먹이제한과 단기 수온하강 조건에서 넙치의 스트레스 반응과 생리학적 대사활성 변화 (Changes of Stress Response and Physiological Metabolic Activity of Flounder, Paralichthys olivaceus Following to Food Deprivation and Slow Temperature Descending)

  • 본 연구는 통상적으로 활어 수송을 위해 사용되는 수온 $15{\pm}2^{\circ}C$ 범위에서 단기 수온 강하와 단기간 먹이제한을 했을 때 넙치의 스트레스 반응과 생리상태를 혈중 생화학 인자와 4종류의 갑상선호르몬의 변화를 통해 파악하고, 이 결과들을 바탕으로 어류의 스트레스 및 생리활성 지표로서 분석 인자들의 효용성을 검토하고자 하였다. 먹이제한 실험은 1년생 넙치 양성어(전장 $32.9{\pm}1.2$ cm, 체중 $348.0{\pm}34.7$ g)를 이용해 8일간 실시하였고, 수온변화 실험은 1년생 양성어(전장 $32.7{\pm}1.2$ cm, 체중 $327.1{\pm}33.6$ g)를 이용해 8일간 수온을 $15.8^{\circ}C$에서 $13.3^{\circ}C$로 하강시키며 실시하였다. 혈중 생화학적 분석 인자는 ALT, AST, TP, GLU, 전해질($Na^+$, $K^+$, $Cl^-$), thyroid hormones (total $T_3$ & $T_4$ free $T_3$ & $T_4$)의 혈중 변화로 이루어졌다. 먹이섭식 제한 결과, AST, ALT 및 전해질에서는 절식기간 동안 어떠한 유의한 변화를 찾아볼 수 없었지만, TP와 GLU는 유의하게 감소하는 경향을 보였다. 갑상선호르몬 농도의 경우, 조사된 4종류 모두 절식기간이 길어지면서 감소하는 경향이 있었으며, 통계적으로 $FT_4$$TT_3$가 개시에 비해 실험 종료시 유의하게 감소하였다. 수온하강 실험의 경우, AST, ALT, TP 및 전해질 및 갑상선호르몬 농도에서 유의한 변화는 없었으나, GLU에서는 온도하강에 따른 농도 감소가 관찰되었다(P<0.05).

국산 및 중국산 백수오(白首烏)의 Triton WR-1339 유발 고지혈증(高脂血症) 흰쥐에 대한 효능 연구 (Effects of Cynanchum spp. on the hyperlipidemia in rats induced by Triton WR-1339)

  • Objective : In order to compare of hyperlipidemia effect of C. wilfordii, C. auriculatum in korea and C. auriculatum in China, it was performed on the hyperlipidemia in rats induced by Triton WR-1339. Method : The 70% MeOH extracts from dried root of C. wilfordii and C. auriculatum in korea and C. auriculatum in china were fractionated into H2O layer and CHCl3 layer. The level of total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, AST and ALT in serum were used to observe the effectiveness of C. wilfordii korean C. auriculatum and chinese C. auriculatum. Results : In the hyperlipidemic rats induced by Triton WR-1339, C. wilfordii and korean C. auriculatum significantly decreased total cholesterol, triglyceride, otherwise significantly increased HDL. And C. wilfordii and chinese C. auriculatum significantly decreased AST and ALT. Conclusion : This study indicate that C. wilfordii korean C. auriculatum and chinese C. auriculatum were effective in the hyperlipidemic rats induced by Triton WR-1339. According to the results, C. wilfordii korean C. auriculatum and chinese C. auriculatum need to considered for the application in hyperlipidemia.

