Dosimetric Analysis of a Phase I Study of PSMA-Targeting Radiopharmaceutical Therapy With [177Lu]Ludotadipep in Patients With Metastatic Castration-Resistant Prostate Cancer

  • Objective: 177Lutetium [Lu] Ludotadipep is a novel prostate-specific membrane antigen targeting therapeutic agent with an albumin motif added to increase uptake in the tumors. We assessed the biodistribution and dosimetry of [177Lu]Ludotadipep in patients with metastatic castration-resistant prostate cancer (mCRPC). Materials and Methods: Data from 25 patients (median age, 73 years; range, 60-90) with mCRPC from a phase I study with activity escalation design of single administration of [177Lu]Ludotadipep (1.85, 2.78, 3.70, 4.63, and 5.55 GBq) were assessed. Activity in the salivary glands, lungs, liver, kidneys, and spleen was estimated from whole-body scan and abdominal SPECT/CT images acquired at 2, 24, 48, 72, and 168 h after administration of [177Lu]Ludotadipep. Red marrow activity was calculated from blood samples obtained at 3, 10, 30, 60, and 180 min, and at 24, 48, and 72 h after administration. Organand tumor-based absorbed dose calculations were performed using IDAC-Dose 2.1. Results: Absorbed dose coefficient (mean ± standard deviation) of normal organs was 1.17 ± 0.81 Gy/GBq for salivary glands, 0.05 ± 0.02 Gy/GBq for lungs, 0.14 ± 0.06 Gy/GBq for liver, 0.77 ± 0.28 Gy/GBq for kidneys, 0.12 ± 0.06 Gy/GBq for spleen, and 0.07 ± 0.02 Gy/GBq for red marrow. The absorbed dose coefficient of the tumors was 10.43 ± 7.77 Gy/GBq. Conclusion: [177Lu]Ludotadipep is expected to be safe at the dose of 3.7 GBq times 6 cycles planned for a phase II clinical trial with kidneys and bone marrow being the critical organs, and shows a high tumor absorbed dose.

Effective Suppression of Methane Production by Chelating Nickel of Methanogenesis Cofactor in Flooded Soil Conditions (담수토양에서 메탄생성반응 보효소 니켈의 킬레이팅에 의한 메탄 생산량의 효과적 저감)

  • BACKGROUND: Methane($CH_4$) is considered as the secondmost potent greenhouse gas after carbon dioxide ($CO_2$). Methanogenesis is an enzyme-mediated multi-step process by methanogens. In the penultimate step, methylated Co-M is reduced by methyl Co-M reductase (MCR) to $CH_4$ involving a nickel-containing cofactor F430. The activity of MCR enzyme is dependent on the F430 and therefore, the bioavailability of Ni to methanogens is expected to influence MCR activity and $CH_4$ production in soil. In this study, different doses of EDTA(Ethylene Diamine Tetraacetic Acid) were applied in flooded soils to evaluate their suppression effect on methane production by chelating Ni of methanogenesis cofactor. METHODS AND RESULTS: EDTA was selected as chelating agents and added into wetland and rice paddy soil at the rates of 0, 25, 50, 75, and $100mmol\;kg^{-1}$ before 4-weeks incubation test. During the incubation, cumulative $CH_4$ production patterns were characterized. At the end of the experiment, soil samples were removed from their jars to analyze total soil Ni and water-soluble Ni content and methanogen abundance. Methane production from 100 mmol application decreased by 55 and 78% in both soils compared to that from 0 mmol. With increasing application rate of EDTA in both soils, water-soluble Ni concentration significantly increased, but total soil Ni and methanogen activities showed negative relationship during incubation test. CONCLUSION: The decrease in methane production with EDTA application was caused by chelating Ni of coenzyme F430 and inhibiting methanogenesis by methyl coenzyme M reductase. Consequently, EDTA application decreased uptake of Ni into methanogen, subsequently inhibited methanogen activities and reduced methane production in flooded soils.

