Comparison in Serum Constituents of Cultured Marine Fishes in Early Summer Season (주요 양식어류의 하절기 혈액성분 비교)

  • This study was performed to obtain the basic data on the serum constituents of several marine fish spesies commonly cultured in Korea. Blood samples taken from six species of fish were analyzed for various components of serum, total protein (TP), albumin (ALB), triglyceride (TRIG), cholesterol (CHOL), glucose (GLC), lipase (LIPA), amylase (AMYL), aspartate transaminoferase (AST), sodium (Na), potassium (K), chloride (CI) and phosphorus (PHOS). The fish used were coho salmon (Oncorhynchus kisutch), rock fish (Sebastes schlegeli), sea bass (Lateolabrax japonicus), olive flounder (Paralichthys olivaceus), rock fish (Sebastes schlegeli), sea bass (Lateolabrax japonicus), olive flounder (Paralichthys olivaceus), parrot fish (Oplegnathus fasciatus) and jack mackerel (Trachurus jaonicus) reared at the Chungmu Experimental Fish Culture Station of KORDI when the water tempetature was ca. 16.5$^{\circ}C$. There were significant differences in TRIG, CHOL, LIPA and AMYZ among the species analyzed. TRIG concentratin were ranged 178~180mg/dl in jack mackeerel and rock fish, 126~159 mg/dl in olive flounder and sea bass, and 102~114 mg/dl in coho salmon and parrot fish, respectively. Jack mackerel showed the highest levels in CHOL (255mg/dl) and GLC(138mg/dl) among species. LIPA levels were recorded 256 U/dl in coho salmon, 41~42 U/dl in parrot fish and rock fisk, and 5~11 U/dl jack mackerel and sea bass, respectively. AMYL activity of coho salmon was measured as 2, 665 U/dl, and that of jack mackerel was 1,210 U/dl while sea bass showed 60 U/dl and parrot fish, olive flounder and rock fish had at most 5 U/dl. On the other hand, there was no significant difference in the concentration of Na and CI. Na and K were proved that they were negatively correlated in all the species. Generally, among blood components, PHOS and CHOL levels were different depending on environmental temperature of each fish species, especially in olive flounder. Rock fish and parrot fish showed high blood concentration of those components during low temperature period while olive flounder and jack mackerel reached high level during their optimal environmental temperature period. The electrolyte concentration and LIPA activity were high during low water temperature period, in general, but TP and ALB concentrations were high during optimal temperature period. The concentrations of TRIG, CHOL and GLC, those which were used as energy sourses, were different among species by season.

Flavor Constituents in Enzyme Hydrolysates from Shore Swimming Crab and Spotted Shrimp (민꽃게 및 꽃새우 효소가수분해물의 풍미발현성분)

  • For the developing natural fisheries flavoring substances using crustacea, the flavor constituents of enzyme hydrolysates from shore swimming crab (crab) and spotted shrimp (shrimp) were investigated. In taste-active compounds of both enzyme hydrolysates, total free amino acid contents of crab and shrimp enzyme hydrolysates were 5,226.7 mg% and 8,757.3 mg%, respectively. The major amino acids were taurine, glutamic acid, proline, asparagine, glycine, alanine, valine, leucine, lysine anserine and arginine. As for ATP related compounds, AMP was the principal component and small amounts of IMP was detected in both enzyme hydrolysates. In the quarternary ammonium bases, betaine was the principal component (593.8mg%), and contents of TMAO and betaine in both samples were 60.7 mg% and 850.0 mg%, 124.1 mg% and 755.9 mg%, respectively. The major components were Na, K, P and Cl in inorganic ions. The major fatty acids of both sample were 14 : 0, 16 : 0, 16 : 1n7, 18 : 1n9, 20 : 5n3 and 22 : 6n3, and composition ration of n3 polyunsaturated fatty acids of were 27.8% and 28.5%, respectively. Total 99~109 volatile compounds were detected as a cooked odor of crab and shrimp enzyme hydrolysates by SDE apparatus/gas chromatography/mass spectrometry. The volatile flavor compounds identified from cooked crab enzyme hydrolysate were composed of 6 acids, 10 alcohols, 7 aldehydes, 11 ketones, 1 ester, 5 phenols, 4 benzenes, 22 hydrocarbons, 1 furan, 21 nitrogen containing compounds and 11 micellaneous compounds. And the volatile flavor compounds indentified from cooked shrimp enzyme hydrolysate were composed of 13 acids, 10 alcohols, 6 aldehydes, 10 ketones, 3 esters, 2 phenols, 5 benzenes, 36 hydrocarbons, 1 furan, 14 nitrogen containing compounds and 8 micellaneous compounds.

