Heterosis Effects of Body Weight and Jumping Height in Rotational Crossing of Two-Subspecies of Mice

  • The present study was conducted to evaluate heterosis effects of body weight and jumping height for successive generations of rotational crossing using two subspecies of mice which are very different in body weight and in genetic relationship from each other. Domesticated laboratory mouse $CF_{{\sharp}1}$ (C) and Yonakuni wild mouse (Y) were used as materials. Two groups of rotational crossing was made according to the parent used at the beginning of crosses, C male$\times$Y female and Y male$\times$C female. These crosses were done to produce the first ($G_1$ and $G_1{^{\prime}}$), second ($G_2$ and $G_2{^{\prime}}$) and third generations ($G_3$ and $G_3{^{\prime}}$) with sire used was alternated. Individual body weights were weighed at 1 (wk1), 3 (wk3), 6 (wk6) and 10 weeks of age (wk10) and jumping heights were measured at six weeks of age (wk6). Only the first litter used. For body weight, results of this study showed that genetic group effects were significant (p<0.01) source of variation at all ages studied. Sex effects were significant (p<0.01) at wk3, wk6 and wk10, but not at wk1. Significant interaction effects (p<0.01) between genetic group and sex were found at wk6 and wk10. The C mice with large maternal effects produced heavier offspring body weight and crosses using sire of this subspecies maintained heavy weight compared to wild Y mouse sire that has small body size. Heterosis tended to exist at the rotational crossing started from Y male C female. For jumping height, effects of genetic group and sex were significant, sire and dam effects (heterosis) exhibited from the first to third generations, and no maternal effects were observed.

Growth Rates of Transgenic Mice Containing Growth Hormone Receptor Gene

  • Transgenic mice containing GH Receptor (GHR) gene fused to metallothionein promoter were analyzed to evaluate effect of GHR expression on growth in vivo. Three founder mice lines contained copies of GHR transgene and transmitted these genes into F$_1$ and F$_2$ progenies. The mRNA expression of transgene was identified using RT-PCR with GHR genes in tissues. To analyze the effects of transgenes on growth performance, body weights of pups were measured at 4, 10 and 14 weeks after birth. The body weight of transgenic mice was higher compared with that of non-transgenic control mice regardless of sex (P<0.05). Body weights between transgenic and non-transgenic mice were increased with aging. Overall, GHR transgenic mice tended to grow about 10 to 15 % faster than non-transgenic mice without any pathological defects.

Isolation and Characterization of Lectins from Stem and Leaves of Korean Mistletoe (Viscum album var. coloratum) by Affinity Chromatography

  • We attempted to isolate and characterize the lectins from stem and leaves of Korean mistletoe (Viscum album var. coloratum) by affinity chromatography. Lectin I was isolated only from stem. Lectin II was not isolated from Korean mistletoe, whereas lectin III was isolated from the stem and leaves. The hemagglutinating activity of lectin I was 16HU and inhibited by D-galactose, lactose, and N-acetyl-D-galactosamine. The lectin I has molecular weight of 60, 000D being composed of two basic subunits with molecular weights of 32, 000D and 28, 000D which are linked by a disufide bond. The lectin III from stem has molecular weight of 66, 000D being two basic subunits which have molecular weights of 34, 000D and 29, 000D and are linked by a disufide bond. The activity of lectin I was stable at the pH range of 4.00-8.50 and at a wide range of temperature (0-42.deg. C). The lectin I showed more potent mitogenic activity to murine lymphocytes than concanavalin A.

새우젓으로부터 혈전과 chitin 분해능을 지닌 균주 Bacillus licheniformis SC082의 분리 및 특성 (Isolation and Characterization of Bacillus licheniformis SC082 Degrading Fibrin and Chitin from Shrimp Jeot-Gal)

