The non infecting, plant associated bacteria have attracted increased attention for stimulating plant growth and as environmental friendly plant protecting agents. Pink-pigmented facultatively methylotrophic bacteria (PPFMs), classified as Methylobacterium spp., are persistent colonizers of plant leaf surfaces. As the leaves of most or all plants harbor PPFMs that utilize leaf methanol as their sole source of carbon and energy, which is a specific attribute of the genus Methylobacterium. Although they are not well known, these bacteria are co-evolved, interacting partners in plant metabolism. This claim is supported, for example, by the following observations: (1) PPFMs are seed-transmitted, (2) PPFMs are frequently found in putatively axenic cell cultures, (3) Low numbers of seed-borne PPFMs correlate with low germinability, (4) Plants with reduced numbers of PPFM show elevated shoot/root ratios, (5) Foliar application of PPFMs to soybean during pod fill enhances seed set and yield, (6) Liverwort tissue in culture requires PPFM-produced vitamin B12 for growth, (7) treated plants to suppress or decrease disease incidence of sheath blight caused by Rhizoctonia solani in rice, and (8) the PPFM inoculation induced number of stomata, chlorophyll concentration and malic acid content, they led to increased photosynthetic activity. Methylobacterium spp. are bacterial symbionts of plants, shown previously to participate in plant metabolism by consuming plant waste products and producing metabolites useful to the plant. There are reports that inform about the beneficial interactions between this group of bacteria and plants. Screening of such kind of bacteria having immense plant growth promoting activities like nitrogen fixation, phytohormone production, alleviating water stress to the plants can be successfully isolated and characterized and integration of such kind of organism in crop production will lead to increased productivity.
Since the efficiency of wastewater treatment using microalgae differs depending on the metabolic characteristics of the species, it is important to understand the characteristics of target algae prior to the application in wastewater treatment. In this study, for the application of Arthrospira platensis to wastewater treatment, which is a filamentous alkaliphilic cyanobacteria, basic species specificity was identified and the possibility of application to wastewater treatment was investigated. As a result of the species specificity investigation, the specific growth rate between pH 7.0 and 11.0 showed the highest value near pH 9 at 0.25/day. The reason for the relatively low growth(0.08/day) at pH 11 was thought to be the CA(carbonic anhydrase) enzyme that is involved in carbon fixation during photosynthesis has the highest activity at pH 8.0 to 9.0, and at pH 11, CA activity was relatively low. In addition, A. platensis showed optimal growth at 400 PPFD(photosynthetic photon flux density) and 30℃, and this means that cyanobacteria such as A. platensis have a larger number of PS-I(photosystem I) than that of PS-II(photosystem II). It was speculated that it was because higher light intensity and temperature were required to sufficiently generate electrons to transfer to PS-I. Regarding the applicability of A. platensis, it was suggested that if a system using the synergistic effect of co-culture of A. platensis and bacteria was developed, a more efficient system would be possible. And different from single cocci, filamentous A. platensis expected to have a positive impact on harvesting, which is very important in the latter part of the wastewater treatment process.

Ursolic acid (UA) and Oleanolic acid (ONA), known as urson, micromerol, prunol and malol, are pentacyclic triterpenoid compounds which naturally occur in a large number of vegetarian foods, medicinal herbs, and plants. They may occur in their free acid form or as aglycones for triterpenoid saponins, which are comprised of a triterpenoid aglycone, linked to one or more sugar moieties. Therefore UA and ON A are similar in pharmacological activity. Lately scientific research, which led to the identification of UA and ONA, revealed that several pharmacological effects, such as antitumor, hepatoprotective, anti-inflammatory, antimicrobial, and anti-hyperlipidemic could be attributed to UA and ONA. Here, we introduced the effects of UA and ONA on acute barrier disruption and normal epidermal permeability barrier function. To clarify the effects of UA and ONA on skin barrier recovery, both flank skin of 8-12 weeks hairless mice were topically treated with samples (2mg/ml) after tape stripping, then measured recovery rate using TEWL on hairless mice. The recovery rate increased in UA and ONA treated groups at 6h more than 20% compared to vehicle treated group (p <0.05). For verifying the effects of UA and ONA on normal epidermal barrier, hydration and TEWL were measured for 1 and 3 weeks after UA and ONA applications (2mg/ml per day). We also investigated the features of epidermis and dermis using electron microscopy (EM) and light microscopy (LM). Both samples increased hydration compared to Vehicle group from 1 week without TEWL alteration (p<0.005). EM examination using Ru04 and OsO4 fixation revealed that secretion and numbers of lamellar bodies and complete formation of lipid bilayers were most prominent (ONA$\geq$UA>Vehicle). LM finding showed that stratum corneum was slightly increased and especially epidermal thickening and flattening was observed (UA>ONA>Vehicle). Using Masson-trichrome and elastic fiber staining, we observed collagen thickening and elastic fiber increasing by UA and ONA treatments. In vitro results of collagen and elastin synthesis and elastase inhibitory experiments were also confirmed in vivo findings. This result suggested that the effects of UA and ONA related to not only skin barrier but also collagen and elastic fibers. Taken together, UA and ONA can be relevant candidates to improve barrier function and pertinent agents for cosmetic applications.
