Supplementary Effect of the High Dietary Fiber Rice on Lipid Metabolism in Diabetic KK Mice (고식이섬유쌀의 급여가 KK 당뇨 마우스의 체내 지질 대사에 미치는 영향)

  • This study was conducted to investigate the supplementary effects of Suwon 464 developed by Rural Development Administration, which has over two times of dietary fiber content compared with normal rice (Ilpum), on lipid metabolism in diabetic mice. We supplied 5 kinds of experimental diets (com starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight, organ weights, and lipids levels of serum, liver and feces were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, 1.4, and 2.0% respectively. Body weight and liver and epididymal fat pad weights were lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentrations of serum triglyceride was lower in SP and SB groups than CO and IP groups. The levels of hepatic total lipid and total cholesterol were significantly lower in SP and SB groups than CO group, and the level of hepatic triglyceride was lower in IB, SP and SB groups than CO group. The levels of total lipid and triglyceride excreted in feces were higher in IB, SP and SB, and the level of total cholesterol in feces was higher in SP and SB groups than CO group. These results suggested that the high dietary fiber rice (Suwon 464) decrease the triglyceride or total cholesterol concentrations of serum and liver by increasing of fecal lipid excretion in diabetic mice. Agarwood Inhibits Histamine Release from Rat Mast Cells and Reduces Scratching Behavior in Mice -Effect of Agarwood on Histamine Release and Scratching Behavior-

  • Objectives: This study was conducted to clarify the effects of agarwood on histamine release from mast cells in rats and on the scratching behaviors in mice. Methods: Histamine release from rat mast cells induced by compound 48/80 or concanavalin A (Con A) and compound 48/80-induced scratching behavior in mice were examined to investigate the effects of agarwood. The hyaluronidase activity and the 3',5'-cyclic adenosine monophosphate (cAMP) levels in mast cells were examined to investigate the mechanisms for the inhibition of histamine release. The correlation between the inhibitory effects of agarwood on histamine release and the content of its typical ingredients, a 2-(2-phenylethyl)chromone derivatives, was analyzed using thin-layer chromatography. Results: Agarwood showed an inhibitory effect on mast-cell histamine release induced by compound 48/80 or Con A without any effect on hyaluronidase activity; this effect involves an increase in the cAMP levels in mast cells. Oral administration of agarwood showed an inhibitory effect on compound 48/80-induced scratching behavior in mice. The inhibitory effects of agarwood on histamine release were quite different, depending on the area where the agarwood was produced, its quality, and its market price. No correlation was found between the inhibitory effects of agarwood on histamine release and the typical ingredients of agarwood, which are 2-(2-phenylethyl)chromone derivatives. Conclusion: These results show that agarwood inhibits histamine release from mast cells partially through an increase in the cAMP levels in cells. We suggest that some active ingredients of agarwood must be effective on oral intake and that agarwood can be used to treat patients with a number of conditions, including urticaria, atopic dermatitis, and bronchial asthma, in which an increase in histamine release occurs. Differences in the pharmacological effects of this crude drug among markets may provide important information for the quality control of this herbal medicine.

Simulation on the Alternation of Limestone for Portland Cement Raw Material by Steel By-products Containing CaO (CaO 함유 철강 부산물을 활용한 시멘트 원료 석회석 대체 시뮬레이션)

  • In this study, to reduce CO2 emission in the cement manufacturing process, we evaluated the limestone that is used as a raw material for cement, substituted with steel slag by the various substituted levels. Based on the chemical composition of each raw materials including limestone, and blast furnace slow cooling slag, converter slag, and KR slag as an alternative raw material, we simulated the optimal cement raw mixture by the substitution levels of limestone. Test results indicated that the steel slags contain a certain level of CaO that can be used as alternative decarbonated raw materials, and it has enough to partially reduce the amount of limestonem. And we estimated the maximum usable levels of each raw material. In particular, it was confirmed that by using a mixture of these raw materials rather than using them one by one, the effect of reducing limestone was increased and CO2 emission from the cement manufacturing process could be reduced.

