고밀도 배양에 있어서 영양강화 방법 및 종류에 따른 rotifer의 지방산 조성의 변화 (Change of fatty acid compositions of rotifer according to enrichment diets and methods in the high density culture)

  • 본 연구는 고밀도 배양시 rotifer의 영양강화 방법 및 영앙강화제에 따른 rotifer의 개체밀도와 지방산 조성을 조사하였다. 첫째 방법은 담수산 Chlorella로 rotifer를 고밀도로 배양한 후 영양강화제인 유지효모, Algamac, Super Selco, 해수산 Chlorella로 2차 영양강화 하였다. 두 번째 방법은 rotifer 반 연속 고밀도 배양중 18시간 동안 담수산 Chlorella를 공급한 후 6시간동안 영양강화제를 공급하였고 대조구로 담수산 Chlorella을 공급하지 않고 24시간 동안 해수산 Chlorella를 공급하였다. 배양수조는 5$\ell$를 이용하여 수온 $28^{\circ}C$를 유지하였고 산소가스를 공급하였다. 반 연속 고밀도 배양 방법에서 24시간 동안 해수산 Chlorella를 공급하였을 때 rotifer 밀도와 용존산소는 안정적으로 유지되었고 rotifer의 n-3 HUFA의 함량이 다른 실험구보다 가장 높게 나타났다. 그러나 2차 영양강화 방법과 반 연속 고밀도 배양 방법에서 유지효모, Algamac 및 Super Selco로 영양강화하였을 때 rotifer의 밀도와 용존산소는 급격히 감소하는 경향을 보였고 이들 영양강화제 가운데 Super Selco가 비교적 높은 n-3 HUFA의 함량i글 보였다. 따라서 본 실험을 통해서 rotifer 고밀도 배양시 안정적인 rotifer 성장과 rotifer의 질적 개선을 위해서는 24시간 해수산 Chlorella를 공급하는 것이 효율적인 것으로 판단된다.

The Production of Lutein-Enriched Eggs with Dietary Chlorella

  • Two experiments were conducted to investigate the dietary effect of chlorella vulgaris on egg production and lutein incorporation into chicken eggs. In Exp. 1, a total of three hundred, 70 wk-old Hy-Line brown layers were divided into six groups with five replicates and fed each experimental diet (corn-SBM based control diet and diets with 0.1, 0.3 or 0.5% chlorella powder and with 0.8 or 2.4% chlorella cultured media) for 6 wk, respectively. The egg production in the groups fed diets containing the chlorella powder and chlorella cultured media were higher than that of the control group (p<0.001). As dietary chlorella levels increased, the yolk color linearly increased. However, there were no significant differences in egg-shell qualities. The layers fed diet with 2.4% chlorella cultured media showed the highest Haugh unit value. In Exp. 2, a total of one hundred-eight 80 wk-old Hy-Line brown layers were assigned into four groups with three replicates per group (9 birds per replicate). The birds were fed one of four experimental diets (0, 0.5, 1.0 or 2.0% chlorella powder) for 4 wk, followed by a 14 d feeding of a withdrawal diet devoid of chlorella powder. At 2 wk, the lutein greatly increased with increasing levels of chlorella powder in birds fed diets containing more than 1%. The maximum incorporation of lutein into eggs was reached after 2 or 3 wk of feeding diets with chlorella powder. After a 7 d withdrawal, the lutein contents of egg yolks in the groups fed diets with more than 1% chlorella powder were still higher than that of control group (p<0.05). No significant differences in the lutein levels were found among groups after a 14 d withdrawal period. These results indicated that the use of chlorella in layer diets was effective in improving egg production and egg quality and for the production of lutein fortified eggs.