Strain Improvement of Aspergillus oryzae for Increasing Productivity of a Proteolytic Enzyme. (고활성 단백질분해효소 생산균주의 개발을 위한 Aspergillus oryzae의 원형질체 융합에 의한 변이)

  • Aspergillus oryzae producing high proteolytic enzyme was isolated from soybean koji and named tentatively A. oryzae O-1. A. oryzae U-1 was obtained by mutation of A. oryzae O-1 with ultraviolet (UV)-irradiation and produced 14 times higher pretense activity compared with A. oryzae O-1. A. oryzae E-1 was acquired by treatment of A. oryzae U-1 with 0.5 M ethylmethanesulfonate (EMS) for 6 min at 3$0^{\circ}C$ and produced 39 times higher proteolytic activity than A. oryzae O-1. With protoplast fusion between A. oryzae O-1 and A. oryzee E-1 in the presence of polyethylenegylcol (PEG)-CaCl$_2$, proteolytic activity was increased to 82 times compared to A. oryzae O-1, and the fusant was named A. oryzae PF. The activities of the cultures containing proteolytic enzymes produced by the strains were determined to be 0.23 U/$m\ell$ for A. oryzae O-1, 3.29 U/$m\ell$ for A. oryzae U-1, 8.91 U/$m\ell$ for A. oryzae E-1, and 19.0 U/$m\ell$ for A. oryzae PF.

Isolation and Characterization of Bacillus subtilis MP56 with Antimicrobial Activity against MDR (Multi Drug Resistant) Strains (다약제내성균에 대한 항균 활성을 가지는 Bacillus subtilis MP56 균주의 분리 및 특성분석)

  • A new Bacillus strain designated as MP56 producing antimicrobial substance has been isolated from the mud flat of Korea. The strain MP56 was found to exhibit broad spectrum of antimicrobial activity against Gram-positive pathogenic microorganisms and MDR (multi drug resistant) strains. The 16S rRNA sequence revealed that the MP56 was closely related to Bacillus subtilis with 99.93% homology. The optimal medium composition for production of antimicrobial substance in the B. subtilis MP56 were 1% mannitol, 1% oat meal, 0.01% $CaCl_2$. Antimicrobial activity of the culture broth against different pathogenic strains was assessed using the antimicrobial spectrum. The result suggests that Bacillus strain MP56 produces high quality antimicrobial substance that might be very useful to control varieties of pathogenic microbial growth.

Analysis of Results and Techniques about Precipitation Enhancement by Aircraft Seeding in Korea (항공기를 이용한 인공증우(설) 기술과 결과분석)

  • National Institute of Meteorological Sciences has conducted a total 54 cloud seeding experiments with a silver iodide and calcium chloride using aircrafts from 2008 to 2018. The goal of the experiments is to improve the techniques of precipitation enhancement in Korea. The cloud seeding experiments using the silver iodide and calcium chloride were 36 and 18 times, respectively. During the cloud seeding experiments of the silver iodide and calcium chloride, the average values of total cloud amount for two kinds of seeding materials were 9.6 for and 8.1, respectively. The cloud type with the highest occurrence was Nimbostratus (Ns)-Stratus (St) (58%) in the silver iodide cloud seeding experiment. It was Altostratus (As)-Stratocumulus (Sc) (44%) in the calcium chloride cloud seeding experiment. Compared to probability of obtaining cloud seeding effect of the experiments using a leased aircraft, the probability using an atmospheric research aircraft increased from 43% to 63% in the silver iodide cloud seeding experiment and from 29% to 75% in the calcium chloride cloud seeding experiment. However, the increasing tendency was only shown during the one year experiment (2018). To get the meaningful statistical tendency of the cloud seeding effects, it is needed to implement many experiments in several years. Further we have to more clearly understand the characteristics of clouds developing in Korea and implement the cloud seeding experiments under a variety of weather conditions in order to develop the optimized precipitation enhancement technology in Korea.

