Development of a Biofungicide Using a Mycoparasitic Fungus Simplicillium lamellicola BCP and Its Control Efficacy against Gray Mold Diseases of Tomato and Ginseng

  • To develop a commercial product using the mycoparasitic fungus Simplicillium lamellicola BCP, the scale-up of conidia production from a 5-l jar to a 5,000-l pilot bioreactor, optimization of the freeze-drying of the fermentation broth, and preparation of a wettable powder-type formulation were performed. Then, its disease control efficacy was evaluated against gray mold diseases of tomato and ginseng plants in field conditions. The final conidial yields of S. lamellicola BCP were $3.3{\times}10^9conidia/ml$ for a 5-l jar, $3.5{\times}10^9conidia/ml$ for a 500-l pilot vessel, and $3.1{\times}10^9conidia/ml$ for a 5,000-l pilot bioreactor. The conidial yield in the 5,000-l pilot bioreactor was comparable to that in the 5-l jar and 500-l pilot vessel. On the other hand, the highest conidial viability of 86% was obtained by the freeze-drying method using an additive combination of lactose, trehalose, soybean meal, and glycerin. Using the freeze-dried sample, a wettable powder-type formulation (active ingredient 10%; BCP-WP10) was prepared. A conidial viability of more than 50% was maintained in BCP-WP10 until 22 weeks for storage at $40^{\circ}C$. BCP-WP10 effectively suppressed the development of gray mold disease on tomato with control efficacies of 64.7% and 82.6% at 500- and 250-fold dilutions, respectively. It also reduced the incidence of gray mold on ginseng by 65.6% and 81.3% at 500- and 250-fold dilutions, respectively. The results indicated that the new microbial fungicide BCP-WP10 can be used widely to control gray mold diseases of various crops including tomato and ginseng.

  • 자몽씨추출물(GSE)과 ZnONP을 아가필름에 동시에 첨가하여 다기능성 복합필름을제조하였다. Agar/GSE/ZnONP복합필름은 특히 자외선차단성과 항균성에 대한 시너지 효과를 나타냈다. 광 차단성은 자외선 파장범위인 280 nm와 가시광선 파장범위인 660 nm에서 유의적인 차이를 보이며 크게 나타났다. 자외선차단성은 agar/GSE와 agar/ZnONP 필름에 비해 agar/ZnONP3%/GSE5% 필름에서의 크게 상승하여 GSE와 ZnONP의 시너지 효과를 확인할 수 있었다. 항균성으로는 그람양성균인 L. monocytogenes와 그람음성균인 E. coli O157: H7 모두 agar/ZnONP3%/GSE5% 필름에서 강한 항균성 및 시너지 효과가 있었다. 자외선 차단성과 항균성은 포장식품의 안전성을 확보하고 유통기한을 연장시키는데 필수적인 특성으로 agar/ZnONP3%/GSE5% 필름은 식품의 유통기한 연장과 식품의 안전성을 위한 포장필름으로 활용 가치가 높을 것으로 사료된다.

  • 중성염에 용해한 후 얻어진 견피브로인 수용액의 pH를 3.0으로 조정하였을 때 피브로인 분자쇄들간에 정전기적 반발력이 감소하여 피브로인 분자쇄 상호간에 결합력이 증가하므로 겔화시간이 단축되었고 적외선분광분석 결과 ${\beta}$-sheet 구조로의 전이가 빨리 일어나고 있음을 확인하였다. 2. Silk fibroin 용액내에 glycerol 함량비가 클수록 겔화의 시간은 크게 단축되었으나 상대적인 수분함량의 증가로 강도는 낮아졌다. 3. Silk fibroin 용액내에 피브로인의 농도가 높을수록 겔의 강도는 낮아졌다. 4. 효소분해된 Silk fibroin 용액을 pH3.0으로 조정 후 실온에서 보관하였을 때 분자량저하에 따른 분자쇄간 결합력 약화로 겔상을 이루지 못하고 흰색의 분말상 침전을 형성하였으나 pH 3.0 조정 후 원심분리하면 겔상을 얻을 수가 있었다. 5. 시차주사열분석결과, 견피브로인 수용액을 등전점 조절하였을 때에는 재결정화에 의한 발열피크가 나타나지 않았고 결정성이 좀더 높았으며 효소처리한 경우에는 분자량감소로 낮은 온도 부근에서 일부 열분해가 일어났으나 저분자화된 분자쇄들이 새로운 결합을 형성하여 열적안정성이 다소 높게 나타났다.

