Enhanced Production of Exopolysaccharides by Fed-batch Culture of Ganoderma resinaceum DG-6556

  • The objectives of this study were to optimize submerged culture conditions of a new fungal isolate, Ganorderma resinaceum, and to enhance the production of bioactive mycelial biomass and exopolysaccharides (EPS) by fed-batch culture. The maximum mycelial growth and EPS production in batch culture were achieved in a medium containing 10 g/l glucose, 8 g/l soy peptone, and 5 mM $MnCl_2$ at an initial pH 6.0 and temperature $31^{\circ}C$. After optimization of culture medium and environmental conditions in batch cultures, a fed-batch culture strategy was employed to enhance production of mycelial biomass and EPS. Five different EPS with molecular weights ranging from 53,000 to 5,257,000 g/mole were obtained from either top or bottom fractions of ethanol precipitate of culture filtrate. A fed-batch culture of G. resinaceum led to enhanced production of both mycelial biomass and EPS. The maximum concentrations of mycelial biomass (42.2 g/l) and EPS (4.6 g/l) were obtained when 50 g/l of glucose was fed at day 6 into an initial 10 g/l of glucose medium. It may be worth attempting with other mushroom fermentation processes for enhanced production of mushroom polysaccharides, particularly those with industrial potential.

Effects of Caloric Restriction on Endocrine Functions and Body Fat Distribution in Overweight Premenopausal Women, Related to their UCP3 (Uncoupling Protein 3) Genotypes

  • A mutation in the promoter region of uncoupling protein 3 (UCF3), specifically the -55C longrightarrow T transition, may influence an individual's energy metabolism and body weight. The objective of this study was to investigate the effect of a weight reduction program on endocrine functions and body fat distribution, related to UCP3 promoter genotype. Ninety overweight pre-menopausal female subjects participated in the weight reduction program at Yonsei University Hospital, and were placed on a calorie-restricted diet (300 kcal less than their daily requirements) for 12 weeks. After 12 weeks, all subjects on the program lost approximately 5% of their initial body weights and had lower Body Mass Index (BMI) values. Among the 90 women, 56 had a normal (without mutation) UCP3 genotype, while 34 women had mutations in the promoter region of UCP3. Despite similar weight reductions in both groups, a significantly higher decrease in abdominal adipose tissue was observed in the normal UCP3 genotype group, compared to the group with mutations. In particular, there was a significant reduction of fat at the lumbar 1 (Ll) level in the without-mutation group. Serum levels of total cholesterol, apolipoprotein Al were significantly decreased in the without-mutation group, by 4.4% and 5.7% respectively. Serum levels of hormones were not significantly changed in both groups artier the intervention. However, in the group without the mutations, the leptin level significantly reduced by 23.4% (p<0.001). Serum free fatty acid (FFA) concentration was significantly increased in the group with mutation following the weight reduction program. On the other hand, FFA responses were shown similar increases in both groups. In conclusion, although no difference was found in the magnitude of weight reduction in both groups, there were significant differences in body fat distribution and in endocrine function between the groups.

Growth and Ginsenoside Content of One Year Old Ginseng Seedlings in Hydroponic Culture over a Range of Days after Transplanting (수경재배 시 1년생 묘삼 이식 후 경과일수에 따른 인삼의 생육 및 Ginsenoside 함량)

  • Background: Ginseng produced by hydroponics can be cultivated without using agricultural chemicals; thus, it can be used as a raw materials for functional foods, medicines, and cosmetics. This study aimed to determine the optimal harvesting time to obtain the highest levels of ginsenoside and ginseng, as this was not previously unknown. Methods and Results: One-year-old organic ginseng seedlings were transplanted and cultivated using hydroponics for 150 days in a venlo-type greenhouse, using ginseng nursery bed soil and a nutrient solution ($NO_3{^-}-N$; 6.165, P; 3.525, K; 5.625, Ca; 4.365, Mg; 5.085, S; $5.31mEq/{\ell}$). Ginsenoside content and fresh and dry weights were higher at 120 days after transplanting than at 30, 60, 90, and 150 days. Total ginsenoside content was 11.86 times higher in the leaf and stem than in the root at 120 days after transplanting. Ginsenosides F1, F2, F3, and F5 were detected in ginseng leaves and stems. These chemical compounds are known to be effective in altering skin properties, including whitening, anti-inflammation, and anti-aging. Conclusions: Optimal harvesting time for ginseng cultivated using hydroponics was 120 days after transplanting when the biomass and ginsenoside content were highest.