Study on Soil Extraction Methods for the Human Health Risk Assessment of Crop Intake Pathway around Abandoned Metal Mine Areas (폐금속광산 지역 농작물섭취경로의 인체위해도 산정을 위한 생물농축계수와 토양분석방법에 관한 연구)

  • Generally, the contribution of crop-intake pathway (CIP) is remarkable in human health assessment (HHA) of heavy metal contamination. Although the crop exposure concentrations (Cp) should directly be used for calculating the average daily dose (ADD) of CIP, the soil exposure concentration (Cs) multiplied by soil-crop bio-concentration factor (BCF) has frequently been used instead of using Cp values. Thus, the BCF values are significant in the HHA, and care should be taken to ensure the reasonable acquisition of BCF values. Meanwhile, the BCF values are known to be significantly affected by analytical methods. Nevertheless, they have been calculated from the concentrations of soil and crop analyzed by only one method: total digestion (aqua regia extraction). For this reason, this study was initiated to seek appropriate soil analysis methods for effective computation of the ADD of CIP. The concentrations of 5 metal contaminants (As, Cd, Cu, Pb, and Zn) in 127 soil samples obtained from 4 abandoned metal mine areas were analyzed by several methods including total digestion and partial digestions using 0.1/1 N HCl, 1M $NH_4NO_3$, 0.1 M $NaNO_3$, and 0.01M $CaCl_2$. The heavy metal concentrations in 127 crop samples (rice grains) were analyzed by total digestion as well. Using the concentrations of soils and crops, the BCF values of each contaminant were calculated according to the kind of soil extraction methods applied. Finally, the errors between Cp and $C_s{\times}BCF$ were computed to evaluate the relevance of each method. The results indicate that the partial extraction using 0.1 N and 1 N HCl was superior or equivalent to total digestion. In addition, the 0.1M $NaNO_3$ method combined with total digestion is recommended for improving the reliability of BCF values.

Effect of Soil Salinity on Nitrate Accumulation of Lettuce (토양 염류집적이 상추의 Nitrate 축적에 미치는 영향)

  • Accumulation of nitrate in edible crops is undesirable due to potential risks to human health. Since nitrate has a role in the osmotic regulation of plants, salt accumulation in soil is expected to stimulate nitrate accumulation in plants. Lettuce (Lactuca sativa L.) was grown in soils of different salinities, 9.69 and $4.49dS\;m^{-1}$, in a greenhouse, and the effect of soil salinity on nitrate accumulation in lettuce was investigated. Content of nitrate in the lettuce increased significantly as soil salinity increased under low light intensity and ample supply of nitrate in root media. Soluble sugar and oxalate contents in lettuce were also significantly higher in the soil of higher salinity. Phosphate, Cl, and $SO_4$ contents in lettuce were not significantly different in soils of different salinities. Among the cations, K content in lettuce was significantly higher in the soil of higher salinity, but Na, Ca, and Mg comtents were not much influenced. Comparing to the lettuce grown in low salinity soil, although the growth of lettuce was decreased by 9% in the soil of higher salinity, nitrate accumulation in the lettuce was increased by 18.6%. These results indicate that higher nitrate content in lettuce of higher salinity soil is a positive accumulation to adapt to the water stress condition. The nitrate accumulation of vegetables grown in plastic film houses is known to be due to the heavy fertilization and low light intensity, but salt accumulation in the soil, which can lower soil water potential, is expected to stimulate the nitrate accumulation further.

Change of Bioavailability in Heavy Metal Contaminated Soil by Chemical Amendment (중금속 오염 농경지에 처리된 안정화제의 적용에 따른 토양 내 생물유효도 변화)

  • Crop safety in heavy metal contaminated agricultural field has been a critical issue in Korea and various remediation methods are proposed for minimizing heavy metal transfer from soil to crops. The main objective of this research was to evaluate remediation efficiency of two chemical amendments, lime and steel slag, and to decide extractant for assessing bioavailability of heavy metals. In order to select optimum extractant for evaluating bioavailability of heavy metals, four different single extractants, HCl, DTPA, $CaCl_2$, $NH_4NO_3$, and sequential extraction method were examined. Both chemical amendments showed high immobilization effect for Cd (66%, $33.62mg\;kg^{-1}$) and Pb (74%, $27.65mg\;kg^{-1}$) in soil by HCl extractant. In terms of heavy metal concentration in rice grains, concentrations for Cd (77%, $0.023mg\;kg^{-1}$) and Pb (82%, $0.039mg\;kg^{-1}$) decreased, with addition of chemical amendments. HCl, DTPA, and sequential extractant showed the higher correlation between heavy metal concentration in soil and crops than others. These results indicated that they could be used for assessing bioavailability of heavy metals.

