Effect of Inonotus obliquus Extract on the Expression MMPs and HAS-2 (차가버섯 추출물의 MMPs 발현 저해 및 HAS-2 발현 증가효과)

  • In order to investigate the possibility of Inonotus obliquus extract as an active ingredient for wrinkle-care cosmetics, we prepared 70 % ethanolic extract of Inonotus obliquus and measured its DPPH radical scavenging activity and elastase inhibitory activity. We also evaluated the effect of Inonotus obliquus extract on expression of MMPs and HAS-2 in fibroblast and HaCaT cell, respectively. Inonotus Obliquus extract showed DPPH radical scavenging activity and elastase inhibitory activity in a dose-dependant manner ($SC_{50}$ = 91 ${\mu}g$/mL, $IC_{50}$=124 ${\mu}g$/mL). Inonotus obliquus extract inhibited the expression of MMP-1 mRNA 50 ~ 79 % at concentrations of 5 ~ 25 ${\mu}g$/mL, and reduced the expression of MMP-2 around 20 % at a concentration of 10 ${\mu}g$/mL. And it also reduced the expression of MMP-9 54 ~ 70 % in tconcentrations of 5 to 25 ${\mu}g$/mL. Furthermore, Inonotus obliquus extract increased HAS-2 mRNA expression in a dose-dependent manner in concentrations of 5 to 25 ${\mu}g$/mL without cytotoxicity. These results suggested that Inonotus Obliquus extract could be useful as an active ingredient for wrinkle-care cosmetics.

Culture study on the hybrid by interspecific crossing between Porphyra pseudolinearis and P. dentata (Bangiales, Rhodophyta), two dioecious species in culture

  • Physiological studies on the hybrid by crossing between two dioecious species, Porphyra pseudolinearis and P. dentata from Korea were conducted at constant temperatures (5, 10, 15, 20, and $25^{\circ}C$), at photon flux densities (10, 20, 40, and $80\;{\mu}mol\;m^{-2}s^{-1}$) under photoperiods (14 L : 10 D and 10 L : 14 D). In the hybrid, higher growth of conchocelis was observed at 20 and $40\;{\mu}mol\;m^{-2}s^{-1}$ under 14 L : 10 D. Conchosporangial branches were produced under $10-80\;{\mu}mol\;m^{-2}s^{-1}$ at only $25^{\circ}C$, and were abundant when the conchocelis was cultured under 10 L : 14 D. Foliose thalli of the hybrid grew well at the conditions of $10-20^{\circ}C$, 10 L : 14 D and $15-20^{\circ}C$, 14 L : 10 D. The foliose thalli grew very slowly at $5^{\circ}C$ and $30^{\circ}C$, respectively. No archeospores were observed at any culture conditions. Spermatangial and zygotosporangial sori were formed at the marginal portion of mature thallus. Zygotospores from the hybrid were released at $10-2^{\circ}C$ under both photoperiods, and gave rise to form conchocelis filament. Monoecious thalli were observed at $10^{\circ}C$ under 14 L : 10 D. Neither monospores nor protothalli were produced from the conchocelis in culture.

The Study of Zn, Cu, Mn, Ni Contents of Serum, Hair, Nail and Urine for Female College Students (일부 여대생의 혈청, 소변, 두발, 손톱 중의 아연, 구리, 망간, 니켈 함량에 관한 연구)