Preparation and Characteristics of Partially Fluorinated-Sulfonated Poly(biphenylene-co-sulfone)ether Membranes for Polymer Electrolyte Membrane Fuel Cell (고분자전해질 연료전지용 부분 불소계 설폰화 Poly(biphenylene-co-sulfone)ether 막의 제조와 특성)

  • A series of partially fluorinated, sulfonated poly(biphenylene-co-sulfone)ether containing perfluorocyclobutane(PFCB) groups were prepared for fuel cell applications through three synthetic steps: synthesis of trifluorovinylether-terminated monomers, thermal cycloaddition and post-sulfonation. Two kinds of trifluorovinylether-terminated monomers were synthesized and statistically copolymerized via thermal cycloaddition to obtain a series of polymers containing 20-60 mol% of biphenyl units(PBS-X). The post-sulfonation of PBS-X was carried out using chlorosulfonic acid(CSA) to obtain copolymers with various sulfonation levels(SPBS-X). All the synthesized compounds, monomers and polymers were characterized by $^1H$-NMR, $^{19}F$-NMR and FT-IR. It was confirmed that the ion exchange capacity(IEC), water uptake and ion conductivity of SPBS-X increased with the increment of sulfonated biphenyl units. Particularly, SPBS-60 showed higher ion conductivity compared to Nafion$^{(R)}$-115 at 25~80 $^{\circ}C$.

Biosorption and Desorption of Heavy Metals using Undaria sp. (미역 폐기물의 중금속 흡탈착 특성)

  • The adsorption and desorption of Pb, Cd, Co, Zn, Cr, Co, Ni, and Mo on the waste Undaria sp. were studied. Except for Pb. the mono adsorption rate for all heavy metals were lower than that of the heavy metals mixed. However, the adsorption capacity of the heavy metals by 1g of biosorption, in mixed heavy metals increased According to FT-IR analysis of the biosorbent after heavy metal biosorption, the replacement of the functional group by the heavy metals ions could be confirmed and the inverted peaks became larger after heavy metals adsorption. The adsorption equilibrium of heavy metals was reached in about 1 hour. The equilibrium parameters were determined based on Langmuir and Freundlich isotherms. The affinity of metals on the biosorbent decreased in the following order: Pb>Cu>Cr>Cd>Co. The desorption rate decreased in the following sequence: NTA>$H_2SO_4$>HCl>EDTA. The desorption rate of heavy metals by NTA increased with increase in the concentration from 0.1 to 0.3% but the desorption rate became constant beyond 0.3%. Therefore, it represented that desorption rate of heavy metals was suitable under optimized condition ($30^{\circ}C$, pH 2 and 0.3% NTA solution) and was fast with 80% or more the uptake occurring within 10 min of contact time.

Liming Effect on Cadmium Immobilization and Phytoavailability in Paddy Soil Affected by Mining Activity (중금속 오염 논토양에서 카드뮴의 부동화와 식물이용성에 대한 석회 시용 효과)

  • BACKGROUND: Many studies associated with cadmium (Cd) immobilization using lime fertilizer have been conducted for several decades. However, these studies did not suggest exact mechanism of Cd immobilization using lime fertilizer and evaluated effect of lime fertilizer on Cd phytoavailability in rice paddy soil under field condition. METHODS AND RESULTS: This study was conducted to determine exact mechanism of Cd immobilization using lime fertilizer and evaluate liming effect on Cd uptake of rice in contaminated paddy soil. $Ca(OH)_2$ was mixed with Cd contaminated arable soil at rates corresponding to 0, 1,000, 2,000, 4,000, and 8,000 mg/kg. The limed soil was moistened to paddy soil condition, and incubated at $25^{\circ}C$ for 4 weeks. $NH_4OAc$ extractable Cd concentration in soil decreased significantly with increasing $Ca(OH)_2$ rate, since $Ca(OH)_2$ markedly increased net negative charge of soil by pH increase, and decreased bioavailable Cd fractions (F1; exchangeable + acidic and reducible Cd fraction). Calculated solubility diagram indicated that Cd solubility was controlled by soil-Cd. $NH_4OAc$ extractable Cd and F1 concentration were negatively related to soil pH and negative charge. $Ca(OH)_2$ was applied at rates 0, 2, 4, and 8 Mg/ha and then cultivated rice in the paddy soil under field condition. Cadmium concentrations in grain, straw, and root of rice plant decreased significantly with increasing application rate of $Ca(OH)_2$. CONCLUSION(S): Alleviation of Cd phytoavailability with $Ca(OH)_2$ can be attributed primarily to Cd immobilization due to the increase in soil pH and negative charge rather than precipitation of $Cd(OH)_2$ or $CdCO_3$, and therefore, $Ca(OH)_2$ is effective for reducing Cd phytoavailability of rice in paddy soil.

