Effect of Growth and Culture Conditions of Paecilomyces japonica and Cordyceps militaris on the Formation of Bioactive Substance

  • To investigated the effect of liquid culture conditions and nutrient sources on the formation with bioactive substance of Paeilomyces japonica and Cordyceps militaris cultivated in the country, the result are as follow; The growth temperature of two mycelia is 25$^{\circ}C$ and the proper temperature for cordycepin growth is around 20$^{\circ}C$. The formation amount of bioactive substance by nutrient sources reached its peak with using 2% glucose and 1% galactose in case of carbon sources and 0.4% inorganic compound in case of nitrogen sources. Also, the ratio of C/N was optimal with 3% glucose 1% peptone. For a natural medium, most grains were sufficient but the soybean oil was superb. The formation amount of protein-binding polysaccharide that are used for anticancer substance was in proportion to the growth rate of mycelium, had lots of aeration and showed a trend of increasing when the acidity lower. and the content of structural protein showed a trend of increasing when the acidity lower. However, the content of the structural hexosamin did not get a great the effect of culture conditions and nutrient sources. The constitution of monosaccharide that organizes a protein-binding polysaccharide greatly changed in proportion to carbon sources. When Paecilomyces japonica cultured in a silkworm larvae for 30 days, the content of cordycepin was 204.5 mg/100mL as a dry weight in the fruiting body, 41.8 mg/100mL in mycelium and larva, and the content for each bottle was average 29.5 mg/100mL. In case of Cordyceps militaris for 45days, the fruiting body was 563.5 mg/100mL, the larva and the mycelium was 86.1 mg/100mL, and the content for each bottle was average 65.0 mg/100mL.

$\Omega$-3계 지방산이 다량 함유된 꽁치 너겟의 텍스쳐 특성에 관한 연구 (Study on the Texture Characteristics of High Omega-3 Fatty Acid Saury Nuggets)

  • The principal objective of this study was to determine the appropriate water content for optimal texture in high omega-3 fatty acid saury nuggets. The approach adopted in this study essentially involved variations in water levels(0{\sim}25%$). The main ingredients of the nuggets included saury mince, mild pizza cheese and hydrated textured soy protein concentrate. The formulated products were molded(dia. 4.5, thickness 1.5 cm, 20 g), battered lightly, flash-fried far 4 min at $160^{\circ}C$ and frozen until they were tested. The frozen nuggets were cooked to $65^{\circ}C$, then subjected to sensory evaluation, texture analysis at 80% deformation, and expressible fluid tests. The moisture contents in the nugget batter and the cooked nuggets were determined. In our sensory evaluation, nugget texture was adjudged the best in the nuggets prepared with a 20% water level(S5). The hardness and chewiness in the nuggets were reduced with increases in the water level. However, when the nuggets contain more than 15% water(S4, S5, S6), we noted no significant differences in the hardness values. The difference in moisture content between the uncooked nugget bat or and the cooked nugget was most profound in the nugget prepared with a water level 25%(S6). The nugget prepared with a water level 25% was also the highest in expressible fluids, and was also highest in internal moisture content. The texture preference of nuggets was correlated positively with moistness(r=0.881) and moisture content(r=0.827), and correlated negatively with firmness(r=-0.870) and cohesiveness(r=-0.839), and these correlations were significant.

푸새 직물의 흡습률에 관한 1연구 (On the rate of absorption of the sized fabrics)

  • This paper aims to examine the variety of the absorption of cotton, T/C and hemp fabrics seperately seperaterly sized by rice, wheat, potato, corn flour and pp.V.A.. Experimental variables occurring in the concentration of sizing agents, the water content of unsized fabrics and an iron temperature show the following results, 1. When fabrics sized, the rate of absorption increases according to the order of rice, corn, pp.V.A., potato, wheat flour for cotton fabrics, of rice, corn, pp.V.A., wheat, potato flour for hemp fabrics and of the rice, pp.V.A., potato, corn, wheat flour for T/C fabrics : rice flour shows an absorption rate highest among all the others mentioned above. 2. To a certain extent, the stronger the concentration of sizing agents, the higher the rate of absorption. 3. The higher fabrics density, the higher absorption rate. 4. The structure and hydrophilic property of the sized fabrics affect the rate of absorption. 5. The fabrics with water content of 20% before sizing it shows the rate of absorption highest. 6. An iron temperature after sizing fabrics shows the rate of absorption highest at the properest at the properest one : $180^{\circ}C$ for cotton, $150^{\circ}C$ for T/C and $200^{\circ}C$ for hemp fabrics.