플랫 플레이트 구조에서 뚫림전단에 대한 전단보강체의 보강능력 (Shear Capacity of Shear Reinforcement for Punching Shear in Flat Plate Structure)

  • 본 연구는 플랫 플레이트의 전단보강체에 관한 연구의 일부이고, 슬래브-기둥 접합부의 수직전단력 실험에서 보강체의 종류에 따라 뚫림전단강도에 대한 영향을 확인하는 것을 목표로 한다. 제안된 4개의 보강체는 s/s bar, c/s bar, wiremesh, CFS이다. 전체 5개의 실험체를 제작하고 실험을 행하였으며, 슬래브의 제원은 $2.63{\ast}2.725.0.18m$이며, 중앙에 0.6${\ast}$0.8m의 단면을 가진 기둥을 가지고 있다. 실험결과를 요약하면 CFS 전단보강체는 콘크리트와의 부착이 충분히 되지 않아서 보강체가 보강력을 발휘하기 전에 보강체와 콘크리트가 분리되어 보강효과를 발휘하지 못하였다. CFS를 제외한 3개의 전단보강체는 슬래브-기둥 접합부의 뚫림전단 저항력을 증가시켜 높은 보강효과를 나타내었다. 전단보강이 되지 않은 실험체에 비하여 강도면에서는 16${\sim}$35%의 증가율을 보였고, 변형능력면에서는 35${\sim}$63%의 증가율을 보였다.

  • $crp^{\ast}$ 유전자가 도입된 대장균 MK2001($crp^{{\ast}1}$}, cya::km)을 숙주로 사용하여 mal 유전자 발현을 촉진시키는 유전자의 하나인 sfs1(sugar fermentation stimulation)의 구조해석 결과에 의하면, 잠정적인 sfs1의 promoter 영역에는 CRP 단백질과의 결합영역으로 보이는 염기배열이 존재하였다. 본 실험에서는 sfs1 유전자의 cAMP-CRP에 의한 발현 조절을 확인하고자, lacZ 와의 융합 유전자를 작성하였다. 작성된 융합 유전자는 cya 결손주인 Tp2010에서 cAMP의 첨가에 의해 ${\beta}-galactosidase$ 활성이 크게 증가하였으며, Western blotting의 실험에서도 같은 결과를 나타냈다. in vivo에서 발현이 확인된 전사산물은 cAMP에 의해 전사 촉진이 일어났으며, CRP의 결합부위로 예상되는 DNA 영역은 cAMP가 존재하면 CRP 단백질과 결합하는 특성을 나타내었다. 이상의 결과로 보아, sfs1 유전자의 발현은 UMP-CRP에 의한 전사촉진 현상을 받는 것으로 나타났다.

국산과 중국산 지실(枳實)의 Triton WR-1339 유발 고지혈증(高脂血症) 흰쥐에 대한 효능 연구 (Lipid lowering activity of Ponciri Fructus and Aurantii Fructus Immaturus on hyperlipemia rats induced by Triton WR-1339.)

  • Objective : In order to elucidate hyperlipidemic effects of the fruits of Poncirus trifoliata and Citrus aurantium, our experimental study was performed on hyperlipidemia rats induced by Triton WR-1339. Method : The hyperlipidemia rats induced by intraperitonial injection of triton WR-1339 were treated with the fruits of P. trifoliata (rare and preparata) and C. aurantium 50 and 200mg/kg. The amount of triglyceride. total cholesterol. HDL-cholesterol, LDL-cholesterol, AST. and ALT levels were measured. Results: The levels of triglyceride. total cholesterol and LDL-cholesterol were significantly reduced in fruits of P. trifoliata (rare and preparata) and C. aurantiu. Simvastatine treated group compared with those of control but the level of HDL-cholesterol was incresed. The level of AST and ALT were not significantly reduced by the treatment to the fruits of P. trifoliata (rare and preparata) and C. aurantium. Conclusion : In the hyperlipidemia rats induced by triton WR-1339. the fruits of P. trifoliata (rare and preparata) and C. aurantium showed a hyperlipidemic effect.