클로렐라를 첨가하여 제조한 두부의 품질특성과 저장성 (Studies on Quality Characteristics and Shelf-life of Chlorella Soybean Curd (Tofu))

  • 클로렐라를 첨가하여 기능성 두부를 제조함에 있어 클로렐라의 첨가량이 두부의 품질에 미치는 영향을 조사하였다. 건조 대두를 기준으로 클로렐라를 0.5-2.0% 첨가하여 제조한 두부의 수율, 물성, 색도, 저장성 및 관능적인 특성을 클로렐라를 첨가하지 않고 제조한 일반두부와 비교하였다. 수율은 클로렐라를 1% 첨가한 경우 일반두부에 비하여 10% 이상 향상되었다. 클로렐라를 첨가함으로써 두부의 물성 중 견고성, 검성과 씹힘성이 증가하였으며, 색도는 L값, a값, b값 모두 첨가량에 비례하여 감소하였다. 관능적 특성은 조직감 이외의 항목에서는 일반두부와 통계적으로 유의적인 차이가 얼었다. 제조한 두부를 $15^{\circ}C$로 보관하면서 pH와 총균수의 변화를 측정한 결과 클로렐라를 0.5-1.0% 첨가한 두부에서 pH하락과 미생물 생육이 효율적으로 지연되었다.

Activity of Early Gene Promoters from a Korean Chlorella Virus Isolate in Transformed Chlorella Algae

  • As a unicellular green alga that possesses many of the metabolic pathways present in higher plants, Chlorelia offers many advantages for expression of heterologous proteins. Since strong and constitutive promoters are necessary for efficient expression in heterologous expression systems, the development of such promoters for use in the Chlorella system was the aim of this study. Proteins encoded by the early genes of algal viruses are expressed before viral replication, probably by the host transcriptional machinery, and the promoters of these genes might be useful for heterologous expression in Chlorella. In this study, putative promoter regions of DNA polymerase, ATP-dependent DNA ligase, and chitinase genes were amplified from eight Korean Chlorella virus isolates by using primer sets designed based on the sequence of the genome of PBCV-1, the prototype of the Phycodnaviridae. These putative promoter regions were found to contain several cis-acting elements for transcription factors, including the TATA, CAAT, NTBBF1, GATA, and CCAAT boxes. The amplified promoter regions were placed into Chlorella transformation vectors containing a green fluorescence protein (GFP) reporter gene and the Sh ble gene for phleomycin resistance. C. vulgaris protoplasts were transformed and then selected with phleomycin. The GFP fluorescence intensities of cells transformed with chitinase, DNA polymerase, and DNA ligase gene promoter-GFP fusion constructs were 101.5, 100.8, and 95.8%, respectively, of that of CaMV 35S-GFP-transformed Chlorella cells. These results demonstrate that these viral promoters are active in transformed Chlorella.

Chlorella의 인산대사에 관한 연구 (Studies on the Phosphate Metabolism in Chlorella, with Special Reference to Polyphosphate)

Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction. Effect of Biostimulator Chlorella fusca on Improving Growth and Qualities of Chinese Chives and Spinach in Organic Farm

  • This study was conducted to investigate the efficacy of freshwater alga, Chlorella fusca on the improvement of growth and qualities in organic spinach and Chinese chives farm. The average height of Chinese chives treated with the chlorella was 3.7 cm smaller than that of the untreated. The leaf width and fresh weight of Chinese chives treated with the chlorella was 0.5 mm wider and 30.3 g heavier than that of the untreated. The commercialization and yield of Chinese chives treated with the chlorella was 11.9% and 18.3%, respectively higher than that of the untreated. Also, the disease severity of gray mold disease of Chinese chives treated with the chlorella was reduced by more than 24.2% when compared with the untreated. The thickness and number of spinach leaves treated with chlorella was 27.9% and 41.8%, respectively higher than that of the untreated. The fresh weight and yield of the spinach treated with the chlorella was 63.6% and 31.5%, respectively higher than that of the untreated. Moreover, the mineral content of K, Ca, Mg, P, Fe, and Mn were recorded higher in the spinach treated with chlorella compared with that of untreated. The results indicated that the freshwater alga, Chlorella fusca is efficient and economical biostimulant in improving plant growth and quality of Chinese chives and spinach in organic farm.

