In vitro propagation and multiple shoot induction of Rhodiola rosea L. by axillary bud culture (홍경천(Rhodiola rosea L.)의 액아배양을 통한 다신초 유도 및 기내 대량증식)

  • An efficient in vitro propagation was established by using axillary bud explants of roseroot (Rhodiola rosea L.), which has been known as a medicinal plant in East Asia. Among various media tested, MS medium supplemented with 1.0 mg/L BA and 1.0 mg/L $GA_3$ was found to be the best for multiple shoot formation (15 axillary shoots per axillary bud). In addition 1/2MS medium containing 50 g/L sucrose was best for shoot elongation (7.8 cm) and increasing total chlorophyll contents (8.64 mg/g) best. Maximum number of roots (17.7 roots per explant) was observed on the medium without plant growth regulators. Propagated plants were successfully acclimatized to ex vitro conditions, with a survival frequency of 97% after 12 weeks. Most rooted shoots grew well and produced viable seeds when grown in vitro culture conditions. Therefore, R. rosea can be effectively propagated in vitro by the system we developed in this study.

Geochemistry of Trace and Rare Earth Elements from Coal-bearing Metapelites of the Ogcheon Supergroup at the Hoenam Area, Korea (회남지역(懷南地域)의 옥천누층군(沃川累層群)에 분포(分布)하는 탄질(炭質) 변성이질암(變成泥質岩)의 미량(微量) 및 희토류원소(稀土類元素) 지화학(地化學))

  • The coal-bearing metapelites from the Hoenam area are interbedded within the Ogcheon Supergroup, which are composed mainly of graphite, quartz, muscovite and associated with trace amount of biotite, chlorite, pyrite, rutile and barite. Although the formation was mined for coal, and the contents of V, U and Mo are a higher grade in coal formations than the host metapelites. The ratios of Al/Na and K/Na in coal formation are very heterogeneous and wide range from 10.28 to 90.91 and from 3.73 to 36.60, respectively. The V content increase with increasing Al and Ba contents, but the U and Mo are not related with other elements. Those are suggested that controlled of mineral compositions in coaly metapelites due to substitution, migration and reequilibrium of elements by regional metamorphism. These coal formation were deposited in basin of marine environments and the REE of these rocks are not influenced with metamorphism and hydrothermal alterations on the basis of Al content versus La, La against Ce, the ratios of La/Ce (0.23 to 0.73) and Th/U (0.03 to 16.6). These rocks also show much variation in $La_N/Yb_N$ (0.53 to 14.19), Th/Yb (0.51 to 6.00) and La/Th (0.15 to 18.92), and their origin is explained by derivation from a mixture of sedimentary and metasedimentary rocks. The wide range in trace and REE element characteristics as Co/Th (0.07 to 3.00), La/Sc (0.04 to 23.48), Sc/Th (0.06 to 7.57), V/Ni (2 to 3319), Cr/V (0.03 to 1.06) and Ni/Co (1.00 to 79.85) of these coaly metapelites argues for inefficient mixing of the various source lithologies during sedimentation.

Dispersion and Enrichment of Potentially Toxic Elements in the Chungjoo Area Covered with Black Shales in Korea (충주지역 흑색셰일 분포지역에서의 잠재적 독성원소들의 분산과 부화)

  • This study had three purposes: (1) to investigate dispersion and enrichment level of potentially toxic elements; (2) to identify uranium-bearing minerals in black shales; and (3) to assess the chemical speciation of heavy metals in soils and sediments. Rock, surface soil and stream sediment samples were collected in the Chungjoo area covered with black shales in Korea. These samples were analyzed for multi-elements using INAA and ICP-AES. The maximum abundance of U in black shales is 56 ppm and radioactivity counts up to 240CPM. Molybdenum, V, Ba, Cu, and Pb are enriched in black shales and most of soils show high concentrations of U, Mo, Ba, Cu, Pb and Zn. Concentrations of potentially toxic elements decrease in the order of mountain soil > farmland soil > paddy soil. Enrichment index of soils and sediments are calculated and higher than 1.0 in the black shale area with the highest value of 6.1. In order to identify U-bearing minerals, electron probe micro analysis was applied, and uraninite and brannerite in black shale were found. Uraninite grains are closely associated with monazite or pyrite with the size of $2{\mu}m$ to $10{\mu}m$ in diameter whereas brannerite occurs as $50{\mu}m$ euhedral grains. With the results of sequential extraction scheme, residual fractions of Cu, Pb and Zn in soils are mainly derived from weathering of black shale but Cu, Pb and Zn in sediments are present as non-residual fractions. Lead is predominantly present as oxidizable phase in soils whereas Zn is in exchageable/water-acid soluble phase in sediments.