Effect of Al Alloy Content on Processing of Reaction-Bonded Al2O3 Ceramics Using Al Alloy Powder

  • The effect of Al content on the processing of reaction-bonded $Al_2O_3$ (RBAO) ceramics using 40v/o ~ 80v/o Al-Zn-Mg alloy powder was studied in order to improve traditional RBAO ceramic processes that use ~ 40v/o pure Al powder. The influence of high Al content in starting $Al_2O_3$-Al alloy powder mixtures on its particulate characteristics, reaction-bonding, microstructure, physical and mechanical properties was revealed. Starting $Al_2O_3$-Al alloy powder mixtures with 40v/o ~ 80v/o Al alloy powder were milled, reaction-bonded, post-sintered, and characterized. With an increasing Al alloy content, the milling efficiency of Al alloy powder was lowered, resulting in a larger particle size after milling. However, in spite of the larger particle size of Al alloy powder, the oxidation, i.e., reaction-bonding, of the Al alloy was successfully completed via solid and liquid state oxidation, in which the activation energy of the oxidation was nearly the same regardless of Al alloy content. After reaction-bonding and post-sintering at $1600^{\circ}C$, RBAO ceramics from 80v/o Al alloy content showed a relative density of ~97% and a flexural strength of 251 MPa compared to ~ 96% and 353 MPa for RBAO ceramics from 40v/o Al alloy content, respectively. The lower flexural strength at 80v/o Al alloy content was due to the weak spinel phase that formed from Zn, Mg alloying elements in Al.

Structural and Optical Properties of Sol-gel Derived MgxZn1-x Thin Films

  • In this report, the structural and optical properties of sol-gel derived $Mg_xZn_{1-x}O$ thin films upon changes in the composition and annealing temperature were investigated. The $Mg^{2+}$ content and the annealing temperature were varied in the range of $0{\leq}x{\leq}0.35$ and $400^{\circ}C{\leq}T{\leq}600^{\circ}C$, respectively. The films exhibited a hexagonal wurtzite structure of a polycrystalline nature. The optical transmittance exceeded 85% and the optical band gap of the film was tuned as high as 3.84 eV at a value of x = 0.35 (annealed at $400^{\circ}C$), which was evidently the maximum $Mg^{2+}$ content for the single-phase polycrystalline $Mg_xZn_{1-x}O$ thin films prepared in this experiment. The optical band gap and photoluminescence emission were tailored to the higher energy side while maintaining crystallinity without a significant change of the lattice constant.

코끼리조개의 성분 조성과 냉동 저장 중의 성분 변화 (Chemical Composition of Korean Geoduck and Changes in Their Composition during Frozen Storage)