  • The purpose of this study was to evaluat the Zn, Cu, Mn and Ni status and correlation of their concentrations in serum, urine, hair and fingernail in female college students. As the results are follows; The mean age of subjects was 22.5 years and height, weight and BMI were 160.1 cm, 51.0 kg and 20.0 respectively. The daily intake of energy 1769.5 kcal and the ratio of carbohydrate, fat and protein for energy is 60 : 20 : 15. The daily intake of energy (88.5%), vitamin B2 (86.1%), Ca (75.4%), Fe (58.3%) and Zn (63.0%) of subjects did not reach to Korean Recommended Dietary Allowance (RDA). The daily mean intake of Zn, Cu, Mn and Ni were 7.56 mg, 2.30 mg, 3.81 mg and 0.18 mg respectively. The concentrations of Zn in serum, urine, hair and nail were 85.6$\mu\textrm{g}$/dL, 391.2$\mu\textrm{g}$/day, 174.6$\mu\textrm{g}$/g and 102.4$\mu\textrm{g}$/g respectively and those Cu were 84.2$\mu\textrm{g}$/dL, 56.6 $\mu\textrm{g}$/day, 20.3$\mu\textrm{g}$/g and 4.3$\mu\textrm{g}$/g respectively. The concentrations of Mn in serum, urine, hari and nail were 0.2$\mu\textrm{g}$/dL, 1.1$\mu\textrm{g}$/day, 1.8$\mu\textrm{g}$/g and 1.6$\mu\textrm{g}$/g respectively and those of Ni were 0.6$\mu\textrm{g}$/dL, 24.5$\mu\textrm{g}$/day, 3.5$\mu\textrm{g}$/g and 3.1$\mu\textrm{g}$/g respectively. The daily intake of Mn showed the positive correlations with concentration of Mn in hair (p<0.05) and the daily intake of Ni was positively correlated with urinary exretion (p<0.05) and hair contents (p<0.001) of Ni. The more detailed studies about these trace mineral status should be required.

Blood Lead, Manganese, Aluminium and Silicon Concentrations in Korean Adults (한국인의 연, 망간, 알루미늄 및 실리콘의 혈중 농도)

  • Objectives : This study was peformed to determine the reference values of blood lead, manganese, aluminium, and silicon in healthy adults. Methods : The subjects were 132 (67 male and 65 female), and classified to three age groups $(\leq39,\;40\sim49,\;and\;50\leq)$. Blood lead, manganese and aluminium were analyzed by atomic absorption spectrophotometer, and blood silicon was analyzed by direct current plasma optical emission spectrometer. Results : Blood lead levels(geometric mean, S.D) were (3.49, 1.70) ${\mu}g/dL$ in male and (3.04, 1.65) ${\mu}g/dL$ in female, but the difference is not significant, and there was no significant difference between age groups. Mean blood manganese level was $0.99{\pm}0.41{\mu}g/dL$, and there was no significant difference between sex or age groups. Mean blood aluminium level was $0.59{\pm}0.35{\mu}g/dL$, and there was no significant difference between sex or age groups. Mean blood silicon level was $54.41{\pm}27.64{\mu}g/dL$ in male and $43.34{\pm}23.51{\mu}g/dL$ in female, and the level in male was significantly higher than that in female (p<0.05). There was significant difference between age groups, and the oldest showed the highest level in male (p<0.05), but no significant difference between age groups in female. Conclusions : Authors hope that this study would provide basic data for determining reference values and evaluating health effects.

Protective Effects of Water dropwort and Jin-do turmeric on the Irradiation in Mouse (마우스에서 미나리와 진도 울금의 방사선조사에 대한 방어효과)

  • This paper searched the affect of Water dropwort and Tumeric on radiation exposure. By experimenting 50 male mouse, ICR type 5 weeks old male 25~30g, 5 mouse was divided in to each group respectively. A : 3 Gy control group, B : 5 Gy control group, C : 3 Gy, FW, D : 5 Gy FW, E : 3 Gy LW, F : 5 Gy LW, G : 3 Gy FT, H : 5 Gy FT, I : 3 Gy LT, J : 5 Gy LT FW : Ate Water dropwort before the radiation exposure. FT : Ate Tumeric before the radiation exposure. LW : Ate Water dropwort after the radiation exposure. LT : Ate Tumeric after the radiation exposure. During the 7 day radiation, Control group (A, B) received physiological saline (500mg/kg/day) in oral administration, LW and LT group (E, F, I, J) respectively received Water dropwort or tumeric (500mg/kg/day) in oral administration after the radiation, FW and FT group (C, D, G, H) respectively received Water dropwort or tumeric (500mg/kg/day) in oral administration before the radiation. As a result, an average leukocyte rate on 5 mouse was 200K/${\mu}L$ on group A. 6.46K/${\mu}L$ on group C which was in normal rate range (1.8-10.7 K/${\mu}L$). An average leukocyte rate was 23.33K/${\mu}L$ on group G. which was not normal rate, but compare to 200K/${\mu}L$, it was closer to normal rate. An average lymphocyte rate was 118.87K/${\mu}L$ on Group A. 1.51K/${\mu}L$ on Group G. which was in normal lymphocyte rate range (0.9-9.3K/${\mu}L$). An average thrombocyte rate was 4000K/${\mu}L$ on Group A. 1107 K/${\mu}L$ on Group C. 2759.6 K/${\mu}L$ on Group G. which was in normal thrombocyte rate range (592-2972 K/${\mu}L$). From this result, it was found that when exposed to radiation, Water dropwort and Tumeric have defensive effect in a preventive level. Also from an animal experiment, it was found that blood corpuscle rate returns to a normal rate. Based on this experiment, diverse research on this field should be conducted.