  • 전통발효식품인 새우젓으로부터 혈전과 chitin 분해력이 우수한 균을 분리하였으며 16S rRNA 염기서열 분석으로 B. licheniformis와 가장 유사한 균주임을 확인하였다. 이 균주를 B. licheniformis SC082로 명명하였고, 최적 생육조건은 $37^{\circ}C$, pH 7.0, 염 농도 6%로 확인되었다. 이 균주의 기질특이성을 조사한 결과, 우수한 혈전분해력을 나타냈으며 1% 농도의 colloidal chitin의 첨가에 의하여 chitinase 활성이 증가됨을 관찰할 수 있었다. 또한 지질 분해능은 없었고 약한 skim milk 분해력을 가지고 있었다. SDS-PAGE와 zymogram 분석 결과, 이 균은 혈전분해효소 isozyme과 chitinase isozyme을 생성하는 균주로 확인되었다. 그 대략적인 분자량은 각각 22.0, 66.0, 72.0 kDa과 55.0, 62.0 kDa이었다. B. licheniformis SC082 균주가 생성하는 혈전분해효소는 pH 9.0 그리고 $50^{\circ}C$까지 안정하게 유지되었고, 이와 더불어, chitinase 활성은 pH 5, $45^{\circ}C$일 때 높게 나타났다. B. licheniformis SC082 균주의 DPPH 전자공여능법에 의한 항산화력은 농도의 증가에 따라 상승되었는데 $20\;{\mu}g$의 균상등액에 대한 항산화력은 약 31%으로 나타났다.

  • 혈합육어 중 연근해 온대산 멸치의 내장과 고등어의 유문수, 그리고 원양 열대산 황다랭이 및 날개다랭이의 유문수를 각각 시료로 하여 trypsin을 분리 정제하였으며, 그 분자량과 아미노산 조성 및 효소의 반응 최적조건에 관하여 비교 분석하였다. 실험결과를 요약하면 다음과 같다. 1. 멸치의 내장과 고등어, 황다랭이 및 날개다랭이의 유문수에서 각각 황산암모늄염석, benzamidine-Sepharose 6B 친화성 크로마토그래피, DEAE-Sephadex A-50 크로마토그래피, Sephadex G-75 겔 여과 크로마토그래피 등의 방법을 혼용하여 고등어로부터는 2종의 trypsin (고등어 trypsin A와 고등어 trypsin B로 명명함)을, 그리고 다른 어류로부터는 각각 1종의 trypsin을 분리 정제하였다. 2. 멸치 trypsin과 고등어 trypsin B는 고등어 trypsin A와 황다랭이 trypsin 및 날개다랭이 trypsin에 비하여 그 고유활성이 월등히 높았다. 3. 이들 혈합육어 trypsin의 분자량은 22kDa-26kDa 범위였다. 4. 이들 trypsin은 공통적으로 glycine, serine, aspartic acid를 많이 함유하고 있었으며, tryptophan, methionine, lysine, tyrosine의 함유량은 적었다. 5. BA-$\rho$-NA기질에 대한 반응 최적조건은 멸치 trypsin은 pH 8.0에서 45$^{\circ}C$, 고등어 trypsin A와 B는 pH 8.0에서 5$0^{\circ}C$, 그리고 황다랭이 trypsin은 PH 9.0에서 55$^{\circ}C$, 날개다랭이 trypsin은 pH 9.0에서 5$0^{\circ}C$였다. 6. 위에 든 실험 결과들은 혈합육 어류의 서식온도가 이들 어류의 trypsin의 반응 최적 온도와는 어느 정도 관계가 있을 것으로 추측되었다.

Differences in Flood-Stress Tolerance among Sprout Soybean Cultivars

  • In this study, the response of 22 soybean cultivar sprouts to flooding stress was investigated. After sprouts were subjected to flooding stress for 10 days at the V4-V5 growth stages, their dry weights were compared. All plants were kept in a greenhouse under natural light conditions, an average daytime temperature of $35.6{\pm}5.3^{\circ}C$ and an average nighttime temperature of $18.2{\pm}1.7^{\circ}C$. Soybeans were grown in a concrete bed filled with silt loam soil. Subjecting plants to flooding stress resulted in a large reduction in plant dry weight, plant height, number of nodes, and number of leaves. Cultivars differed significantly in their responses to flooding stress, as indicated by these characteristics (p<0.05). Soybean cultivars were classified into three groups based on their degree of flood tolerance: strong, moderate, and weak. Hannamkong, Namhaekong, Sobaegnamulkong, and Sorogkong had strong tolerance for flood conditions. Tawonkong, Pureunkong, Eunhakong, Myeongjunamulkong, Doremikong, Saebyeolkong, Paldokong, Sowonkong, Pungsannamulkong, Dagikong, Dachaekong, and Anpyeongkong had weak tolerance for flood conditions.