  • 우리나라 동해안 연안에서만 서식하고 있는 코끼리조개의 식품학적인 자료를 얻기 위하여 계절적 성분 변화와 냉동 저장 중의 지질, VBN, TMAO 및 TMA, Total creatine, $NH_2$-N 그리고 단백질 변화를 분석하여 비교 검토한 결과는 다음과 같다. 1. 계절적 성분 변화는 수분이 평균 79.9%이며, 7월이 82.2%로 가장 높고, 10월이 78.1%로 가장 낮았다. 조단백질은 평균 13.7%로, 7월이 12.3%로 가장 낮고 1월에 16.2%로 높았다. 조지방은 평균 1.1%, 회분은 평균 1.4%로서 지방보다 높았다. 2. 휘발성 염기질소의 변화는 저장 첫날에 내장을 포함한 육, 생육, blanching한 육에서 1.6mg/100g, 0.6mg/100g, 1.1mg/100g이었으며, 저장 90일에는 13.5mg/100g, 12.0mg/100g, 10.1mg/100g으로 blanching한 육에서 휘발성 염기질소량의 증가가 적었다. 3. 저장 중의 TMAO의 변화는 내장을 포함한 것과 생육이 저장 첫날에 2.4mg/100g, 2.8mg/100g이던 것이 저장 90일에 0.1mg/100임으로 감소하였고, blanching한 육도 저장 기간 동안 감소하였다. 한편, TMA 함량의 변화는 내장을 포함한 육, 생육, blanching한 육에서 저장 기간에 따라 증가하였다. 4. 동결 저장 중 아미노태질소의 변화는 내장을 포함한 육이 저장 초기에 24.1mg/100g, 생육 17.8mg/100g, blanching한 육 14.2mg/100g이던 것이 저장 90일에 각각 93.1mg/100g, 59.3mg/100g, 62.0mg/100g으로 증가하였다. 5. total creatinine의 변화는 내장을 포함한 육이 12.3mg/100g, 생육 10.5mg/100g, blanching한 육 8.3mg/100g으로서 내장을 포함한 육에서 많았으며, 저장 90일에는 143.3mg/100g, 104mg/100g, 151mg/100g으로 증가하였다. 6. 동결 저장 중의 과산화물 값(peroxide value)의 변화는 저장 초기에 내장을 포함한 육과 생육, blanching한 육에서 각각 0.41meq/kg, 0.38meq/kg, 0.44meq/kg이던 것이 증가를 보이다가, 저장 90일에는 3.72meq/kg, 3.72meq/kg, 1.44meq/kg으로 증가하였으며, 특히 내장을 포함한 육에서의 과산화물 값의 증가가 높았다. 그러나 전체적인 함량은 적었다. 7. 총지질의 지방산 조성 중 포화 지방산은 내장을 포함한 육이 39.3%, 생육 37.0%, blanching한 육 39.6%이며, 주요 지방산은 $C_{14}$ : 0, $C_{16}$ : 0, $C_{18}$ : 0였으며, Monoenoic acid은 내장을 포함한 육이 19.8%, 생육 22.2%, blanching한 육 22.4%이고, 주요 구성 지방산은 $C_{16}$ : 1, $C_{18}$ : 1, $C_{20}$ : 1이었다. 그리고 polyenoic acid은 내장을 포함한 육이 40.9%, 생육 40.8%, blanching한 육 38.0%로서 내장을 포함한 육의 함량이 높았으며, 주요 구성 지방산은 $C_{20}$ : 5, $C_{22}$ : 6이었다. 동결 저장 중의 변화는 포화 지방산이 내장을 포함한 육, 생육, blanching한 육이 각자 39.3%, 36.9%, 39.6%에서 저장 90일에 44.8%. 41.9%, 42.6%로 증가하였으며, 반대로 불포화 지방산은 내장을 포함한 육, 생육, blanching한 육이 40.9%, 40.8%, 38.0%에서 저장 90일에 31.4%, 34.3%, 34.0%로 감소하였으며, Monoenoic acid의 변화는 동결 저장 기간 중 약간 증가하였다. 8. 중성 지질의 지방산 조성 중 포화 지방산은 내장을 포함한 육이 45.2%, 생육 33.7%, blanching한 육 35.5%이며, Monoenoic acid은 내장을 포함한 24.4%, 생육 25.1%, blanching한 육 23.8%이고, Polyenoic acid은 내장을 포함한 육 30.4%, 생육 41.2%, blanching한육 40.7%이며, 주요 구성 지방산은 $C_{14}$:0, $C_{16}$:0, $C_{18}$:0, $C_{16}$:1, $C_{18}$:1, $C_{20}$:1 그리고 $C_{20}$:5, $C_{22}$:6이고, 저장 중의 변화는 총 지질과 같이 불포화 지방산의 변화가 가장 심했다. 9. 당지질의 지방산 조성은 $C_{16}$:0, $C_{18}$:0, $C_{20}$:1, $C_{20}$:5, $C_{22}$:6이 가장 많고 그 외 $C_{14}$:0, $C_{18}$:1, $C_{22}$:2도 주요 구성 지방산이었다. 저장 중의 변화는 포화 지방산이 저장 기간 중에 증가하였으며, 반대로 불포화 지방산은 감소하였다. 10. 인지질의 지방산 조성은 중성 지질과 같이 $C_{16}$:0, $C_{18}$:0, $C_{18}$:1, $C_{20}$:5, $C_{22}$:6이며, 동결 저장 중의 변화는 당지질과 마찬가지로 포화 지방산의 증가에 상용하여 불포화 지방산의 감소가 있었으며, 특히 고도 불포화 지방산이 저장 중에 변화가 가장 심했다. 11. 단백질의 조성은 sarcoplasmic protein이 37%, myofibrillar protein이 29%, alkali-soluble이 22%, stroma가 12%로 나타났고, 저장 중에는 sarcoplasmic과 myofibrillar가 약간 감소하였으며, 반대로 alkali-soluble과 stroma는 저장 기간 중 증가하였다.

Comparison of Productivity and Feed Value of Silage Corn according to the Cutting Height

  • Corn silage is extensively utilized in ruminant feeding on a global scale, with substantial research efforts directed towards enhancing its nutritional worth and managing moisture content. The purpose of this study was to assess the impact of normal cutting height and elevated cutting height on whole-crop corn silage. Corn was harvested at heights of 15 cm and 45 cm above the ground, respectively, 45 days after heading. The harvested corn was cut into 2-3 cm lengths and packed into 20-liter plastic silos in triplicate. The results showed that dry matter (DM), crude protein (CP), water soluble carbohydrates (WSC), and in vitro dry matter digestibility (IVDMD) of C45 were significantly higher than those of the control, while the neutral detergent fiber (NDF) was significantly lower in C45 (p<0.05). The C15 had higher yields than C45 (p<0.05). There was no significant difference in the total digestible nutrients (TDN) yield of whole-crop corn silage. The increase in cutting height resulted in a larger change in moisture content and NDF per centimeter. After 60 days-ensiling, C45 showed significantly lower NH3-N concentrations. Moreover, C45 had significantly higher lactic acid concentration, lactic acid/acetic acid ratio, and lactic acid bacteria count compared to the control. Mold was not detected and the yeast count was less than 2 log10 cfu/g fresh matter in both control and C45. In summary, C45 improved the feeding value and fermentation quality of whole-crop corn silage at the expense of forage productivity.