Bioequivalence of Glycomin Tablet to Glucophage Tablet (Metformin HCl 500 mg) (굴루코파지 정(염산메트폴민 500 mg)에 대한 그리코민 정의 생물학적 동등성)

  • Metformin is an oral antihyperglycemic agent used in the therapy of noninsulin-dependent diabetes mellitus and does not cause hypoglycemia at the therapeutic dose. Its mechanism of action may involve an increased binding of insulin to its receptors and glucose uptake at the post-receptor level. The purpose of the present study was to evaluate the bioequivalence of two metformin tablets, Glucophage (Daewoong Pharmaceutical Co., Ltd.) and Glycomin (Ilsung Pharmaceuticals Co., Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). The metformin release from the two metformin tablets in vitro was tested using KP VII Apparatus II method with various dissolution media (pH 1.2, 4.0, 6.8 buffer solution and water). Twenty four normal male volunteers, $23.75{\pm}1.96$ years in age and $68.77{\pm}10.41\;kg$ in body weight, were divided into two groups with a randomized $2{\times}2$ cross-over study. After one tablet containing 500 mg as metformin was orally administered, blood was taken at predetermined time intervals and the concentrations of metformin in serum were determined using HPLC with UV detector. Besides, the dissolution profiles of two metformin tablets were very similar at 떠1 dissolution media. The pharmacokinetic parameters such as $AVC_t,\;C_{max}\;and\;T_{max}$ were calculated. The ANOVA test was performed for the statistical analysis of the logarithmically transformed $AVC_t\;and\;C_{max}$, untransformed $T_{max}$. The results showed that the differences in $AVC_t,\;C_{max}\;and\;T_{max}$ between two tablets based on the Glucophage were 0.09%, 6.09% and -8.22%, respectively. There were no sequence effects between two tablets in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log(0.8) to log(1.25) $(e.g.,\;log(0.94){\sim}log(1.09)\;and \;log(1.01){\sim}log(1.15)$\;for\;AVC_t\;and\;C_{max},\;respectively)$, indicating that Glycomin tablet is bioequivalent to Glucophage tablet.

Neuroprotective Effects of Medicinal Herbs in Organotypic Hippocampal Slice Cultures (뇌해마의 장기양 조직배양을 이용한 한약물의 뇌신경세포손상 보호효능 연구)

  • Objectives : For the screening of neuroprotective effects of medicinal herbs, the complex system of animal models suffer some disadvantages in controlling critical parameters such as blood pressure and body temperature. Additionally, application of drugs to the appropriate brain area sometimes is difficult, due to poor permeability though the blood brain barrier, and so potential protective effects might be masked. Methods : Organotypic hippocampal slice culture (OHSC) method has the advantages of being relatively easy to prepare and of maintaining the general structure, including tissue integrity and the connections between cells. Drugs can easily be applied and neuronal damage can easily be quantified by using tissues and culture media. This study demonstrates neuroprotective effects of Puerariae radix (葛根, PR), Salviae miltiorrhizae radix (丹蔘, SR), Rhei rhizoma (大黃, RR), and Bupleuri radix (柴胡, BR). These were screenedand compared to MK-801, antagonist of NMDA receptors, by using OHSC of 1 week-old Sprague-Dawley rats. Oxygen/glucose deprivation (OGD) were conducted in an anaerobic chamber $(85%\;N_2,\;10%\;CO_2\;and\;5%\;H_2)$ in a deoxygenated glucose-free medium for 60 minutes. Water extracts of each herbs were treated to culture media with $5\;{\mu}g/ml$ for 48 hours. Results : Neuronal cell death in the cultures was monitored by densitometric measurements of the cellular uptake of propidium iodide (PI). PI fluorescence images were obtained at 48 hours after the OGD and medicinal herb treatment. Also TUNEL-positive cells in the CAI and DG regions and LDH concentrations in culture media were measured at 48 hours after the OGD. According to measured data, MK-801, PR, SR and BR demonstrated significant neuroprotective effect against excessive neuronal cell death and apoptosis induced by the OGD insult. Especially, PR revealed similar neuroprotective effect to MK-801 and RR demonstrated weak neuroprotective effect. Conclusions : These results suggest that OHSC can be a suitable method for screening of neuroprotective effects of medicinal herbs. Effects of Unripe Black Raspberry Water Extract on Lipid Metabolism and Oxidative Stress in Mice (복분자 미숙과 물 추출물이 마우스의 지질대사 및 산화적 스트레스에 미치는 영향)