The Anti-Obesity Activity of Syzygium aromaticum L. in High-Fat Diet-induced Obese Mice (고지방식이로 유도된 비만 마우스에서 정향(丁香)의 항비만 효과)

  • Objectives : This study aims to analyze the anti-obesity effect of Syzygium aromaticum L. (SA) in obese mice made by a 60% high-fat diet (HFD). Methods : The antioxidant activities of SA were evaluated in vitro. To assess the anti-obesity effect of SA, male C57BL/6 mice were divided into five groups: Normal, Control, GC100 (Garcinia cambogia 100 mg/kg/day), SA100 (SA 100 mg/kg/day), SA200 (SA 200 mg/kg/day). All groups underwent a 6-week regimen of HFD and oral administration, except for the Normal group. Subsequently, we performed blood analysis, western blotting, and histopathological staining. Results : SA demonstrated effectiveness in antioxidant measurements. SA treatment resulted in a significant decrease in body weight gain, along with reductions in liver and epididymal fat weights. Serum triglyceride (TG), total cholesterol (TC), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and leptin levels were reduced with SA treatment. Moreover, in the SA100 group, the reduction of both TG and TC synthesis was caused by inhibiting the sterol regulatory element-binding transcription factor 1 (SREBP-1) and sterol regulatory element-binding transcription factor 2 (SREBP-2) through the Sirtuin 1 (Sirt1)/phospho-AMP-activated protein kinase (p-AMPK) pathway. Furthermore, SA treatment at a dose of 100 mg/kg reduced the accumulation of lipid droplets in the liver and the adipocyte size of the epididymal fat. Conclusion : Our research reveals the anti-obesity effects of SA by demonstrating its ability to inhibit body weight gain and lipid accumulation, suggesting that SA might be promising for obesity treatment.

Effects of Feeding Induced Molting on the Performance, Egg Quality, and Visceral Organs in Laying Hens (산란계에서 급이환우가 산란계의 생산성과 계란의 품질 및 주요 장기에 미치는 영향)

  • This study was conducted to compare the effect of feeding molting and fasting molting on the performance, egg quality, and visceral organs in laying hens for animal welfare. Eighty one 62-wk-old White Leghorn hens that egg production was over 80% and average weight was $1.6{\pm}0.3\;kg$ were used in this study. Treatments were control (non-molt treatment), feeding molt treatment, and fasting molt treatment. The three treatments were administered to three replicate group of nine hens wherein each group. All treatment groups were fed the basal diet (CP 15%, ME 2,700 kal/kg) for two weeks as the adaptation period. Induced molt diets contains low CP (6.7%) and low energy (2,200 kal/kg). Test periods were 14 days for feeding molting and 10 days for fasting molting. Egg production decreased to be 0% at 10 days of feeding molting treatment, but at 2 or 3 days of fasting molting treatment. Egg production restarted after 19 days ending molt at feeding molting treatment, while after 24 days at fasting molting treatment. On the egg quality was improved at molting treatments (p<0.05) except egg yolk. Egg shell tissue was crowded at molting treatment to compare to control. Liver weights, heart weight, and oviduct weight of laying hens decreased at molting treatments (p<0.05). Finally, feeding molting might could be replaced fasting molting on the welfare and further studies were needed about molting program.