HIP처리한 초미립 WC-(5~20) mass% Co 초경 합금의 미세조직과 기계적 성질 (Microstructures and Mechanical Properties of HIPed Submicron WC-(5~20) mass% Co Cemented Carbides)

  • The microstructures and mechanical properties of submicron WC-Co cemented carbides were investigated in relation to cobalt content. To inhibit the WC grain growth during sintering, VC was added as a inhibitor in each alloy with 3 mass% to the cobalt content. The WC-(5, 8, 10, 15, 20) mass% Co compacts were sintered at $1400^{\circ}C$ for 30 min in vacuum. Some of WC-(5, 8, 10) mass% Co sintered compacts were HIPed with 120 atm at 130$0^{\circ}C$ for 1 hr. The shrinkages of all HIPed alloys were increased without depending on the cobalt contents and the sintered densities of them. The relative densities of the alloys were increased with the cobalt content and HIPing. The less the cobalt content, the larger the WC grain. Many contiguities of WC grains were found in WC-5 mass% Co alloy. The sizes and numbers of pores in the alloys were decreased by HIPing. And also the strength and the hardness of each alloy were increased. The maximum hardness was about 18.95 GPa in the WC-5 mass% Co alloy HIPed and the maximum transverse-rupture strength (T.R.S.) 3.2 GPa in the WC-20 mass% Co alloy sintered.

Distribution, Content and Molecular Heterogeneity of Gastrin-Releasing Peptide in Rat Pancreas

  • Although importance of intrapancreatic neurons containing gastrin-releasing peptide (GRP) in control of exocrine secretion has been raised, the nature of GRP in the pancreas is unclear. Thus, the present study was undertaken to see distribution, content and molecular heterogeneity of immunoreactive GRP in the rat pancreas. Content of immunoreactive GRP in the rat pancreas was $2.99\;{\pm}\;0.66$ ng/g wet tissues determined by radioimmunoassay. Immunoreactive GRP was most abundantly expressed in the duodenal part among 3 parts of the pancreas; duodenal, body and splenic part. Vagotomy failed to change the content of immunoreactive GRP in the pancreas. Three distinct forms of immunoreactive GRP, very identical to GRP-27, bombesin-24 and neuromedin C, were observed in the rat pancreas by using reversed phase $C_{18}$ HPLC and Sephadex G-50 superfine column chromatography. Cell bodies of neurons containing immunoreactive GRP were scattered in pancreatic connective tissues and their nerve fibers innervated pancreatic acini and large ducts as determined by immunohistochemistry. The present results suggest that three distinct forms of GRP exist in intrapancreatic GRPergic neurons, which exert a stimulatory role in pancreatic exocrine secretion in rats.

슬러리법에 의한 탄소섬유보강 시멘트복합체의 제조에서 보강섬유와 계면결착제와의 상관특성 (Characteristics Correlations Between Fiber-Reinforced and Interfacial Adhesion in Carbon fiber reinforced Cement composite Prepared by Slurry Method.)

  • The objective of the study is to examine the characteristic correlations between reinforcing carbon fiber and interfacial adhesion agent since the interfacial adhesion strength between reinforcing carbon fiber and matrices is believed to be an essential element influencing the physical properties in carbon fiber reinforced cement composite using slurry method. The integrity of interfacial adhesion between reinforcing fiber and cement not only affects the quality of fiber reinforced cement composite but also influences to a large degree the physical properties of the cement composite when producing carbon fiber reinforced cement composite using slurry method. Having analyzed the physical properties 1.e., water content, tensile strength, flexural strength and flexural toughness of carbon fiber reinforced cement composite specimens, C-PAM(cation polyacrylamide) was determined to be an optimum interfacial adhesion agent. The study has also demonstrated that interfacial adhesion strength varies largely on the content and type of the reinforcing fiber. Judging from magnified view of the tensile shear cross-section using VMS(video microscope system), interfacial adhesion strength between reinforcing fiber and matrices is affected by the type of interfacial adhesion agent. According to the result of the experiments, C-PAM was determined to be an ideal interfacial adhesion agent when using carbon fiber in producing carbon fiber reinforced cement composite with the optimum content of carbon fiber being established.