The pyrenoid ultrastructure and distribution of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and Rubisco activase in the chloroplasts of Chlamydomonas reinhardtii was studied using the immunogold localization technology with electron microscopy. There were several tubular thylakoids invading in the pyrenoid matrix to form several spokewise channels. The connections between pyrenoid matrix and stroma of chloroplast were the partial of channels. The starch sheath surrounding the pyrenoid was separated into several parts by the connections in transection. Some thylakoids were packed together near the connections in one side of the pyrenoid. Those special structures might be used to transport substance between pyrenoid and stroma of chloroplasts. With the antibody raised against the large subunits of Rubsico from C. protothecoides, the result of the gold immunolocalization of Rubisco in Chlamydomonas reinhardtii showed most of the gold particles heavily labeled the pyrenoid matrix, as well as the starch sheath matrix, and very few in the stroma of chloroplasts. The gold particle density was 880.00 $\pm$ 164.32, 190.00 $\pm$ 152.39 and 9.60 $\pm$ 5.37 ${\mu}m^{-2}$ in pyrenoid matrix, starch sheath and stroma region of chloroplast respectively (background: 5.67 $\pm$ 1.53 ${\mu}m^{-2}$). 99.59% of the total Rubiscos was calculated to be concentrated in the pyrenoid matrix and starch sheath by spatial densities. The gold immunolocalization of Rubisco activase also showed that Rubisco activase was mainly concentrated in the periphery of the pyrenoid and the starch sheath (the density was as high as 229.69 $\pm$ 96.96 ${\mu}m^{-2}$). There were very few gold particles located in the stroma of chloroplasts. These results indicated that pyrenoid surface and starch sheath was the site for Rubisco activation and $CO_2$ fixation, which supported the suggestion that pyrenoids perform photosynthesis function.

The ocean is a significant sink for atmospheric anthropogenic CO2, absorbing one-third of the total CO2 emitted by human activities. In return, oceans have experienced significant declines in seawater pH and the aragonite saturation state also called ocean acidification. This study evaluates the distribution of aragonite saturation state, an indicator to assess the potential threat from ocean acidification, by combining newly obtained data from the west coast of South Korea with previous datasets covering the Yellow Sea, East Sea, northern South China Sea, and southeast coast of South Korea. In general, offshore waters absorb atmospheric CO2; however, most of the collected water samples show aragonite oversaturation. On the southeast coast, the aragonite saturation state was significantly affected by river discharge and associated variables, such as freshwater input with nutrients, seasonal stratification, biological carbon fixation, and bacterial remineralization. In summer, hypoxia and mixing with relatively acidic freshwater made the Jinhae and Gwangyang Bays undersaturated with respect to aragonite, possibly threatening marine organisms with CaCO3 shells. However, widespread aragonite undersaturation was not observed on the west coast, which receives considerable river water discharge. In addition, occasional upwelling events may have worsened the ocean acidification in the southwestern part of the East Sea. These results highlight the importance of investigating site-specific ocean acidification processes in coastal waters. Along with the above-mentioned seasonal factors, the dissolution of atmospheric CO2 and the deposition of atmospheric acidic substances will continue to reduce the aragonite saturation state in Korean waters. To protect marine ecosystems and resources, an ocean acidification monitoring program should be established for Korean waters.
To elucidate the physiological characteristics of 2 japonica type and 2 tongil type of rice cultivars, photosynthesis, respiration and diurnal changes of photosynthesis rate had been investigated 6 times at each growing stage. The ratios of photosynthetic part of tongil types were higher than those of japonica types at tillering stage. The highest photosynthetic activity was measured on August 1, before heading in each growing stage, and efficiency of photosynthesis showed its the highest on July 25. Diurnal photosynthetic curve at heading stage reached the peak at 11-12 AM, but after 2 weeks from heading stage, the peak apperred at 1-2 PM. Conversion of respiration into photosynthesis for $CO_2$ fixation started 30~60 minutes later than the standard sunrise time and reconversion for carborhdrate accmulation began 50~60 minutes earlier than the standard sunset time. Photosynthesis showed positive correlation with chlorophyll content and root activity. Harvest index of tongil type cultivars were significantly higher than those of japonica type cultivars.