Quality Evaluation of Cheongpomuk Added with Chlorella Powder

  • This study examined the quality characteristics to evaluate the optimal mixing ratio of a chlorella powder in Cheongpomuk. The total polyphenol and total flavonoids contents of Cheongpomuk and chlorella powder was 279.90 ㎍/mL, 489.50 ㎍/mL and 48.79 ㎍/mL, 687.85 ㎍/mL respectively. The DPPH free radical scavenging activities and ABTS radical scavenging activities of chlorella powder was 65.39%, 91.28% respectively. The L values (58.85~26.23), a values (-0.44~-5.84) of the Hunter's color values decreased and b value (-10.98 ~4.12) increased significantly in proportion to the amount of chlorella powder. The total polyphenol contents (55.00~734.20 ㎍/mL), total flavonoids contents (41.00~88.29 ㎍/mL), DPPH free radical scavenging activities (35.10~65.50%) and ABTS radical scavenging activities (50.29~92.86%) of Cheongpomuk increased significantly in groups of chlorella powder. In the physical properties, the hardness (135.93~103.93 g/㎠), chewiness (6.76~3.76 g), springiness (4.92~4.36 %), and gumminess (147.70~94.23) of Cheongpomuk decreased significantly in proportion to the amount of chlorella powder. The Cheongpomuk containing 15% chlorella powder showed a high score with regard to color, flavor, moistness, chewiness and overall acceptance. The appropriate addition amount of chlorella powder for making Cheongpomuk is 15%. This study indicate that Cheongpomuk treated with chlorella powder had the highest functional component and antioxidant activity.

Carbon Dioxide Mitigation by Microalgal Photosynthesis

  • Algal growth studies of Chlorella strains were conducted in a batch mode with bench type experiments. Carbon dioxide fixation rates of the following green microalgae were determined: Chlorella sp. H84, Chlorella sp. A2, Chlorella sorokiniana UTEX 1230, Chlorella vulgaris, and Chlorella pyrenoidosa. C. vulgaris, among other strains of microalgae, showed the highest growth rate (1.17 optical density/5 days). Cultivating conditions for C. vulgaris that produced the highest growth rate were at concentrations of 243 ${\mu}g\;CO_2$/mL, 10 mM ammonia, and 1 mM phosphate, with an initial pH range of 7-8.

Isolation of a Calcium-binding Peptide from Chlorella Protein Hydrolysates

  • To isolate a calcium-binding peptide from chlorella protein hydrolysates, chlorella protein was extracted and hydrolyzed using Flavourzyme, a commercial protease. The degree of hydrolysis and calcium-binding capacity were determined using trinitrobenzenesulfonic acid and orthophenanthroline methods, respectively. The enzymatic hydrolysis of chlorella protein for 6 hr was sufficient for the preparation of chlorella protein hydrolysates. The hydrolysates of chlorella protein were then ultra-filtered under 5 kDa as molecular weight. The membrane-filtered solution was fractionated using ion exchange, reverse phase, normal phase chromatography, and fast protein liquid chromatography to identify a calcium-binding peptide. The purified calcium-binding peptide had a calcium binding activity of 0.166 mM and was determined to be 700.48 Da as molecular weight, and partially identified as a peptide containing Asn-Ser-Gly-Cys based on liquid chromatography/electrospray ionization tandem mass spectrum.

  • Wir wollten beobachten, ob die Chlorophyllbnahme von Chlorella luteoviridis(211/2b) bei Glukose-Zugabe durch Stickstoffmangel hervorgerufen worden ist, wie viele Leute bei mineralsalzmangel(Aach (1952, Fogg 1956 Eyster et al (1958), oder Photooxydationschlorose (Kandler 1958) vermuteten. Chlorella luteoviridis bildete nicht Chlorophyll bei Glukosezugabe wie bei Autotrophkultur, obwohl Stickstoffsynthese bei Autotrophkultur stark gef rdert worden ist. Bei Nitratanzucht mit Glukose (Mixotroph) waren die Zellvermehrung und Kohlenhydratbildung nicht so stark wie bei der Glukoseanzucht mit NH4Cl2 trotzdem nahm Chlorophyll ab. Chlorella luteoviridis bildete Chlorophyll im Dunkeln und St rkeakkumulation erschien nicht bei Glukoseanzucht.