  • 본 연구는 청소년 스트레스 호르몬 변화를 보기위한 연구로 컬러 컨설팅 환경개선에 따른 스트레스 호르몬 변화를 연구하였다. 청소년 스트레스 연구들에 따르면, 아동이나 청소년도 성인만큼 스트레스를 경험하고 있고 청소년들의 스트레스는 정서적 부적응과 신체적 건강에도 영향을 미친다. 스트레스가 지속되면 부신피질에서 스테로이드 호르몬인 코티솔도 분비하는 근거를 통해 환경변화로 생긴 코티솔 분비를 측정하였다. 코티솔 분비를 통한 스트레스 반응 정도 측정은 침해성을 최소화하기 위한 간편한 방법인 타액검사로 실시하였으며 비 침습적인 방법으로 특별한 장비나 도움이 없이 수집할 수 있었고 부작용이 적은 효율적 검사였다. 연구 결과 전체 학생(46명)의 코티솔은 환경개선 사전($0.121{\pm}0.087{\mu}g/dL$)에 비해 사후($0.096{\pm}0.058{\mu}g/dL$)에 감소하였고(t=1.718, p=.093) 중고등학생(36명)의 코디솔은 사전($0.137{\pm}0.092{\mu}g/dL$)에 비해 사후($0.104{\pm}0.062{\mu}g/dL$)에 감소하였으며, 유의수준 10%에서 통계적으로도 유의미한 차이를 보였다(t=1.750, p=.089). 따라서 환경 개선 전에 비해 환경 개선 후 청소년들의 스트레스가 감소되었음을 알 수 있다. 하지만 초등학생(10명)의 코티솔은 사전($0.064{\pm}0.023{\mu}g/dL$)에 비해 사후($0.065{\pm}0.016{\mu}g/dL$)에 변화를 보이지 않아, 유의수준 5%에서 통계적으로 유의미한 변화는 나타나지 않았다(t=-0.121, p=0.906). 따라서 후두엽이 발달하는 중학생 시기는 환경 변화에 민감하며 색채 자극에 크게 영향을 미치는 시기인 만큼 본 연구에서 환경전후로 검사한 코티솔 분비의 측정값도 감소함을 볼 수 있었다. 하지만 본 연구는 환경 변화에 코티솔 분비량에 유의미한 변화를 나타내지 않은 초등학생들은 스트레스 호르몬 변화에는 차이가 없었지만 환경변화의 자극이 없었다고 볼 수는 없다.

Antioxidative Activities of Aronia melanocarpa Fruit and Leaf Extracts (아로니아 베리 열매 및 잎 추출물의 항산화 활성)