Applicability of Artificial Light Source and Newly Developed Growing Medium for Lettuce Cultivation in a Closed-type Plant Production System (밀폐형 식물생산시스템에서 상추재배를 위한 인공광원과 신개발 배지의 적용)

  • This study was conducted to evaluate the growth characteristics of lettuce (Lactuca sativa L.) as affected by artificial light sources and different growing media in a closed-type plant production system (CPPS). The lettuce seeds were sown in the 128-cell plug tray filled with 5 different growing media such as urethane sponge (US), rock-wool (RW), Q-plug (QP), TP-S2 (TP) and PU-7B (PU). The germination rate of lettuce seeds was examined during 12 days after sowing. On the 13 days after sowing, the lettuce seedlings were transplanted in a CPPS with temperature $25{\pm}1^{\circ}C$ and nutrient solution (EC $2.0dS{\cdot}m^{-1}$, pH 6.5) using recirculating deep floating technique system. The light sources were set with FL (fluorescent lamps) and combined RB LEDs (red : blue = 7 : 3) with $150{\pm}10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD and a photoperiod of 14/10 hours (light/dark). The initial germination rate of lettuce was the highest in TP. The final germination and mean daily germination were the significantly highest in RW, QP and TP. The plant height, leaf length, leaf width, leaf area, and fresh and dry weights of shoot were the greatest in QP irradiated with RB LED. The number of leaves, fresh and dry weights of root and SPAD were the greatest in QP and TP irradiated with RB LED. The root length was the longest in TP irradiated with RB LED. Therefore, these results indicate that RB LED was effective for the growth of lettuce and it was also found that the QP and TP were effective for the germination and growth of lettuce in a CPPS. In addition, we confirmed the applicability of the newly developed growing medium TP for the lettuce production in a CPPS.

The Protective Effects of Ethanol Extract of Wild Simulated Ginseng on Carbon Tetrachloride Induced Acute Hepatic Injury in Mouse (사염화탄소 유발 급성 간독성 생쥐모델에서 산양삼 에탄올 추출물의 간 보호 효과)

  • The wild simulated ginseng (WSG) has been effectively used in folk medicine as a remedy against hepatic disease, hypertension and arthritic disease. However, there is still lack of scientific proof about its antioxidant capability. The present study has been conducted to evaluate the protective role of the WSG ethanol extract in the CCl4-induced oxidative stress and resultant hepatic disfunction in ICR mice. The electron donating abilities and IC50 of WSG etnanol extract were 76.86 ${\pm}$ 1.06% and 33.3 ${\mu}g$/mL (that of ascobic acid was 16.5 ${\mu}g$/mL), respectively. Total antioxidant status of WSG extract was 2.13 ${\pm}$ 0.06 mmoL/mg, while the values of ascorbic acid and BHT were 3.63 ${\pm}$ 0.06 and 3.12 ${\pm}$ 0.02, respectively. ICR mice (aged 3weeks) were fed for 4 weeks on AIN-93M diet and had free access to food and water. The animals were divided into three groups: normal group (intraperitoneally (i.p) injected with PBS at 100 ${\mu}L$/mouse), group C; CCl4-induced and without any treatment. (i.p injected only PBS, 100 ${\mu}L$ /mice), group G; CCl4-induced and treated with WSG (i.p injected with 5 mg WSG extract per mouse, suspended in 100 ${\mu}L$ phosphate buffer). After the i.p. injection of WSG or PBS (5 times for 7weeks), all mice were administered CCl4 in olive oil at the last day of the experiment, except for normal group. The normal group was administered only olive oil. Determination of plasma triglyceride, total cholersterol, fasting glucose and GPT activity was performed using automatic blood analyzer. To evaluate the protective effect against the oxidative stress, DNA fragmentation and TBARS were determined in blood leucocytes and RBC and hepatocyte, respectively. Body and organs weights and food intake did not show significant differences among the groups. Blood total cholesterol of group G was similar to that of normal group, which was the lowest in group C. The fasting blood glucose level was the highest in normal group (205.20 ${\pm}$ 135.24), which were decreased in group C (134.2 ${\pm}$ 79.31) and group G (126.48 ${\pm}$ 77.05). TBARS values in a red blood cell and hepatic tisuue homogenate were lower in group G comparing to the group C. DNA% in tail, tail length (TL) and tail moment (TM) of blood leucoocytes showed the highest values in group C (20.11 ${\pm}$ 2.47, 17.36 ${\pm}$ 2.58, 94.11 ${\pm}$ 12.29) and they were significantly diminished in group G (9.63 ${\pm}$ 1.19, 7.04 ${\pm}$ 1.50, 38.64 ${\pm}$ 7.60). In conclusion, wild simulated ginseng might be a protective agent against the oxidative stress.