Water Vapor and Thermal Transmission Properties of Hybrid Yarns Fabrics for High Emotional Garments -Water Vapor and Heat Transport according to Experimental-Method- (고감성 의류용 복합사 직물의 수분증기 및 열이동 특성 -실험방법에 따른 수분증기 및 열이동-)

  • Water vapor and thermal transmission properties of high emotional garments are important to evaluate wear comfort; in addition, the measuring methods of these properties are also critical for breathable and warm suit fabrics. In this study, the water vapor and thermal properties of composite yarn fabrics made of CoolMax, Tencel, and Bamboo fibers with filaments were measured and compared according to the measuring method. Water Vapor Transmittance (WVT) of the fabric woven by the sheath/core composite yarn in the warp direction was the highest due to the small staple fiber volume in the sheath/core yarn structure and high air voids in the sheath/core yarn fabrics. This property was also the highest in fabrics woven by bamboo staple yarns in the weft direction, and was the lowest on hi-multi filament fabrics. However, water vapor resistance ($R_{ef}$) of these fabrics by KSK ISO 11092 showed the opposite results to the water vapor transmittance method ($CaCl_2$ method); in addition, its correlation coefficient was low. The correlation coefficient between $R_{ef}$ and the drying rate was 0.719; therefore, the measurement mechanism of $R_{ef}$ is analogous to the drying property measurement. The thermal conductivity of the fabrics woven with compact staple yarn showed a high value; however, the hi-multi filament fabric showed low thermal conductivity. Therefore, fiber characteristics affect thermal properties more than yarn structure. The correlation between thermal property and moisture transport was also low. This study showed that: water vapor transmittance was active at the loose yarn structure, dry heat transport was vigorous at the compact yarn structure, and heat transport was affected more by fiber characteristics than yarn structure. In conclusion, sheath/core composite yarns were relevant to the high absorptive cool suit along with siro-fil and CoolMax/Bamboo staple yarns that were relevant to the heat diffusive cool suit.

Factor Analyses for Water Quality Indicators of Streams, Ground Water, and Reservoir in Agricultural Small Catchments of the Han River Basin

  • The principal indicators contributing to water qualities was screened by factor analyses, based on the monitored chemical parameters of water quality for various water resources from 1995 to 1999 in the small agricultural catchments of the Han River Basin. Water samples of streams, groundwaters, and reservoirs were taken four times a year from upper (Daegwanryong), middle (Dunnae and Chunchon) and lower (Guri) reaches of Han River Basin. In these areas, the respective type of farming practiced was alpine agriculture and livestocks raising, typical upland and paddy cultivation, and intensive cropping in the plastic film house. Water quality was monitored for twenty-one water quality parameters, including pH, EC, SS, T-N, T-P, COD, cations, anions, and heavy metals. pH, EC and COD of the stream waters were suitable for the Korea irrigation water quality guidelines. However, T-N and T-P concentrations of water samples in four catchments far exceeded the irrigation water guideline. Concentrations of canons and heavy metals in Wangsuk stream in Guri area were higher than those in streams in other areas. Factor analysis revealed that significant correlation was observed for 81 pairs out of 231 water quality indicators of stream water among the $21\;{\times}\;21$ cross correlation matrix of stream water quality indicators. The first factor accounted for 27.01% of the total variation in stream water quality indicators, and high positive factor loadings were shown on EC, K, Na, $NH_4\;^+-N$, $PO_4\;^{3-}$, $SO_4\;^{2-}$, and COD. Fifty-three water quality indicator pairs were significant out of 190 ground water quality parameters. The first factor accounted for 28.54% of the total variation in ground water quality indicators, and high loadings were revealed on EC, Ca, Mg, K, Na, $NH_4\;^+-N$, and $SO_4$. Twenty-nine pairs of reservoir water quality indicators were significant out of 66 pairs. The first factor accounted for 37.06% of the total variation in reservoir water quality indicators, and high loadings were shown on EC, Mg, K, Na, SS, T-P, Cl, and COD. These results demonstrate that EC was the first factor contributing to water quality.