Anabaena variabilis A TCC 29413. a photoautotrophic and nitrogen fixing cyanobacteria. was investigated on the environmental factors regulating the growth and nitrogen lixation activity. A good growth of cyanobacteria] cells was observed due to nitrogen t1xation by the heterocyst differentiation in nitrogen free Allen and Arnon (]/8) medium. The nitrogenase activity was appeared to be in proportion to the cell growth lor 6 days then drastically decreased in the later growth period when the nitraTe was accumulated to high level in the culture to cause the inhibition. The optima] conditions lilr the cell growth and nitrogenase activity of A. varillbili.l were anaerobic. IO.OO0 lux. $30^{\circ}C$ and pH 8 with the nitrogen Cree minimal medium. The activity was significantly inhihited by the low concentrations of ammonium and nitrate. but was stimulated b) the ]ow Ieve] of phosphate and carbonate sources. The treatments of several toxic heavy metals showed strong inhibition of the cell growth and nitrogenase activity by O.3~10 ppm in the order of $Hg^{2+}$ > $Cd^{2+}$ > $Co^{2+}$ > $Zn^{2+}$ > $Ph^{2+}$, and the concentrations for 50% inhibition of the maximum activity were 0.41. 0.47. 0.5 L 0.66 and 8.1 ppm. respectively. The addition of carbohydrates (0.5~ 1.0%) in the dark condition stimulated the growth and activity in the order of sucrose > fructose > glucose.
In the clothes industry, there are lack of experts including technical designers who can analyze the fit of clothes. This study is to provide practical data available for fit analysis education by distinguishing the differences in standards and aspects of garment fit between experts and novices, through the eye-tracking technology to quantify the sense of fit. For this study, two groups were organized; one composed of 7 experts with over 15 year-experience including technical designers and patternmakers, and the other composed of 7 novices who are students majoring in clothing. Wearing the goggle type eye-tracker Tobii Pro Glasses 2, the participants in the experiments were required to conduct fit analyses for a pair of pants on a live model. After those experiments, they were required to check the items for fit analysis and assess the importance level of them on a questionnaire. The differences between the two groups in the ratios of total visit count and total visit duration by each AIO(Area of Interest) of clothes were analyzed through non-parametric statistical test. The results of eye tracking experiments showed that experts focused on center front and back line, crotch area, and side seam, while novice's fixation points were dispersed around the pants. The survey results showed that the experts put importance on the center line position and its verticality, front-back proportion of side seam line, and front-back proportion of waist line, 71.4~100% of whom checked them, while 14.3% of the novices checked them.
Natural abundances of stable isotopes of nitrogen and carbon (${\delta}^{15}N$ and ${\delta}^{13}C$) are being widely used to study N and C cycle processes in plant and soil systems. Variations in ${\delta}^{15}N$ of the soil and the plant reflect the potentially variable isotope signature of the external N sources and the isotope fractionation during the N cycle process. $N_2$ fixation and N fertilizer supply the nitrogen, whose ${\delta}^{15}N$ is close to 0%o, whereas the compost as. an organic input generally provides the nitrogen enriched in $^{15}N$ compared to the atmospheric $N_2$. The isotope fractionation during the N cycle process decreases the ${\delta}^{15}N$ of the substrate and increases the ${\delta}^{15}N$ of the product. N transformations such as N mineralization, nitrification, denitrification, assimilation, and the $NH_3$ volatilization have a specific isotope fractionation factor (${\alpha}$) for each N process. Variation in the ${\delta}^{13}C$ of plants reflects the photosynthetic type of plant, which affects the isotope fractionation during photosynthesis. The ${\delta}^{13}C$ of C3 plant is significantly lower than, whereas the ${\delta}^{13}C$ of C4 plant is similar to that of the atmospheric $CO_2$. Variation in the isotope fractionation of carbon and nitrogen can be observed under different environmental conditions. The effect of environmental factors on the stomatal conductance and the carboxylation rate affects the carbon isotope fractionation during photosynthesis. Changes in the environmental factors such as temperature and salt concentration affect the nitrogen isotope fractionation during the N cycle processes; however, the mechanism of variation in the nitrogen isotope fractionation has not been studied as much as that in the carbon isotope fractionation. Isotope fractionation factors of carbon and nitrogen could be the integrated factors for interpreting the effects of the environmental factors on plants and soils.
Succinic acid is an important precursor in industries producing biopolymers, pharmaceutical and food additives and green solvents. However, due to the high price of petroleum and the global $CO_2$ emission, the biological production of succinic acid from renewable biomass is a novel process due to the fixation of $CO_2$ into succinate during fermentation. In this study, aqueous two phase systems based on imidazolium ionic liquids/$K_2HPO_4$ were used as an effective separation and concentration process for succinic acid. Experimental results show that aqueous two phase systems can be formed by adding appropriate amount of imidazolium ionic liquids to aqueous $K_2HPO_4$ solutions in the presence of succinic acid. It can be found that the ability of imidazolium ionic liquids for phase separation followed the order [HMIm][Br]${\fallingdotseq}$[OMIm][Br]>[BMIm][Br]>[EMIm][Br]. The maximum value of extraction efficiency for succinic acid was about 90% and the amount of coextracted water into top phase is proportional to the chain length of cation in imidazolium ionic liquids. It was concluded that the aqueous two phase systems composed of imidazolium ionic liquids/$K_2HPO_4$ was effective for the selective extraction and concentration of succinic acid.
