both AcMRJP1 and AcMRJP2 was determined. Both the AcMRJP1 (3663 bp) and AcMRJP2 (3963 bp) genes contained six exons and five introns, where all boundaries conformed to the GT/AG rule. AcMRJP1 and AcMRJP2 cDNAs were cloned into pET17b, and both the recombinant (r) AcMRJP1 (47.9 kDa) and rAcMRJP2 (51.7 kDa) were expressed in the insoluble form. Western blot analysis and N-terminal sequencing of the solubilized proteins revealed successful expression of rAcMRJP1 and rAcMRJP2 in vitro. The yields of the purified rAcMRJP1 and rAcMRJP2 were approximately 20 and 8mg protein per liter of the flask culture, respectively.

  • The authors carried out an experiment to measure the hydrodynamic resistance of the gear and the efficiency of otter board of the midwater trawl, which is the same used in the former experiment in this series of studies. The whole resistance of the gear was measured by a 10 ton scale recording tension meter, and they were analyzed into the shearing force and the drag with relevant factors determined in the former experiments. The results obtained can be summarized as follows: 1. The whole drag of gear T(kg) and the drag of net R sub(N) (kg) can be expressed as T=2.15 v super(1.12). R sub(N)=1.96 v super(1.01) (v: towing speed in m/sec) 2. The formula of estimating the drag of net deduced by Koyama's method can be expressed as R sub(N)=4.3$\times$d/l$\times$abv 3. The shearing force and the drag of otter board is about 19 to 22% and 5 to 7% of the drag of net, respectively. Whereas, the shearing coefficient and the drag coefficient calculated by the resistance and the opening of gear are 1.5 and 0.42, respectively.

Pharmacokinetic Characteristics of Levosulpiride in Relation to the Genetic Polymorphism of MDR1: From Knockout Mouse to Human

  • The purposes of this study were to clarify the involvement of P-glycoprotein (P-gp) in the efflux of levosulpiride in knockout mice that lack the mdr1a1b gene and to evaluate the relationship between the genetic polymorphisms in MDR1 gene (exon 21) and levosulpiride disposition in healthy Korean subjects. After oral administration ($10\;{\mu}g/g$) of levosulpiride to mdr1a/1b(-/-) and wild-type mice, plasma and brain samples were obtained at 45 min. We also investigated the genotype for MDR1 (exon 21) gene in humans using a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. A single oral dose of 25 mg levosulpiride was administered to 58 healthy subjects, who were based on the MDR1 genotype for the G2677T SNP. Blood samples were taken up to 36 hr after dosing. The concentrations of levosulpiride in mouse plasma and brain were statistically significant difference between the two animal groups (P<0.05). In addition, the average brain-to-plasma concentration ratio (Kp) of levosulpiride was 3.4-fold (P<0.01) higher in the mdr1a/1b(-/-) mice compared with the wild-type mice. We also found that the values of $AUC_{0-{\infty}$, partial AUC ($AUC_{0-4h}$) and $C_{max}$ were significantly different between homozygous 2677TT subjects and the subjects with at least one wild-type allele (GG and GT subjects, P=0.012 for $AUC_{0-{\infty}$; P=0.008 for $AUC_{0-4h}$; P=0.038 for $C_{max}$). The results confirm that levosulpiride is a P-gp substrate in vivo, and clearly demonstrate the effect of SNP 2677G>T in exon 21 of the MDR1 gene on levosulpiride disposition.

Associations of ABCB1 and XPC Genetic Polymorphisms with Susceptibility to Colorectal Cancer and Therapeutic Prognosis in a Chinese Population

  • Associations between ABCB1 and XPC genetic polymorphisms and risk of developing colorectal cancer (CRC) as well as clinical outcomes in CRCs with chemotherapy were investigated. A case-control study was performed on the ABCB1 C3435T, G2677T/A and XPC Lys939Gln polymorphisms in 428 CRC cases and 450 hospitalbased, age and sex frequency-matched controls using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) assays. We observed that the ABCB1 3435CT or CC+CT variants were significantly linked with increasing risk of developing CRC (adjusted OR (95% CI): 1.814 (1.237-2.660), P=0.0022; adjusted OR (95% CI): 1.605 (1.117-2.306), P=0.0102, respectively). Moreover, the distribution frequency of XPC AC genotype or AC+CC genotypes also showed a tendency towards increasing the suscepbility for CRC (P=0.0759 and P=0.0903, respectively). Kaplan-Meier curves showed that the ABCB1 C3435T variant was associated with a tendency toward longer progression-free survival (PFS) (n=343, Log-rank test: P=0.063), and the G2677T/A variant genotypes (GT+TT+GA+AA) with a tendency for longer OS in postoperative oxaliplatin-based patients (n=343, Log-rank test: P=0.082). However, no correlation of the XPC Lys939Gln polymorphism was found with PFS and OS in patients with postoperative oxaliplatin-based chemotherapy (n=343). Our study indicated that ABCB1 polymorphisms might be candidate pharmacogenomic factors for the prediction of CRC susceptibility, but not for prognosis with oxaliplatin chemosensitivity in CRC patients.