TLR9 Expression in Uterine Cervical Lesions of Uyghur Women Correlate with Cervical Cancer Progression and Selective Silencing of Human Papillomavirus 16 E6 and E7 Oncoproteins in Vitro

  • Background: Cervical cancer is listed as one of high-incidence endemic diseases in Xinjiang. Our study aimed to evaluate the expression of TLR9 in uterine cervical tissues of Uyghur women and examine associations with clinicopathological variables. We further characterized the direct effects of TLR9 upon the selective silencing of human papillomavirus (HPV) E6 and E7 oncoprotein expression in HPV 16-positive human cervical carcinoma cells treated with siRNA in vitro. Materials and Methods: Immunohistochemistry was applied to evaluate TLR9 expression in 97 formalin-fixed paraffin-embedded cervical samples from Uyghur women; 32 diagnosed with cervical squamous cell carcinomas (CSCC), 14 with low-grade cervical intraepithelial neoplasias (CINI), 10 medium-grade (CINII), 24 high-grade (CINIII), and 17 chronic cervicitis. $BLOCK-iT^{TM}$ U6 RNAi Entry Vector $pENTR^{TM}$/U6-E6 and E7 was constructed and transfected the entry clone directly into the mammalian cell line 293FT. Then the HPV 16-positive SiHa human cervical carcinoma cell line was infected with RNAi recombinant lentivirus. RT-PCR and Western blotting were used to determine the expression of TLR9 in both SiHa and HPV 16 E6 and E7 silenced SiHa cells. Results: Immunohistochemical staining showed that TLR9 expression was undetectable (88.2%) or weak (11.8%) in chronic cervicitis tissues. However, variable staining was observed in the basal layer of all normal endocervical glands. TLR9 expression, which was mainly observed as cytoplasmic staining, gradually increased in accordance with the histopathological grade in the following order: chronic cervicitis (2/17, 11.8%)

$CH_{3}CN$ 감지를 위한 $SnO_{2}/Al_{2}O_{3}/Pd$ 후막소자의 제조 및 그 특성 (Fabrication and Characteristics of $SnO_{2}/Al_{2}O_{3}/Pd$ Thick Film Devices for Detection of $CH_{3}CN$ Vapor)

  • $CH_{3}CN$ 감지를 위한 최적 모물질은 $CH_{3}CN$의 억분해 온도와 생성량을 적외선 흡수 스펙트럼으로부터 비교함으로써 선정되었다. $SnO_{2}$ 표면에서 $CH_{3}CN$$130^{\circ}C$에서부터 열분해되기 시작하여 $300^{\circ}C$에서는 많은 양의 생성물을 생성하였다. 산화반응에 의한 $CH_{3}CN$$CO_{2}$, $NH_{3}$$H_{2}O$로 열분해되었으며, $320^{\circ}C$에서부터 $N_{2}O$가 생성되기 시작하였다. $SnO_{2}$ 감지소자의 $CH_{3}CN$에 대한 감지특성은 $CH_{3}CN$과 금속산화물과의 산화반응으로 인해 생성된 흡착종에 의해 영향을 받았다. 감지물질표면과의 반응에서 생성된 흡착종은 CO, $NH_{3}$, $H_{2}O$$NO_{x}$ 등이었다. $NO_{x}$의 생성량은 감지특성에 큰 영향을 나타냄을 알 수 있다. 170 ppm의 $CH_{3}CN$에 대한 $SnO_{2}$의 감도와 동작온도는 각각 70% 정도와 $300^{\circ}C$이었다. 0.2wt% Pd 첨가된 $SnO_{2}/Al_{2}O_{3}/Pd$ 감지소자는 $CH_{3}CN$에 대해 높은 감도를 나타내었으며, 응답시간은 약 10초이었다.