  • In this study, the antioxidative effects of Aronia melanocarpa fruit and leaf extracts were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of the ethylacetate and aglycone fractions of fruit extracts were 16.29 ${\mu}g/mL$, and 12.29 ${\mu}g/mL$, respectively. The free radical scavenging activity of fruit extract was higher than that of leaf extracts. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethylacetate and aglycone fractions of fruit extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay showed 2.86 ${\mu}g/mL$, and 1.80 ${\mu}g/mL$, respectively. ROS scavenging activity of the aglycone fraction of fruit extracts was similar to that of L-ascorbic acid (1.50 ${\mu}g/mL$). The ROS scavenging activity of fruit extracts was higher than that of leaf extracts. The cellular protective effects of aglycone fraction of fruit extracts (${\tau}_{50}$ = 72.3 min) on the $^1O_2$-induced cellular damage of human erythrocytes especially were increased in a concentration dependent manner (5 ~ 50 ${\mu}g/mL$). ${\tau}_{50}$ (72.3 min) of the aglycone fraction showed 1.9 times higher than (+)-${\alpha}$-tocopherol (38 min), known as lipophilic antioxidant at 10 ${\mu}g/mL$. These results incidicate that A. melanocarpa fruit extracts have higher antioxidant effects than leaf extracts and could be applicable to functional cosmetics materials for antioxidants by protecting skin exposed to solar UV radiation against ROS including $^1O_2$.

  • Effects of two auxins, 2,4-dichlorophenoxyacetic acid (2,4-D) and a-napthaleneacetic acid (NAA) on nicotine production during callus culture of a wild tobacco (Nicotiana glauca) were investigated using a high performance liquid chromatography (HPLC). The high concentration ($11.5\;\mu\textrm{M}$)of 2,4-D and NAA had peaks of nicotine contents at 4th and 2nd week, respectively. Thereafter, the concents decreased and the nicotine was metabolized to other alkaloids. The low concentration ($1.5\;\mu\textrm{M}$) of 2,4-D on the medium supplemented with 0.1 mM of L-aspartic acid or L-arginine inhibited nicotine production. However, the low NAA promoted it only when the medium was supplemented with L-aspartic acid. From these results, it could be concluded that both auxins exhibit different action mechanisms on nicotine production pathway and the low NAA promotes the activities for the pathway with L-aspartic acid as a precursor.cursor.

Development and Growth of Larvae of Four Bivalve Species (4종 조개류 유생의 발생과 성장)

  • Exposure to air and increased temperature induced successful spawning in Mytilus edulis, M. coruscus Crassostrea gigas and Pinctata fucata martensii. Developmental durations required for an egg to attain D-shaped larva and the D-shaped larva to reach pediveliger stage were estimated in these bivalves. Size of fertilized eggs was the largest (70.3 ${\mu}m$) in M. coruscus and the smallest (45.3 ${\mu}m$) in P. fucata martensii. At 17$^{\circ}C$, M. edulis and M. coruscus attained D-shaped larval stage within 48 hours after fertilization but those of C. gigas and B. fucata martensiii within 24 and 22 hours at 21 and 26$^{\circ}C$, respectively. The development duration required for a D-shaped larva to attain pediveliger stage was the longest (27 days) in M. coruscus and ranged between 20 and 22 days for the others. The shell length of the pediveliger was the longest (274.9 ${\mu}m$) in C. gigas and smallest (190.9 ${\mu}m$) in P. fucata martensii Length and height of larval shell was highly correlated with each other in all the 4 species. The shell height of C. gigas was more than the shell length beyond the size of 100 ${\mu}m$ shell length. However, shell length of the others was always longer than shell height at the larval stage.

Extraction and Purification of Acanthoside-D from Acanthopanax chilsanensis (지리산 오갈피나무로부터 Acanthoside-D의 추출 및 정제)

  • Acanthoside-D, contained in Acanthopanax chilsanensis is known as a ginseng-like substance. This work was focused to set up analytical and preparative conditions for Acanthoside-D purification. The ethanol extract from the powder of the trunk of Acanthopanax chilsanensis was partitioned with hexane. A $\mu$-Bondapak $C_{18}$ (3.9$\times$300 mm, $10\mu\textrm{m}$) column was used to separate Acanthoside-D from the trunk of Acanthopanax chilsanensis. From the experimental results, the mobile phase used for isolating Acanthoside-D from the extract was water/acetonitrile/methanol=80/14/6 %(v/v). The flow rate of mobile phase was 1.0 mL/min, and UV wavelength was fixed at 210 nm. Finally on a semi-preparative column (3.9$\times$ 300 mm, $15\mu\textrm{m}$, Lichrospher 100RP-18) with the same mobile phase composition, the allowable maximum injection volume increased to 250 $\mu\ell$