Microwave 처리가 알찜의 품질 특성에 미치는 영향 (Characteristics of egg coagulates cooked conventionally or by microwaves)

  • Comparisions were made for cooking times, internal temperatures, thiamin contents and textural properties of various portion weights of egg mixture cooked in a microwave oven at high and/or low power levels and in a conventional double boiler. The mean internal temperatures of conventionally cooked egg mixtures were 76.4~80.7$^{\circ}C$. When cooking was made by steaming, the mean internal temperature were comparable among samples. With the microwave cooking, the range of mean temperature was 83.8~96.4$^{\circ}C$, and they were significantly higher than the conventionally cooked egg mixtures. The hardness determination was conducted using universal testing machine and a taste panel. Samples cooked with steam or with larger portion weight had softer texture than samples cooked by microwaves or smaller amount, respectively. Mean internal temperature was significantly(r=0.99, p<0.05) correlated with mechanical hardness determination. Statistical analyses indicated no significant difference in the thiamin content among various treatments.

  • A new class of soluble PPV derivatives containing dimethyldodecylsilylphenyl unit as a pendant was synthesized by Gilch polymerization method. The resulting electroluminescent (EL) polymers showed good solubility, good film-forming ability onto the ITO substrate, and exhibited an amorphous morphology due to dimethyldodecylsilylphenyl branched group linked to the polymer backbone. The weight average molecular weights and polydispersities of the present EL polymers were in the range of 8.0-80.0 x $10^{4}$ and 2.67-7.80, respectively. The resulting EL polymers revealed a high thermal stability of up to $355-410^{\circ}C$. Their glass transition temperatures were in the range of $104-251^{\circ}C$. The emission colors could be tuned from green to orange-red colors by changing the MEH-PPV contents in copolymer systems. The turn-on voltages of the EL polymers were in the range of 1.8-4.0 V.

Characteristics of Chitosanases from Aspergillus fumigatus KB-1

  • Two chitosanases produced by Aspergillus fumigatus KB-1 were purified by ion exchange and size exclusion chromatographies. Molecular weights of chitosanases were 111.23 kDa (chitosanase I) and 23.38 kDa (chitosanase II). The N-terminal amino acid sequence of chitosanase II was determined as follows: YNLPNNLKQIYDKHKGKXSXVLAKGFTN. The optimum pH of the chitosanase I and II was 6.5 and 5.5, respectively. The optimum temperatures were $60^{\circ}C$ for chitosanase land $70^{\circ}C$ for chitosanase II. Hydrolysis products of two chitosanases were analyzed by HPLC and GPC. Chitosanase I hydrolyzed substrate to glucosamine. Chitosanase II produced chitooligosaccharides.

  • This research was conducted to determine the influence of physiological and chemical properties of root substrates on the growth of the daughter plants of 'Maehyang' strawberry produced by bag culture. The daughter plants produced by stock plants during bag culture were individually separated and grown in 10-cm diameter plastic pots containing six different formulations of root substrates: a) 50% peatmoss plus 50% vermiculite (5:5 by volume, A), b) 70% peatmoss plus 30% perlite (7:3, B), c) 70% coir dust plus 30% perlite (7:3, C), d) mixture of 35% coir dust, 35% peatmoss, and 30% perlite (3.5:3.5:3.0, D), e) mixture of 20% rice hull, 70% coir dust, and 10% perlite (2:7:1, E), and f) 30% rice hull plus 70% coir dust (30:70, F). The container capacity and air filled porosity of the growing medium varied greatly among the six substrate formulations evaluated. The substrates E and F had less container capacity and higher air-holding spaces than the rest of the formulations. Therefore, these two formulations (E and F) may cause a problem in water management during the production of healthy daughter plants. The substrate formulations A, B, and D retained higher nitrogen (N) concentrations than other formulations containing coir dust or rice hull. The substrate formulations E and F which contained rice hull had lower N, phosphorus (P), and potassium(K) concentrations than other substrate formulations containing coir. The quality of the daughter plants grown in all six different substrate formulations was good with the crown diameters at around 10 mm. Fresh weights of the daughter plants grown in substrate formulations A, C, and D were higher than those obtained from B, E, and F. Dry weights of the daughter plants showed a similar trend. The daughter plants having high fresh and dry weights and increased crown diameter are in demand by the industry. For this reason, the substrate formulations A, C and D can readily be used as potting mixes during the production of 'Maehyang' strawberry transplants utilizing the bag culture system.
