  • The effect of cultural media and growth regulators on multiple plant regeneration from leaf explants of Heloniopsis orientalis (Thunb.) Tanaka was evaluated. The highest percentage of shoot and root formation were 20 and 30% on MS medium treated at 3.0 mg $l^{-1}$ of zeatin, respectively. Also 67 and 33% of high shoot formation appeared on 1/2 MS and 5 culture medium treated at zeatin 1.0 and 3.0 mg $l^{-1}$ respectively. With MS treated at 0.5 mg $l^{-1}$ of 2,4-D 1/2 MS and B5 culture media treated at each 1.0 and 3.0 mg $l^{-1}$ of zeatin the highest ratios of plant produced were 100, 280 and 310 % respectively relative to the other treatments. Generally, there was highest possibility for multiple propagation of Heloniopsis orientalis (Thunb.) C. Tanaka with B5 culture media supplemented 3.0 mg $l^{-1}$ of zeatin.

  • The effects of the different temperatures on the amylase activity and growth rate of the rye seedling grown in the solutions containing radioactive sulfur- 35 were studied. The amylase activity of the coleoptiles obtained from the seedlings grown in the solutions of S-35, at 14$^{\circ}C$, appeared to be strongly stimulated in comparison to the control, but the culture temperatures of 22$^{\circ}C$ and 3$0^{\circ}C$ showed the decrease in the amylase activity. The amylase activity of the grains treated with the low intensity of the ratioactive material didn't show clear changes, at any culture temperatures, but the amylase activity of the grains treated with the high intensity of S-35, 50$\mu$c, showed definite decline at the elevated culture temperature, 3$0^{\circ}C$. Similar effects was also found in the growth of the seedlings. However, we would consider the effects of the radioactive materials on the acticity of the amylase and the growth of the seedlings are resulted from the accumulation of the much amount of the radioactive materials, and this accumulation rate depends upon actually the elevation of the culture temperature.

  • The experiments tvere carried out to different temperature which affected growth, yield, proximate and antitumor activity in the Codonopsis lanceolata. Growth of aerial part and subterranean partwere best at 2$0^{\circ}C$ and lower 3$0^{\circ}C$. Components of fat, protein and fiber were best at 3$0^{\circ}C$. Wild C.fonceoforo had higher contents of fat, protein, fiber and ash as compared with cultur'ed C.lanceolataArginin was predominant amino acid in both wild and cultivated C.laceolata No significant differ-ence in the mineral contents was found between the wild root and the cultivated at 30'c inbiotronroom. No minerals difference in the contents was found between the cultivated temperature.The content of elements of inorganic metal differs according to the part. C. lanceolata were subjectedto preliminary antitumer screening test with Sarcoma 180 ascites and screening on V-79 cell. Thisexperiments were conducted in accordance with the total packed cell volume method and cytotoxicity method.

Note on the Species of Ceramium (Ceramiace Rhodophyta) from Ullungdo Island, Korea

  • The taxonomic note was given to the species of Ceramium from Ullungdo Island. Five species, Ceramium aduncum Nakamura, C. codii (Rhichards) G. Mazoyer, C. flaccidum (Harvey exK tzing) Ardissone, C. paniculatum Okamura, and C. tenerrimum (Martense) Okamura were collected during this study. C. aduncum was collected in the intertidal and upper subtidal zones from December to Feburary. They had a narrow internodal space showing 1/3-1/5 times as large as nodal band. C. codii had the spermatangial mother cells developed on abaxial cortical cells of cortical band C. paniculatum collected in summer and winter had a creeping thallus and alternative branching type in gross morphology. The axial cell of C. flaccidum showed 2.3-2.8 L/B ratio, smaller than those of plants from Chejudo Island.

Development of a radiation thermometer for the adaptive steel plant (제철소 적응형 복사온도계 개발)

  • In this research, we have worked about the radiation thermometer which can be applied to the hazard circumstance such as steel plant. In the results, we have developed radiation thermometer of a measuring range $500-1500^{\circ}C$, accuracy ${\pm}0.1%$, repeatability ${\pm}0.1%$, resolution $0.2^{\circ}C$. We performed extensive field test for 6 months at the hot strip mill in steel plant. Through the test, we have confirmed the reliability of the developed pyrometer.