Xanthomonas campestris pv. sesami에 의한 참깨의 세균성잎마름병 (Bacterial Blight of Sesame Caused by Xanthomonas campestris pv. sesami)

  • 2000년 수원, 홍천, 연천 지역의 참깨 포장에서 새로운 병해가 발견되었다. 처음에는 잎에 수침상의 작은 점무늬를 형성하다가 점점 커져 괴사가 일어나고 주위로 황화되었다. 심하게 감염된 경우는 병든 잎은 떨어졌다. YDC 배지에서 병원세균을 순수 분리 하였을 때 Xanthomonas 속 세균의 전형적인 특징인 노란색 색소를 띤 세균이 형성되었다. 분리된 세균을 $10^{8}cfu/ml$로 현탁한 후 3주 동안 자란 참깨 잎에 분무 접종하였을 때 처음과 같은 증상이 재현되었다. 분리된 세균은 대조균주 X. campestris pv. sesami LMG865와 지방산 조성 및 다양한 탄소원 이용정도를 이용하여 비교하였을 때 $100\%$의 가능성으로 X. campestris pv. sesami로 동정되었다. 분리된 병원균은 $18\~36^{\circ}C$에서 생장이 양호하였으며, $27^{\circ}C$에서 가장 우수하였다. 이 보고는 국내에서 참깨의 세균성잎마름병 최초 보고이며, P. syringae pv. sesami에 의한 세균성점무늬병과 외부적인 병징으로 구별하기가 매우 어렵다.

열화율이 상이한 밀랍지의 초임계유체추출 탈랍처리효과 비교분석 (Comparisons of Effectiveness of the Supercritical Fluid Extraction Dewaxing on the Beeswax-Treated Paper with Different Aging Degrees)

  • This study aims to investigate the applicability of supercritical fluid extraction as a dewaxing technique to restore the beeswax-treated volume of the Annals of the Joseon Dynasty in various deterioration and damage conditions. Thus, this study analyzed the dewaxing efficiency and changes in physical and morphological properties before and after dewaxing, by applying the optimal supercritical fluid extraction dewaxing condition ($70^{\circ}C$, 40 MPa, $CO_2$, Co-solvent 20% DCM, 2 hour) to 3 kinds of beeswax-treated paper with different deterioration rates (dry artificial aging of 10, 20 and 30 days at $120^{\circ}C$). After dry artificial aging at $120^{\circ}C$ for 30 days, the average molecular weight of the beeswax-treated specimen was $1.856{\times}10^5g/mol$, showing deterioration about 80% of the beeswax-treated paper before dewaxing. It was a similar level to the molecular weight of the bees-waxed volume of the Annals of the Joseon Dynasty that has a higher degree of damage. As a result of analyzing the dewaxing efficiency through FT-IR analysis, this study discovered that it was possible to effectively dewax beeswax-treated paper in the range of deterioration 20 to 80% with this supercritical fluid extraction dewaxing technique applied. As a result of analyzing changes in the physical and morphological properties before and after dewaxing, the viscosity tended to decrease to a slight degree, and since no morphological deformation or damage of stencil fibers was found, it was concluded possible to conduct stable dewaxing through this supercritical fluid extraction technique.

초등학생의 Dental IQ 수준과 구강보건 관리실태 (Dental IQ and Oral Health Care Status of Elementary School Students)