Effect of Non-starch Polysaccharides and Resistant Starch on Mucin Secretion and Endogenous Amino Acid Losses in Pigs

  • Generally, dietary fibre (DF) includes lignin, non-starch polysaccharides (NSP) and resistant starch (RS). In monogastric species, low levels of dietary fibre in the diet are associated with various diseases and high levels reduce nutrient digestibilities. In this study, the effects of different types and levels of NSP (soluble: $\beta$-glucan, insoluble cellulose) and resistant starch on mucin secretion and endogenous nitrogen and amino acid losses in pigs were investigated. A total of 25 five-week-old weaner pigs (9.5 kg${\pm}$1.5 kg), were randomly allocated to each of five experimental diets. Different levels of purified barley $\beta$-glucan (BG) extract (5 or 10% of $Glucagel^{(R)}$ $\beta$-glucan, providing 4 or 8% of BG in the diet), and resistant starch (RS) (8.3 or 16.6% of Hi-$Maize^{TM}$, providing 5 or 10% RS in the diet) were substituted for wheat starch in a purified diet in which enzymatically-hydrolysed casein was the sole source of protein. The diets were fed for 21 days. No statistically significant difference between treatments (p>0.05) was observed for growth performance and organs weights. No difference in ileal starch digestibility was observed between pigs on the cellulose or $\beta$-glucan diets. However, as the level of resistant starch in the diet increased the ileal starch digestibility decreased (p<0.05). The inclusion of resistant starch in the diet (5 or 10%) did not increase mucin production when compared with the cellulose-only diet. However, as the level of beta-glucan in the diet increased, both crude mucin in the digesta dry matter and per kg dry matter intake increased (p<0.05). Pigs fed the diet containing 8% of beta-glucan had higher endogenous loss flow than those fed the diets including 5 or 10% of resistant starch or 4% of $\beta$-glucan. In conclusion, dietary inclusion of resistant starch increased the level of starch reaching the large intestine without any effect on mucin secretion, or endogenous nitrogen or amino acid losses content in the small intestine. The addition of $\beta$-glucan to a diet containing cellulose increases both mucin secretion and endogenous amino acid and nitrogen losses in the small intestine.

Association of Insulin-Like Growth Factor-I (IGF-I) Gene Polymorphism with Serum IGF-I Concentration and Body Weight in Korean Native Ogol Chicken

  • IGF-I is involved in the regulation of growth and differentiation in mammals, but its role as a modulator of growth and metabolism in poultry is poorly understood. And, no studies have so far been reported for the comparison between serum IGF-I concentration and body growth in the egg type or the dual purposes (meat and egg type) chicken including the Korean Native Ogol Chicken (KNOC). Therefore, in order to improve the body growth and meat production of the KNOC, this study was conducted for the identification of the polymorphism of IGF-I gene and for its possible association with both body weight and IGF-I concentration. The RFLP patterns for IGF-I gene were identified by the PstI restriction enzyme. The frequencies of +/+, +/-, and -/- genotype were 16.9%, 51.7%, and 31.4%, respectively. Any statistical significance was not observed in all variations except for sex variation (p<0.01) by covariate quadratic model. The significant effect of the IGF-I genotype on body weight by sex indicates that there are different physiological characteristics in gender. Although the body weights of male KNOCs in most ages were not significant, there was a tendency of KNOCs with +/+ IGF-I genotype to be heavier than those with any other genotypes. But all IGF-I genotypes in female did not influence on body weight. The ANOYA revealed no significant effects of IGF-I genotypes on serum IGF-I concentration but sex effect was highly significant on the IGF-I concentration at 20 and 40 weeks (p<0.01). Although the +/+ genotype, in gender, tended to express a higher IGF-I concentration than the other genotypes at all ages in males, a statistical difference among the genotypes was not found except for 60 weeks (p<0.05). Furthermore, since body weight and IGF-I genotypes are associated, it is possible to improve KNOC to a meat type breed if a continuous selection can be made for the body weight and/or IGF-I traits.