Construction of Linkage Map Using RAPD and SSR Markers in Soybean (Glycine max)

  • Linkage maps based on molecular markers are valuable tools in plant breeding and genetic studies. A population of 76 RI lines from the mating of A3733 and PI437.088 was evaluated with Random Amplified Polymorphic DNA(RAPD) and Simple Sequence Repeats (SSR) markers to create soybean molecular linkage map, 302 RAPD and 21 SSR markers were genetically linked and formed forty linkage groups. These linkage groups spanned a genetic distance of 1,775 cM. The average distance between markers was 5.5 cM.

  • Genomic and cDNA clones containing the RAT3 gene involving in Agobacterium-mediated plant transformation were identified using plant DNA flanking the righ border of a T-DNA rescued from the rat3 mutant as hy-bridization probe. Two highly homologous cDNA clones were identified; one (RAT3-1) weakly hybridized with the probe whereas another (RAT3-2) strongly hybridized with the probe. Both Rat3-1 and Rat3-2 proteins contain a putative signal peptide for secretion. The deduced molecular weights of encoded proteins are 15 kDa. The results of genomic DNA blot analysis and DNA sequencing indicated that RAT3-1 and RAT3-2 exist as single copy genes and they were arranged side by side with just 600 bp distance between them. RAT3-1 was disrupted by the integration of T-DNA into the 3 untranslated region in rat3 mutant. A BLAST search showed that both RAT3-1 and RAT3-2 proteins have homology with only the C-terminal region of $\beta$-1,3-glucanase homologues from Triticum aestivum and Arabidopsis thaliana. Thses $\beta$-1,3-glucanase homologues contain an unusually long C-terminal region with no sig-nificant homology to other $\beta$-1,3-glucanase.

  • Relative degree of resistance of citrus to Xanthomonas axonopodis pv. citri, the causal bacterium of canker, was investigated. Growth rate of a bacterium in leaf tissues after infiltration, disease incidence, and percent of lesion area were compared. By using growth rate[(GR=(At - A$\sub$t-1/)/A$\sub$t-1] host plants were differentiated into susceptible and resistant. Growth rates reached to peak at 40 hrs after inoculation and then declined. The growth rate in leaf tissues of a moderately susceptible cultivar, Citrus sinensis vu. Lane late(sweet orange), was the highest, and those of C. unshiu ${\times}$ C. sinensis(kiyomi), C. junos(yuzu), [(Citrus. unshiu x C. sinensis) x C. reticulata] (shiranuhi), and C. unshiu(satuma mandarin) were similar. This result indicates that the growth rate of the bacterium in leaf tissues can be effectively used for evaluation of disease resistance for citrus plants to X. axonopodis pv. citri. The disease on sweet orange occurred earlier than relatively resistant citrus plants tested. The percent of lesion area on leaf was also higher in sweet orange than those of satsuma mandarin, shiranuhi and kiyomi, and yuzu. The disease severity was highest on sweet orange and followed by kiyomi, shiranuhi, satsuma mandarin, and yuzu.

  • During this study, number of fan by ambient air temperature that condenser pressure satisfies steam turbine exhaust pressure condition with intervals of $3^{\circ}C$ within the 150 MW thermal power plant site temperature range of $-17.1^{\circ}C$ to $36.7^{\circ}C$ was reviewed. An air cooled condenser changes its operating pressure influenced by cooling air circulation amount by atmospheric temperature and number of fan. For stable power plant operation, these were confirmed to maximize a quantity of air-cooled condenser fans at above or equal from design ambient temperature and to reduce an amount of circulating air to an air cooled condenser by depending on a quantity of fan considering exhaust pressure operation condition of a steam turbine at below design ambient temperature.

Ascospore Infection and Colletotrichum Species Causing Glomerella Leaf Spot of Apple in Uruguay

  • Glomerella leaf spot (GLS) caused by Colletotrichum spp. is a destructive disease of apple restricted to a few regions worldwide. The distribution and evolution of GLS symptoms were observed for two years in Uruguay. The recurrent ascopore production on leaves and the widespread randomized distribution of symptoms throughout trees and orchard, suggest that ascospores play an important role in the disease dispersion. The ability of ascospores to produce typical GLS symptom was demonstrated by artificial inoculation. Colletotrichum strains causing GLS did not result in rot development, despite remaining alive in fruit lesions. Based on phylogenetic analysis of actin, ${\beta}$-tubulin and glyceraldehyde-3-phosphate dehydrogenase gene regions of 46 isolates, 25 from fruits and 21 from leaves, C. karstii was identified for the first time causing GLS in Uruguay and C. fructicola was found to be the most frequent (89%) and aggressive species. The higher aggressiveness of C. fructicola and its ability on to produce abundant fertile perithecia could help to explain the predominance of this species in the field.
