Cryopreservation of Embryogenic Callus in Sweetpotato cv. 'Yulmi' (고구마품종 '율미' 배발생 캘러스의 초저온 동결보존)

  • Cryopreservation of embryogenic callus derived from apical meristem culture was attempted by slow prefreezing method (two-step method) with various cryoprotectants in sweetpotato cv. 'Yulmi' Precultured embryogenic calli on medium containing 10 mg/L ABA prior to slow prefreezing in liquid nitrogen indicated higher survival rate than 1.0 mg/L ABA preteatment. The cryoprotectant comprising 1.28 M DMSO in 0.4 M sucrose solution gave the best survival (over 46%) of sweetpotato cells exposed to liquid nitrogen as determined by TTC reduction and FDA staining method. Cryopreserved calli cultured on MS medium with 1.0 mg/L 2,4-D were grown for 4 weeks in the dark and induced embryos after another 4 weeks. They were subcultured on MS medium supplemented with 0.1 mg/L 2,4-D+0.1 mg/L kinetin for 2 weeks and regenerated into normal plantlets in MS basal medium.

Correlation between Cyclin D1, p53, Bcl-2 Protein Expression and Prognosis in Primary, Resected Stage IIIA Non-Small Cell Lung Cancer (NSCLC) (완전 절제된 제IIIA기 비소세포폐암에서 Cyclin D1, p53, Bcl-2 단백질 발현의 의의)

  • Background: Lung cancer formation is a multistage process involving activation of protooncogene and inactivation of tumor suppressor genes. We evaluate the significance of cyclin D1, p53, bcl-2 gene mutations in patients with curatively resected stage IIIA non-small cell lung cancer(NSCLC). Material and Method: One hundred consecutive cases of stage IIIA lung cancers from patients operated on curatvely between 1990 and 1995 for which adequate paraffin blocks and clinical history were available. Immunohistochemical studies were performed on the representative tissue sections from each case by the labelled streptovidin- biotin method. Sections for cyclin D1, p53, Bcl-2 immunostaining were pretreated in a microwave oven for 10 to 20 minutes in citrate buffer before immunostaining. The overnight incubation with NCL-cyclin D1-GM for cyclin D1, with clone DO-7 for p53, with clone 124 for bcl-2 was done. Mean follow-up was 24.1 months (range 2-84 months) after operation. Result: One hundred cases of lung cancers were composed of 56 cases of squamous cell carcinoma, 37 cases of adenocarcinoma, 5 cases of adenosquamous cell carcinoma, and 2 cases of large cell carcinoma. The 5-year survival was 32.1%. The positive expression rate of cyclin D1 was 35%, p53 was 56%, and bcl-2 was 17%. But there were no correlation between cyclin D1, p53, Bcl-2 protein expression and survival. Conclusion: These observation indicate that cyclin D1, p53, bcl-2 protein overexpression might be implicated in the oncogenesis of non-small cell lung carcinomas but they have no usefulness as a prognostic marker.

  • The SW-centered battlefield management information system R&D project takes a long period of 5-10 years or more by applying a complex and rigid batch acquisition strategy. In order to solve this problem, it is necessary to institutionalize a rapid and flexible battlefield management information system R&D project management procedure applying agile development methodology, and a government project management organization and contract management method to support it In this study, we analyzed the case of applying the Agile development method centered on Scrum to the US SW-centered weapon system R&D project and the characteristics and problems of the battlefield management information system R&D project in Korea, and suggested improvement measures as follows. First, the battlefield management information system R&D model applies the hybrid development method, and the system requirements analysis and system structure design use the existing waterfall development procedure, and the agile method is applied from the SW requirements analysis to the system integration stage. Second, flexible adjustment of performance, schedule, and cost by organizing an Agile IPT in which military (requirements) - DAPA (project management) - developer - functional specialized organizations (test and evaluation, quality, government research institutes, etc.) participate. Third, improving the Basic Order Agreement so that it can be applied to agile R&D.

  • In this paper, we suggest a 3D vision inspection method which use only one camera. If we have the database of pattern and can recognize the object, and also estimate the rotated shape of the parts, we can inspect the parts using only one image. We used the 3D database and the 2D geometrical pattern matching, and the rotation transition theory about the algorithm. As the results, we could have the capability of the recognition and inspection of the rotated object through the estimation of rotation an81e. We applied our suggested algorithm to the inspection of typical IC and capacitor, and compared our suggested algorithm with the conventional 2D inspection method and the feature space trajectory method.

  • 'Gesture' is the most intuitive means of communication except the voice. Therefore, there are many researches for method that controls computer using gesture input to replace the keyboard or mouse. In these researches, the method of user detection and main body parts estimation is one of the very important process. in this paper, we propose user objects detection and main body parts estimation method on inaccurate depth information for pose estimation. we present user detection method using 2D and 3D depth information, so this method robust to changes in lighting and noise and 2D signal processing 1D signals, so mainly suitable for real-time and using the previous object information, so more accurate and robust. Also, we present main body parts estimation method using 2D contour information, 3D depth information, and tracking. The result of an experiment, proposed user detection method is more robust than only using 2D information method and exactly detect object on inaccurate depth information. Also, proposed main body parts estimation method overcome the disadvantage that can't detect main body parts in occlusion area only using 2D contour information and sensitive to changes in illumination or environment using color information.