  • Salmonella is one of the major pathogenic bacteria that cause food poisoning. This study investigated whether heat-killed as well as live Lactobacillus protects host animal against Salmonella infection. Live and heat-killed Lactobacillusacidophilus was administered orally to Sprague-Dawley rats for 2 weeks before the rats were inoculated with Salmonella. Rise in body temperature was moderate in the group that was treated with heat-killed bacteria as compared to the Salmonella control group. The mean amount of feed intake and water consumption of each rat in the heat-killed bacteria group were nearly normal. The number of fecal Salmonellae was comparable between the live and the heat-killed L. acidophilus groups. This finding shows that L. acidophilus facilitates the excretion of Salmonella. Moreover, the levels of pro inflammatory cytokines, including tumor necrosis factor (TNF)-alpha and interleukin (IL)-1 beta, in the heat-killed L. acidophilus group were significantly lower when compared to the levels in the Salmonella control group. These results indicate that nonviable lactic acid bacteria also could play an important role in preventing infections by enteric pathogens such as Salmonella.

Anti-osteoarthritis Effects of the Combination of Boswellia serrata, Curcuma longa, and Terminalia chebula Extracts in Interleukin-1β-stimulated Human Articular Chondrocytes

  • In this study, extracts of Boswellia serrata gum resin, Curcuma longa rhizome, and Terminalia chebula fruit were combined in different ratios, and their anti-osteoarthritis effects were compared to determine which combination had the best synergistic effect. B. serrata, C. longa, and T. chebula extracts in a 2:1:2 ratio exhibited higher antioxidative activity in scavenging DPPH radicals than did the individual extracts alone or the other extract combinations. Additionally, the 2:1:2 combination significantly improved the levels of enzymatic antioxidants and antioxidant-related proteins. Moreover, this same combination ratio decreased the protein levels of matrix metalloproteinase (MMP) 3 and MMP13 in interleukin-1β-stimulated human articular chondrocytes (HCHs) and increased those of aggrecan and collagen type II alpha 1 chain (COL2A1). Analysis of the underlying mechanisms revealed that the 2:1:2 combination significantly inhibited the phosphorylation of nuclear factor kappa B (NF-κB) p65, extracellular regulated protein kinase (ERK), and p38 mitogen-activated protein kinase (MAPK). Therefore, the 2:1:2 combination of these three plant extracts has the best potential for use as an effective dietary supplement for improving joint health compared with the individual extracts and their other combination ratios.

Rapid Changes in CO Concentration Levels at Seven Roadside Locations in Seoul before and after 2000

  • Concentrations of carbon monoxide (CO) in air were analyzed from seven urban roadside (U-RS) stations in Seoul, Korea over an 11-year period (1998-2008). As noticeable changes in CO levels coincided with the Natural Gas Vehicle Supply (NGVS) program in the year 2000, its concentration data were examined between 1998-2000 (period I) and 2001-2008 (period II). The mean values of CO from all 7 stations for the whole study period were recorded as $1.19{\pm}0.22$ ppm with large drops in its mean from $1.83{\pm}0.43$ ppm in period I to $0.94{\pm}0.21$ ppm in period II. As such, CO levels between the two periods undertook the mean reduction rate of $46.9{\pm}16.2%$ (range of 18.3 to 59.9%). According to the analysis of longterm trends of CO, its concentrations in the U-RS stations decreased very rapidly with the annual mean reduction rate of 9.5% $yr^{-1}$. A line of evidence collected in this study suggests that the implementation of the NGVS program has been quite effective to induce the alterations in CO levels in all urban roadside locations throughout the city of Seoul.

Improvement of Growth of Potato (Solanum tuberosum L. cv. Dejima) Plants at In Vitro and Ex Vitro and Energy Efficiency by Environmental Control with Growth Stage in Photoautotrophic Micropropagation System (광독립영양 기내 미세증식 시스템에서 생육단계별 환경조절을 통한 감자의 기내 및 기외 생육과 에너지 효율 향상)