  • Intramolecular proton transfer equilibria of acetamide and methyl carbamate have been studied theoretically by MNDO MO method. For both substrates, carbonyl-O protonated tautomer was found to be the most stable form, the next most stable one being N-protonated form. Gas phase proton transfers take place by the 1,3-proton rearrangement process and in all cases have prohibitively high activation barriers. When however one solvate water molecule participates in the process, the barriers are lowered substantially and the process proceeds in an intermolecular manner through the intermediacy of the water molecule via a triple-well type potential energy surface; three wells correspond to reactant(RC), intermediate(IC) and product complex(PC) of proton donor-acceptor pairs whereas two transition states(TS) have proton-bridge structure. General scheme of the process can be represented for a substrate with two basic centers(heteroatoms) of A and B as, $$ABH\limits^+\;+\;H_2O\;{\to}\;ABH\limits^+{\cdots}{\limits_{RC}}OH_2\;{\to}\;AB{\cdots}H\limits_{TS}^+{\cdots}{\limits_{1}}OH_2\;{\to}\;AB{\cdots}{\limits_{IC}}H\limits^+OH_2\;{\to}\;BA{\cdots}H\limits_{TS}^+{\cdots}{\limits_{2}}OH_2\;{\to}\;BA H\limits^+{\cdots}{\limits_{PC}}OH_2\;{\to}\;BAH\limits^+\;+\;H_2O$$ Involvement of a second solvate water had negligible effect on the relative stabilities of the tautomers but lowered barrier heights by 5∼6 Kcal/mol. It was calculated that the abundance of the methoxy-O protonated tautomer of the methyl carbamate will be negligible, since the tautomer is unfavorable thermodynamically as well as kinetically. Fully optimized stationary points are reported.

  • Three PEA (Pulverised Fuel Ash) samples, which were fresh, 17 and some 40 years weathered, were collected from two major British power plants. Batch leaching tests with these samples using distilled water and simulated industrial leachate showed higher amounts of element liberation from fresh ash, including Ca, Na, K, S (as $SO^{2-}_4$, $Cr_{total}$, Cu, Li Ni, Mo and CI and this seems to indicate their surface association and easier dissolution when contact with water. On the contrary Mg, Al, Ba, Si, V, As and Se do not show such readily leachable concentrations and these elements might be more associated with glass fraction in PFA particle rather than surface. Although element concentrations in the weathered ash are much lower than those in the initial leachate from the fresh ash, elements are still detected as resonable concentrations, with rather constant levels and this seems to demonstrate the element release from unstable glass phase of PFA particle. Fe, Ca, $Cr_{total}$, Cu, Ni, Zn and Hg were removed from the synthetic leachate by PFA and this is also confirmed by gain in solid PFA. The order of element retention is Meaford weathered ash > Drax weathered ash > Drax fresh ash in decreasing order and this conforms with the degree of weathering. Namely, the more wethered, the more wethered, the more effective in metal retention from the synthetic leachate.

  • The Cretaceous metal ore deposits in the Gyeongsang basin of Gyeongsangbuk-do are characterized by the formation of metallogenic provinces which show zonal distribution pattern around Yeonil province where pneumatolytic type is dominated and hydrothermal type are distributed in the order of decreasing temperature type outward. Some Cretaceous granitic rocks include zoned alkali feldspars which reflect rapid variation of $H_2O$ during emplacement and crystallization of the water-saturated granitic magma. The ore deposits are considered to be originated from upward transportation of ore solution from the excess of water exhausted from uprising magma, which seems to be intimately related to the fact that the majority of the ore deposits in Daegu area are cummulated around the granites including zoned alkali feldspars. In order to collect geochemical data necessary for geochemical exploration in the study area, certain trace elements were chosen as pathfinders from monzonite and soil in the vicinity of Dalsung Tungsten Mine by studying the dispersion patterns of trace elements: Ba and Sr show trends to decrease toward ore deposit while Cu, Pb, and Mo increase. Around mining area there are distributed apparently Equisetum arvense Linne and Mentha sachinensis Kudo which may be used as index plants. In the viewpoint of geologic structure, the trends of the ore veins in contact aureole around the Palgongsan granite body correspond with the pre- and syn- plutonism joint pattern in hornfels in the area.