The Small GTPase CsRAC1 Is Important for Fungal Development and Pepper Anthracnose in Colletotrichum scovillei

  • The pepper anthracnose fungus, Colletotrichum scovillei, causes severe losses of pepper fruit production in the tropical and temperate zones. RAC1 is a highly conserved small GTP-binding protein in the Rho GT-Pase family. This protein has been demonstrated to play a role in fungal development, and pathogenicity in several plant pathogenic fungi. However, the functional roles of RAC1 are not characterized in C. scovillei causing anthracnose on pepper fruits. Here, we generated a deletion mutant (𝜟Csrac1) via homologous recombination to investigate the functional roles of CsRAC1. The 𝜟Csrac1 showed pleiotropic defects in fungal growth and developments, including vegetative growth, conidiogenesis, conidial germination and appressorium formation, compared to wild-type. Although 𝜟Csrac1 was able to develop appressoria, it failed to differentiate appressorium pegs. However, 𝜟Csrac1 still caused anthracnose disease with significantly reduced rate on wounded pepper fruits. Further analyses revealed that 𝜟Csrac1 was defective in tolerance to oxidative stress and suppression of host-defense genes. Taken together, our results suggest that CsRAC1 plays essential roles in fungal development and pathogenicity in C. scovilleipepper fruit pathosystem.

Identification of a Novel Single Nucleotide Polymorphism in Porcine Beta-Defensin-1 Gene

  • Porcine beta-defensin-1 (PBD-1) gene plays an important role in the innate immunity of pigs. The peptide encoded by this gene is an antimicrobial peptide that has direct activity against a wide range of microbes. This peptide is involved in the co-creation of an antimicrobial barrier in the oral cavity of pigs. The objective of the present study was to detect polymorphisms, if any, in exon-1 and exon-2 regions of PBD-1 gene in Large White Yorkshire (LWY) and native Ankamali pigs of Kerala, India. Blood samples were collected from 100 pigs and genomic DNA was isolated using phenol chloroform method. The quantity of DNA was assessed in a spectrophotometer and quality by gel electrophoresis. Exon-1 and exon-2 regions of PBD-1 gene were amplified by polymerase chain reaction (PCR) and the products were subjected to single strand conformation polymorphism (SSCP) analysis. Subsequent silver staining of the polyacrylamide gels revealed three unique SSCP banding patterns in each of the two exons. The presence of single nucleotide polymorphisms (SNPs) was confirmed by nucleotide sequencing of the PCR products. A novel SNP was found in the 5'-UTR region of exon-1 and a SNP was detected in the mature peptide coding region of exon-2. In exon-1, the pooled population frequencies of GG, GT, and TT genotypes were 0.67, 0.30, and 0.03, respectively. GG genotype was predominant in both the breeds whereas TT genotype was not detected in LWY breed. Similarly, in exon-2, the pooled population frequencies of AA, AG, and GG genotypes were 0.50, 0.27, and 0.23, respectively. AA genotype was predominant in LWY pigs whereas GG genotype was predominant in native pigs. These results suggest that there exists a considerable genetic variation at PBD-1 locus and further association studies may help in development of a PCR based genotyping test to select pigs with better immunity.