  • This study was conducted to evaluate the effect of optimized environment conditions with growth stage in photoautotrophic micropropagation on the growth of potato (Solanum tuberosum L. cv. Dejima) plantlets and energy efficiency. Optimum environment conditions at each stage were decided in our previous study. For the evaluation of optimized environment control, potato plantlets were cultured under four different conditions: photoautotrophic optimum conditions of photosynthetic photon flux density (PPFD) and $CO_2$ levels with growth stage (POG), photoautotrophic constant condition with average PPFD and $CO_2$ levels (PCA), photoauototrophic constant condition with maximum PPFD and $CO_2$ levels (PCM), and photomixotrophic conventional condition with 3% sucrose (PMC) as control. As a result, environment control with growth stage (POG) significantly promoted all the growth characteristics such as the number of nodes and unfolded leaves, shoot height, shoot diameter, and fresh and dry weights of potato grown in vitro. In addition, based on dry weight consumed electricity and $CO_2$ were the lowest in POG suggesting the highest energy efficiency among the treatments. After transferring potato plantlets to greenhouse, the plantlets under POG showed vigorous growth, which was pretty similar with those under PMC. The accumulations of dry matter in POG were 4.7 times in vitro and 3.8 times in greenhouse as much as those in the conventional control (PCM). Thus, we concluded that in vitro environment control with growth stage induced vigorous growth of potato plantlets both in vitro and in greenhouse with less energy consumption.

Memory-improving effect of formulation-MSS by activation of hippocampal MAPK/ERK signaling pathway in rats

  • MSS, a comprising mixture of maesil (Prunus mume Sieb. et Zucc) concentrate, disodium succinate and Span80 (3.6 : 4.6 : 1 ratio) showed a significant improvement of memory when daily administered (460 mg/kg day, p.o.) into the normal rats for 3 weeks. During the spatial learning of 4 days in Morris water maze test, both working memory and short-term working memory index were significantly increased when compared to untreated controls. We investigated a molecular signal transduction mechanism of MSS on the behaviors of spatial learning and memory. MSS treatment increased hippocampal mRNA levels of NR2B and TrkB without changes of NR1, NR2A, ERK1, ERK2 and CREB. However, the protein levels of pERK/ERK and pCREB/CREB were all significantly increased to $1.5{\pm}0.17$ times. These results suggest that the improving effect of spatial memory for MSS is linked to MAPK/ERK signaling pathway that ends up in the phosphorylation of CREB through TrkB and/or NR2B of NMDA receptor.

Effect of Low Adapted Temperature and Medium Composition on Growth and Erythropoietin (EPO) Production by Chinese Hamster Ovary Cells

  • Temperature and medium composition were changed with the aim of increasing growth and erythropoietin (EPO) production in EPO-producing Chinese hamster ovary (CHO) cells. We used the CHO cell line, IBE, and its derivative, CO5, which over-expresses the first two enzymes of the urea cycle, carbamoyl phosphate synthetase I (CPS I) and ornithine transcar-bamoylase (OTC). When supplements were added to the medium at $33\;^{\circ}C$, the growth of IBE and CO5 cells increased by $27\%\;and;26\%$, respectively and the maximum yield of EPO was increased by $40\%$ in both cell lines. The absolute EPO concentration in the CO5 cells was always $55{\sim}60\%$ higher than in the IBE cells. In addition, when the two cell lines were continuously cultured with supplements at $33\;^{\circ}C$ until their growth rates approached those at $37\;^{\circ}C$, the growth rates of both IBE and CO5 cells increased by $54\%$ and their maximum EPO levels increased by up to $73\%\;and\;56\%$, respectively. Therefore, the growth and EPO expression levels of CO5 cells increased 2.2-fold and 2.6-fold, respectively, compared to those of the IBE cells. These results indicate that adaptation to lower temperature as well as medium supplementation could be important for improving cell growth and EPO production.

Electrochemical Corrosion Properties of Amorphous Co-Nb-B Alloys Prepared by Melt-Spinning Method (액체급냉법에 의해 제조한 비정질 Co-Nb-B 합금의 전기화학적 부식 특성)

  • This study was undertaken to examine the electrochemical corrosion properties of an $Co_{80}Nb_{10}B_{10}$ and $Co_{82}Nb_8B_{10}$ amorphous alloys prepared by melt-spinning method under various conditions. The potentiodynamic polarization responses at various levels of pH (pH 2, pH 7, pH 12) showed that the corrosion current rate of $Co_{80}Nb_{10}B_{10}$ alloy is lower than that of $Co_{82}Nb_8B_{10}$ alloy in all levels of pH, implying the general corrosion resistance of the alloy with higher Nb content is better than that with higher Co content. The pitting potential of $Co_{80}Nb_{10}B_{10}$ alloy was also better than that of $Co_{82}Nb_8B_{10}$, evidenced by the higher pitting potential. Nb is thought to be effective in increasing the protectiveness of the passive film and hence to improve the corrosion resistance of Co-Nb-B alloys.
