Improvement on the Antioxidant Activity of Instant Noodles Containing Enzymatic Extracts from Ecklonia cava and Its Quality Characterization (감태 효소 추출물을 이용한 즉석 국수의 항산화성 개선 및 품질 특성)

  • This study was conducted to improve the antioxidative activity of instant noodles containing enzymatic extracts from Ecklonia cava (EEC). EEC has relatively better antioxidative activity than extracts from other indigenous plants in Jeju Island. The EEC (2.5 mg/mL) had 82.5% for the hydroxy radical, 78.4% for the DPPH radical, and 64.9% for the superoxide anion radical scavenging activities, and 65.2% for the cell viability (100 ${\mu}g/mL$). According to the texture of the dough, the DPPH free radical scavenging of uncooked instant noodles, sensory evaluation of cooked instant noodles, and turbidity of the cooking drip, the optimal EEC concentration was 1.8% for the instant noodles. The major amino acids in the instant noodles with EEC were glutamic acid (24.2%), proline (10.2%), valine (10.0%), and isoleucine (12.3%). The zinc and iron in the instant noodles were enhanced by adding 1.5-1.8% EEC. The antioxidant activity of instant noodles with EEC was 75.4% for the hydroxy radical, 74.1% for the DPPH radical, and 51.2 % for the superoxide anion radical scavenging activities.

Biological Activities of Flavonoid Glycosides Isolated from Angelica keiskei (신선초에서 분리된 flavonoid glycosides의 생리활성)

  • Recently, much attention has been focused on plant antioxidants, because they are expected to protect against oxidative damage, possibly preserving biological functions of cells. Antioxidant compounds were isolated from Angelica keiskei through extraction with 80% EtOH, and fractionations were carried out sequentially with n-hexane, chloroform, ethyl acetate, n-butanol, and water. Two active compounds were isolated from ethyl acetate fraction by silica gel column chromatography, and were identified as isoquercitrin ($quercetin-3-O-{\beta}-D-glucose$) and hyperoside ($quercetin-3-O-{\beta}-D-glucose$). Isoquercitrin and hyperoside showed strong antioxidative potency, as revealed by evaluation of their ABTS, DPPH, OH, and $H_{2}O_{2}$ radical-scavenging activities, and ex vivo DNA damage-protecting effects.

The Isolation and Antioxidative Effects of Vitexin from Acer palmatum

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental factors are critical players in cellular damage and aging. In order to develop a new antiphotoaging agent, this work focused on the antioxidant effects of the extract of tinged autumnal leaves of Acer palmatum. One compound was isolated from an ethyl acetate soluble fraction of the A. palmatum extract using silica gel column chromatography. The chemical structure was identified as apigenin-8-C-beta-D-glucopyranoside, more commonly known as vitexin, by spectral analysis including LC-MS, FT-IR, UV, $^{1}H-$, and $^{13}C-NMR$. The biological activities of vitexin were investigated for the potential application of its anti-aging effects in the cosmetic field. Vitexin inhibited superoxide radicals by about $70\%$ at a concentration of $100\;{\mu}g/mL$ and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals by about $60\%$ at a concentration of $100\;{\mu}g/mL$. Intracellular ROS scavenging activity was indicated by increases in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20\;mJ/cm^2$ in cultured human dermal fibroblasts (HDFs) after the treatment of vitexin. The results show that oxidation of 5-(6-)chloromethyl-2',7'-dichlo-rodihydrofluorescein diacetate ($CM-H_{2}DCFDA$) is inhibited by vitexin effectively and that vitexin has a potent free radical scavenging activity in UVB-irradiated HDFs. In ROS imaging using a confocal microscope we visualized DCF fluorescence in HDFs directly. In conclusion, our findings suggest that vitexin can be effectively used for the prevention of UV-induced adverse skin reactions such as free radical production and skin cell damage.

  • Numerical simulations are performed at atmospheric pressure in order to understand the effect of additives on flame speed, flame temperature, the radical concentration, the NOx formation in freely propagating $CH_4/O_2/N_2$ flames. The additives used are carbon dioxide and hydrogen chloride which have a combination of physical and chemical behavior on hydrocarbon flame. In the flame established with the same mole of methane and additive, $CO_2$ addition significantly contributes toward the reduction of flame speed and flame temperature by the physical effect, whereas addition of HCl mainly does by the chemical effect. The impact of HCl addition on the decrease of the radical concentration is about 1.6-1.8 times as large as $CO_2$ addition. Hydrogen chloride addition is higher on the reduction of EINO than $CO_2$ addition because of the chemical effect of HCl.