  • In this study, hyaluronic acid (HyA) - Gelatin (Gel) hydrogels were prepared at ratio of 15:85 with the goal of obtaining a high uniform porosity and porous biocompatibility scaffold for bone tissue engineering applications. In order to develop a proper scaffold for bone implant application, a HyA-Gel hydrogel loaded in sponge Biphasic Calcium Phosphate (BCP) was prepared. To assay the cytocompatibility and cell behavior on the HyA-Gel hydrogel and HyA-Gel/BCP scaffold, cell attachment and spreading of MSCs seeded on the scaffolds were studied. An invivo study was performed for HyA-Gel/BCP scaffolds after 1 and 3 months implantation. Our results provide a novel and simple method to obtain an adequate scaffold for osteoblast cells and indicate that HyA-Gel hydrogel and HyA-Gel/BCP scaffold could be a good candidate for bone tissue engineering scaffolds.

In vitro shoot regeneration and genetic transformation of the gerbera (Gerbera hybrida Hort.) cultivar 'Gold Eye'

  • This research was conducted to improve the cold tolerance of the gerbera cv. Gold Eye by introduction of the Arabidopsis $Ca^{2+}/H^+$ antiporter gene (CAX1) via Agrobacterium-mediated transformation. Prior to genetic transformation, we optimized a combination of plant growth regulators; $1.0mgl^{-1}$ 6-Benzyladenine (BA) and $0.1mgl^{-1}$3-indole-acetic acid (IAA) were found to lead to proper in vitro shoot regeneration from petiole explants. In addition, $50mgl^{-1}$ kanamycin was determined to be the minimal concentration useful for selection of putative transgenic plants. In this study, transgenic gerbera expressing the Arabidopsis $Ca^{2+}/H^+$ antiporter gene (CAX1) were obtained using the optimized concentrations. We expect that introduction of the gene to the cultivar will improve cold tolerance, which will be important in the winter months.

  • Computational investigation was conducted to study the effects of the shape parameter of shroud on the performance of the outdoor unit of an air-conditioners. For this study the Design of Experiment(4-factor 3-level) was created and the an automatic program was composed using VBA to reduce the load of pre-process for CFD. The estimated mathematical equation was produced from this analysis and it was found that the gap between fan and shroud affects more heavily than the other parameters. As a result, the composition of the best parameters was verified that its flow rate was increased by 10 percents and sound pressure level was reduced by 1.2 dBA compare with the worst. And finally, a kind of Propeller fan with blades which were attached to the shroud, so-called 'ProBeller Fan' was introduced in this study.

  • The effect of oxygen-deficiency on the charge distributions and orbital energies for small copper oxide clusters representing the superconducting materials $YBa_2Cu_3O_x (6{\leq}x{\leq}7)$ were investigated by the extended Huckel molecular orbital (EHMO) method with the tight-binding model. Our calculations show +3 oxidation state of Cu(1) in the $CuO_3$ chain and +2 or +1 of Cu(2) in the $CuO_2$ layers for $YBa_2Cu_3O_7$ with the nominal charge of $Cu_3$ = +7 (or +5), while for $YBa_2Cu_3O_6$ +1 oxidation state of Cu(1) and +3 (or +2) of Cu(2) in the $CuO_2$ layers with the nominal charge of $Cu_3$ = +7 (or +5). For $Cu_3O_{12}$ cluster representing $YBa_2Cu_3O_7$ with the nominal charge of $Cu_3$ = +7 the Cu(2) $d_{{x^2}-{y^2}}$ orbitals in the $CuO_2$ layers is a typical Jahn-Teller $d^9$ system with the partial hole and the Cu(1) $d_{{_z2}-{_y2}}$ orbital in the $CuO_3$ chain contains hole occupancy. For $Cu_3O_{10}$ cluster representing $YBa_2Cu_3O_6$ with the nominal charge of Cu = +5 the orbital character of the highest partially occupied MO (HPOMO) and the lowest completely unoccupied MO (LCUMO) of $Cu_3O_{12}$ representing $YBa_2Cu_3O_7$ with the nominal charge of $Cu_3$ = +7 is reversed, and the character of Cu(1) $d{{x^2}-{y^2}}$ orbital of LCUMO of the $Cu_3O_{12} $cluster is vanished. It is suggested that the local crystal field environment of Cu(1) by the oxygens in the Cu(1) chain may play a vital role in conductivity and superconductivity, either alone or through cooperative electronic coupling with the Cu(2) layers in $YBa_2Cu_3O_7.$.