Comparison of 3D Reconstruction Methods to Create 3D Indoor Models with Different LODs

  • A 3D indoor model becomes an indiscernible component of BIM (Building Information Modeling) and GIS (Geographic Information System). However, a huge amount of time and human resources are inevitable for collecting spatial measurements and creating such a 3D indoor model. Also, a varied forms of 3D indoor models exist depending on their purpose of use. Thus, in this study, three different 3D indoor models are defined as 1) omnidirectional images, 2) a 3D realistic model, and 3) 3D indoor as-built model. A series of reconstruction methods is then introduced to construct each type of 3D indoor models: they are an omnidirectional image acquisition method, a hybrid surveying method, and a terrestrial LiDAR-based method. The reconstruction methods are applied to a large and complex atrium, and their 3D modeling results are compared and analyzed.

Measurement Guideline of Fresnel-Field Antenna Measurement Method

  • In this letter, a parametric analysis of the Fresnel-field antenna measurement method is performed for a square aperture. As a result, the optimum number of Fresnel fields for one far-field point is guided as $M_{opt}=N_{opt}=D^2/{\lambda}R+5$, where D is the antenna diameter, ${\lambda}$ is the wavelength, and R is the distance between the source antenna and the antenna under test. For the aperture size 5 ${\leq}$ $L_x/{\lambda}$ ${\leq}$ 20, the tolerable distances for gain errors of 0.5 dB and 0.2 dB can be guided as $R_{0.5\;dB}$ ${\approx}$ $1.2Lx/{\lambda}$ and $R_{0.2\;dB}$ ${\approx}$ $2.0L_x/{\lambda}$, where $L_x$ is the lateral length of the square aperture. The tolerable distances for 20 ${\leq}$ $L_x/{\lambda}$ ${\leq}$ 200 are also proposed. This measurement guideline can be fully utilized when performing the Fresnel-field antenna measurement method.

A New Linear Explicit Camera Calibration Method (새로운 선형의 외형적 카메라 보정 기법)

  • Vision is the most important sensing capability for both men and sensory smart machines, such as intelligent robots. Sensed real 3D world and its 2D camera image can be related mathematically by a process called camera calibration. In this paper, we present a novel linear solution of camera calibration. Unlike most existing linear calibration methods, the proposed technique of this paper can identify camera parameters explicitly. Through the step-by-step procedure of the proposed method, the real physical elements of the perspective projection transformation matrix between 3D points and the corresponding 2D image points can be identified. This explicit solution will be useful for many practical 3D sensing applications including robotics. We verified the proposed method by using various cameras of different conditions.

Evaluation of internal fit of interim crown fabricated with CAD/CAM milling and 3D printing system

  • PURPOSE. This study is to evaluate the internal fit of the crown manufactured by CAD/CAM milling method and 3D printing method. MATERIALS AND METHODS. The master model was fabricated with stainless steel by using CNC machine and the work model was created from the vinyl-polysiloxane impression. After scanning the working model, the design software is used to design the crown. The saved STL file is used on the CAD/CAM milling method and two types of 3D printing method to produce 10 interim crowns per group. Internal discrepancy measurement uses the silicon replica method and the measured data are analyzed with One-way ANOVA to verify the statistic significance. RESULTS. The discrepancy means (standard deviation) of the 3 groups are $171.6\;(97.4){\mu}m$ for the crown manufactured by the milling system and 149.1 (65.9) and $91.1\;(36.4){\mu}m$, respectively, for the crowns manufactured with the two types of 3D printing system. There was a statistically significant difference and the 3D printing system group showed more outstanding value than the milling system group. CONCLUSION. The marginal and internal fit of the interim restoration has more outstanding 3D printing method than the CAD/CAM milling method. Therefore, the 3D printing method is considered as applicable for not only the interim restoration production, but also in the dental prosthesis production with a higher level of completion.

A Study on the Visual Preference in Enclosed Spaces Based on the Paired Comparison Method (쌍체비교기법을 통한 중정의 시각적 선호에 관한 연구)

  • The purpose of this study was to investigate desirable width / height ratio of enclosed space and to offer basic information for the design of enclosed spaces. The data of this study were gathered from the experiment through simulation. Photographic sampling of 7 campus sites located in Kwanak campus of Seoul National University and slides of perspective drawings of enclosed spaces with various D/H ratio (12, 1, 2, 4, 6, 9, 12)were used for simulation. A response format was made using the method of paired comparison and students from various departments were randomly selected for evaluation. In the method of analysis of data Thurstone's equation of Comparative Judgement was used for calculating preference score. A1so non-linear regression analysis was used for finding out relations between preference and D/H ratio. The other method of data analysis were correlation, Chi - test. And the results were summarized as follows. 1. In sketch simulation the ratio of D/H 4 got the highest preference store. This result suggests that desirable D/H ratio of enclosed space is D/H 4. And at the lower range of D/H ratio, as ratio increases, the preference score increases. At the higher range of D/H ratio, however, as ratio increases, the preference score starts to decrease from the cardinal point of D/H 4. 2. In campus photograph simulation preference is influenced by slope, elevation change of floor, trees, lawn area, familiarity, space arrangement, and exterior shape of building besides D/H ratio. 3. The preference virtue was not significantly affected by order effect. This result suggests that order effect can be ignored in the study of visual preference. 4. There are so many factors related to preference that it is difficult to explore those factors without scientific information based on scientific method. We must carry out study for scientific approach of planning and design based on precise, complete simulation technique.