Development and Characterization of PCE-to-Ethene Dechlorinating Microcosms with Contaminated River Sediment

  • An industrial complex in Wonju, contaminated with trichloroethene (TCE), was one of the most problematic sites in Korea. Despite repeated remedial trials for decades, chlorinated ethenes remained as sources of down-gradient groundwater contamination. Recent efforts were being made to remove the contaminants of the area, but knowledge of the indigenous microbial communities and their dechlorination abilities were unknown. Thus, the objectives of the present study were (i) to evaluate the dechlorination abilities of indigenous microbes at the contaminated site, (ii) to characterize which microbes and reductive dehalogenase genes were responsible for the dechlorination reactions, and (iii) to develop a PCE-to-ethene dechlorinating microbial consortium. An enrichment culture that dechlorinates PCE to ethene was obtained from Wonju stream, nearby a trichloroethene (TCE)-contaminated industrial complex. The community profiling revealed that known organohalide-respiring microbes, such as Geobacter, Desulfuromonas, and Dehalococcoides grew during the incubation with chlorinated ethenes. Although Chloroflexi populations (i.e., Longilinea and Bellilinea) were the most enriched in the sediment microcosms, those were not found in the transfer cultures. Based upon the results from pyrosequencing of 16S rRNA gene amplicons and qPCR using TaqMan chemistry, close relatives of Dehalococcoides mccartyi strains FL2 and GT seemed to be dominant and responsible for the complete detoxification of chlorinated ethenes in the transfer cultures. This study also demonstrated that the contaminated site harbors indigenous microbes that can convert PCE to ethene, and the developed consortium can be an important resource for future bioremediation efforts.

Cosmological Tests using Redshift Space Clustering in BOSS DR11

  • We analyze the clustering of large scale structure in the Universe in a model independent method, accounting for anisotropic effects along and transverse to the line of sight. A large sample of 690,000 galaxies from The Baryon Oscillation Spectroscopy Survey Data Release 11 are used to determine the Hubble expansion H, angular distance D_A, and growth rate GT at an effective redshift of z=0.57. After careful bias and convergence studies of the effects from small scale clustering, we find that cutting transverse separations below 40 Mpc/h delivers robust results while smaller scale data leads to a bias due to unmodelled nonlinear and velocity effects. The converged results are in agreement with concordance LCDM cosmology, general relativity, and minimal neutrino mass, all within the $68{\backslash}%$ confidence level. We also present results separately for the northern and southern hemisphere sky, finding a slight tension in the growth rate -- potentially a signature of anisotropic stress, or just covariance with small scale velocities -- but within $68{\backslash}%$ CL.

  • The battery of an electric vehicle is a key part of the energy supply to operate the vehicles. There are many factors affecting battery life such as charging method, discharge rate, and ambient temperature those are requires systematic monitoring and management. To solve the issues like environmental problems and fuel consumption reduction the battery needs more performance improvement. In this study, it was analyzed the thermal characteristics and securing battery stability for electric vehicle battery cooling system. The simulation test was operated using GT-suite software with several conditions like cooling capacity 1, 2 and 4 kW, cooling flow rate 5, 10, 20 and 30 LPM, and battery initial temperatures 40, 35, and 30℃ at the temperature of ambient 25℃. The results shown that the case of cooling flow rate at 20 LPM was most efficient among all above conditions.

  • 최근의 R&D동향은 각 분야에서 발전해온 기술들을 집약하여 다른 분야로 그 영역을 확대 적용함으로써 융 복합기술의 신규 영역창출을 도모하고, 고유영역의 한계를 극복하는 신 패러다임의 기술시대로 진입하고 있다. 신재생에너지원을 수자원 설비에 적용하는 수상태양광발전 융합기술은 저수지 공유수면 개발 이용을 통한 국토의 효율적 활용, 수자원을 이용한 친환경에너지생산 및 녹조류 제거, 어류번식 등을 할 수 있는 고부가가치 하이테크 기술로 기존의 제한된 수자원설비의 이용기술 영역을 탈피하여, 신재생에너지, IT, BT기술이 융합된 GT기술로 미래형 친환경 저수지 실증기술로 개발이 가능한 분야이다. 이 기술이 상용화 될 경우 K-water가 보유한 다목적댐 저수지 유역면적의 1(%)만 개발해도 352(MW)정도의 친환경에너지를 생산 할 수 있을 것으로 추정하고 있다. 국토가 협소한 우리나라의 경우 그동안 태양광개발로 인해 임야 및 농지 등의 환경훼손이 심각한 수준에 이르고 있어 신재생에너지개발이 다른 양상의 환경파괴를 일으키고 있다. 수상 태양광발전기술은 자연과 어우러진 친환경융합기술로 태양광부문의 태양광모듈기술의 비약적 발전과 가격 경쟁력확보, 주변기술 등이 획기적으로 개발 될 경우 친환경수자원 분야의 미래성장 동력으로 자리매김 할 수 있을 것으로 판단된다.