  • The ethanolic extract of chestnut (Castanea crenata S. et Z., Fagaceae) inner shell (CISE) and one of its components, ellagic acid (EA), were evaluated for their protective effects against 1, 1-diphenyl-2-picryl hydrazine (DPPH) free radical generation and hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line. CISE and EA were shown to possess the free radical scavenging effect against DPPH radical generation, significantly. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from Chinese hamster lung (CHL) cell, assessed by single cell gel electrophoresis assay and 8-hydroxy -2'-deoxy guanosine (8-OH-2'dG) assay. Furthermore, topical application of CISE [$12.5\%$(w/w) cream] and ellagic acid [$1.0\%$(w/w) cream] for 14 days potently inhibited malondialdehyde (MDA) formation of mouse dorsal skin (a marker of lipid peroxidation) induced by ultraviolet B exposure. Therefore, CISE and its component, ellagic acid, may be the useful natural antioxidants by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage when topically applied.

Experimental Investigation on Premixed Combustion Characteristics with Suction & Blow Fans (Suction과 blow fan을 이용한 연소기내의 부분 예혼합화염 연소 특성에 관한 실험적 연구)

  • We measured emission indices for $NO_x$, CO, temperature and radical characteristics for partially premixied flames formed by suction & blow fans air condition. At sufficiently high levels of partial premixing a double flame structure consisting of a rich premixed inner flame and outer diffusion flame was established similar to that previously observed in premixed flames. $NO_x$, Temperature. CO concentration were experimented with approximately constant air flow rate and decreasing equivalence ratios. The reduction in $NO_x$, and temperature at suction condition as compared with that for blow condition was approximately 20%, but on the contrary, CO emission was increased. In addition, We measured temperature distributions and found that temperature increased continuously with increasing partial premixing. We also estimated CH, $C_2$ radical intensity. CH and $C_2$ radicals provide evidence that, for the present measurement, CH and $C_2$ radicals intensity was associsated with their premixed component. And we observed stronger $C_2$, CH radicals intensity at suction conditions than blow conditions.

  • One of the chronic problems in the issue of global warming is the emission of greenhouse gases. Carbon dioxide (CO2), which accounts for the highest proportion of various greenhouse gases, has been continuously researched by humans to separate it. From this point of view, a poly(vinyl alcohol) (PVA)-based copolymer with acrylic acid monomer was utilized in a gas separation membrane in this study. We employed a free radical polymerization to fabricate PVA-g-PAA (VAA) graft copolymer. It was utilized in the form of a composite membrane on a polysulfone substrate. The proper amount of acrylic acid reduced the crystallinity of PVA and increased CO2 solubility in separation membranes. In this perspective, we suggest the novel approach in CO2 separation membrane area by grafting and solution-diffusion.

Detection of gamma irradiated South Sea cultured pearls (감마선 조사된 남양진주의 검지)

  • We have been performed on $Co^{60}$ ${\gamma}$-ray irradiation of South Sea cultured pearls, with the absorbed dose of 0.1~100 kGy at room temperature. In addition, it was investigated electron paramagnetic resonance (EPR) and amino acid analysis (AAA) for detection of Gamma irradiated South Sea cultured pearls. It was observed that the irradiated South Sea cultured pearls revealed additional free radical peak such as $CO_2^-$, at a g-factor of $2.001{\pm}0.002$ in EPR spectra. From the amino acid analysis (AAA), it was shown that some of amino acid in the protein of the nacre destroyed after ${\gamma}$-ray; glutamic acid residue by 11.43 %, alanine by 3.11 %, and histidine by 43.75 %. It was useful to detect the irradiated South Sea cultured pearls by EPR measurement in our study.

  • Tetracarbonylhydridoferrate, $HFe(CO)^-_4$, generated by the reaction of $Fe(CO)_5$ with alkaline solution, is a good reducing agent for mono-dehalogenation of gem-dihalocyclopropanes. It also acts as a good reducing catalyst under phase transfer reaction conditions. 1,1-Dibromo-2-phenylcyclopropane and 1,1-dichloro-2-phenylcyclopropane were reduced to the corresponding mono-dehalogenated products in excellent yields. Thermodynamically stable trans-l-bromo-2-phenyl cyclopropane was formed as the major product over the cis-isomer, trans/cis=3/2. The 1-bromo-2-phenyl cyclopropane radical intermediate was formed by single electron transfer from $HFe(CO)^-_4$. Dissociation of bromide anion, followed abstraction of hydrogen radical from alcoholic solvent would lead to the formation of the stable trans-isomer. The further mechanistic aspects were discussed.