  • Objectives: Oral health care program for the elementary school children (ESOHCP) should be met the first priority because dental caries are highly sensitive and prevalent among them. This study was performed to enhance the efficiency of ESOHCP, and their oral health promotion. Methods: The Subjects were 346 students (3rd grade 163, 6th grade 185) of one elementary school in Busan. Dental IQ and actual oral health status; the decayed, mixing and filling teeth were checked through questionnaire and oral health examination from April to June 2002. Dental IQ was presented out of one hundred, and data analysis was done using SAS (ver 8.1) program. Results: The students' cognitive level about the causative and preventive factor of dental caries was relatively high. However, the practical aspects of preventive behavior - tooth brushing; three times per day (20%), three minutes per time (22%), oral health examination; one time per three months (10%) left much to be desired. And, only 27% of the subjects had experienced in school oral health education. The mean level of dental IQ was 79 out of 100, and 51 % and 42% of them had the decayed and teeth with filling, respectively, with the rate of DMFT 82%, DT 43%, FT 57%. The level of dental IQ was higher in case of having his (her) own tooth brush (p=0.072), standard tooth brushing (p<0.001) three times per day, post meal, three minutes per time, present experience of oral health examination (p<0.001) and dental clinic visit (p<0.001). The grade of caries was more serious in case of 6th grade (p=0.059), an absence of his (her) own tooth brush (p= 0.090), present experience of oral health examination (p=0.021), and an absence of regular dental clinic visit (p=0.003). The frequency of oral health examination (γ= 0.620), tooth brushing; times per day (γ=0.445), post meal (γ=0.355), expending times per brushing (γ=0.352), right cognition to the treatment of caries (γ=0.401), positive attitude to dental treatment (γ=0.387), the frequency of dental clinic visit for the past one year (γ=0.152) showed significant correlation with dental IQ. In the multivariate analysis, dental IQ was influenced by the frequency of oral health examination, right cognition to the cause of caries, times of teeth brushing per day, right cognition to the treatment of caries, etc., with adjusted R2=0.857. Conclusion: Though the students' cognitive level about the causative and preventive factor of dental caries was high, the practical aspects of preventive behavior left much to be desired. Ant the current ESOHCP considered to be still inefficient. However, frequent dental visits had apparent correlation with hish level of dental IQ. The specialized and practical program should be needed for the efficient ESOHCP. Harmonized effort from educational, health and dental society is essential.

Synthesis and Properties of Arylacetylene Resins with Siloxane Units

  • A series of arylacetylene resins with siloxane units were synthesized by the condensation reactions of m-diethynylbenzene magnesium reagents with various $\alpha,\omega$-bis(chloro)dimethylsiloxanes. These resins are liquids and are miscible with common organic solvents at room temperature. The structures of the resins were characterized by FT-IR, $^1H$ NMR, $^{13}C$ NMR, $^{29}Si$ NMR, and gel permeation chromatography (GPC). The thermal behaviors of the resins were examined with differential scanning calorimetry (DSC). These resins have good processability. They can be thermally cross-linked through the ethynyl groups to produce cured resins. The thermal and thermooxidative stabilities of the cured resins were studied by thermogravimetric analysis (TGA). The cured resins possess high thermal and thermooxidative stability. Their decomposition occurs at above $500^{\circ}C$ in both $N_2$ and air. With increasing the length of siloxane units in the resins, the thermal stability of the cured resins decreases in $N_2$. When the cured resins were sintered above $1450^{\circ}C$ under argon, hard and glassy SiOC ceramics were obtained. These SiOC ceramics have the decomposition temperatures at 5% weight loss above $800^{\circ}C$ in air.

The Influence of Plasma Surface Modification on Frictional Property of Natural Rubber Vulcanizates

  • 라디오 주파수(13.56 MHz) 무전극 종형 플라즈마 반응기를 이용하여 천연고무 가교체의 표면을 클로로디플루오로메탄으로 처리하였다. FT-적외선 분광분석으로 표면개질 정도를 정성적으로 조사하였다. 플라즈마 처리표면의 마찰힘은 플라즈마 처리시간 증가에 따라 감소하였다. 고무표면에 에틸렌글리콜과 물을 떨어뜨려 접촉각을 측정한 결과 플라즈마 처리에 따라 감소하는 것으로 미루어 플라즈마 개질에 따라 표면극성이 증가하는 것을 확인하였다. 유리판 표면을 동일조건으로 플라즈마 처리한 경우는 극성의 감소만이 확인되었다. 표면자유에너지의 London 비극성 및 극성요소를 계산하는데 있어서 기하평균법과 조화평균법이 유용한 것으로 확인되었다. 평균방법에 관계없이 플라즈마 처리시간이 증가함에 따라 표면자유에너지는 증가하였다 그러나 조화평균법으로 계산된 자유에너지가 기하평균법으로 계산된 값에 비해 상대적으로 높았다. 플라즈마 표면개질은 마찰의 계면, 히스테리시스, 점성요소들에 영향을 미침으로써 마찰계수를 변화시키는 것으로 나타났다.