  • We examined the effects of unripe black raspberry water extract (UBR-W) on lipid metabolism and oxidative stress in mice. C57BL/6J mice were divided into 4 groups: those administered a control diet (CTL), high-fat diet (HFD), UBR-W and simvastatin for 12 weeks. In the HFD group, LDL cholesterol were significantly higher than in the CTL group. However, the UBR-W treated group showed dose-dependent reduction of plasma LDL levels. Hepatic total lipid, TC, and malondialdehyde were significantly increased in hyperlipidemic mice. However, supplementation with either UBR-W or simvastatin effectively reduced these lipid profiles and lipid peroxidation. UBR-W increased mRNA expression of the LDL receptor, sterol regulatory element binding protein 2 (SREBP2), 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase and ATP-binding cassette transporter A1 (ABCA1) compared to that observed in the HFD group. In addition, UBR-W and simvastatin showed significantly reduced oxidized LDL uptake by the scavenger receptor CD36. These results suggest that UBR-W is useful for treatment and prevention of hyperlipidemia and lipid peroxidation.

The impact of municipal waste disposal of heavy metals on environmental pollution: A case study for Tonekabon, Iran

  • Municipal solid waste disposal is considered as one of the most important risks for environmental contamination which necessitates the development of strategies to reduce destructive consequences on the ecosystem as related especially to heavy metal accumulation. This study investigates heavy metal (i.e., As, Cd, Co, Cr, Cu, Mn, Ni, Pb, Zn) accumulation in the Tonekabon region, NW of Iran that is related to city waste disposal and evaluates the environmental impact in the Caspian Sea coastal region. For this purpose, after performing field studies and collecting 50 soil specimens from 5 sites of the study area, geochemical tests (i.e., inductively coupled plasma mass spectrometry, atomic absorption spectroscopy and x-ray fluorescence) were conducted on the soil specimens collected from the 5 sites (named as Sites A1, A2, A3, A4 and A5) and the results were used to estimate the pollution indices (i.e., geo-accumulation index, normalized enrichment factor, contamination factor, and pollution load index). The obtained indices were utilized to assess the eco-toxicological risk level in the landfill site which indicated that the city has been severely contaminated by Cu, Mn, Ni, Pb and Zn. These levels have been developed along the stream towards the nearshore areas indicating uptake of soil degradation. The heavy metal contamination was classified to range from unpolluted to highly polluted, which indicated serious heavy metal pollution in the study area as related to municipal solid waste disposal in Tonekabon.

Estimation of the methane generation rate constant using a large-scale respirometer at a landfill site

  • The objective of this study is the evaluation of the performance of a large-scale respirometer (LSR) of 17.7 L in the determination of the methane generation rate constant (k) values. To achieve this objective, a comparison between anaerobic (GB21) and LSR tests was conducted. The data were modeled using a linear function, and the resulting correlation coefficient ($R^2$) of the linear regression is 0.91. This result shows that despite the aerobic conditions, the biodegradability values that were obtained from the LSR test produced results that are similar to those from the GB21 test. In this respect, the LSR test can be an indicator of the anaerobic biodegradability for landfill waste. In addition, the results show the high repeatability of the tests with an average coefficient of variance (CV) that is lower than 10%; furthermore, the CV for the LSR is lower than that of the GB21, which indicates that the LSR-test method could provide a better representation of waste samples. Therefore, the LSR method allows for both the prediction of the long-term biodegradation potential in a shorter length of time and the reduction of the sampling errors that are caused by the heterogeneity of waste samples. The k values are $0.156y^{-1}$ and $0.127y^{-1}$ for the cumulative biogas production (GB21) and the cumulative oxygen uptake for the LSR, respectively.