Vibrational Analysis of Ferrocyanide Complex Ion Based on Density Functional Force Field

  • Vibrational properties of ferrocyanide complex ion, $[Fe(CN)_6]^{4-}$ , have been studied based on the force constants obtained from the density functional calculations at B3LYP/$6-31G^{\ast\ast}$ level by means of the normal mode analysis using new bond angle and linear angle internal coordinates recently developed. Vibrations of ferrocyanide were manipulated by twenty-three symmetry force constants. The angled bending deformations of C-Fe-C, the linear bending deformations of Fe-C${\equiv}$N and the stretching vibrations of Fe-C have been quantitatively assigned to the calculated frequencies. The force constants in the internal coordinates employed in the modified Urey-Bradley type potential were evaluated on the density functional force field applied, and better interaction force constants in the internal coordinates have been proposed. The valence force constants in the general quadratic valence force field were also given. The stretch-stretch interaction and stretch-bending interaction constants are not sensitive to the geometrical displacement in the valence force field.

Hepatoprotective Activity of Thespesia populnea Bark Extracts against Carbon Tetrachloride-Induced Liver Toxicity in Rats

  • In the present study, Thespesia populnea (Malvaceae) bark was extracted with methanol and water. The extracts were vacuum dried to yield the respective methanol (MET) and aqueous extract (AET). The extracts were evaluated for hepatoprotective activity against carbon tetrachloride $(CCl_{4})$ induced liver damage at 2 dose levels (250 and 500 mg/kg). The biochemical parameters observed in serum were total bilirubin, alkaline phosphatase (ALP), serum glutamate oxaloacetate transaminase (SGOT), serum glutamate pyruvate transaminase (SGPT) levels and total protein. Aspartate transaminase (AST), alanine transaminase (ALT) and total protein levels in liver were also evaluated. Histopathological study on the liver tissue was also performed. The extracts exhibited dose dependent reduction in total bilirubin, ALP SGOT, SGPT, AST, ALT and increase in total protein (serum and liver) levels. The extracts also exhibited only mild hepatocytic damage compared to the $CCl_{4}$ Treated group. MET was found to exhibit higher hepatoprotection than AET.

충돌과냉수분류(衝突過冷水噴流)의 비등열전달(沸騰熱傳達)에 관한 연구(硏究) (A Study on Boiling Heat Transfer in a Impinging Subcooled Water Jet System)

  • This paper describes the boiling heat transfer phenomena to be divided into three regions, nonboiling, nucleate boiling and burn-out in the impinging subcooled water jet system. In the nonboiling region, Nusselt number is a function of Prandtl number, Reynolds number and ${\Delta}T_{sub}/T_{ast}$ In the nucleate boiling region, the heat flux increases with increment of the nozzle exit velocity. But the degree of liquid subcooling does not affect the shape of the nucleate boilng curve. The dimensionless correlations can be expressed in the form of $q{\ell}/K_f{\Delta}T_{ast}=C(Bo{\cdot}C_p{\cdot}{\Delta}T_{sat}/Vo^2)^m{\cdot}(Re/We)^n$. The burn-out heat flux increases linearly with increment of the nozzle exit velocity, but independs of degree of subcooling and the supplementary water height.

큰실말에서 분리된 식이섬유가 고지방식이로 유도된 흰쥐의 콜레스테롤에 미치는 영향 (Effects of Cladosiphon Okamuranus Dietary Fiber on Cholesterol in High Fat Diet-Fed Rats)

  • Cladosiphon okamuranus is edible brown algae cultured commercially and massively at Okinawa coast. Dietary fiber occupying most of C. okamuranus isn't digested and absorbed by digestive enzymes of human. But it is known to prevent and treat constipation as metabolic disease. This study was to investigate the effects of dietary fiber extracted from C. okamuranus on the damage of liver and the blood cholesterol level in the high fat diet-fed rats. The effects were measured by the levels of aspartate aminotransferase (ALT), alanine aminotransferase (AST), total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL) and high density lipoprotein (HDL) in sera. The levels of ALT, AST, TC, TG and LDL were significantly reduced in the C. okamuranus-treated group compared with the high fat diet group. But HDL level was markedly increased. The results showed that the dietary fiber extracts from C. okamuranus have the inhibitive effects of cholesterol biosynthesis in the high fat diet-fed rats.
